Structural Basis For Ternary Complex Formation Between Tau, Hsp90, And Fkbp51, Alexander Steven Barrett

The accumulation of the microtubule associated protein tau has been implicated in several neurological disorders; however, its interaction with chaperones along its normal degradation pathway remains largely uncharacterized at single residue resolution. In this study, nuclear magnetic resonance (NMR) spectroscopy was used to probe the interaction between tau, the molecular chaperone Hsp90, and the immunophilin FKBP51. Resonance intensity changes were observed for specific residues in the heteronuclear single quantum coherence (HSQC) spectra of 15N-labeled tau in the presence of Hsp90 and/or FKBP51. Analysis of the HSQC spectra identified the two hydrophobic hexapeptide motifs located at residues V275 - K280 and …

Environmental temperature exhibits profound effects on the activity and ecology of ectotherms through its impact on muscle contractile physiology. While the performance of locomotor behaviors powered by muscle contraction directly decreases by at least 33% over a 10°C drop in body temperature, chameleons are known to feed, presumably with high performance, at body temperatures where sympatric lizard species remain inactive. I propose that ballistic movements that are powered by the recoil of preloaded elastic and collagenous tissues are less thermally dependent than movements that rely on direct muscular power. Recirculating aquaculture systems (RAS) produce aquaculture products on land with minimal discharge of waste products and minimal water loss. High costs associated with waste treatment for RAS have triggered the growth of integrated aquaculture systems (IAS) which incorporate macrophytes (aquatic plants) into the treatment train. The objective of this research was to examine a pilot scale inland marine IAS with three different methods for solids treatment: a sand filter followed by a plant bed, only a plant bed, and geotextile bags. Signal transduction and stage-specific gene expression are essential components of Plasmodium falciparum development. In this study, the putative phosphatase PF13_0027 is investigated as a critical component of intraerythrocytic development contributing to maturation of the late trophozoite. This putative phosphatase was identified during the course of a large-scale insertional mutagenesis project by insertion of the piggyBac (pB) element, containing a human dihydrofolate reductase (hDHFR) drug selection cassette into the open reading frame (ORF) preventing expression and attenuating parasite development. The goal of this study is to explore specialized and popular cultural models of trachoma, and the interaction between the trachoma elimination program and its target audience in one trachoma hyper-endemic intervention community. Eighty four million people worldwide, mainly children, are infected with active bacterial trachoma. For some, this will lead to painful and progressive corneal opacity and eventual blindness. The disease is most commonly spread by person-to-person contact or by flies, and affects very specific populations living in resource-poor areas such as rural Niger, which has one of the highest prevalence rates worldwide.

Relative timing ratios are a useful measure for determining the temporal regularities of speech. The timing intervals that make up these ratios are thought to be important when creating the motor plan for an utterance (Weismer & Fennell, 1985). In fact, these ratios have been shown to be remarkably stable, even when speakers deliberately increase their rate (Tuller & Kelso, 1984; Weismer & Fennell, 1985). Despite exposure of educational disparities for students of color, as well as the notion that educational training rarely discusses race and racism, there continues to be a lack of discourse on race, racism, and anti-racism in educational leadership. Subsequently, it is important to challenge deficit thinking and encourage further examination of the deeply-rooted foundation of oppression. The study explored personal narratives of white educational leaders who oppose racial inequity to heighten awareness about conceptualizations of race, racism, and anti-racism. Protein tyrosine nitration (PTN) is a posttranslational modification resulting from oxidative/nitrosative stress that has been implicated in a wide variety of disease states. Characterization of PTN is challenging due to several factors including its low abundance in a given proteome, preferential site modification, multiple target site proximity within unique peptide sequences, and analytical method and instrument limitations. Current analytical techniques are insufficiently sensitive to identify endogenous nitration sites without incorporation of either nitrotyrosine or target protein enrichment. The peripheral immune system plays a role in delayed neural injury after stroke. This response originates from the spleen as splenectomy prior to middle cerebral artery occlusion (MCAO) in rats significantly reduces infarct volume in the brain. This research is based on the hypothesis that inhibiting the splenic response will reduce neurodegeneration after stroke. Studies in animals have implicated lymphocytes as the immune cell type that is detrimental following MCAO. Interferon gamma (IFNγ) has been identified as a pro-inflammatory cytokine that is also detrimental following stroke. Harmful Algal Blooms (HABs) in the Gulf of Mexico (GOM) are natural phenomena that can have negative impacts on marine ecosystems on which human health and the economy of some Gulf States depends. Many of the HABs in the GOM are dominated by the toxic dinoflagellate Karenia brevis. Non-toxic phytoplankton taxa such as Scrippsiella sp. also form intense blooms off the Mexican coast that result in massive fish mortality and economic losses, particularly as they may lead to anoxia.

Many of the open problems of current interest in probability and statistics involve complicated data

sets that do not satisfy the strong assumptions of being independent and identically distributed. Often,

the samples are known only empirically, and making assumptions about underlying parametric

distributions is not warranted by the insufficient information available. Under such circumstances,

the usual Fisher or parametric Bayes approaches cannot be used to model the data or make predictions.

Anti-Parasitic And Anti-Bacterial Agents: Studies On 1,4-Dihydropyridines And 2,4-Diaminoquinazolines, Kurt Steven Van Horn

Thirty-three 1,4-dihydropyridine diastereomeric pairs were synthesized and the structure-activity relationship studied in a Plasmodium falciparum in vitro model. Twenty-nine of these derivatives contained a 6-position oxygen, with 2.31, 2.32, 2.52 and 2.53 having single and double digit nanomolar activities. This SAR study revealed some insightful information about the 1,4-dihydropyridine substitution pattern. Substitution at the 7-position other than 3,4-dimethoxy severely reduced the activity. 4-phenyl substitution with 2- or 4- halo or methyl formed active compounds while substitution at the 3-position or with methoxy or conjugated aryl systems resulted in inactive compounds. This dissertation explores a body of films produced after the events of 9/11, and while examining this specific point of departure, the author presents the argument on the vast cultural relevancy of the omnipresent zombie. These films are interrogative and complex, offering the viewing audience a rich tapestry of interwoven meanings. Furthermore, the author suggests that the zombie trope has, in fact, left the genre altogether, reinserted into a style of films he labels as "non-zombie appropriation." The objective of the present study is to address some important questions related to prostate cancer treatments and survivorship among White and African American men. It is commonly understood that the risk of developing prostate cancer is higher in African American men than the other races. However, using parametric analysis, this study demonstrates that this perception is a "myth" not a "reality". The study further identifies the existence of racial/ethnic disparities by comparing the average mean tumor size, the median of survival time, and the survival function between White and African American men. The Assessment Of An In-Vitro Model For Evaluating The Role Of Parp In Ethanol-Mediated Hepatotoxicity, Jayme Coyle

This investigation assesses the role of poly(ADP-ribose) polymerase in ethanol-mediated hepatotoxicity using the untransfected HepG2 hepatocellular carcinoma line, an established, well-characterized toxicological model. HepG2 cells were treated with ethanol at concentrations between 100 mM and 800 mM, and assessed for markers of cytotoxicity. PARP-1 activity in total cell protein lysates was quantified as a proxy of apoptotic induction at six hours. Our results demonstrated a 1.43-fold AST activity increase in culture medium isolates of cells exposed to 800 mM without significant effect on cellular viability. PARP-1 activity varied greatly and results for enzyme activity remained inconclusive. The results suggest a …

Pedicle screw-rod instrumentation is considered a standard treatment for spinal instability, and titanium is the most common material for this application. Cobalt-chromium has several advantages over titanium and is generating interest in orthopedic practice. The aim of this study was to compare titanium versus cobalt-chromium rods in posterior fusion, with and without transverse connectors, through in vitro biomechanical testing and determine the optimal configuration.

Our laboratory recently reported that treatment with the d-amino acid containing peptide HYD1 induces necrotic cell death in multiple myeloma (MM) cell lines. Due to the intriguing biological activity and promising in vivo activity of HYD1, we pursued strategies for increasing the therapeutic efficacy of the parent linear peptide. These efforts led to the development of a cyclized peptidomimetic, MTI-101, with increased in vitro activity and robust in vivo activity as a single agent using two myeloma models that consider the bone marrow microenvironment. Attrition rates for anticancer drugs are much higher than any other therapeutic area. Only 5%#37; of the agents that demonstrate anticancer activity in the preclinical stages of development demonstrate clinical efficacy in phase III trials. This high attrition rate becomes alarming when we consider that the cost of research and development can amount to 1 billion dollars. To exacerbate this problem, many new cancer drugs are being discontinued, withdrawn or suspended. Patient survival post liver transplant (LT) is important to both the patient and the center's accreditation, but over the years physicians have noticed that distant patients struggle with post LT care. I hypothesized that patient's distance from the transplant center had a detrimental effect on post LT survival. I suspected Hepatitis C (HCV) and Hepatocellular Carcinoma (HCC) patients would deteriorate due to their recurrent disease and there is a need for close monitoring post LT. Recent studies suggest that some cases of prenatal depression may be associated with reactivation of latent infections of the herpesvirus family. The possible relationships among stress, prenatal depression, and herpes viral reactivation in pregnancy are understudied and the molecular pathways such as the neuroimmune biogenic amine pathway are unidentified. Chronic stress shifts the T helper-1 cell (Th1) cytokine profile to a Th2 profile, which favors virus induced pathogenesis and survival. Pregnancy is also associated with a similar Th2 dominance. PANcreatic-DERived Factor (PANDER), or FAM3B, is a 235-amino acid protein strongly expressed within and secreted from the endocrine pancreas. Research surrounding PANDER has revealed a large role for the protein in maintaining glucose homeostasis, as evidenced by several Ad-PANDER overexpressing murine models, our lab's pancreas-specific PANDER transgenic overexpressor, and most recently our mixed genetic C57/129J PANDER knockout (PANKO) mouse. However, PANDER's overall role in glycemic regulation and glucose homeostasis has yet to be studied in a purebred C57BL/6J PANDER knockout model. January of 2010 brought record-breaking cold temperatures to Florida. Such freeze events can upset vulnerable populations of marine life and other species that rely on stable water temperatures. Sea turtles are one group of species that are particularly susceptible to abrupt drops in water temperature. When water temperatures drop below 10°C, a mass hypothermic stunning, or cold-stunning, event for sea turtles can be expected, with many debilitated turtles washing onshore with a very limited time window to be rehabilitated (Foley et al. 2007).
