Localization Of Five Target Proteins In Tachyzoites Of Toxoplasma Gondii, Abigail M. Kaiser

Five target genes were selected in Toxoplasma gondii tachyzoites for localization studies. These five genes, detected through proteomics studies, included TgME49_227450, TgME49_223080, TgME49_262390, TgME49_230940, and TgME49_269620. Localization of these five target proteins is a first step to confirm their interaction with TgCrk2 and understand their function and role in TgCrk2 regulation of the tachyzoite cell cycle. Gene models for the targets were analyzed using ToxoDB and ApE analysis tools. Endogenous tagging constructs were created for each target. Transgenic parasites were created. Background: Pre-exposure prophylaxis (PrEP) is a daily pill that can reduce a person’s chances of acquiring HIV. HIV testing and counseling is a critical point during which non-clinical staff could intervene, discuss and/or refer clients for PrEP. However, not all HIV testing/counseling staff take part in PrEP implementation in the same way. This study investigated: 1) the underlying PrEP implementation subgroups of staff who perform HIV testing and 2) PrEP implementation as a function of key constructs from the Consolidated Framework for Implementation Research (CFIR).

Chronic lymphocytic leukemia (CLL) is the most common leukemia in developed countries. It is characterized by the accumulation of CD5+ B lymphocytes in the peripheral blood, bone marrow, and lymphoid tissues of affected patients. Patients experience lymphadenopathy, fatigue, and are severely immunocompromised. Management of disease and symptoms is possible with chemo-immunotherapy for higher risk patients; however, CLL remains incurable unless bone marrow transplant is performed. Identification of novel targets and optimization of current therapeutics is therefore still necessary. Human Papillomavirus (HPV) is the most common sexually transmitted infection (STI) in the US. Infection with low-risk HPV (i.e., 6 and 11) can cause genital warts, and persistent infection with high-risk HPV types (i.e., HPV 16 and 18) can progress to cancer. Currently, there is an HPV vaccine that is recommended for boys and girls, aged 11 to 12. Healthy People 2020 established a national objective of 80% completion of HPV vaccination among children aged 13 to 15 years old. The failure of lung cancer treatments has been attributed partly to the development of drug resistance, however the underlying cellular and molecular mechanisms are poorly understood. Sampling of airborne dust in industry is influenced by the potential health effects associated with human exposure. Health effects depend on mass concentration and particle size which influences the site of pulmonary deposition. Occupational diseases tend to be associated with dust deposition in specific regions of the respiratory tract. The ACGIH size selective TLVs are expressed in three forms: Inhalable Particulate Matter, Thoracic Particulate Matter, and Respirable Particulate Matter. Americans of all ages experience overweight and obesity; however, increasing rates occur during young adulthood. Climatic variations, together with large-scale environmental forces and human development affect the quality of coastal recreational waters, creating potential risks to human health. These environmental forces, including increased temperature and precipitation, often promote specific vector-borne diseases in the Caribbean and Gulf of Mexico. Human activities affect water quality through discharges from urban areas, including nutrient and other pollutants derived from wastewater systems. Resilience has been measured using qualitative and quantitative metrics in engineering,economics, psychology, business, ecology, among others. This dissertation proposes a resilience metric that explicitly incorporates the intensity of the disruptive event to provide a more accurate estimation of system resilience. A comparative analysis between the proposed metric and average performance resilience metrics for linear and nonlinear loss and recovery functions suggests that the new metric enables a more objective assessment of resilience for disruptions with different intensities. Leishmaniasis is a disease caused by obligate intracellular parasites of the genus Leishmania, including 20 species that are pathogenic to humans. Female sand fly is the known vector that can transmit the disease. Visceral leishmaniasis is the severe form of the disease that affects internal organs and can be fatal with inappropriate diagnosis or treatment. Leishmania donovani is the causative agent of visceral leishmaniasis. Approximately 350 million in 89 countries are at risk of infection. Trauma as an official diagnosis first entered the DSM in 1980 and literary theorists began employing the term to discuss literature not too long after. Since the 1990s, theorists have largely focused on twentieth-century trauma literature with Holocaust and Modernist texts garnering much of the critical interest. Yet, Victorian life was also marked by trauma-causing events. From railway catastrophes, to industrial accidents, to premature deaths, and infectious diseases, Victorians reckoned with wounds to the mind through their lived experience. The malaria pre-erythrocytic stages have been identified as an ideal therapeutic target, but complex in vitro models for Plasmodium vivax and Plasmodium falciparum lack the efficiency needed for rapid screening and evaluation of new vaccines and drugs, especially targeting the P. vivax hypnozoite. To address this challenge, we employed a multi-parameter approach using “omics’” to identify pre-erythrocytic targets and biomarkers, guide phenotypic therapeutic screening, and study parasite functionality with innovative bioassays using highcontent screening. There has been considerable interest in the development of antimicrobial agents which mimic the natural HDPs, and among them biodegradable polymers are newly discovered drug candidates with ease of synthesis and low manufacture cost compared to synthetic host defense peptides. In 2015 Dr. Pratiksha Vaghela started the “Stop Poking Me” campaign which was aimed at curbing the increase in the number of needlesticks at the James A. Haley Veteran Affairs Hospital (JAHVA). The data for needlesticks was collected by the Occupational Medicine Clinic (OMC) between Oct 2013 and Oct 2016. We then obtained the original data from Dr. Vaghela’s project and compared the data to assess whether the new implementations have truly decreased the number of needlesticks.
