This dissertation explores the culture of sport among African American male football players as well as African American perspectives on sport and success. A case study of six African American, Division 1 FBS (Football Bowl Subdivision) collegiate student athletes was conducted along with seventeen supplemental interviews with community members, parents, coaches and former athletes and fans. The participants answered questions that explored education, success, identity construction, ethnicity and sport. Archival data was also reviewed framing the discussion on football in Florida, links between education and sport participation and African American male academic achievement. Epithelial ovarian cancer (EOC) is a leading cause of morbidity and mortality among women in the United States, and the etiology is incompletely understood. Common, low penetrant genetic variants such as single nucleotide polymorphisms (SNPs) likely contribute to a significant proportion of EOC. We examined whether SNPs in two understudied yet biologically important types of genes, mitochondrial-related and miRNA-related genes, may contribute to EOC susceptibility using data from a large, homogeneous study population of 1,815 EOC cases and 1,900 controls (frequency-matched on age-group and race/ethnicity) genotyped through stage 1 of an ongoing genome-wide association study. Background: Helicobacter pylori (H. pylori) infection is a significant risk factor for peptic ulcer (PU) and gastric cancer (GC). Apart from the virulent CagA genotype of H. pylori environmental and dietary factors influence disease outcomes. There have been no studies addressing these factors in Western India. Hence, we conducted a case control study enrolling PU, GC patients and controls at Pune, India.

Methods: Risk factors for PU and H. pylori infection were assessed in participant interview. H. pylori status was assessed from stool by monoclonal antigen detection. To understand treatment effect, we followed 100 H. pylori positive patients.

Altered DNA methylation may lead to suboptimal fetal programming, increasing the risk of adverse pregnancy outcomes such as small for gestational age (SGA); however, few studies have examined the associations between DNA methylation, prenatal exposures, and fetal outcomes. Cross-sectional data from a larger, ongoing study were used to assess the impact of prenatal smoking on gene specific methylation of umbilical cord blood derived DNA and to investigate the association between gene-specific methylation and risk of SGA. Knowledge of many life history parameters are essential to properly assess and manage fish species. Although the snowy grouper, Epinephelus niveatus, is a commercially valuable fish, which is harvested throughout the Gulf of Mexico, little is known about its age, growth, and reproduction from this region. In this study, snowy grouper from the northern and eastern regions of the Gulf of Mexico were examined primarily using commercially-derived samples that were collected between 1984 and 2004. Understanding emerging viruses is critical for disease monitoring and prediction; however, surveys of novel viruses are hindered by the lack of a universal assay for viruses. Viral metagenomics, consisting of viral particle purification and shotgun sequencing, is a powerful technique for discovering viruses in a wide variety of sample types. However, current protocols are not effective on tissue samples (e.g., lungs, livers and tumors), where they are hindered by the high amount of host nucleic acids which limits the percentage of sequences that originate from viruses. Ovarian cancer (OC) is the second most common gynecologic cancer; however it is responsible for the most gynecologic cancer-related deaths. Apoptosis evasion is an important mechanism in OC tumorigenesis, and the prototypic anti-apoptotic protein, B-cell lymphoma 2 (Bcl-2), is often overexpressed in OC tumors. Gaining a better understanding of the mechanism(s) behind Bcl-2 overexpression and potential extra-anti-apoptotic functions of Bcl-2 could elucidate the importance of elevated Bcl-2 in OC. The complex interaction between predators and their prey is rarely preserved in the fossil record. However, predation of marine mollusks by drilling gastropods leaves a diagnostic hole in the shell of the prey, possibly allowing for quantitative analysis of this ecological interaction. Drilling frequency, as measured in marine mollusks both in the Modern and fossil record, has been heralded as a potential opportunity to quantify these ecological interactions and use these values in the testing of hypotheses.

Phosphorylation of proteins is a reversible and very important regulatory mechanism that occurs in both prokaryotes and eukaryotes. Chromatin Unfolding By Cdt1 Regulates Mcm Loading Via Opposing Functions Of Hbo1 And Hdac11-Geminin, Philip G. Wong

The efficiency of metazoan origins of DNA replication is known to be enhanced

by histone acetylation near origins. Although this correlates with increased MCM

recruitment, the mechanism by which such acetylation regulates MCM loading is

unknown. We show here that Cdt1 induces large-scale chromatin decondensation that is

required for MCM recruitment. This process occurs in G1, is suppressed by Geminin, and

requires HBO1 HAT activity and histone H4 modifications. HDAC11, which binds Cdt1

and replication origins during S-phase, potently inhibits Cdt1-induced chromatin

unfolding and re-replication, suppresses MCM loading, and binds Cdt1 more efficiently

in the presence of Geminin. We also …

Peptide a-amidation is known as a signature of bioactivity due to the fact that half of the bioactive peptides found in the nervous and endocrine systems are a-amidated and that most known a-amidated peptides are bioactive. a-Amidated peptides are produced by the oxidative cleavage of glycine-extended precursors. Peptidylglycine a-amidating monooxygenase (PAM) is the only known enzyme responsible for catalyzing this reaction and its sole physiological function is to convert glycine extended prohormones to their a-amidated forms. High levels of PAM are found in certain tissues with no corresponding level of amidated products suggesting the presence of undiscovered a-amidated peptide hormones. …

global climate have created proposals for the reduction of carbon emissions from large

Familial history is the strongest risk factor for developing ovarian cancer (OC), and a significant contributor to breast cancer risk. Most hereditary breast cancers and OCs are associated with mutation of the tumor suppressor Breast and Ovarian Cancer Susceptibility Gene 1 (BRCA1). Studying risk-associated BRCA1 truncation mutations, such as the founder mutation 185delAG, may reveal signaling pathways important in OC etiology. Laser-Induced Breakdown spectroscopy (LIBS) and Laser-Induced Thermal Emission (LITE) emission based laser remote sensing were investigated with the application to the remote measurements of trace chemical species. In particular, UVvisible LIBS and Mid-IR LITE systems were developed and measurements of remote targets and chemical surfaces were studied. The propagation through the atmosphere of the multi-wavelength backscattered LIBS and LITE optical spectrum with atmospheric absorption effects on the returned lidar signal was investigated. An enhanced model of the atmospheric effects on emission-based laser-remote sensing was developed and found to be consistent and in agreement with our experimental results.

This dissertation explores the hydrologic controls on salinity within mangroves and lagoons at sites in Florida and Mexico. The main objective of this research is to better understand hydrologic controls on mangrove ecosystem structure and develop ideas that will be useful to land managers attempting to regulate and conserve these critical habitats. This study was conducted at sites in Ft. Pierce, FL and Costalegre on the central Pacific coast of Mexico.

Studies of food insecurity have frequently focused on rural dwellers as vulnerable

populations. However, during the ‘global food crisis’ of 2007-2008, riots in more than

50 countries visibly demonstrated the vulnerability of urban populations to food

insecurity due to rapidly rising food prices. This study examines factors associated with

participation in an urban garden project (UGP), utilizing surveys (n=61) and in-depth

household interviews (n=37) to examine food security and dietary diversity of households

in urban Lesotho.

Households that participated in the garden project were more food insecure and

The analysis of craniometric data collected from skeletal remains, combined with

archaeological data, can provide very valuable information pertaining to biological

distance and gene flow among prehistoric populations through space and time. The goal

of this study was to examine microevolutionary change among prehistoric populations in

south Florida based on the degree of cranial variation among populations at seven

prehistoric sites. It was expected that as time progressed, microevolutionary forces

caused significant changes in the crania of the various populations based on the relative

Despite an increase in the number of Ecuadorian medical professionals, health outcomes in rural areas of the country have steadily declined over the past decade. Using a political economic framework and data collected from interviews with Ecuadorian doctors, government officials, policy makers, and local rural populations, this thesis investigates how the interplay among medical migration, educational structures, and public health policy contributes to growing health disparities between urban and rural dwellers. Angiogenesis by either normal or neoplastic cells involves a delicate balance of both angiogenic and angiostatic regulators. In the ovary, normal physiological angiogenesis occurs around the developing follicle and corpus luteum in response to hormonal shifts. Interestingly, carcinomas arising from the ovary are usually highly vascularized and are commonly clinically observed to produce cyst fluids or ascites which contain both angiostatic and/or angiogenic regulators. Melanoma is an aggressive disease with few effective treatment options. Non-toxic, anti-tumor therapies and prophylactic approaches are currently being investigated to identify treatment options that will control and remove late-stage melanoma.

The overall goal of this project was to establish an effective delivery method for a plasmid encoding human interleukin (phIL-15) into mouse melanoma cells (B16.F10) using the gene transfer technique electroporation (EP)1. The EP delivery phIL-15 was optimized using an in vitro 3D tumor model. Plazas and patios were important spaces for expressing power and social identity in prehispanic Mesoamerica. However, plazas can be analytically problematic, because they were often kept clean of material debris. Previous geoarchaeological studies of anthropogenic soils and sediments have shown that specific activities leave characteristic chemical signatures on prepared earthen surfaces. The research presented here uses soil chemical residue analysis and excavation data to examine use patterns in the North Plaza of Palmarejo, Honduras during the Late Classic period.
