Palmitoylation refers to the covalent attachment of fatty acids, such as palmitate, onto the cysteine residues of proteins. This process may subsequently alter their localization and function. Nearly all of the enzymes that catalyze palmitoylation, zDHHC protein acyl transferases (PATs), are implicated in neurological disorders, infectious diseases, and cancer in humans. Of particular interest to those who study palmitoylation are Ras family GTPas and zDHHC9-GCP16, the zDHHC PAT that palmitoylates Ras proteins. Erf2-Erf4 is the zDHHC PAT that palmitoylates Ras proteins in Saccharomyces cerevisiae. Currently, there are no methods to therapeutically target palmitoylation for the treatment of disease. One …

Medical certification of workers for respirator use is an important activity of occupational medicine health professionals. Spirometry is a diagnostic tool to evaluate respiratory distress/insufficiency that may affect respirator use. In this study, we analyzed the pulmonary function data of 337 workers from different occupations which required medical evaluation to wear a respirator. The American Thoracic Society and National Fire Protection Association criteria were used to evaluate employees. Of 337 workers who were cleared for respiratory use on the basis of medical questionnaires for respirator compliance, 14 (4.15%) failed to pass respirator compliance on the basis of NFPA criteria and …

Filariasis and onchocerciasis are debilitating diseases affecting 120 million people globally. The massive socio-economic impact of these diseases energized the international community to declare a goal of eliminating filariasis 2020. This resulted in a dramatic increase in the efforts to eliminate filariasis and onchocerciasis, employing a strategy of mass drug administration (MDA). However, these programs rely upon the small arsenal of drugs. This leaves these programs vulnerable to failure in the face of developing resistance and local intolerance to the current drug regimens. This dissertation presents an investigation on the capabilities of Near-Field Microwave Microscopy (NFMM) for the characterization of surface and subsurface materials. Subsurface characterization refers to the detection, differentiation and imaging of dielectric, and metallic features that are coated with an insulating layer. The design, simulation and modeling, and testing of a dielectric resonator (DR)-based NFMM and a coaxial transmission line resonator-based NFMM are discussed in detail in this work. Additionally, materials differentiation and imaging capabilities of each microscope are examined using several bulk samples, liquids, GaAs MMIC circuits, and gold/glass testing patterns.

PANcreatic-DERived factor (PANDER, FAM3B) has been shown to regulate glycemic levels via interactions with both pancreatic islets and the liver. Although PANDER is predominantly expressed from the endocrine pancreas, recent work has provided sufficient evidence that the liver may also be an additional tissue source of PANDER production. At physiological levels, PANDER is capable of disrupting insulin signaling and promoting increased hepatic glucose production. As shown in some animal models, strong expression of PANDER, induced by viral delivery within the liver, induces hepatic steatosis. However, no studies to date have explicitly characterized the transcriptional regulation of PANDER from the liver. …

This dissertation explores the intersection of new media and narrative, as it is presented through a cinematic aesthetic. The narrative language of film is analyzed through the theoretical framework of Bakhtin’s concepts of Heteroglossia, Chronotope, and Dialogism. Bakhtin’s ideas of classifying language act as strong tools for demonstrating how cinematic narrative can inform and alter the perception of its spectators. Lev Manovich’s principles of New Media, specifically Variability, Modularity, and Automation are also utilized to demonstrate how cinema is a constantly evolving paradigm.

This dissertation is dedicated to the research and investigation of novel enzymes and the methods used to study them, with physiological roles ranging from isoprenoid biosynthesis to neurotransmitter production. Using a combination of bioinformatics, recombinant cloning, enzymology, and proteomics, we have contributed to the understanding and exploration of several human illnesses, including malaria, cancer, and endocrine dysfunction.

Phenotypic plasticity is the ability of a given genotype to exhibit different phenotypes in response to environmental variables, which can impact population level processes. Plasticity of ecologically-relevant traits is important to an organism’s environmental response; however, the underlying mechanisms of plasticity are largely unknown. Ecological epigenetics may offer mechanisms (e.g. DNA methylation) underlying phenotypic plasticity. Epigenetics can be defined as the underlying molecular mechanisms that allow one genotype to exhibit different phenotypes. For this dissertation I studied phosphorus (P) sorption dynamics in the shallow groundwater of the southern Everglades. In particular, I examined how the ambient water type governs soluble reactive P (SRP) availability through adsorption/desorption reactions with the aquifer matrix. Chapter 2 investigated how P sorption dynamics of the mangrove root zone sediment are affected by high bicarbonate brackish groundwater compared to both fresh groundwater and saltwater. The results from chapter 2 show that the sediment exhibited exceptionally low sorption efficiency in the high bicarbonate brackish water, which would allow ambient water SRP concentration to be maintained at a higher level. …

This study investigates how heterosexual young people understand and manage risks related to meeting sexual partners online in the United States. The purpose of this study is to help inform the development of culturally-appropriate sexual risk communication and health promotion messages for young people by linking public health knowledge of adolescent sexual health and eHealth with anthropological theories of risk. With qualitative data from two rounds of semi-structured interviews and two group interviews with university students in central Florida, this study shows how young people experience and prioritize more social-emotional risks in meeting online-to-offline sexual partners compared to physical risks. …

North American river otters are considered common in the state of Florida, but their populations have not been studied since the 1980’s. Since that time, Florida’s human population has more than doubled, and many natural areas of Florida have been developed. The aim of this study was to determine the presence or absence of river otters at locations in Florida which they historically inhabited. Diel vertical migration (DVM) can offer motile phytoplankton a competitive advantage over other phytoplankton species. DVM has been well documented in the harmful dinoflagellate, Karenia brevis; however, the extent to which changes in nutrient regimes and density stratification modify migration patterns has not been thoroughly investigated. A 2 m experimental column was constructed with sensors and sampling ports at four depths to obtain fluorescence and temperature measurements and discrete water samples. Concussion is an injury that disproportionately affects children and adolescents and has the potential to negatively impact school performance. Currently, a significant proportion of youth with concussion go unreported due to a lack of effective concussion surveillance procedures. The objective of this study is to investigate land-use changes and water quality trends within the headwaters of the Alafia River watershed. Water quality data were obtained from the Environmental Protection Commission of Hillsborough County (EPCHC). Eleven water quality parameters selected for analysis included: temperature (˚C), dissolved oxygen (DO), percent saturation of DO, conductivity, pH, total phosphorous (TP), total nitrogen (TN), ammonium, chlorophyll-a (uncorrected), fecal coliforms, and enterococci. Limestone dissolution in karst environments is likely due to geochemistry of the water, the actions of microbial communities, and the effect of water flow. We explored the rate of limestone dissolution and will examine here the microbial communities associated with the limestone. A conduit within the brackish cave, Double Keyhole Spring, on the coast of central west Florida was the site of the experiment. PVC pipes (5cm x 16cm) were filled with crushed limestone that was screened to a 1.9cm – 2.54cm size range. This dissertation research explored mechanisms of human milk (HM) expulsion by describing the effects of skin to skin contact (SSC) on maternal basal oxytocin (OT) secretion among 20 premature mothers with hospitalized premature infants. This one-group, repeated measures design consisted of: 1) daily SSC with covariant data via self-report diary and 2) maternal salivary OT with and without SSC at 4 time points were collected over a 7 day time frame. Results indicate that mean levels of basal OT increase over time (M 234 pg/ml, SD 108 pg/ml time point 1; M 257 pg/ml, SD 125 pg/ml time point 3). …

Although the prevalence of Autism Spectrum Disorder (ASD) is currently 1 in 68 (Centers for Disease Control, 2015) and research in this area is growing, high-functioning individuals on the spectrum are often overlooked. This is because of their relatively milder symptoms. The recent collapse of Asperger Syndrome (AS) with autism in the most recent version of the Diagnostic and Statistical Manual (DSM-5; APA, 2013) also has obscured the differences that may exist between those with higher vs. lower levels of functioning. The OGFC mixture design strongly depends on the Optimum Binder Content (OBC) which represents if the mixture has sufficient bonding between the aggregate and asphalt binder. In this research I first examine how Americans’ perceptions of what it takes to get ahead are influenced by their income and then compare those perceptions to measured levels of intergenerational socio-economic mobility. By better understanding these relationships I hope to gain insight into the paths people see to upward mobility, how this varies by income, and to what extent this belief is reflected in past mobility measurements. Additionally, I compare perceptions of what it takes to get ahead with responses regarding attitudes towards public assistance. Anaerobic digestion (AD) is a biotechnology that employs natural microbial metabolism under oxygen-free conditions to stabilize organic waste. AD has been shown to be the most environmentally sustainable technology for treating the organic fraction of municipal solid waste (OFMSW), as it allows for the recovery of energy and nutrients from the waste. AD of OFMSW also saves landfill space and reduces leachate generation and fugitive methane emissions from landfills. Because nitrogen isotopes are fractionated along the soil-plant-insect-bat-guano pathway, it may be possible to reconstruct environmental and climatic changes reflected in the nitrogen isotopic composition of guano. A 1.5-m core of bat guano from Zidită Cave (western Romania) provides a record of climatic and anthropogenic influence on the regional nitrogen cycle and paleoenvironmental controls on nitrogen transforming processes. Increasing and decreasing trends of nitrogen isotopic composition (δ15N values) correspond well with changes in the influence of farming practices, deforestation, and forest expansion. Here we present a novel Smartphone-based colorimeter and demonstrate its application to the measurements of glucose and protein concentrations in biological samples. The key innovation of our approach was to combine powerful image processing encoded into a mobile phone application with a low cost 3D printed sample holder that allowed to control lighting conditions and significantly improved sensitivity. Different solutions with protein and glucose concentrations ranging from 0 to 2000 mg/dL were prepared and analyzed using our system. Dissolved Nutrient Distributions In The Gulf Of Mexico Following The Deepwater Horizon Oil Spill, Ashley Ann Parks

The Deepwater Horizon (DWH) oil spill in the Gulf of Mexico (GoM) in the spring of 2010 introduced 4.4 million of barrels of oil to the ecosystem. Some biodegradation of the oil occurs when microorganisms, particularly bacteria, metabolize the oil as a carbon source. During this process, the microbes also require nutrients for energy. An introduction of oil at this magnitude has the ability to induce large blooms of microbes, which in turn can affect nutrient concentrations. Microbial petroleum degradation decreases nutrient concentrations, whereas the microbial assimilation and decay of organic matter increase nutrient concentrations. This study assessed whether any …

The purpose of this study was to investigate the extent to which the use of social stories that integrate a child’s particular restricted and repetitive behaviors results in differential social outcomes compared to the use of social stories that do not integrate restricted and repetitive behaviors. A non-concurrent multiple baseline experimental design across participants was used to examine the effects of two Social Story interventions on the frequency of appropriate social behaviors made by participants in a school setting.
