The X chromosome linkage group is conserved in placental mammals. However, X chromosome morphological differences, due to internal chromosome rearrangements, exist among mammalian species. We have developed bovine chromosome painting probes for Xp and Xq to assess segment homologies between the submetacentric bovine X chromosome and the acrocentric sheep and goat X chromosomes. These painting probes and their corresponding DNA libraries were developed by chromosome micromanipulation, DNA micropurification, microcloning, and PCR amplification. The bovine Xp painting probe identified an interstitially located homologous segment in the sheep and goat Xq region, most probably resulting from chromosome inversion. Ten type II (microsatellite) …

Seventy-seven polymorphic microsatellites were analysed in offspring of three elite sires that were part of the foundation of an experimental population selected for twinning rate at the US Meat Animal Research Center, Clay Center, Nebraska. All females were assessed for ovulation rate by rectal palpation of corpora htea over 8-10 consecutive oestrous cycles from approximately 12 to 18 months of age, and associations between ovulation rate and sire allele were examined in each of the three sire groups. A newly developed rapid microbial ATP bioluminescence test IR-mATPI was shown to be an adequate means to assay the microbial load of poultry carcasses. This assay utilizes differential extraction and filtration to separate somatic from microbial ATP in a very rapid timeframe. The assay requires approximately 5 min to complete; approximately 3.5 min to sample and 90 s analytical time. Correlation coefficient (r) between aerobic colony counts and R-mATP test results In (n= 329) was 0.82. Post-test probabilities to correctly classify carcasses with different levels of microbial contamination were as high as 98% for samples of ≥ 3.5 …

Lean and adipose beef carcass tissues inoculated with Brochothrix thermosphacta (BT) (approx. 4·50 log10 cfu cm-2) were left untreated (U) or treated with 100 µg ml-1 nisin (N), calcium alginate (A) or 100 µg ml-1 nisin immobilized in a calcium alginate gel (AN). Tissue samples were refrigerated after treatments and bacterial populations and nisin activity were determined at 0, 1, 2 and 7 d. U, A and N treatments of lean and adipose tissues did not suppress bacterial growth (>6 log10 cfu cm-2 by day 7) while treatments of lean and adipose …

Effectiveness Of A Steam-Vacuum Sanitizer For Reducing Escherichia Coli 0157:H7 Inoculated To Beef Carcass Surface Tissue, W. J. Dorsa, C. N. Cutter, G. R. Siragusa

A steam-vacuum sanitizer reduced aerobic plate counts associated with bovine faecal cantamination from 5·5 log10 cfu cm-2 to 3·0 ± 0·21 log10 cfu cm-2 on beef carcass short plates. The same beef carcass short plates inoculated with 7·6 ± 0·09 log10 cfu cm-2 Escherichia coli 0157:H7 in faeces, yielded an average residual level of E. coli 0157:H7 of 2·1 ± 0·21 log10 cfu cm-2 after steam-vacuum treatments. This study deomonstrates the effectiveness of a steam vacuum sanitizer for removing E. coli 0157:H7 from beef carcasses.

Six bacterial sampling methods that might be used for rapid sampling of beef carcasses were evaluated in two separate studies. In Study 1, bacterial recovery from uninoculated beef rounds was 2·6, 2·3, 2·1 and 1·3 log10 cfu cm-2, respectively for excision (EX), and swabbing with cheesecloth (CC), sponge (SP) and cotton-tipped wooden swabs (CS). For Study 2, beef tissue was inoculated with bovine faeces at different levels and the mean recovery was 3·7, 3·0, 3·1 and 3·1 log10 cfu cm-2, respectively for EX, and swabbing with SP, griddle screen (GS) and 3M mesh (M). …

The Rio Grande Delta of Texas is one of the most diverse ecological areas in Texas. The area is favorite among ornithologists due to the presence of many species that winter there or migrate through the area. Similarly, the Delta harbors several rare mammals, reptiles, amphibians, and a host of invertebrates, which are found nowhere else in North America. Most ecologists attribute this unique faunal diversity to the great variety of vegetation that characterizes the Rio Grande Delta. Accurately identifying the vegetation is therefore an important aspect of field research performed in the region.

Recent occurrences and reports of gray wolves (Canis lupus) in North Dakota have generated public interest and created demands on management agencies. We summarized reports of wolves in North Dakota received from the public and government personnel, and evaluated suitability of habitat for wolves. We suggest that in recent years dispersing wolves have sporadically occurred in all quarters of the state except the southwest, and non-dispersing wolves have occurred in the Turtle Mountains region in north-central North Dakota. Habitat data from the 1,500 km2 Turtle Mountains physiographic region, bisected by the North Dakota/Manitoba border, suggest the potential …

She writes in first person with short, easy-reading chapters about her life, generally in chronological order from childhood to present. The anecdotes are concise, witty, and spiced with humor in her characteristic, blissful confidence. There are many unique black-and-white photographs reproduced, as well as pen-and-ink illustrations mostly by her artist-daughter, Elva Hamerstrom Paulson. How politico-military force will be employed in future war is of major concern to U.S. national security and, because of its operational leadership in this area, to the Army's Force XXI campaign plan. It is envisioned that such force will qualitatively change because of the introduction of advanced technology. This technology will extend the West's continuing mastery of the application of political violence.
