in the Hind HI site of ORF A. Since pKM3 had two Hind HI sites (one is in the multiple cloning site of the vector pJM103 and the other is in ORF A) the ORF A fragment was first transferred into vector pGEM5Z which does not have any Hind in sites and then the kanamycin cassette was inserted. Protoplast transformation into B. megaterium PV361 yielded no kanamycin resistant colonies and controls were positive in three experiments, therefore, it can be concluded that ORF A was inactivated. To test whether the inactivated ORF A could then be complemented, PV361 containing a …

Effect Of Media Constituents On In Vitro Culturing Of Cowpea (Vigna Unguiculata) Shoot Tip And Leaf Disk Explants, Mohanjeet S. Brar, Edwin J. Anderson, Teddy E. Morelock, Ronald W. Mcnew

Journal of the Arkansas Academy of Science

Cowpea is an important legume food crop that is commonly grown in Arkansas and numerous other southern states. The application of biotechnological approaches for the improvement of U.S. cowpea genotypes is currently not possible due to the lack of a regeneration and transformation system. Therefore, the first priority of our research efforts is the development of a plant regeneration system that will facilitate plant transformation studies. In an effort to optimize the media requirements for tissue culturing cowpea, we evaluated the in vitro response of shoot tip and leaf disk explants to various levels of Murashige and Skoog (MS) macro …

Impact Of Amateur Educational Video Viewing On Language Acquisition Of Preschool Children, Jennifer L. Radcliffe Jan 1997

Impact Of Amateur Educational Video Viewing On Language Acquisition Of Preschool Children, Jennifer L. Radcliffe

Masters Theses

Child language-learning is a complex process that varies for each individual child. Researchers have found that children learn from viewing professional television programs specifically designed for children, such as Sesame Street. Professional television programming principles can be adopted to produce amateur educational videos. Video may also be a viable option to provide repetitive models to improve vocabulary acquisition within speech-language pathology. The purpose of this study was to measure the effect of direct stimulation with amateur educational videos on a child's general and specific vocabulary acquisition.

A six-week experiment included an initial two-week control period, a two-week experimental period …

The Treatment Of Vegetable Processing Waste Water Using A Pilot Scale Sequencing Batch Reactor Activated Sludge Process, Peter Hassett Jan 1997

The Treatment Of Vegetable Processing Waste Water Using A Pilot Scale Sequencing Batch Reactor Activated Sludge Process, Peter Hassett

Theses

The SBR process is a discontinuous, unsteady state process and has developed popularity in many countries assisted by reliable control technology for example programmable logic control. A typical SBR installation achieves equalisation, aeration and clarification of batches of wastewater on a timed sequence in one basin. This is in contrast to a volume sequenced process such as the conventional continuous stirred tank reactor where individual treatment objectives are met in separate basins. Advantages include, flexability, minimum energy input, consistent effluent quality and inhibition of filament organism growth by properly controlling substrate concentration and aeration sequences. Nitrification, denitrification and Bio-P removal …

So Long Sweetheart - State Farm Fire & (And) Casualty Co. V. Gandy Swings The Pendulum Further To The Right As The Latest In A Line Of Setbacks For Texas Plaintiffs., Timothy D. Howell

St. Mary's Law Journal

Some legal commentators would analogize the description of the prisoner of The Pit and the Pendulum to modern tort law and policy. Like a pendulum, tort law is always in motion, swinging between two ideologies: those being the plaintiff-oriented rights of the 1970s-80s, and the tort-reforms which began in the mid-80s. This trend continued until the 1990s and the decision of State Farm Fire & Casualty Co. v. Gandy where Texas plaintiffs began experiencing setbacks. The Gandy decision likely curtails the use of the “sweetheart deal” a practice, in which an insured defendant first settles with the plaintiff, then assigns …

Understanding Audience Understanding, Paul A. Soukup Jan 1997

Understanding Audience Understanding, Paul A. Soukup

Communication

Communication study has approached the issue of audience understanding of messages from the perspective of the message and from that of the audience. On the one hand, the "powerful-message" construct paints the audience as passive recipients of the meaning presented in the media. On the other hand, the "active audience" construct places most interpretive power in the audience, stressing their selectivity of messages, their use of the media, their social positions, and their ability to generate new messages based on the media. A middle position sees audience understanding emerge from an interaction between messages and audience members.

Using The Internship As A Tool For Assessment: A Case Study, Beverly Graham, Pamela G. Bourland-Davis, Hal W. Fulmer Jan 1997

Using The Internship As A Tool For Assessment: A Case Study, Beverly Graham, Pamela G. Bourland-Davis, Hal W. Fulmer

Journal of the Association for Communication Administration

This article presents a case study involving public relations internship letters that were used to generate a checklist for the assessment process. The case study suggests three key points about the usefulness of internships in assessment. First, the internship can be translated into empirical results, greatly assisting those individuals who directly shape the public relations program. This empiricism should also be advantageous when confronting non-program administrators (deans, vice-presidents) with the need for additional human and physical resources. Second, this case study highlights the significant confluence that occurs between the program administrator (the faculty typically), the students of the program, and …

A "Glimpse" Of The Status Of The Discipline: Internal Identity Of Speech Communication At A Small College, Barbara J. Adler Jan 1997

A "Glimpse" Of The Status Of The Discipline: Internal Identity Of Speech Communication At A Small College, Barbara J. Adler

Journal of the Association for Communication Administration

This article discusses the results of a survey administered to faculty and administration at a small U.S. liberal arts college associated with the Lutheran Church, to assess the attitudes toward the speech communication discipline at the college. Speech communication discipline was assessed according to its level of respect, its overall status and its identity as a reputable academic field. Speech communication received its highest ranking for the descriptor applications based, matching the perception that the field is dominated by utility courses that teach students how to give a speech.
