Group B Streptococcus (Streptococcus agalactiae; GBS) is a leading cause of neonatal sepsis worldwide. Intrapartum antibiotics in GBS-colonized mothers significantly reduced the incidence of early-onset sepsis occurring in the first week of life; however, there are currently no prophylactic interventions for late-onset sepsis, occurring within the first 3 months of life. GBS is a pathobiont that primarily colonizes the gastrointestinal tract and can cross the intestinal barrier via both the paracellular and transcellular routes, causing invasive disease. Neonatal susceptibility to GBS intestinal translocation is mediated by inadequate epithelial barrier function and microbiome immaturity; however, the mechanisms by which GBS exploits …

This dissertation examines Indigenous women’s access to and beliefs regarding gender-based violence (GBV) services and the Guatemalan legal system. It illuminates the legal, structural, and institutional challenges and power dynamics associated with seeking support and justice for women in Guatemala, as well as why and how women decide to seek formal assistance. This research also investigates changes in attitudes and beliefs regarding women’s rights, gender roles, and formal and informal forms of support for GBV survivors. Chimeric antigen receptor (CAR) T-cell therapy has revolutionized the treatment of patients with relapsed/ refractory large B cell lymphoma (LBCL). Despite its promising efficacy, a subset of patients experiences primary resistance or relapse. In Chapter 2, we explore the tumor microenvironment (TME)-induced resistance mechanisms in patients with LBCL. Analysis of the intratumoral immune infiltrates in pre-infusion tumor biopsies revealed that the levels of immunoregulatory macrophages were elevated in patients who responded poorly to axicabtagene ciloleucel (axi-cel). Using preclinical mouse B cell tumor models, we found that the CAR T-cell-produced interferon-gamma (IFN-γ) enhanced the expression of inducible nitric oxide synthase (iNOS, …

My dissertation is titled "The Determinants of Women's and Children's Health in Bangladesh." The first chapter studies the impact of a Bangladeshi schooling program on child immunization rates. The program provided a stipend and other incentives to attend secondary school, significantly increasing rural girls' education. I hypothesize that this would lead to higher immunization rates in the next generation since better-educated mothers have better knowledge of health practices. The study uses a regression model ("difference-in-differences"), which is based on differential exposure to the 1994 Female Secondary School Stipend Program (FSSSP) by birth cohort and rural residence. Virologic suppression is an essential part to a successful regimen for Persons with HIV (PWH) and the 95-95-95 initiative. Yet, PWH experiencing episodes of low-level viremia (LLV) may be more prone to experience virological failure (VF). Emergency responders face a significant risk of exposure to blood-borne pathogens (BBPs), such as HIV and hepatitis, due to the nature of their work environment. This study investigates this issue utilizing a dual approach of data analysis and online surveys to assess exposure rates, risk perceptions, factors that contribute to risk and prevention practices. In addressing issues ranging from personal protective equipment (PPE) usage to levels of concern surrounding blood and bodily fluid contact, the findings underscore the need for tailored safety measures, comprehensive education, and improved prevention strategies.

Urbanization poses significant challenges to wildlife, including amphibians. The present study uses a multi-parameter approach that incorporates veterinary techniques to investigate the current health status of the adult southern leopard frogs (Lithobates sphenocephalus) in Tampa Bay, Florida, amidst urban stressors including nutrient loading, fragmented habitats, and light and noise pollution. Novel values for fluctuating asymmetry (e.g., variation between left and right bilateral traits; FA), body condition (CF), stress (e.g., neutrophil:lymphocyte (N:L) ratio), and inflammation (e.g., neutrophil morphologic abnormalities, parasite load) are reported. While infections can often be asymptomatic in immunocompetent individuals, they can pose significant risks to those who are immunocompromised, as well as pregnant women. The COVID-19 pandemic has brought a wealth of new information regarding the virus's origin and treatment. Regrettably, the vast amount of online information has resulted in the proliferation of misinformation, as the World Health Organization reported. To investigate whether journalists may have contributed to this problem, a qualitative analysis was conducted on 15 news articles from top-tier media outlets. Using NVivo software, the articles were analyzed to identify linguistic words and visuals that fell into coding categories based on Behrendt's (2008) contextual analysis for evaluating text. This dissertation demonstrates that a critical, clinically applied anthropology is possible by testing a hypothesis that a syndemic intervention, and use of a structural vulnerability assessment tool, can achieve improved healthcare outcomes. A critical, clinically applied anthropology integrates social scientists into healthcare delivery, alongside biomedical providers, through the co-creation of new diagnostic and patient care pathways that utilize anthropological methods (ethnographically informed care, syndemics, thematic/mixed methods data analysis) and advance anthropological theory (biomedicine and culture, structural violence, structural competency/vulnerability, ontology, assemblage theory, and entanglements) to decrease healthcare inequities.

Lung cancer is the first cause of cancer-related deaths in both men and women with an overall five-year survival rate of 28%. Although immune checkpoint blockers (ICBs) are currently FDA-approved for the treatment of non-small cell lung cancer (NSCLC), only 17-20% of patients achieve durable responses by the induction of immunologic memory. The lack of response in most patients can be attributed to the tumor-intrinsic or tumor-extrinsic immune resistance mechanisms. In the United States, it remains the third leading cause of death in 2022. Malaria, caused by the apicomplexan Plasmodium spp. , is a major public health issue that impacts over one-third of the world’s population ,with Plasmodium vivax accounting for over 130 million clinical cases annually. The circumsporozoite protein (CSP) is the most abundant molecule on the surface of Plasmodium sporozoites and is considered a leading pre-erythrocytic stage vaccine candidate. CSP is essential for sporozoite maturation, migration, and invasion. Anti-CSP antibodies interrupt sporozoite migration and infection of hepatocytes thus reducing liver-stage burden. RTS,S/AS01, vaccine targeting malaria caused by Plasmodium falciparum, used in moderate to high malaria transmission, provides hope for pre-erythrocytic (PE) subunit malaria vaccine development. Despite this vaccine advance, efforts to develop an efficacious vaccine against the neglected parasite Plasmodium vivax have lagged. In addition, there exists a gap in our understanding of PE antigens that can induce broadly-neutralizing antibodies. In most practical applications, the nonlinear dynamic constraint $f$ is unknown analytically. The problem of approximating $f$ directly from data measurements generated by the system is a main goal of this manuscript. Transmission of the deadly malaria parasite Plasmodium falciparum from human to mosquito is achieved by specialized intraerythrocytic sexual forms called gametocytes. Circulating in the vertebrate host, relatively few intraerythrocytic gametocytes are picked up during a bloodmeal to continue sexual development in the mosquito vector. Human-to-vector transmission thus represents an infection bottleneck in the parasite's life cycle for therapeutic interventions to prevent malaria. Though the crucial regulatory mechanisms leading to gametocyte commitment have recently come to light, networks of genes that control sexual development remain to be elucidated. Malaria is a parasitic illness caused by the Plasmodium spp., with an estimated 247 million malaria cases reported in 2021 across 84 endemic countries. Treatment of the deadliest malaria parasite, P. falciparum, relies on artemisinin combination therapies (ACTs). However, while ACTs have greatly decreased malaria mortality, this progress is threatened due to artemisinin (ART) partial resistance and ACT failure in Southeast Asia and Sub-Saharan Africa. SNPs to the Kelch13 (K13) gene are a well-characterized marker for ART resistance. This study demonstrates for the first time that HDAC8 function is critical for MCL survival, and abrogating its activity in human primary NK cells does not interfere with NK IgG antibody directed ADCC therapies ex vivo. Human NK cells, isolated from healthy donors, are highly resistant to HDAC8 inhibitor treatment with PCI-34051. Even at the highest concentration, 20uM, no toxicity was observed. Conversely, MCL cell lines representative of the aggressive MCL subtype, classical MCL, were especially sensitive to PCI-34051, an HDAC8 selective inhibitor, treatment.
