Contributions Of Appetitive And Aversive Motivational Systems To Decision-Making, Heather E. Soder Nov 2017

Contributions Of Appetitive And Aversive Motivational Systems To Decision-Making, Heather E. Soder

USF Tampa Graduate Theses and Dissertations

Optimal decision-making entails outcome evaluation, comparing received costs and benefits with predicted costs and benefits. The anterior cingulate cortex (ACC), a brain area with major connections to the appetitive and aversive motivation systems, may provide the neural substrate of this evaluation process. One way to measure the relative contribution of these systems on decision-making is to measure individual differences in risk-taking behaviors. For individuals who make risky choices, this evaluation step may be biased: some show a preference for immediate, short-term rewards (increased appetitive system), while devaluing the long-term consequences of their choices (decreased aversive system). However, most studies supporting …

The Suffering South: 1878 Yellow Fever Narratives And Post-Reconstruction Southern Identity, Jessica Wells Nov 2017

The Suffering South: 1878 Yellow Fever Narratives And Post-Reconstruction Southern Identity, Jessica Wells

USF Tampa Graduate Theses and Dissertations

The Suffering South offers a cultural history of a yellow fever epidemic that swept through the Mississippi Valley in 1878. It argues that the yellow fever narratives created during this epidemic constituted a discursive attempt by Southerners to renegotiate Southern identity and social hierarchy following the Civil War and Reconstruction. White Southerners, in particular, used the epidemic as an occasion to foster a return to a more traditional foundation of white supremacy and patriarchy as the basis for Southern identity and belonging. The narratives written by these Southerners, in which they described their experiences with yellow fever and the effects …

Antibiotic resistant bacteria and antibiotic resistance genes are released into aquatic ecosystems through hospital waste, residential sewer lines and animal agricultural waste streams. Animal agriculture accounts for approximately 70% of antibiotic use in the United States (3). In agricultural ecosystems, …

Evaluating Small-Scale Simulation For Training Firearm Safety Skills, Trevor Maxfield Nov 2017

Evaluating Small-Scale Simulation For Training Firearm Safety Skills, Trevor Maxfield

USF Tampa Graduate Theses and Dissertations

There is limited research using small-scale simulation in applied behavior analysis. We used small-scale simulation to train firearm safety skills to 3 to 5-years-old children and assessed whether the skills generalized to the natural environment through in situ assessment. Three participants completed the training and all participants learned the safety skills from simulation training. Two of the participants acquired the safety skills after the first simulation training and the third participant required one booster training before demonstrating the safety skills in the natural environment.

How Did I Get Here? Testing The Translation Of The Morris Water Maze And The Influence Of Hemoglobin A1c On Spatial Navigation Performance, Colleen Pappas Nov 2017

How Did I Get Here? Testing The Translation Of The Morris Water Maze And The Influence Of Hemoglobin A1c On Spatial Navigation Performance, Colleen Pappas

USF Tampa Graduate Theses and Dissertations

Changes in cognitive status occur with aging and significant attention has been placed on developing interventions to possibly delay cognitive decline and identifying risk factors that exacerbate cognitive deficits. One issue that arises when studying interventions is that they do not always effectively translate from animal models to human subjects. When testing potentially modifiable risk factors related to cognitive impairment, more sensitive metrics could help in identifying targets for intervention at earlier time-points. Therefore, the aims of the current dissertation were twofold. The first study examined the ability to translate between species using a common behavioral paradigm, the Morris water …

Bioleaching Potential Of Filamentous Fungi To Mobilize Lithium And Cobalt From Spent Rechargeable Li-Ion Batteries, Aldo Lobos Nov 2017

Bioleaching Potential Of Filamentous Fungi To Mobilize Lithium And Cobalt From Spent Rechargeable Li-Ion Batteries, Aldo Lobos

USF Tampa Graduate Theses and Dissertations

Demand for lithium (Li) and cobalt (Co) is on the rise, due in part to their increased use in rechargeable Li-ion batteries (RLIB). Current recycling processes that utilize chemical leaching efficiently recover in Li and Co from the cathode material in spent batteries; however, these processes are costly and emit hazardous waste into the environment. Therefore, a more sustainable process for recycling Li and Co is needed, and bioleaching may provide a solution. Fungal bioleaching has been shown in previous studies to effectively mobilize metals (Pb, Al, Mn, Cu, and Zn) from mine tailings, electronic scrap, and spent batteries with …

Predicting Hand Surface Area From A Two-Dimensional Hand Tracing, Myles O'Mara Nov 2017

Predicting Hand Surface Area From A Two-Dimensional Hand Tracing, Myles O'Mara

USF Tampa Graduate Theses and Dissertations

Recent occupational health studies have focused on dermal exposure at the hands, but have been unable to accurately express dose without knowing the HSA. There is no standard method to calculate HSA, though some researchers have derived HSA formulas based on dimensions from a Taiwanese population. This research paper describes a shortcut method to estimate the hand surface area (HSA) of a human hand from a two-dimensional hand tracing, and repeated a Taiwanese HSA study in order to explore the viability of its HSA formula in an American university population. A sample of nine adult men and nine adult women, …

Thermodynamic And Kinetic Aspects Of Hen Egg White Lysozyme Amyloid Assembly, Tatiana Miti Nov 2017

Thermodynamic And Kinetic Aspects Of Hen Egg White Lysozyme Amyloid Assembly, Tatiana Miti

USF Tampa Graduate Theses and Dissertations

Deposition of protein fibers with a characteristic cross-β sheet structure is the molecular marker associated with human disorders ranging from Alzheimer's disease to type II diabetes and spongiform encephalopathy. Given the large number of non-disease related proteins and peptides that have been shown to form amyloid fibrils in vitro, it has been suggested that amyloid fibril formation represents a generic protein phase transition. In the last two decades it has become clear that the same protein/peptide can assemble into distinct morphologically and structurally amyloid aggregates depending on the solution conditions. Moreover, recent studies have shown that the early stage, oligomeric …

Re-Placing The Plantation Landscape At Yulee’S Margarita Plantation, Katherine M. Padula Oct 2017

Re-Placing The Plantation Landscape At Yulee’S Margarita Plantation, Katherine M. Padula

USF Tampa Graduate Theses and Dissertations

U.S. Senator David Levy Yulee’s Margarita sugar plantation flourished from 1851 to 1864 in Homosassa, Citrus County, Florida. The plantation was abandoned in 1864 and memory of its precise location slowly faded, as the physical evidence of its existence deteriorated. Today, the only plantation structure known to be still standing is the sugar mill, preserved as part of the Yulee Sugar Mill Ruins Historic State Park (CI124B). The remainder of the plantation, including its boundaries, remains unknown. Perhaps at least partly owing to this absence, the mill’s interpretive signage provides an unfortunate univocal historical interpretation of the site and lacking …

Identifying New Treatment Options And Risk Factors For Type 2 Diabetes: The Potential Role Of Thymoquinone And Persistent Organic Pollutants, Shpetim Karandrea

USF Tampa Graduate Theses and Dissertations

Type 2 Diabetes Mellitus (T2DM) is a metabolic disorder characterized by chronic hyperglycemia, which develops as a consequence of peripheral insulin resistance and defective insulin secretion from pancreatic β-cells. A high calorie diet coupled with physical inactivity are known risk factors for the development of T2DM; however, these alone fail to account for the rapid rise of the disease. Recent attention has turned to the role of environmental pollutants in the development of metabolic diseases. PBDEs (polybrominated diphenyl ethers) are environmental pollutants that have been linked to the development of type 2 diabetes (T2D), however, the precise mechanisms are not …

Stable Isotopes In The Eye Lenses Of Doryteuthis Plei: Exploring Natal Origins And Migratory Patterns In The Eastern Gulf Of Mexico, Brenna A. Meath

USF Tampa Graduate Theses and Dissertations

Stable isotope analysis is an emerging tool to examine trophic pathways and migratory patterns of marine organisms. Squid are widely distributed in coastal and deep water regions of the Gulf of Mexico. Stable isotope ratios of carbon and nitrogen found within cephalopod tissues can provide information on both trophic level and habitat of their food sources. More recently, ontogenetic changes in stable isotope ratios within squid eye lenses have been documented. Concentric layers of crystallin proteins are added to the lens as the squid ages; the center of the lens contains the oldest layer and the youngest layers are on …

Validation Of Chimeric Viruses In Plaque Reduction Neutralization Test In Arboviral Disease Diagnostics, Jasmine Boykin Oct 2017

Validation Of Chimeric Viruses In Plaque Reduction Neutralization Test In Arboviral Disease Diagnostics, Jasmine Boykin

USF Tampa Graduate Theses and Dissertations

The plaque reduction neutralization test (PRNT) is a confirmatory diagnostic assay that is used to confirm a variety of diseases. The performance of PRNT requires the use of infectious wild type viruses, which increases the risk of laboratory acquired infections. For instance, eastern equine encephalitis (EEEV) is a highly virulent pathogen used in PRNT that can result in potentially fatal neurological diseases among humans and equines. Therefore, arboviral PRNT must be performed in Biosafety Level 3 (BSL-3) containment facilities and may require select agent approved scientists, like in the case of EEEV. These stringent requirements restrict the ability of public …

USF Tampa Graduate Theses and Dissertations

Malaria is a parasitic disease that is caused by the plasmodium parasite. Plasmodium infection has affected man for thousands of years. With advances in drug discovery over the past century, malaria has evolved to possess resistance to most mainline therapeutics. This war of drug discovery vs plasmodium evolution continues to be fought to this very day, with attempts to eradicate malaria worldwide. Frontline treatments such as chloroquine, artemisinin, and atovaquone/proguanil have all seen parasitic resistance in strains of P. vivax as well as P. falciparum. While plasmodium possesses resistance to most classes of anti-malarials, the 8-aminoquinoline (8-AQ) class has …

The tumor microenvironment is composed of cellular and stromal components such as tumor cells, mesenchymal cells, immune cells, cancer associated fibroblasts and the supporting extracellular matrix. The tumor microenvironment provides crucial support for growth and progression of tumor cells and affects tumor response to therapeutic interventions. To better understand tumor biology and to develop effective cancer therapeutic agents it is important to develop preclinical platforms that can faithfully recapitulate the tumor microenvironment and the complex interaction between the tumor and its surrounding stromal elements. Drug studies performed in vitro with conventional two-dimensional cancer cell line models do not optimally represent …

Role Of Viruses Within Metaorganisms: Ciona Intestinalis As A Model System, Brittany A. Leigh Sep 2017

Role Of Viruses Within Metaorganisms: Ciona Intestinalis As A Model System, Brittany A. Leigh

USF Tampa Graduate Theses and Dissertations

Marine animals live and thrive in a literal sea of microorganisms, yet are often able to maintain specific associations that are largely dictated by the environment, host immunity and microbial interactions. Animal-associated microbiomes include bacteria and viruses that vastly outnumber host cells, especially in the gut environment, and are considered to be integral parts of healthy, functioning animals that act as a metaorganism. However, the processes underlying the initial establishment of these microbial communities are not very well understood. This dissertation focuses on the establishment of a well-known developmental animal model, Ciona intestinalis (sea squirt), to study the establishment and …

Freshwater Wetland Creation In A Changing Urban Environment: Designing For Long-Term Viability, Aaron Thomas Ryan Brown

USF Tampa Graduate Theses and Dissertations

Urbanization and wetland mitigation are increasingly common in coastal watersheds with expanding populations. These mitigation wetlands are intended to offset the functional and structural losses experienced when natural systems are degraded or destroyed. In the Tampa Bay watershed, urbanization is both expanding into the upper reaches of the watershed and intensifying in previously-developed areas, resulting in the creations of hundreds of freshwater mitigation wetlands. This dissertation utilized an existing database of mitigation wetlands, publicly available data, and field surveys to investigate the relationship between constructed wetlands and their surroundings and also determine how design affects wetland condition over time. The …

The Effects Of Loneliness On Consumers’ Digital Engagement With Social Media Ads, Yu Qin Jul 2017

The Effects Of Loneliness On Consumers’ Digital Engagement With Social Media Ads, Yu Qin

USF Tampa Graduate Theses and Dissertations

Social media has become an integral part of consumers’ lives and a popular platform for interactive marketing communication. In an age boasting of unprecedented digital connectivity, loneliness (defined as perceived social isolation) has, paradoxically, becoming widespread, affecting consumers’ preferences for products and brands in the marketplace. The present research extends the feelings-as-information theory and develops a conceptual framework in which loneliness modifies people’s cognitive processing style and induces concrete thinking in the context of evaluating social media messages. Using a mixed methods research approach, I first take a close look through the lens of a qualitative study at what it …

Subtropical Benthos Vary With Reef Type, Depth, And Grazing Intensity, Kara R. Wall Jul 2017

Subtropical Benthos Vary With Reef Type, Depth, And Grazing Intensity, Kara R. Wall

USF Tampa Graduate Theses and Dissertations

Marine epibenthic communities are influenced by both pre- and post-recruitment processes. For instance, the larval supply and cues that influence settlement (pre-recruitment), as well as the growth and mortality of individuals (post-recruitment), may differ across reef type and depth. Determining the relative influence of these processes is important to understanding how epibenthic communities can develop in a region. Using both a recruitment experiment that controlled grazing by urchins and in situ photographic surveys of epibenthic communities, this study examined the recruitment and composition of epibenthos on natural limestone and artificial reefs in the eastern Gulf of Mexico (eGOM). In the …

Osteopontin Plays A Pivotal Role In In Increasing Severity Of Respiratory Syncytial Virus Infection, Viviana Sampayo-Escobar Jul 2017

Osteopontin Plays A Pivotal Role In In Increasing Severity Of Respiratory Syncytial Virus Infection, Viviana Sampayo-Escobar

USF Tampa Graduate Theses and Dissertations

The molecular mechanisms underlying susceptibility to severe respiratory syncytial virus (RSV) infection remain poorly understood. Herein, we report on the role of osteopontin (OPN) in regulation of RSV infection in human epithelial cells and how interleukin-1 beta (IL-1β), a cytokine secreted soon after RSV infection, when persistently expressed can induce OPN expression leading to increased viral infection. We first compared OPN expression in two human epithelial cell lines: HEK-293 and HEp-2. In contrast to HEp-2, HEK-293 expresses low levels of pro-caspase-1 resulting in decreased IL-1β expression in response to RSV infection. We found a correlation between low IL-1β levels and …

Regulation Of Extracellular Signal-Regulated Kinase By Histone Deacetylase 6, Jheng-Yu Wu Jul 2017

Regulation Of Extracellular Signal-Regulated Kinase By Histone Deacetylase 6, Jheng-Yu Wu

USF Tampa Graduate Theses and Dissertations

Extracellular signal-regulated kinases 1/2 (ERK1/2) are important kinases regulating cell proliferation and cell migration, and have been established as therapeutic targets for cancer treatment. Previously, we found that ERK1 phosphorylates histone deacetylase 6 (HDAC6) to regulate its enzymatic activity. However, whether HDAC6 reciprocally modulates ERK1 activity is unknown. Here, we have discovered that ERK1/2 are acetylated proteins and shown that HDAC6 manipulates ERK1’s kinase activity via deacetylation. We demonstrated that both ERK1 and ERK2 interact with HDAC6 physically. We showed that the acetylation level of GST-ERK1/2 increased in a dose- and time-dependent manner upon treatment with a pan-HDAC inhibitor, Trichostatin …

USF Tampa Graduate Theses and Dissertations

In the United States, approximately 3% of infants are diagnosed with a major birth defect each year. Whereas prevention efforts have led to decreases in some birth defects, the cause of approximately 70% of birth defects remains unknown. More recently, there has been increasing concern regarding exposures to environmental agents, such as air pollution during pregnancy, and the risk of birth defects. Over the past decade, there has been an increase in research studies examining the association between air pollution and birth defects. The results have been inconsistent, with some studies reporting that higher levels of exposure from the same …

Viruses In Marine Animals: Discovery, Detection, And Characterizarion, Elizabeth Fahsbender Jul 2017

Viruses In Marine Animals: Discovery, Detection, And Characterizarion, Elizabeth Fahsbender

USF Tampa Graduate Theses and Dissertations

Diseases in marine animals are emerging at an increasing rate. Disease forecasting enabled by virus surveillance presents a proactive solution for managing emerging diseases. Broad viral surveys aid in disease forecasting by providing baseline data on viral diversity associated with various hosts, including many that are not associated with disease. However, these viruses can become pathogens due to expansion in host or geographic range, as well as when changing conditions shift the balance between commensal viruses and the host immune system. Therefore, it is extremely valuable to identify and characterize viruses present in many different hosts in a variety of …

Distribution Of Polybrominated Diphenyl Ethers Among Demographic Categories, Giorvanni Merilis Jul 2017

Distribution Of Polybrominated Diphenyl Ethers Among Demographic Categories, Giorvanni Merilis

USF Tampa Graduate Theses and Dissertations

Polybrominated Diphenyl Ethers (PBDEs) are flame retardants widely used within the United States in various products such as plastics, electronics, textiles and furniture. With an increase in production and usage, PBDEs have recently emerged as a contaminant of concern. Due to their chemical structure, PBDEs have the propensity to bioaccumulate in mammals. In fact, elevated PBDE concentrations have been recorded in human breast milk. Due to the potential widespread exposure to PBDEs, this study investigates human blood concentrations of PBDEs generated through the 2003-2004 National Health and Nutrition Examination Survey. Through the use of statistical modeling, a comparison of mean …
