Viruses Found In Raw Sewage And Their Potential To Indicate Fecal Pollution In Coastal Environments, Erin M. Symonds Jun 2008

Viruses Found In Raw Sewage And Their Potential To Indicate Fecal Pollution In Coastal Environments, Erin M. Symonds

USF Tampa Graduate Theses and Dissertations

The presence of pathogenic viruses in coastal environments is an important tool in evaluating water quality and health risks. Millions of viruses are excreted in fecal matter and bacterial indicators do not correlate with the presence of pathogenic viruses. Enteroviruses have been used to identify fecal pollution in the environment; however, other viruses shed in fecal matter could be used to indicate fecal pollution. The purpose of this research is to develop a baseline understanding of the diversity of viruses found in raw sewage and to assess their presence in the marine environment. PCR was used to detect adenoviruses, herpesviruses, …

Immunomodulation As A Potential Therapeutic Approach For Alzheimer’S Disease, William Veljko Nikolic Jun 2008

Immunomodulation As A Potential Therapeutic Approach For Alzheimer’S Disease, William Veljko Nikolic

USF Tampa Graduate Theses and Dissertations

Alzheimer's disease (AD) is the most prevalent form of progressive dementia and is characterized by the accumulation of amyloid beta (Aß) peptide in the brain and in the cerebral vessels forming cerebral amyloid angiopathy (CAA). As previously reported, an active immunization strategy of mice with Aß1-42 peptide results in decreased Th1 and increased Th2 cytokine responses as well as an effectively clearance of CNS Aß. This approach has also yielded favorable results for many patients, unfortunately, a small percentage of these study participants developed severe aseptic meningoencephalitis likely secondary to CNS invasion of activated T-cells. We have previously demonstrated …

The Relationship Between Individual Differences In Cognitive, Social And Personality Development And The Increase In Complexity Of Children’S Alcohol Expectancies, Nicole M. Bekman Jun 2008

The Relationship Between Individual Differences In Cognitive, Social And Personality Development And The Increase In Complexity Of Children’S Alcohol Expectancies, Nicole M. Bekman

USF Tampa Graduate Theses and Dissertations

The current study aimed to simultaneously examine cognitive, social and personality development in a cross-sectional sample of 3rd, 4th and 5th grade children to explore the interplay among these processes and how they relate to changes in children's understanding of alcohol. To replicate previous work, this study comprehensively examined relative increases in types of expectancies as a function of development. Results demonstrated that children in higher grades held more positive, negative and sedating expectancies of alcohol and positive alcohol expectancies increased more than negative alcohol expectancies. Improved performance on cognitive measures were associated with positive alcohol …

The Role Of Extracellular Matrix And Matrix-Degrading Proteases In Neonatal Hypoxic-Ischemic Injury, Christopher C. Leonardo Jun 2008

The Role Of Extracellular Matrix And Matrix-Degrading Proteases In Neonatal Hypoxic-Ischemic Injury, Christopher C. Leonardo

USF Tampa Graduate Theses and Dissertations

Improvements in medical care over recent decades have increased the number of premature and low birth weight infants that survive hypoxic-ischemic (H-I) insults. Because there is a rising incidence in diseases associated with these events, it is critical to develop effective therapies to treat the various resulting neuropathies. Extracellular matrix constitutes the majority of brain parenchyma. Lecticans and matrix-degrading proteases including ADAMTSs (a disintegrin and metalloproteinase with thrombospondin repeats) and matrix metalloproteinases (MMPs) exert effects on cell viability and may be associated with either protective or destructive processes after H-I. Both ADAMTSs (Cross et al. 2006; Tian et al. 2007) …

Error Related Negativity In Parkinson’S Disease: A Test Of The Validity Of Mesencephalicdopamine Contributions To Ern, Craig A. Siders Jun 2008

Error Related Negativity In Parkinson’S Disease: A Test Of The Validity Of Mesencephalicdopamine Contributions To Ern, Craig A. Siders

USF Tampa Graduate Theses and Dissertations

A model proposed by Holroyd and Coles (2002) stating that error related negativity (ERN) is caused by a decrease in mesencephalic dopamine output to the ACC was tested. A group of individuals with Parkinson's disease (N = 16) and an age and education matched group free from neurological disorder (N = 16) completed a card guessing task where the magnitude of monetary penalties and rewards for incorrect and correct answers was varied by block. Individuals with Parkinson's disease were tested after an overnight washout from dopamingeric medications.

The amplitude of the mid-frontal negativity elicited by feedback was analyzed with spatial …

Yeats, Eliot, And Apocalyptic Poetry, Nancy Helen Fletcher May 2008

Yeats, Eliot, And Apocalyptic Poetry, Nancy Helen Fletcher

USF Tampa Graduate Theses and Dissertations

Yeats and Eliot merit comparison because they wrote poetry that has been described as apocalyptic in the same historical period and in the same general geographic area but described entirely different visions. These particular works of Yeats and Eliot are appropriate because they represent two widely varying viewpoints on the causes, nature and desirability of what each author felt would be the coming apocalypse. Therefore, more can be learned by comparing the very different outlooks of the poems than by considering each poem separately.

Yeats sees humanity as both the victim and the beneficiary of a series of inescapable historical …

High-Rise Neighborhood: Rethinking Community In The Residential Tower, Benjamin Hurlbut May 2008

High-Rise Neighborhood: Rethinking Community In The Residential Tower, Benjamin Hurlbut

USF Tampa Graduate Theses and Dissertations

Within the United States, a growing sense of detachment exists. Conditions in both urban and suburban contexts have created a sense of social detachment where spaces do not exist which encourage social interaction. Without this social activity, neighbors become almost a disposable commodity as relationships never fully develop. This thesis will be an examination of environments which do and do not foster community relationships and an implementation of community into an urban multi-family residence.

A major part of the problem is the recent move to the suburbs, but urban buildings also exhibit a lack of spaces which help encourage a …

Analysis Of The Role Of Bhlh/Pas Proteins In Aryl Hydrocarbon Receptor Signaling, Edward J. Dougherty May 2008

Analysis Of The Role Of Bhlh/Pas Proteins In Aryl Hydrocarbon Receptor Signaling, Edward J. Dougherty

USF Tampa Graduate Theses and Dissertations

The aryl hydrocarbon receptor (AHR) is a basic helix-loop-helix PER/ARNT/SIM (bHLH-PAS) transcription factor that binds ligands typified by 2,3,7,8-tetracholordibenzo-p-dioxin, translocates to the nucleus, dimerizes with the aryl hydrocarbon nuclear translocator (ARNT) and associates with specific cis xenobiotic response elements to activate transcription of genes involved with xenobiotic metabolism. AHR-mediated signal transduction has been evaluated thoroughly in the C57BL/6J mouse model system. This model system, however, may not be the most accurate model for human comparisons as the AHRb-1 allele carried by C57BL/6J contains a point mutation that prematurely truncates the receptor at 805 amino acids, while the AHRb-2 …

Corona Ion Deposition: A Novel Non-Contact Method For Drug And Gene Delivery To Living Systems, Niraj Ramachandran

USF Tampa Graduate Theses and Dissertations

Application of corona ions produced in air to B16F10 murine melanoma cells in vitro and in animal models resulted in the transport of molecular therapeutics across the cell membrane. This work presents the development of new methods for drug and gene delivery based upon similar principles as the traditional electrode driven membrane destabilization processes known as electroporation. This was achieved with non-contact corona ion deposition that temporarily increased the permeability of cell membranes.

Interaction of corona charge with biological cells was studied and their potential for molecular delivery was established. Molecular delivery was first demonstrated in vitro using tracer molecules …

A sixth condition attenuated the level of the 3,000 ms, 95 dB, 0.2-24 kHz signal in 3 dB increments until accuracy reached 75%. Subjects performed above …

Black Beauty As Antebellum Slave Narrative, Bonnie L. Blossom Apr 2008

Black Beauty As Antebellum Slave Narrative, Bonnie L. Blossom

USF Tampa Graduate Theses and Dissertations

Published in November 1877, Black Beauty is one of the most popular and enduring works of the nineteenth and twentieth centuries. The book, in which the eponymous narrator relates his life's story, sold well following its publication in England and in the United States; by 1985, sales were estimated at over forty million. While usually regarded as entertaining, Black Beauty has a strong crusading purpose: Anna Sewell herself said she wrote to improve the treatment of horses.

This study springs from an intuitive notion. While reading the 1845 Narrative of the Life of Frederick Douglass, I could not shake …

Boundary And Longing: Narrative Modes In Mary Shelley’S Frankenstein And The Last Man, Marie Hendry Apr 2008

Boundary And Longing: Narrative Modes In Mary Shelley’S Frankenstein And The Last Man, Marie Hendry

USF Tampa Graduate Theses and Dissertations

Boundary and desire surround the relationships in Mary Shelley's Frankenstein and The Last Man. The narrative modes of Captain Robert Walton relate his separation to the rest of the world and his need for companionship. Yet, not any companionship will satisfy his longing for connection with a human being; his search revolves around the need of common understanding. This further separates the character of Lionel in The Last Man from humanity in that he is unable to find anyone left on earth after a series of plague, war, and atmospheric anomalies apparently wipe out the human race. His survival …

Crystal Engineering Of Flavonoids, Padmini Kavuru Apr 2008

Crystal Engineering Of Flavonoids, Padmini Kavuru

USF Tampa Graduate Theses and Dissertations

Crystal engineering is attracting attention in the pharmaceutical industry because the design of new crystal form of drugs can improve their stability, bioavailability and other relevant physical characteristic properties. Therefore, crystal engineering of nutraceuticals such as flavonoids by exploring their hydrogen bonding interactions can generate novel compounds such as pharmaceutical cocrystals. Flavonoids are polyphenolic secondary plant metabolites that are present in varying levels in fruits, vegetables and beverages. The "French paradox", low cardiovascular mortality rate in spite of high intake of saturated fat among the Mediterranean populations made flavonoids an appropriate target for therapeutic researchers.

The work herein deals with …

Isolation And Functional Characterization Of A Dioxin-Inducible Cyp1a Regulatory Region From Zebrafish (Danio Rerio), Gary T. Zeruth

USF Tampa Graduate Theses and Dissertations

Cytochrome P4501A1 (CYP1A1) is a phase I bio-transformation enzyme involved in the metabolism of xenobiotics via the oxygenation of polycyclic aromatic hydrocarbons (PAHs) including the carcinogen, benzo(a)pyrene. Induction of the CYP1A1 gene is regulated at the transcriptional level and is ligand dependent with the prototypical 2,3,7,8,-tetrachlorodibenzo-p-dioxin (TCDD) being the most potent known inducer of CYP1A1 transcription. This process is mediated by the AHR/ARNT signaling pathway whereby ligand binds AHR in the cytoplasm allowing its translocation to the nucleus where it binds with its hertrodimerization partner, ARNT and subsequently binds DNA at cognate binding sites termed xenobiotic …

USF Tampa Graduate Theses and Dissertations

The earliest attempt to form a confederation among the British colonies in North America occurred with the creation New England confederation in 1643. For the next one hundred thirty-four years various attempts would be made to organize British America colonies into a confederacy. "Smole Trifeles": The Itinerant in British America moves beyond traditional histories which analyze how the social and political elite worked to bring the colonies together. This work focuses on the peddlers, the hawkers, petty chapmen, and the itinerant preachers who roamed the primitive roads and highways of early America. These wanderers knit together a nation and helped …

“Pleasant Episodes” Of Gastronomy: Food And Drink In F. Scott Fitzgerald’S The Beautiful And Damned, Melissa Faith Dullaghan Apr 2008

“Pleasant Episodes” Of Gastronomy: Food And Drink In F. Scott Fitzgerald’S The Beautiful And Damned, Melissa Faith Dullaghan

USF Tampa Graduate Theses and Dissertations

This thesis explores the motif of gastronomy in Fitzgerald's critically undertreated second novel, The Beautiful and Damned. Within the discussion of the leisure class, Fitzgerald scholars often focus on Jay Gatsby's parties, but they seem to neglect Anthony Patch and company's fancy for food and drink in Ivy League supper clubs of Manhattan, vaudeville theaters, and houses of languor in Upstate New York. Building upon George J. Searles's article "The Symbolic Function of Food and Eating in F. Scott Fitzgerald's The Beautiful and Damned," this thesis examines the meaning of Fitzgerald's pervasive "prandial allusions" and character psychology with …

Movement, distribution, and habitat use of juvenile bull sharks were examined in two studies using manual and passive acoustic telemetry. Research was conducted in the Caloosahatchee River, which serves as nursery habitat for this species, and is highly impacted due to anthropogenic alterations in water quality and quantity via dams and locks. Manual tracking yielded fine-scale results for eight individuals on home range size, rate of movement, swimming depth, linearity, direction of travel, tidal influence, diel pattern, as well as correlation with environmental variables. Changes in salinity, temperature, dissolved oxygen, turbidity, and pH played a role on the distribution of …

The Role Of Rala And Ralb In Cancer, Samuel C. Falsetti Apr 2008

The Role Of Rala And Ralb In Cancer, Samuel C. Falsetti

USF Tampa Graduate Theses and Dissertations

Ras genes are frequently mutated in human cancers and present compelling targets for therapeutic intervention. While previous attempts to directly inhibit oncogenic Ras function have largely been unsuccessful use of targeted agents to inhibit the three primary oncogenic pathways activated by mutated Ras: RalGEF-Ral, PI3K-Akt and Raf- MEK-Erk, is an area of intense investigation. Here, we describe the ability of a novel pharmacological inhibitor of geranylgeranyltransferase I, GGTI-2417, to inhibit Ral prenylation and localization. We further used a Ral rescue system to selectively preserve RalA and RalB function and localization during GGTI-2417 treatment and determine the precise roles for inhibition …

Micro-Autoradiographic Fusion Tomography, James Merker Apr 2008

Micro-Autoradiographic Fusion Tomography, James Merker

USF Tampa Graduate Theses and Dissertations

Two-dimensional (2-D) micro-autoradiography is typically used to identify the location of a radio-labeled ligand bound to a cellular target in tissue sections. Data, such as a histological image, combined with the autoradiographic data provide a spatial relationship of the radiolabel to cellular structures. However, the disadvantage of 2-D imaging is that it only provides a local distribution of the radiolabel within a tissue slice, and not a volumetric regional distribution in the structure of interest. The development of 3-D autoradiographic/histological visualizations would provide important information not otherwise apparent, such as the ability to visualize the distribution of the labeled agents …

Impact Of West Nile Virus On The Natural History Of St. Louis Encephalitis Virus In Florida, Christy L. Ottendorfer Apr 2008

Impact Of West Nile Virus On The Natural History Of St. Louis Encephalitis Virus In Florida, Christy L. Ottendorfer

USF Tampa Graduate Theses and Dissertations

The emergence of West Nile virus (WNV) has raised important questions about the capacity of the public health infrastructure to implement surveillance and control programs for WNV and other emerging or re-emerging arboviruses in the United States. Florida's mild climate supports year round enzootic transmission of WNV, St. Louis encephalitis virus (SLEV), and Eastern Equine Encephalitis virus (EEEV). It is unknown what effect the establishment of WNV (in 2001) will have on SLEV transmission in Florida, where these closely related flaviviruses share amplifying hosts, habitats, and vectors.

An Arbovirus Isolation Network was formed to obtain and characterize arbovirus strains collected …

Quebrada Communities In The Palmarejo Valley, Northwest Honduras, William A. Klinger Apr 2008

Quebrada Communities In The Palmarejo Valley, Northwest Honduras, William A. Klinger

USF Tampa Graduate Theses and Dissertations

The spatial relationships that exist between ancient and modern settlement and natural resources have the potential to suggest ways in which humans organized themselves into communities. This study evaluates the applicability of the concept, "quebrada community," for understanding human-environmental relationships in northwest Honduras during the Late Classic period (AD 650-900). Existing archaeological, quantitative, and geological evidence for quebrada communities are linked with spatial data on two contemporary local communities, Palmarejo and Palos Blancos. A geographic information system (GIS) is constructed and implemented in order to achieve this goal. It is argued that there are specific relationships that exist between ancient …

Cost-Effectiveness Of Epidural Steroid Injections To Treat Lumbosacral Radiculopathy In Chronic Pain Patients Managed Under Workers’ Compensation, Sheila Mohammed

USF Tampa Graduate Theses and Dissertations

No conclusive evidence exists to determine that epidural steroid injections (ESIs) provide lasting improvements in chronic pain due to herniated discs, in the Workers' Compensation population. Recently, an article by Armon et.al was published by the American Academy of Neurology, which stated that the routine use of ESIs is not recommended and that further studies are needed to elucidate this controversy (Armon, Argoff, Samuels, & Backonja, 2007).

In 1998, back pain in the United States was estimated to have incurred total health-care expenditures of $90.7 billion. Medicare part B. claims in 1999 for 40.4 million individuals amounted to $49.9 million …

Counting Or Discounting Television Information: An Examination Of Viewer Perceptions About Old-Age From A Cognitive Processing Perspective Of Cultivation Effects, Lynda Pasteur Apr 2008

Counting Or Discounting Television Information: An Examination Of Viewer Perceptions About Old-Age From A Cognitive Processing Perspective Of Cultivation Effects, Lynda Pasteur

USF Tampa Graduate Theses and Dissertations

Cultivation theory (Gerbner & Gross, 1976) suggests that the cumulative effect of heavy exposure to television's underrepresentation and negative portrayal of people 65+ as sexless, insignificant, and comical can cause people to assume such television-like perceptions of the age group in the real world. This study's purpose was to explore not only television's cultivation effect on viewers' perceptions of the number and nature of people 65+, but also the psychological processes that are expected to mediate this effect. As an extension of Shrum, Wyer, and O'Guinn's (1998) study on the role of source discounting in cultivation, this quasi-experiment employed three …

Victorian Perspectives On The Supernatural: The Imaginary Versus The Real In Two Brontë Novels, Crystal Sidell Apr 2008

Victorian Perspectives On The Supernatural: The Imaginary Versus The Real In Two Brontë Novels, Crystal Sidell

USF Tampa Graduate Theses and Dissertations

The Victorians obsessed over the supernatural and this fascination with the otherworldly emerges in the literature of the day. With this thesis, I look at two nineteenth century novels that exhibit supernatural phenomena: Charlotte Brontë's Villette (1853) and Emily Brontë's Wuthering Heights (1847). Both novels, I propose, utilize this aspect of the gothic tradition to enhance their characters' psychological realism.

With Villette, I examine the supernatural as a fabricated experience. First, I study the protagonist's psyche and show how her emotional state directly contributes to the appearance of fantastic material. Specifically, I examine Lucy Snowe's childhood experiences in Bretton …

Digitally Implemented Interactive Fiction: Systematic Development And Validation Of “Mole, P.I.”, A Multimedia Adventure For Third Grade Readers, Denise Haunstetter Apr 2008

Digitally Implemented Interactive Fiction: Systematic Development And Validation Of “Mole, P.I.”, A Multimedia Adventure For Third Grade Readers, Denise Haunstetter

USF Tampa Graduate Theses and Dissertations

"Interactive fiction" has been used to describe many of today's multimedia products. In reality, there is not a universal understanding of what interactive fiction is or what it should entail. The meaning of "interactive" is often interpreted in different ways. Many stories are considered to be interactive because they are placed on the computer. Meanwhile, such stories may lack most of the essential qualities for good literature. Interaction fiction should be upheld to the same standards as traditional texts. Following this belief, this research covers the underlying theories of interactive fiction, examples of misleading "interactive fiction" studies, and guidelines for …

Forty-eight (30 Female, 18 Male) subjects were instructed to indicate the direction of an offset over 620 trials. Subjects were primed with either negative, positive or neutral photographs before making the discrimination on all test trials. Feedback regarding offset performance was provided on each trial. Despite initial pilot data indicating that subjects primed with negative, arousing images improved performance over those primed with either neutral or positive images, there was ultimately no reliable advantage for any of the …

Metallopeptides As Model Systems For The Study Of Cu(Ii)-Dependent Oxidation Chemistry, William Maung Tay Apr 2008

Metallopeptides As Model Systems For The Study Of Cu(Ii)-Dependent Oxidation Chemistry, William Maung Tay

USF Tampa Graduate Theses and Dissertations

Copper is one of the essential metal ions for aerobic organisms. Two well known functions of copper in the biological systems are electron transfer and molecular oxygen interaction. Thus, this metal can be found in haemocyanin, an oxygen carrier protein, and superoxide dismutase, an enzyme that involves in electron transfer. In addition, having a positive redox potential allows copper to be involved in redox chemistry. It is the redox properties of copper that are responsible for many important biochemical processes. Although the copper-containing oxidases have been well studied over the years, certain mechanistic details such as reaction intermediates remain to …
