Staphylococcus aureus is a human pathogen of serious concern because of its ability to cause serious and wide-ranging disease. Of particular concern is the ability of S. aureus to cause bacteremia (SAB), allowing the bacteria to disseminate and spread to various organs. This study addressed this issue through the phenotypic characterization of two methicillin-resistant S. aureus (MRSA) clinical isolates taken from patients with sustained complicated bacteremia at Tampa General Hospital, referred to as SA1097 and SA1098. Capillaries, the smallest blood vascular networks, are formed by narrow and elongated endothelial cell (EC)-lined tubes with associated pericytes anchored to the abluminal tube surface. Our laboratory demonstrated that dynamic interactions between ECs and pericytes during capillary tube network assembly are required for basement membrane matrix deposition and maturation. We have also shown that key EC-derived factors promote pericyte recruitment, proliferation, and pericyte-induced basement membrane formation during capillary assembly. The lymphatic vasculature maintains tissue fluid homeostasis by absorbing interstitial fluid to form lymph and transporting it back to the bloodstream. Collecting lymphatic vessels have lymphatic valves (LVs) that prevent the retrograde flow of lymph generated by hydrostatic pressure from the blood vasculature. Backflow and accumulation of lymph leads to a lymphatic pathology called lymphedema. This implicates the involvement of defective LVs in the pathogenesis of lymphedema and warrants the study of lymphatic valve development and maintenance. Incels (short for “involuntary celibates) are an online community of men who blame women for their reputed sexual and romantic failures. The misogyny of their worldview, called the blackpill, has inspired multiple instances of real-world violence against women. Alarmingly, we know little about the temporal processes whereby an incel goes from joining an online community to becoming deeply aligned (i.e., “fused”) with the group. This project sought to address this by scraping incels’ most popular web forum and using text analysis to identify psychological mechanisms that could be responsible for incels’ fusion. The study investigates substance use among Transitional Aged Youth (TAY), particularly focusing on Electronic Dance Music (EDM) events in the Tampa Bay area. TAY, defined as individuals aged 18-29, experience significant psychological, biological, and developmental changes, which can lead to increased vulnerability to substance use. The study examines the prevalence and patterns of substance use, specifically ketamine and cocaine, among TAY at EDM events. Exploratory qualitative data collection method was utilized for the study. Periodontal disease remains a global public health concern. Despite the availability of preventive and therapeutic strategies, the multifactorial nature of periodontitis complicates its understanding and management. Periodontal disease is associated with an increased risk of chronic conditions, including ischemic heart disease (IHD), gestational hypertension, respiratory diseases such as COPD and bronchitis, and various cancers, including kidney and pancreatic cancers. Moreover, individuals with diabetes, autoimmune diseases like lupus and Crohn’s disease, and osteoporosis are more likely to develop periodontitis. Quantifying and understanding trends in freshwater biodiversity has become more urgent as rapid biodiversity loss continues. TET2 is a dioxygenase that regulates gene expression and hematopoiesis by oxidizing 5-methylcytosine residues on DNA to 5-hydroxymethylcytosine. TET2 mutations are common genetic events in hematological disorders. For example, TET2 is the most frequently mutated gene in Chronic Myelomonocytic Leukemia and occurs in about 33% of other myeloid malignancies. It is also the second commonest mutation in clonal hematopoiesis of indeterminate potential (CHIP), a common premalignant condition in the elderly. TET2 mutations arise in founding clones and are implicated in many non-hematological sequelae seen in CHIP like atherosclerotic disease, chronic liver disease, and severe microbial infections.

Many proteins contain fragments that become structured upon binding to specific targets or through self-association. Notable examples of such disorder-to-order transitions include amyloidogenic proteins like amyloid-β peptide (Aβ) and tau peptide in Alzheimer's disease, α-Synuclein in Parkinson's disease, and hIAPP in type 2 diabetes, and so on. These regions can aggregate into amyloid fibrils through β -sheet stacking, contributing to amyloidosis and related diseases. Non-sewered sanitation systems (NSSS) offer innovative solutions to sanitation challenges in areas lacking basic facilities, utilizing advanced treatment technologies for on-site water reuse. The NEWgenerator (NG), an advanced NSSS developed at the University of South Florida under the Bill & Melinda Gates Foundation’s Reinventing the Toilet Challenge, integrates an anaerobic membrane bioreactor (AnMBR), an ion exchange-based nutrient capture system (NCS), and an electrochlorinator for final disinfection. As an atom-economic way of carbon-nitrogen bond formation, hydroaminations are great tool for make amines, hydrazines, and other nitrogen-containing heterocycles. Specifically, radical hydroamination is a useful approach for functionalization of unactivated olefins, especially in sterically hindered environments. Diversification remains a challenge for radical hydroaminated products, since the nitrogen sources are carefully designed. Chapter 1 provides an introduction to protein intrinsic disorder, outlining the historical context and fundamental concepts that highlight the importance of intrinsically disordered proteins (IDPs) and IDPRs in various biological processes. Chapter 2 focuses on the rapid prediction and analysis of protein intrinsic disorder. We introduce RIDAO (Rapid Intrinsic Disorder Analysis Online), a high-efficiency web-based tool that integrates multiple disorder predictors. The biochemistry of human tissues can provide information for geoprofiling (geographic birthplace or migration patterns) of an unidentified decedent. Utilizing isotope analyses (both stable and radiogenic) from teeth, bone, nails, and/or hair reveals specific isotopic information that correlates to diverse geographic regions reflecting an individual’s birthplace and migration patterns. Isotope analysis of enamel and bone has been employed in bioarchaeology for decades to reconstruct the diet, health, and migration patterns of past populations. Circadian rhythms are crucial in regulating various physiological processes, including mood regulation, and disruptions in these rhythms have been linked to several neuropsychiatric disorders, such as depression, bipolar disorder, and schizophrenia. FKBP51, a protein involved in stress responses and glucocorticoid receptor function has emerged as a potential regulator of the circadian clock. FKBP51’s interaction with glucocorticoid receptors impacts the transcriptional regulation of circadian-related genes. This report mainly discusses the development of RET Proteolysis targeting chimera PROTAC molecules based on Selpercatinib (LOXO-292) and Sulfono-γ-AApeptide PROTAC. A PROTAC molecule is a bifunctional molecule composed of two active domains and a linker. It works by inducing selective protein intracellular degradation. This technology was first discussed by Kathleen Sakamoto, Ray Deshaies and Craig Crews in 2001. To understand the dynamics of cancer, mathematical oncologists have developed models of tumor growth and treatment response. Some models are mechanistic and approach tumor growth at the cell-scale, focusing on the evolution of cancerous cells within the ecology of normal tissue, and are often simulated with agent-based modeling. Other models are more clinically motivated and model tumor growth operating at the organ-scale, using patient data to predict treatment response, and are often simulated with partial differential equations. Recent decades have seen increased rates of ice mass loss from Antarctic ice sheets, through thinning of many ice shelves around the periphery of the continent. The most vulnerable are the glacial systems that are marine-terminating and grounded below sea level, a characteristic that exposes these systems to sea level rise and warming oceans. Enhanced basal melting under the influence of warm ocean water masses can lead to thinning and retreat of glaciers, and ultimately to the acceleration of grounded ice loss. Coiled-coil-helix-coiled-coil-helix domain containing 2 (CHCHD2) is a mitochondrial protein that plays important roles in cristae structure, oxidative phosphorylation, and apoptosis. Multiple mutations in CHCHD2 have been associated with Lewy body disorders (LBDs), such as Parkinson’s disease (PD) and dementia with Lewy bodies (DLB), with the CHCHD2T61I mutation being the most widely studied. We generated the first transgenic mouse models expressing human CHCHD2WT and the PD-linked CHCHD2T61I mutation driven by the mPrP promoter. CHCHD2T61I Tg, but not CHCHD2WT Tg mice exhibit perinuclear mitochondrial aggregates, neuroinflammation, and have impaired long-term synaptic plasticity associated with synaptic dysfunction. Since the FDA approval of IL-2 in cancer treatment in the 1980s, immunotherapy has emerged as the standard of care for several tumor types (e.g., melanoma, lung, bladder, colorectal). However, approximately 70 percent of cancer patients, independent of prior treatment or staging, administered immunotherapy will not achieve complete responses and will exhibit disease progression. Several factors have been identified to contribute to treatment failure, including the accumulation of intratumoral immunosuppressive cell populations, insufficient tumor-infiltrating anti-tumor immune populations, or the occurrence of immunoediting on tumor cells. The response to replication stress is critical for ensuring the stability of replication forks and preserving genome integrity. To aid this response, several factors are recruited to stalled replication forks to prevent irreversible collapse of the replication machinery. Replication fork proteomic analyses of stalled forks reveal enrichment of DCAF14, a component of the Cullin4-RING (CRL4) E3 ubiquitin ligase complex, that mediates replication fork protection by blocking nuclease-mediated digestion of nascent DNA. Group B Streptococcus (Streptococcus agalactiae; GBS) is a leading cause of neonatal sepsis worldwide. Intrapartum antibiotics in GBS-colonized mothers significantly reduced the incidence of early-onset sepsis occurring in the first week of life; however, there are currently no prophylactic interventions for late-onset sepsis, occurring within the first 3 months of life. GBS is a pathobiont that primarily colonizes the gastrointestinal tract and can cross the intestinal barrier via both the paracellular and transcellular routes, causing invasive disease.
