N-Thiolated β-lactams represent a novel family of antibacterial agents, whose in vitro activity is confined largely to Staphylococcus species, including multidrug-resistant forms of S. aureus. N-Thiolated β-lactams have recently been shown to possess intriguing biological activities which are addressed in Chapter II. Current development of nanoparticles as a new drug delivery vehicle is described in Chapter III. Chapter IV and V described the current research in our laboratories focusing on the synthesis and characterization of emulsified polyacrylate/polyacrylamide nanoparticle antibacterials for drug delivery of water-insoluble antibiotics. The West Florida Shelf (WFS), typically characterized as being oligotrophic, is one of the most productive continental shelves in the United States. In addition to supporting a large fishing industry, the WFS also supports high biomass blooms of the toxic dinoflagellate Karenia brevis. Because of the large ecological and economic impacts these blooms have on the area, the ECOHAB: Florida program was developed to gain a better understanding of red tides and their initiation, maintenance, and dispersal. The Florida panther (Puma concolor coryi) is a relatively well studied species, but some aspects of its habitat requirements remain poorly understood. While it has been well established that the most important threat to panthers include limited habitat area and continued habitat loss and fragmentation, the importance of roads in this context has not been determined. The goal of this research is to determine the influence of roads on the movement patterns of the Florida panther. This dissertation presents the theory and practice of designing, synthesizing and using peptidomimetics to disrupt protein-protein interactions. Our general strategy is to design and synthesize peptidomimetics that will mimic peptide secondary structures (alpha-helices and beta-sheets). Chapter One is a theoretical examination of the feasibility of using beta-sheet mimics called molecular clamps to inhibit substrate-receptor interactions by blocking the substrate rather than the receptor or enzyme. Industrialization and "development" during the last 200 years have led to an increase of pesticides, an intensified use of synthetic chemicals, higher levels of environmental pollution, and more exposure to hazardous working conditions. Environmental toxins, many of which are endocrine disruptors, are stored in fat tissue, increasing reproductive health risks for both women and men. Women’s bodies are particularly vulnerable as sites for creating, growing, feeding, and nurturing the next generation. Image analysis was used to measure basal areas of selected colonies of Montastraea annularis and Porites astreoides, following the colonies over a three-year period from 2002 to 2004. Existing digital images of permanently-marked quadrats in the Caye Caulker Marine Reserve, Belize, were selected based on image quality and availability of images of selected quadrats for all three years. Annual growth rates were calculated from the basal-area measurements. Mean growth rates (radial skeletal extension) for M. annularis and P. astreoides were 0.02 cm yr-1 and -0.20 cm yr-1, respectively. Basal area measurements demonstrated a large degree of variability. Electric cell-substrate impedance sensing (ECIS) pioneered by Giaever and Keese is suitable for continuous, automatic and real-time cell attachment analysis. ECIS is a novel electrical method to study, in real time, many of the activities of animal cells when grown in tissue culture. These include morphological changes, cell locomotion, and other behaviors directed by the cell's cytoskeleton. One of the most direct ECIS measurements is that of the attachment and spreading behaviors of cells. This work is a comparative analysis of the works of the Ukrainian author Oksana Zabuzhko (Field Work in Ukrainian Sex) and the Algerian writer Assia Djebar (Women of Algiers in Their Apartment). Although the lives of Algerian and Ukrainian women were shaped by different historical and social forces, discourses and traditions, common themes exist in their writings because of their common postcolonial background. Both authors examine the relations of women to history in the postcolonial setting, the problem of inscribing women into history, and the double oppression women experienced during colonial times (as colonized subjects and as gendered subjects).

A study of how mediators interact with the catabolic pathways of microbes was undertaken with a view towards improving the performance of microbial fuel cells. The use of mediators is known to improve the power density in microbial fuel cells, but this work suggests that no single mediator is ideally suited to the task. Instead, a carefully selected mixture of two targeted mediators (Methylene Blue and Neutral Red) might be optimal. To test this hypothesis, a yeast-catalyzed microbial fuel cell was built and empirically evaluated under different mediation conditions while keeping all other parameters constant. This study investigated the mental health of high school students enrolled in the International Baccalaureate (IB) High School Diploma Program (n =139) in a large, southeastern high school. Mental health was assessed using both positive indicators (life satisfaction, academic achievement, academic self-efficacy) and negative indicators (psychopathology) indicators of adolescent social-emotional and school functioning. Findings from this study include that IB students perceive more stress than their general education peers, yet maintain mental health that is equivalent or superior to that of their general education peers. The role of coping in predicting mental health outcomes in IB students was also investigated. …

Where lead pollutes urban soils, both human and animals risk exposure. This exposure gives rise to similar health risks across species. A group of 50 outdoor living cats from inner city Tampa, Florida were tested for blood lead concentration (BLC). Most cats had no measurable lead loads, 14 percent had levels less than or equal to 6 micrograms per decaliter. Soil samples were taken from the home location of each cat. None of these samples, which ranged from 2.6 microgram per gram to 170 micrograms per gram, had hazardous levels of lead. Overall, BLCs were lower than expected. In addition, …

The purpose of this study was to investigate the relationship between residential segregation (the physical separation of Blacks and Whites in residential contexts) and adverse perinatal outcomes (low birth weight, preterm delivery and small for gestational age births) in Florida, Georgia and Louisiana. The study determined the independent effect of the level of residential segregation on the likelihood of adverse perinatal outcomes after controlling for contextual and individual factors. This dissertation examined the potential protective effects of both active A-beta immunotherapy and dietary omega-3 fatty acid administration to AD transgenic mice. The Earth's most extensive living space is found in the bathypelagic zone of the oceans, yet research in these areas is scant. The micronekton of the bathypelagic zone in the eastern Gulf of Mexico (EGOM) was investigated with the goals of comparing its community structure and trophic interactions with those of the well-studied overlying mesopelagic micronekton. Significant changes in faunal structure were found, including shifts in dominant families as well as species. Compared to the mesopelagic zone, the bathypelagic community had increased abundance and biomass contributions from the Gonostomatidae, Oplophoridae,and Eucopiidae, with a simultaneous decrease in the importance of the …

The hearing abilities of elasmobranch fishes were examined in response to several types of stimuli using auditory evoked potentials (AEP). Audiograms were acquired for the nurse shark, Ginglymostoma cirratum, the yellow stingray, Urobatis jamaicensis, in a controlled environment using a monopole underwater speaker. A dipole stimulus was used to measure the hearing thresholds of the horn shark, Heterodontus francisi, and the white-spotted bamboo shark, Chiloscyllium plagiosum. The dipole experiments yielded much lower thresholds than any other experiment, suggesting that this type of sound specifically stimulated the macula neglecta by creating a strong velocity flow above the head of the shark. …

Analysis of the genomic sequences of Streptococcus mutans UA159 and Group B Streptococcus (GBS) strains Streptococcus agalactiae NEM316 and S. agalactiae 2603V/R indicated the presence of two putative collagenase genes in each organism. smcol1 from S. mutans was previously cloned and analyzed and the results indicated that the enzyme belonged to the U32 family of collagenases/peptidases. This enzyme shares homology with the prtC of Porphyromonas gingivalis, one of the principal examples of the U32 family of peptidases.
