Nature, Culture And Big Old Trees: Human Relationships With Ceiba (Ceiba Pentandra) And Live Oak (Quercus Virginiana) In The Landscapes Of Guatemala And Louisiana., Katharine Anderson

The ceiba in Guatemala and the live oak in Louisiana are examples of two trees that attain great size and age within the cultural landscape. Humans have adopted both as symbolic trees; they protect, encourage and even plant them. As the trees age, they become historic landmarks, are indicators of important places, and give character to the landscape. Although each tree is native within portions of each study area, both have taken advantage of human disturbance to expand beyond their original habitats. Edwardsiella ictaluri is the causative agent of enteric septicemia of catfish (ESC), which is the most serious disease affecting the channel catfish industry. Killed vaccines have been inconsistent in providing protection against ESC, and no live attenuated vaccines have been described. The purpose of this study was to develop a nutritionally attenuated mutant strain of E. ictaluri and evaluate its virulence and vaccine efficacy. An E. ictaluri genomic library was constructed, and a clone containing the purA gene was isolated and sequenced. This dissertation traces the evolution of Protestant missions in Central America from the colonial period to the present, focusing on the agents, direction, and methods of Protestant diffusion as well as the changing criteria for site selection. Chapters are arranged chronologically and progress from the macro-scale, Central America, to the micro-scale, six towns in southwestern Honduras (La Esperanza, Intibuca, Yamaranguila, San Juan, Erandique and Gracias). The chapters dealing with Central America outline when and where different mission boards have worked, as well as the geographical, economic, political and theological considerations driving site selection. Goat sperm cells were used as a model to evaluate sperm-mediated gene transfer. Fresh and frozen-thawed goat sperm cells were co-incubated with radio-labelled foreign DNA. After 10 extensive washings, the compacted sperm retained foreign DNA. Confocal microscopy confirmed that the position on the sperm cell responsible for binding of foreign DNA was located in both the anterior and the posterior heads of caprine sperm cells. Furthermore, confocal longitudinal sections of the sperm cells showed that foreign DNA had penetrated into goat sperm nuclei. The mechanism of fish protein solubilization by acid hydrolysis at elevated temperatures was investigated. Rate expressions for the solubilization were determined using pilot scale data with two different rate determination methods. A postulated mechanism was verified and estimated rates were evaluated on five independent verification experiments. A functional relationship between nonprotein byproduct concentration and whiteness of fish protein hydrolysate (FPH) was developed. Initial and set-point operating variables were calculated to produce FPH with acceptable color characteristics for food use. A batch reactor was used for duplicate hydrolysis trials conducted at three acid and three temperature levels for four hours. Samples …

The historical geography of Trujillo, Honduras is told through its changing hinterlands. This dissertation uses world systems to approach and understand the historical geography of Trujillo, however, the focus is on this peripheral area of the world system, rather than on the core. The coast of the western Caribbean experiences economic boom and bust cycles. The periodization of Trujillo's historical geography is based on the city's boom and bust periods, using primary documents from archives in Central America and Spain. Trujillo's first boom period lasted from its founding in 1525 until 1550, based on gold exports from the interior. During …

This work revises earlier interpretations of antebellum American humorists and sets forth a new model for understanding their accomplishments. Rooted in historical thinking, this study integrated biography, publishing history, critical responses, and a penetrating analysis of the sketches of several major figures. The first third examined Seba Smith, Joseph C. Neal, and Augustus Baldwin Longstreet. A devotion to their communities in Maine, Philadelphia, and Georgia, respectively, and preference for writing warm, genial selections remained important shared influences while cultural differences between the societies in which each matured and wrote made their humor diverge significantly. Seventy percent reduced fat Cheddar cheese was manufactured with homogenized cream and sweet cream buttermilk powder in an effort to improve flavor and texture characteristics. Manufacturing procedures, as well as the chemical, physical and sensory attributes. Pasteurized cream was homogenized (15.8 MPa) with and without the addition of buttermilk powder. The cream was added to pasteurized skim milk to produce two vats of cheese, one with and one without buttermilk powder. After milling, each vat was split to produce a washed curd and normal curd cheese. The four cheeses were sampled at week one, and months one, two and four. …

The focus of this dissertation is the relative tempo of molecular evolution among organisms of varying levels of relatedness. Because this is a rapidly expanding area of research, Chapter 1 is devoted to a review of the current state of knowledge on rates of molecular evolution. In Chapter 2, I present an analysis of relative rates of evolution among several distantly related rodents. The heterogeneity in rate of evolution of the cytochrome b gene observed here cannot be explained by metabolic rate, body size, generation time, or nucleotide-generation time. Since before Spanish Contact, the Pech Indians have occupied a large portion of northeastern Honduras. Like other native American populations, they have suffered significant territorial reductions and cultural alterations at the hands of European colonists and modern ladino immigrants. Utilizing the methodologies of cultural geography, ethnohistory, and ethnogeography, the Pech, formerly known as the Paya, are scrutinized to illustrate the process by which indigenous peoples are reduced and incorporated into a developing national setting. Hepatitis A virus (HAV), the causative agent of food and water borne acute hepatitis worldwide, is very difficult and expensive to propagate in tissue culture. Large scale production of HAV proteins in E. coli by recombinant DNA techniques would be invaluable for antisera production, biochemical analyses and in developing sensitive diagnostic tests. Therefore, PCR amplified DNA fragments encoding the HAV capsid proteins were cloned individually into prokaryotic expression vectors for production of either recombinant fusion or non-fusion proteins. Cloning and expression of HAV capsid genes, which induced toxic effects in certain host E. coli, was tolerated better by protease-deficient host …

This research was conducted to obtain a better understanding of the amino acid needs of finishing pigs. Twelve experiments were conducted using the urea nitrogen method to determine the lysine requirement of gilts and barrows. Two experiments were conducted to determine the optimum ratio of total sulfur amino acids to lysine for finishing pigs (74 to 111 kg); and three experiments were conducted to evaluate the net energy content of low crude protein, crystalline amino acid supplemented diets for finishing pigs (70 to 117 kg). Alligator mississippiensis contains all of the components of the hepatic microsomal mixed function oxidase system. In general, basal and induced levels of P450 specific content, immunochemically detected abundance and examined activities are less than in mammals. In alligator at least two classes of P450s are inducible by xenobiotics: those induced by 3-methylcholanthrene (3MC) and those induced by phenobarbital (PB). PB and 3MC induce multiple P450 isoforms as detected by western blot and enzymatic analyses. The induced level of alkoxyphenoxazone O-dealkylation (AROD) was 10- to 100-fold lower in alligator than in rat, with the exception of phenylbenzyloxyphenoxazone O-dealkylation. In contrast to …

The main goal of the study was to provide the basic technical frame work for the use of native fish species as environmental sentinels for potential human exposure to genotoxic agents in general and to 1,2-dichloroethane (ethylene dichloride) in particular. First, LC/MS/ESI/ID and LC/MS/MS/ESI/ID methods for detection and quantification of S- (2-(N$\sp7$-guanyl)ethyl) glutathione DNA adduct from biological samples was developed. The methods were tested by exposing groups of rats with three dose levels of 1,2-dibromoethane. They allowed detection of $\sim$1 adduct in 10$\sp7$ bases. Field cage studies were conducted to measure the effects of late season beet armyworm, Spodoptera exigua (Hubner), infestations (0, 1, 3, and 6 egg masses per 5.1 row m) on defoliation, fruit damage, and yield of cotton. Light penetration through the canopy was significantly higher in infested plots. Although a trend for increased numbers of damaged fruiting forms with increases in egg mass density was observed, there were no significant differences between infested and control plots. There were no significant differences in the cumulative number of shed fruiting forms or yield between infested and control plots. Field tests were conducted …
