Characterization Of An Octopamine Receptor Gene In Drosophila Melanogaster., Kelley Lynn Colvin

The biogenic amine octopamine is a versatile signaling molecule that acts in essential physiological functions in invertebrates. Its roles in neurotransmission, neurohormone regulation and neuromodulation are achieved through interaction with G-protein coupled receptors. Such invertebrate receptors are structurally and functionally analogous to vertebrate adrenergic receptors. A human brain β2-adrenergic receptor cDNA clone was used as a hybridization probe to identify a Drosophila gene encoding an octopamine/tyramine receptor. This 367 base pair fragment of Drosophila genomic DNA, homologous to the putative sixth and seventh transmembrane regions of adrenergic receptors, was used to isolate an octopamine/tyramine receptor 3.3 Kb cDNA clone from …

Zinc is essential for normal growth, however, very little is known about the mechanisms by which zinc influences growth. ALP, an ectoenzyme expressed in high levels in osteoblasts, is a zinc-containing metalloenzyme and is important in bone formation. Preliminary studies showed that low levels of zinc could increase ALP specific activity in human osteosarcoma, SaOS-2 cells. Current studies were designed to characterize this zinc effect, test possible mechanisms, and determine whether the effect is physiologically relevant. Incubation with zinc resulted in a time and dose dependent increase in ALP specific activity in the extracts and conditioned media of SaOS-2 cells, …

Bacterial cells contain various restriction-modification (R-M) systems to protect themselves from incoming foreign DNA. Most bacteria have at least one such system. In this study, the restriction genes and the location of the genes on the chromosome were examined in three enteric bacteria. Salmonella typhimurium LT2, Klebsiella pneumoniae GM 236, and Klebsiella pneumoniae M5al. The 98 minute region of the Salmonella typhimurium LT2 chromosome comprises at least two R-M systems. From genetic data this region was previously thought to be only one minute in length but was found to be larger than two minutes using a physical mapping technique, pulsed …

Several animal models have been proposed to develop pulpal inflammation. Procedures recommended in these models have many variables and have not produced consistent results. The purpose of this study was to create pulpal inflammation using the Nd:YAG laser.

Forty-five cuspids of 12-18 month old beagle dogs were lased at 5, 7.5, and 10 watts (W) for 20 seconds with a 600 micron contact probe. Five cuspids were not lased but were used as controls. Following laser application, the teeth were extracted at intervals of 3, 7, and 30 days. They were then fixed in 10% formalin, decalcified, sectioned, and stained …

Supreme Court decisions have vacillated between two incompatible readings of the Fifth Amendment guarantee that no person "shall be compelled in any criminal case to be a witness against himself." The Court sometimes sees this language as affording defendants and suspects a right to remain silent. This interpretation - a view that countless repetitions of the Miranda warnings have impressed upon the public - asserts that government officials have no legitimate claim to testimonial evidence tending to incriminate the person who possesses it. Although officials need not encourage a suspect to remain silent, they must remain at least neutral toward …

A limited number of studies have been conducted to assess the perceptions of risk of medical students and residents for acquiring Human Immunodeficiency Virus (HIV) or Hepatitis B Virus (HBV) via percutaneous (needlestick) injuries while performing clinical training. The purpose of this study is to investigate these perceptions of risk, and to determine whether there is an association between a high perception of risk and a low incidence of needlestick injury. Interleukin-10 (IL-10) is a recently described pleiotropic cytokine secreted mainly by type 2 helper T cells. Previous studies have shown that IL-10 suppresses cytokine expression by natural killer (NK) and type 1 T cells, thus down-regulating cell-mediated immunity and stimulating humoral responses. We here report that injected IL-10 protein is an efficient inhibitor of tumor metastasis in experimental (B16-F10) and spontaneous (M27 and Lox human melanoma) metastasis models in vivo at doses that do not have toxic effects on normal or cancer cells. Brush border membrane vesicles from larvae of the tobacco hornworm, Manduca sexta, contain protein bands of 85 and 120 kDa which react directly with streptavidin conjugated to alkaline phosphatase. The binding could be prevented either by including 10 μM biotin in the reaction mixture or by prior incubation of the brush border membrane vesicles with an activated 60- to 65-kDa toxin from Bacillus thuringiensis HD-73. The ability of B. thuringiensis toxins to recognize biotin-containing proteins was confirmed by their binding to pyruvate carboxylase, a biotin-containing enzyme, as well as to biotinylated ovalbumin and biotinylated bovine serum albumin but not …

Spatial And Temporal Variability In The Distribution Of Nitrate In An Agriculturally-Impacted Aquifer, Cass County, Michigan, Caroline Lovetere

Ground-water sampling in a shallow unconfined aquifer within a predominantly agricultural area in southwestern Michigan indicated spatial and temporal variability in the distribution of nitrate. The unconfined aquifer, which is about 40 feet thick, consists of glacially-deposited sand and gravel with interbedded clay lenses and is separated from a lower semi-confined aquifer by a 3-to 26-feet-thick till layer.

The site of the study was a 47-acre agricultural field, characterized by corn and alfalfa hay production, to which hog manure and commercial fertilizers were frequently applied. The shallow water table and relatively permeable glacial sands in the vadose zone combined to …

When the Muslim kingdom of Valencia was conquered between 1233-1245 during the Reconguista, by James I, a new era was ushered in for the resident Muslims and Jews, because they would now be ruled by a minority Christian government. King James, in an attempt to control the newly settled Christians, issued the law code the Furs of Valencia. Additionally the Furs also established Christian legal superiority over the Muslims and Jewish populations, too.

Substance abuse treatment outcome is an important area for clinical research. Furthermore, it is important to identify sub-populations which may require additional treatment services in order to improve outcome. Cocaine dependent individuals have been identified as a group that provides many challenges to clinicians and researchers due to the high incidence of relapse. The current study explored the differences in treatment retention between persons diagnosed with either cocaine or alcohol dependence. A method is proposed to analyze the behavior of sequential algorithms. Most current methodologies are in the form of semantic analysis, domain decomposition or fixed point approximation. These tools, though useful in design, verification and testing, are not general enough to characterize the behavior at run-time without the aid of a correct set of specifications. Elders frequently reside in extended care facilities and experience feelings of loneliness, depression, disorientation, and helplessness (McQuillen, 1985). The Book of Mormon annals open in an ancient Near Eastern context. The archaeological-historical context is carefully outlined here within a systematic chronology that is tied to fixed, absolute dates of recorded astronomical events—particularly those from cuneiform eponym calendars. The resultant matrix allows those early Book of Mormon events to be understood in a rational, familiar, and meaningful way—that is, in a biblical context.
