PANcreatic DERived factor (PANDER, FAM3B) is a member of a superfamily of FAM3 proteins that are uniquely structured and strongly expressed from the endocrine pancreas and co-secreted with insulin. Unique animal models available to our lab have indicated that PANDER can induce a selective hepatic insulin resistant (SHIR) phenotype whereby insulin signaling is blunted yet lipogenesis is increased. The complexity of the biological networks involved with this process warranted the logical approach of employing quantitative mass spectrometry based proteomic analysis using stable isotope labeling of amino acids in cell culture (SILAC) to identify the global proteome differences between the PANDER …

Angelman syndrome (AS) is a rare genetic and neurological disorder presenting with severe developmental delay, ataxia, epilepsy, and lack of speech. AS is associated with a neuron-specific loss of function of the maternal UBE3A allele, a gene encoding an E3 ubiquitin ligase. Currently, no cure exists for this disorder; however, recent research using an AS mouse model suggests that pharmacological intervention is plausible, and can alleviate some of the detrimental phenotypes reported in AS patients.
Although there is no curative treatment for AS, seizure medication and behavioral therapies are most commonly prescribed in order to minimize symptoms. However, these options …

The advent of powerful wearable devices and smartphones has enabled a new generation of “in-the-wild” user studies, adaptive behavioral intervention strategies, and context measurement. Though numerous proof-of-concept studies continue to push the limitations of what a behavioral scientist can do with these technologies, there remains a major methodological roadblock separating behavioral theory and application. Climate change is expected to impact species by altering infectious disease outcomes, modifying community composition, and causing species to shift their phenology, body sizes and range distributions. The caudal solitary complex (cSC) is a cardiorespiratory integrative center in the dorsal medulla oblongata that plays a vital role in the central CO2-chemoreceptive network. Neurons in this area respond to hypercapnic acidosis (HA) by a depolarization of the membrane potential and increase in firing rate, however a definitive mechanism for this response remains unknown. Likewise, CO2-chemoreceptive neurons in the cSC respond to hyperoxia in a similar fashion, but via a free radical mediated mechanism. The purpose of this research was to evaluate the capability and performance of the University of South Florida’s (USF) Human Exposure Chamber (HEC) using aerosols in the thoracic range. The goals of this research were two-fold: to obtain an average particle size of 10 µm (thoracic-size range) inside the chamber during dust production and to test for evenness of dust concentration within the chamber. The long history of racism has created cultural barriers that prevent some Black Americans from seeking cancer treatment. Fatalism, physician mistrust, low levels of social support and self-efficacy, are the most cited cultural barriers in the literature. Black Americans’ religious beliefs and church involvement have historically helped them in their struggle against racism. A quantitative and a qualitative comparison studies examine the role of Black cancer survivors’ religious beliefs in their fight against cancer. In response to public opposition to the insanity defense, the Guilty But Mentally Ill(GBMI) verdict was enacted with the intention of limiting the number of insanity acquittals and alleviating the public’s concerns. Prior research suggests, however, that many jurors are making verdict decisions with limited knowledge of the dispositional consequences of the GBMI and NGRI verdicts. Further, jurors may erroneously assume that the GBMI verdict is a compromise between a NGRI and guilty verdict, which mitigates punishment. In reality, the dispositional consequences of a GBMI verdict are equivalent to or more restrictive than a guilty verdict. The current study examined …

Animal models of ocular hypertension are important for glaucoma research but come with experimental costs. Available methods of intraocular pressure (IOP) elevation are not always successful, the amplitude and time course of IOP changes are unpredictable and irreversible, and IOP measurement by tonometry is laborious. This dissertation focuses on the development and implementation of two novel systems for monitoring and controlling IOP without these limitations. There are no historic records that describe the events and peoples in the region during establishment of the Spanish missions in the Tallahassee area to the east. Among increased calls for "new narratives" of Haiti, this thesis seeks to honor Haitian traditions of intellectualism and resistance, centering on the life and legacy of martyred professor Janil Lwijis in post-earthquake student social movements. Based on oral histories with student activists at the State University of Haiti (UEH), this work explores student protest in Haiti through the voices, often at odds, of those en lutte; it explores how Janil is invoked and remembered, and argues that oral history can contribute to activist research and pose a challenge to dominant narratives. Most in vitro systems employ the standard cell culture maintenance conditions of 95 % air with 5 % CO2 to balance medium pH, which translates to culture oxygen tensions of approximately 20 % - above the typical ≤ 6 % found in most tissues. The current investigation, therefore, aims to characterize the effect of maintenance and toxicant exposure with a particular focus on the α,β-unsaturated aldehyde, acrolein, in the presence of physiologically relevant oxygen tension using a differentiated H9c2 cardiomyoblast subclone. Individuals with Traumatic Brain Injury (TBI) often need to be taught independent living skills in order to reintegrate into community settings. This study examined the use of video modeling to teach culinary skills to three individuals with TBI. Video modeling is easily accessible, inexpensive, and not reliant on an additional person to directly teach skills. For all three participants, video modeling resulted in increases in cooking skills using a task analysis created for each food item prepared. For one participant, the skills maintained over two weeks and generalized to a novel food. For another participant video modeling was insufficient in …

Disinfection is an essential process in the treatment of municipal wastewater before the treated wastewater can be discharged to the environment. Hillsborough County's Northwest Regional Water Reclamation Facility (NWRWRF) in Tampa, Florida, currently uses ultraviolet (UV) light for disinfection. However, this method has proven expensive to implement and maintain, and may not be effective if the light transmission is poor. For these reasons, Hillsborough County is considering switching from UV light to sodium hypochlorite for disinfection. Between 1988 and 2014, otter trawls, seine nets, and plankton nets were deployed along the salinity gradients of 18 estuaries by the University of South Florida and the Florida Fish and Wildlife Research Institute (FWRI, a research branch of the Florida Fish and Wildlife Conservation Commission). The purpose of these surveys was to document the responses of aquatic estuarine biota to variation in the quantity and quality of freshwater inflows that were being managed by the Southwest Florida Water Management District (SWFWMD).

Science education research continues to demonstrate improved learning with active-learning techniques compared to lectures. However, the question of which active-learning methods are the most effective for learning complex scientific principles in various context still remains. Models are commonly used in activities that allow students to simplify complex systems and understand how components interact. I investigated the outcomes for student learning and engagement of two model-based activities - concept models and game simulations. In southeastern North America, the Woodland period (ca. 1000 B.C. to A.D. 1050) was arguably witness to the first early village societies, and Kolomoki—located in southwestern Georgia—is among the largest villages during this interval. Though archaeologists recognize these communities as seminal developments in the course of human history, little attention has been paid to how they develop and vary internally. This thesis seeks to address these issues by focusing on the development and social construction of the early village community at Kolomoki. Invasive species can severely disrupt biological communities through their interactions with native organisms, yet little is known about the response of marine predators to the establishment of a competitive invasive fish. Fish can have complex life histories and use multiple habitats and resources throughout their life span. Consequently, their life histories are often poorly understood. The Atlantic Tarpon, Megalops atlanticus, is a large, typically migratory, elopomorph fish that is both ecologically and economically important. Eutrophication is defined as the ‘over enrichment’ of a water body from nutrients, resulting in uncontrolled growth of primary producers, leading to periods of oxygen depletion from decomposition of the algal organic matter. According to the 2010 Water Infrastructure Needs and Investment (a U.S. Congressional Report), 40% of U.S. water bodies are contaminated with pollutants, including nutrients. Non-point sources of nutrient pollution are a major cause of this reduction in water quality. One way to decrease eutrophication is to manage nutrients found in stormwater runoff, before they reach a receiving water body.

Idiopathic Pulmonary Fibrosis (IPF) is a deadly disease of unknown origin, which causes 80,000 deaths every year in the US and Europe combined. Unknown etiology and late diagnosis, combined with limited treatment options, contribute to a dismal survival rate of 3-5 years post diagnosis. Although molecular mechanisms underlying IPF pathogenesis and progression have been studied for over two decades, lack of in vivo models that recapitulate chronic, progressive, and irreversible nature of IPF have contributed to limited therapeutic success in clinical trials. Heterologous expression systems such as COS-7 cells have demonstrated the profound effects of KCNAB1-3 or Kvβ1-3 proteins on voltage gated potassium channels (Kv) channels. Indeed, in the presence of these β-subunits transiently expressed Kv channels are often modulated in multiple ways. Kv channel membrane expression is often increased in the presence of β-subunits. In addition, non-inactivating Kv currents suddenly become fast-inactivating and fast-inactivating channels become even faster. While much research has demonstrated the profound effects the β-subunits in particular the Kvβ1 subunit have on transiently expressed Kv currents little to date is known of the physiological role it may play. …
