In this thesis, we explored the differential effects of sodium-glucose cotransporter-2 (SGLT2) inhibitors on cardiovascular and renal health, utilizing both clinical data and molecular insights. Focusing on three commonly prescribed SGLT2 inhibitors—Dapagliflozin, Canagliflozin, and Empagliflozin—we employed the Food and Drug Administration Adverse Event Reporting System (FAERS) database to assess adverse events, including myocardial infarction, cardiac failure, tachycardia, acute kidney injury (AKI), renal failure, urinary tract infections, and fungal infections. This analysis was complemented by transcriptomic data from kidney cell lines treated with these drugs, processed through NCBI’s GEO2R and Limma programs, to reveal the gene expression profiles linked to these …

The literature review reveals a gap in secondary English Language Arts (ELA) teachers’ perception of students’ moral intelligence (MI). While many character education studies examine the student perspective, this study contributes to the limited research on secondary ELA teachers’ perceptions of students’ moral intelligence and how it can be cultivated. This dissertation addresses that gap by collecting and analyzing ELA teachers’ lived experiences and perceptions of students’ MI, as well as exploring how ELA teachers and schools can support students’ moral development by integrating moral and ethical topics into instructional practices. Wastewater stabilization ponds, such as facultative and maturation lagoons, have been shown to be effective processes to manage conventional water quality parameters found in domestic wastewater. The main goal of this thesis is to explore whether wastewater stabilization ponds can simultaneously reduce pathogens to safe levels while supporting agricultural activities through nutrient recovery. A large dataset for water quality of wastewater lagoon effluent in the U.S. was obtained at the state level and combined into regional southern and northern locations. The purpose of this dissertation was to experiment with qualitative research methodologies while exploring the complexities of doctoral education, research practices, philosophies of inquiry, and other topics intersecting with(in) educational scholarship. My vision for this work was fueled by postmodernist critiques of humanist education, frustrations with the methodological status quo pervasive in educational research, and deep concerns about the unsustainability of performance-focused teaching and learning.

The processes of diversification, selection, dispersal, and stochastic drift can combine in many ways to assemble ecological communities. A key focus in ecology is to describe and quantify the relative influences of these processes for different systems and explain the mechanisms by which these processes produce patterns of community structure. To accomplish this goal, ecologists have developed several frameworks that seek to integrate processes that occur locally andregionally. The metacommunity concept treats local communities as pieces of a broader landscape that are connected through dispersal of their constituent organisms. Lane departure crashes are one of the most prevalent accidents in Florida and the United States. Various forms of rumble strips have been employed to mitigate and decrease these incidents. The Florida Department of Transportation (FDOT) recently adopted a new sinusoidal rumble strip pattern for statewide implementation of audible and vibratory treatment (AVT). Recent research indicates sinusoidal rumble strips are highly effective in alerting drivers audibly and through tactile vibrations, thus aiding lane departure prevention. Metabolic alterations play a critical role in shaping tumor growth and development. Many metabolic enzymes are shown to play an important role in different aspects of tumor development, growth, and metastasis. One such enzyme that we are investigating in this project is the Phosphoglycerate Dehydrogenase (PHGDH). The PHGDH gene, encoding the rate-limiting enzyme of the serine synthesis pathway, is located at chromosome locus 1p12 and frequently amplified in melanoma. PHGDH is required for the proliferation of melanoma cells with chromosome 1p12 amplification. However, the role of PHGDH in chromosome 1p12 non-amplified melanoma is poorly understood. Families are often the forgotten partners in education and in educational research. It is important to consider input from families regarding their children’s education. Their voices are crucial ones that should be heard more often. The research is currently lacking information about families’ perceptions about the education of their children with significant disabilities. Capillaries, the smallest blood vascular networks, are formed by narrow and elongated endothelial cell (EC)-lined tubes with associated pericytes anchored to the abluminal tube surface. Our laboratory demonstrated that dynamic interactions between ECs and pericytes during capillary tube network assembly are required for basement membrane matrix deposition and maturation. We have also shown that key EC-derived factors promote pericyte recruitment, proliferation, and pericyte-induced basement membrane formation during capillary assembly. The lymphatic vasculature maintains tissue fluid homeostasis by absorbing interstitial fluid to form lymph and transporting it back to the bloodstream. Collecting lymphatic vessels have lymphatic valves (LVs) that prevent the retrograde flow of lymph generated by hydrostatic pressure from the blood vasculature. Backflow and accumulation of lymph leads to a lymphatic pathology called lymphedema. This implicates the involvement of defective LVs in the pathogenesis of lymphedema and warrants the study of lymphatic valve development and maintenance. Incels (short for “involuntary celibates) are an online community of men who blame women for their reputed sexual and romantic failures. The misogyny of their worldview, called the blackpill, has inspired multiple instances of real-world violence against women. Alarmingly, we know little about the temporal processes whereby an incel goes from joining an online community to becoming deeply aligned (i.e., “fused”) with the group. This project sought to address this by scraping incels’ most popular web forum and using text analysis to identify psychological mechanisms that could be responsible for incels’ fusion. Empathy, care, and prosocial behavior are essential to social group cohesion, and antisocial and violent criminal behavior, on the other pole, can cause serious disruptions in group cohesion and negatively impact society as a whole. As an atom-economic way of carbon-nitrogen bond formation, hydroaminations are great tool for make amines, hydrazines, and other nitrogen-containing heterocycles. Specifically, radical hydroamination is a useful approach for functionalization of unactivated olefins, especially in sterically hindered environments. Diversification remains a challenge for radical hydroaminated products, since the nitrogen sources are carefully designed. In the first chapter of this dissertation, we report a highly diversifiable radical hydroamination reaction with diazirine as a nitrogen source and its potential applications in synthesis of several pharmaceutical interested compounds. Alcohol consumption tends to occur in social contexts and has been shown to increase prosocial behaviors and facilitate social connection. Simultaneously, socially excluded drinkers tend to increase or continue problematic levels of alcohol consumption when alone, possibly consuming alcohol as a replacement for desired but unavailable social connection. This investigation attempted to examine the relationship between loneliness (i.e., lack of perceived social connection) and subsequent alcohol consumption in solitary versus social contexts using ecological momentary assessment (EMA) techniques. The purpose of this study is to explore the experiences and perceptions of teachers who use arts-integrated instruction and their directors of K-2 students before, during, and after the unique period of online instruction during the COVID-19 pandemic, which began in March of 2020. The biochemistry of human tissues can provide information for geoprofiling (geographic birthplace or migration patterns) of an unidentified decedent. Utilizing isotope analyses (both stable and radiogenic) from teeth, bone, nails, and/or hair reveals specific isotopic information that correlates to diverse geographic regions reflecting an individual’s birthplace and migration patterns. Isotope analysis of enamel and bone has been employed in bioarchaeology for decades to reconstruct the diet, health, and migration patterns of past populations. Delay discounting has long been associated with substance misuse across both animal subjects and human participants. The purpose of the proposed study was to evaluate if and when changes in delay discounting occur during a residential treatment program for alcohol use. Participants were asked to complete the 5-trial adjusting amount discounting task (Koffarnus & Bickel, 2014) and an electronic version of the Du et al. (2002) delay discounting task repeatedly across 21 days while in residential treatment. A PROTAC molecule is a bifunctional molecule composed of two active domains and a linker. It works by inducing selective protein intracellular degradation. This technology was first discussed by Kathleen Sakamoto, Ray Deshaies and Craig Crews in 2001. Reasoning over natural text is highly nuanced, and interpretations can vary widely depending on cultural background, financial status, age, gender, or even mood. This doctoral dissertation seeks to not only mimic human reasoning behaviors but also improve the task used in natural language processing (NLP) to capture naturalistic reasoning, known as the Natural Language Inference (NLI) task. Since the FDA approval of IL-2 in cancer treatment in the 1980s, immunotherapy has emerged as the standard of care for several tumor types (e.g., melanoma, lung, bladder, colorectal). However, approximately 70 percent of cancer patients, independent of prior treatment or staging, administered immunotherapy will not achieve complete responses and will exhibit disease progression. Several factors have been identified to contribute to treatment failure, including the accumulation of intratumoral immunosuppressive cell populations, insufficient tumor-infiltrating anti-tumor immune populations, or the occurrence of immunoediting on tumor cells. Group B Streptococcus (Streptococcus agalactiae; GBS) is a leading cause of neonatal sepsis worldwide. Intrapartum antibiotics in GBS-colonized mothers significantly reduced the incidence of early-onset sepsis occurring in the first week of life; however, there are currently no prophylactic interventions for late-onset sepsis, occurring within the first 3 months of life. GBS is a pathobiont that primarily colonizes the gastrointestinal tract and can cross the intestinal barrier via both the paracellular and transcellular routes, causing invasive disease. The focus of this dissertation was simulating trophic dynamics in the pelagic Gulf of Mexico (GOM) with an emphasis on ecosystem-based fishery management (EBFM) strategies for large pelagic species. Fisheries management in the United States largely utilizes single-species assessments, although managers are increasingly interested in EBFM approaches to better account for ecosystem effects on stocks such as predator/prey dynamics. Production in the surface layer of the pelagic zone is variable with environmental conditions and prey is often concentrated in patches by hydrodynamic features. Tumor-immune interactions shape a developing tumor and its tumor immune microenvironment (TIME) resulting in either well-infiltrated, immunologically inflamed ‘hot’ tumor beds, or ‘cold’ immune deserts with low levels of infiltration. The pre-treatment immune state of the TIME is associated with treatment outcome; immunologically hot tumors generally exhibit better responses to radio- and immunotherapy than cold tumors. However, radiotherapy is known to induce opposing immunological consequences, resulting in both immunostimulatory and inhibitory responses. In fact, it is thought that the radiation induced tumoricidal immune response is curtailed by subsequent applications of radiation. Progressive muscle relaxation (PMR) has been shown to produce desired outcomes by decreasing disruptive behavior in the classroom and increasing academic engagement. The integration of technology into the educational setting alleviates some of the burden on educators and enhances the learning experience for students. This study evaluated the effectiveness of brief video-based PMR to increase academic engagement in the classroom with three elementary school students diagnosed with autism spectrum disorder (ASD).
