The use and storage of hazardous chemicals at U.S. military facilities often adversely affect the groundwater when contaminants infiltrate the subsurface as a result of leaks and accidental spills. These contaminants, if not located and remediated in a relatively short time, may move and settle unpredictably, essentially creating a source some distance from the original leak or spill. An example of this phenomenon is found with migrating dense nonaqueous phase liquid (DNAPL) contaminants. Although various methods for estimating the present day locations of these migrated contaminants are in use, accurately pinpointing the source of contaminants remains a difficult problem in …

The purpose of this study was to determine if a model existed which significantly increased the researcher's ability to accurately explain whether or not a recruited student will enroll based upon current recruitment strategies and demographic characteristics. The population for this study was defined as all prospective freshmen students who were recruited to attend Louisiana State University in the fall of 1995 and fall 1996. A random sample of the population was drawn from the population of prospective high school graduating seniors on the admissions data base for the years 1994-96. Women continue to be underrepresented as faculty at U.S. universities, especially in the physical sciences and engineering. The belief that only women can adequately serve as role models and mentors for other women may be a roadblock to progress for women in disciplines where they are underrepresented. This study of women faculty investigates how female career commitment is influenced by academic discipline and mentoring. The survey responses of sixty-six tenure-track women faculty were used to obtain data for both science or engineering and non-science or engineering academic disciplines at three public Ph.D. granting research universities. The nitroreduction of 4NQO and five structural analogs were studied with respect to radical production and mutagenicity. In the umu assay, 4NQO was the most mutagenic analog. Addition of microsomal and cytosolic protein from induced and uninduced rat liver samples decreased 4NQO mutagenicity in the standard strain (Salmonella typhimurium TA 1535/psk1002) 4NQO mutagenicity was also found to be decreased in the nitroreductase-deficient strain (Salmonella typhimurium TA1535NR/psk1002). In several strains of the Protox$\rm\sp{TM}$ assay 4NQO yielded the highest induction rate as compared to other compounds tested. In the presence of ascorbate 4NQO cytotoxicity, but not mutagenicity, was substantially enhanced. Oxygen consumption …

Introductions of exotic aquatic plants raise concerns for the health of assemblages of native species. The possible influence of the establishment of the exotic Eurasian watermilfoil Myriophyllum spicatum on fish and invertebrate assemblages in the Lake Pontchartrain estuary was examined with a microhabitat approach. Fish and invertebrate assemblage structure and population responses were compared among macrophyte species and unvegetated areas. Native macrophytes included wild celery Vallisneria americana and wigeongrass Ruppia maritima. Additionally, the relationships of the common submerged macrophytes to environmental and temporal variables were analyzed. Edwardsiella ictaluri is the causative agent of enteric septicemia of catfish (ESC), which is the most serious disease affecting the channel catfish industry. Killed vaccines have been inconsistent in providing protection against ESC, and no live attenuated vaccines have been described. The purpose of this study was to develop a nutritionally attenuated mutant strain of E. ictaluri and evaluate its virulence and vaccine efficacy. An E. ictaluri genomic library was constructed, and a clone containing the purA gene was isolated and sequenced. The Congressional declaration of national coastal policy in Section 303 of the Federal Coastal Zone Management Act (CZMA) of 1972 charges the coastal states to meet the objective of coastal resource protection. Vegetation dieback is an important component of the wetland loss process in coastal Louisiana. To determine the factors causing vegetation dieback in a Louisiana salt marsh and oligohaline marsh, field monitoring and manipulative field experiments were conducted in both marsh types. In the salt marsh, a three-year field study was conducted in which biotic and edaphic characteristics were measured and the effect of elevation on four salt marsh plant species was tested in a manipulative field experiment. Goat sperm cells were used as a model to evaluate sperm-mediated gene transfer. Fresh and frozen-thawed goat sperm cells were co-incubated with radio-labelled foreign DNA. After 10 extensive washings, the compacted sperm retained foreign DNA. Confocal microscopy confirmed that the position on the sperm cell responsible for binding of foreign DNA was located in both the anterior and the posterior heads of caprine sperm cells. Furthermore, confocal longitudinal sections of the sperm cells showed that foreign DNA had penetrated into goat sperm nuclei. Billions of dollars are spent each year on training in an effort to increase productivity so businesses can stay competitive. However, little is known about factors that impact the trainee's decisions to use what they learned in these training programs, back on the job. Since before Spanish Contact, the Pech Indians have occupied a large portion of northeastern Honduras. Like other native American populations, they have suffered significant territorial reductions and cultural alterations at the hands of European colonists and modern ladino immigrants. Utilizing the methodologies of cultural geography, ethnohistory, and ethnogeography, the Pech, formerly known as the Paya, are scrutinized to illustrate the process by which indigenous peoples are reduced and incorporated into a developing national setting. Hepatitis A virus (HAV), the causative agent of food and water borne acute hepatitis worldwide, is very difficult and expensive to propagate in tissue culture. Large scale production of HAV proteins in E. coli by recombinant DNA techniques would be invaluable for antisera production, biochemical analyses and in developing sensitive diagnostic tests. Therefore, PCR amplified DNA fragments encoding the HAV capsid proteins were cloned individually into prokaryotic expression vectors for production of either recombinant fusion or non-fusion proteins. Three experiments are reported that attempt to further our knowledge of the preparation and production of rapid sequential aiming movements. Specifically, these experiments examined possible reasons why in sequential aiming responses, the second movement segment (MT2) is consistently performed more slowly than first and third movement segments (MT1 and MT3). In the first experiment, participants struck 1, 2, or 3 targets in sequence without the benefit of visual feedback and without time stress to determine if MT2 is slowed due to a visually based on-line trajectory-correction process. The results showed that MT2 was not slower than MT1 under these condition. …

Alligator mississippiensis contains all of the components of the hepatic microsomal mixed function oxidase system. In general, basal and induced levels of P450 specific content, immunochemically detected abundance and examined activities are less than in mammals. In alligator at least two classes of P450s are inducible by xenobiotics: those induced by 3-methylcholanthrene (3MC) and those induced by phenobarbital (PB). PB and 3MC induce multiple P450 isoforms as detected by western blot and enzymatic analyses. The induced level of alkoxyphenoxazone O-dealkylation (AROD) was 10- to 100-fold lower in alligator than in rat, with the exception of phenylbenzyloxyphenoxazone O-dealkylation. In contrast to …

The main goal of the study was to provide the basic technical frame work for the use of native fish species as environmental sentinels for potential human exposure to genotoxic agents in general and to 1,2-dichloroethane (ethylene dichloride) in particular. First, LC/MS/ESI/ID and LC/MS/MS/ESI/ID methods for detection and quantification of S- (2-(N$\sp7$-guanyl)ethyl) glutathione DNA adduct from biological samples was developed. The methods were tested by exposing groups of rats with three dose levels of 1,2-dibromoethane. They allowed detection of $\sim$1 adduct in 10$\sp7$ bases. This study provides a description and analysis of the effects of a mathematics reform initiative on classrooms in two rural communities. It describes the sites, details interactions that take place in the classroom, and enumerates teachers' perceptions of the change. A mixed methodology was used to support the findings. Qualitative data were collected through classroom observations and interviews and quantitative data were gathered through questionnaires. Quantitative results indicated that teachers who participate in the training have the opportunity to learn more about new teaching strategies in mathematics and are likely to change their teaching practices in mathematics.
