This dissertation is dedicated to the research and investigation of novel enzymes and the methods used to study them, with physiological roles ranging from isoprenoid biosynthesis to neurotransmitter production. Using a combination of bioinformatics, recombinant cloning, enzymology, and proteomics, we have contributed to the understanding and exploration of several human illnesses, including malaria, cancer, and endocrine dysfunction.
Our first project involved studying the enzymes responsible for N-acylarylalkylamide biosynthesis in Bombyx mori. Very little is known how these potent signaling molecules are produced in vivo, however, one possible pathway is the direct conjugation of an acyl-CoA to a corresponding …

The vivid memory of an emotional event, as well as memory for incidental details associated with the arousing event, has been referred to collectively as a “flashbulb memory”. An important aspect of flashbulb memory in people is that an emotional event enhances memory of contextual details, such as the weather, or clothes one was wearing at the time of the event. Therefore, an emotional event not only produces a detailed memory of the event, itself, but also enhances memory for contextual details that would otherwise not be remembered. This study investigated the use of instructive feedback for modeling early literacy skills. Instructive feedback is defined as the presentation of additional information during the positive feedback phase of learner trials. Thus, it is a way of modeling additional information when students respond to a trial correctly. Previous studies have demonstrated that instructive feedback can facilitate students’ observational learning of skills, such as sight words, numerals, and vocabulary. Instructive feedback has yet to be investigated when modeling early literacy skills.

The purpose of this study was to assess if Behavioral Skills Training (BST) can be used to train a sticker to function as a discriminative stimulus (Sᴰ) for engaging in household poison safety skills and assess whether this skill generalized to untrained household chemicals that bear the Sᴰ in the form of a sticker. Three typically developing children ages 3 and 5 and their parents participated in this study which took place in their homes. Oil from the Deepwater Horizon blowout reached the Gulf of Mexico coast in the summer of 2010 and potentially exposed species living in those areas to toxic chemicals. The purpose of this study is to examine otoliths from Red Drum (Sciaenops ocellatus) for evidence of oil exposure that could be related to reduced growth rates. Because otolith growth and somatic growth are directly related, differences in annulus measurements can indicate differences in annual somatic growth, which is a good indicator of overall fish condition, and translates into changes in survival and lifetime reproductive potential. The present study had two main objectives. The first was to determine the degree of understanding of climate change, sea level and sea level rise among middle school students. Combining open-ended questions with likert-scaled questions, we identified student conceptions on these topics in 86 students from 7th and 8th grades during 2012 and 2013 before and after implementing a Curriculum Unit (CU). Additional information was obtained by adding drawings to the open-ended questions during the second year to gauge how student conceptions varied from a verbal and a visual perspective. Perhaps, under the consciousness of today, “neoliberalism” has defined our world during the previous and current centuries more than any other socioeconomic system. But the evolution of this ideology, which initially aimed to enhance, or rather, reinvent capitalism and individual freedom, has, in essence, induced an unrecognized problem. I argue that neoliberalism is the catalyst for much of the hostility in this globalized society where tensions and poverty are casualties of individual and corporate prosperity. Because of this revelation, I argue that neoliberalism inadvertently instills violence that is both unseen and gendered. In order to formulate my argument, I introduce …

North American river otters are considered common in the state of Florida, but their populations have not been studied since the 1980’s. Since that time, Florida’s human population has more than doubled, and many natural areas of Florida have been developed. The aim of this study was to determine the presence or absence of river otters at locations in Florida which they historically inhabited. Forty-six sample sites where otters were historically found were obtained from the Florida Museum of Natural History Mammals Master Database (FMNH MMD). The goal of this thesis was to adopt a reverse flow injection analysis (rFIA) technique to the fluorometric analysis of the reaction o-phthaldialdehyde (OPA) with ammonium, allowing accurate measurements of ammonium concentrations lower than the detection limit of the widely used indophenol blue (IPB) colorimetric method while accounting for the background fluorescence of seawater. Ammonium is considered an essential nutrient for primary productivity, especially in the nutrient depleted surface ocean where as the most reduced form of dissolved inorganic nitrogen, it is readily assimilated via metabolic pathways. Concussion is an injury that disproportionately affects children and adolescents and has the potential to negatively impact school performance. Currently, a significant proportion of youth with concussion go unreported due to a lack of effective concussion surveillance procedures. Laboratory-based tasks of impulsivity and related constructs can be useful in predicting alcohol use. Performance on these tasks is generally assumed to reflect traits that remain stable across situations. However, several studies have indicated that manipulations of state-like variables (e.g., mood or stress) can influence levels of impulsivity demonstrated on the tasks. Furthermore, environmental context (in the form of physical setting, or contextual cues) has a demonstrable effect on tasks relevant to alcohol-related risky behaviors (e.g., ad lib drinking tasks). Telehealth provides an alternative delivery form of parent training that allows practitioners to disseminate information and feedback at a distance. Telehealth can be as effective as in-person training (Wacker et al., 2013a; Wacker et al., 2013b; Vismara et al., 2009). The objective of this study is to investigate land-use changes and water quality trends within the headwaters of the Alafia River watershed. Water quality data were obtained from the Environmental Protection Commission of Hillsborough County (EPCHC). Eleven water quality parameters selected for analysis included: temperature (˚C), dissolved oxygen (DO), percent saturation of DO, conductivity, pH, total phosphorous (TP), total nitrogen (TN), ammonium, chlorophyll-a (uncorrected), fecal coliforms, and enterococci. This work explores the role that ideas about Africa played in the development of a specifically American identity among free blacks in the United States, from the early nineteenth century to the Civil War. Previous studies of the writings of free blacks in the Revolutionary period, and of the American Colonization Society (ACS), which was devoted to removing them back to an African homeland, have suggested that black discussions of Africa virtually disappeared after 1816, when the colonization movement began. The impact of human activity on ecosystems is an issue at the forefront of global concern. Marine ecosystems are a particular concern, given their importance for human sustenance. Through the removal of species that are highly susceptible to the effects of overfishing, global fisheries have been driven to near collapse in recent decades. This dissertation research explored mechanisms of human milk (HM) expulsion by describing the effects of skin to skin contact (SSC) on maternal basal oxytocin (OT) secretion among 20 premature mothers with hospitalized premature infants. This one-group, repeated measures design consisted of: 1) daily SSC with covariant data via self-report diary and 2) maternal salivary OT with and without SSC at 4 time points were collected over a 7 day time frame. Results indicate that mean levels of basal OT increase over time (M 234 pg/ml, SD 108 pg/ml time point 1; M 257 pg/ml, SD 125 pg/ml time point 3). …

The purpose of this study was to evaluate the use of auditory feedback for teaching individuals with intellectual disabilities the “Mississippi Cha Cha Slide.” Participants consisted of six males ages 35 to 61. During baseline, line dance skills were low for all participants. During the auditory feedback intervention, the trainer used a clicker to reinforce dance steps and forward chaining to chain movements into a sequence. Once auditory feedback was implemented, line dance skills increased substantially for all participants. Generalization assessments for four of the participants resulted in performance levels similar to baseline and demonstrate the need for future training …

Anaerobic digestion (AD) is a biotechnology that employs natural microbial metabolism under oxygen-free conditions to stabilize organic waste. AD has been shown to be the most environmentally sustainable technology for treating the organic fraction of municipal solid waste (OFMSW), as it allows for the recovery of energy and nutrients from the waste. AD of OFMSW also saves landfill space and reduces leachate generation and fugitive methane emissions from landfills. Because nitrogen isotopes are fractionated along the soil-plant-insect-bat-guano pathway, it may be possible to reconstruct environmental and climatic changes reflected in the nitrogen isotopic composition of guano. A 1.5-m core of bat guano from Zidită Cave (western Romania) provides a record of climatic and anthropogenic influence on the regional nitrogen cycle and paleoenvironmental controls on nitrogen transforming processes. Increasing and decreasing trends of nitrogen isotopic composition (δ15N values) correspond well with changes in the influence of farming practices, deforestation, and forest expansion. Here we present a novel Smartphone-based colorimeter and demonstrate its application to the measurements of glucose and protein concentrations in biological samples. The key innovation of our approach was to combine powerful image processing encoded into a mobile phone application with a low cost 3D printed sample holder that allowed to control lighting conditions and significantly improved sensitivity. Different solutions with protein and glucose concentrations ranging from 0 to 2000 mg/dL were prepared and analyzed using our system. The recent global economic downturn has stimulated a growing interest among scholars in how employees interpret and respond to the circumstance of being overqualified. However, the overqualification literature has been hindered by uncertainty regarding the extent to which employees’ perceptions of being overqualified are based in reality. The present study sought to address this concern by proposing and testing a theoretical model of objective overqualification, perceived overqualification, job satisfaction, and well-being using a cross-sectional sample of full-time employees who had recently graduated from college. Structure based drug design is a rapidly advancing discipline that examines how protein targets structurally interact with small molecules, or known inhibitors, and then uses this information to lead inhibitor optimization efforts. In the case of novel inhibitors, protein structural information is first obtained via X-ray crystallography, NMR studies, or a combination of both approaches. Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by two major pathological hallmarks, amyloid plaques and neurofibrillary tangles. The accumulation of amyloid-β protein (Aβ) is an early event associated with synaptic and mitochondrial damage in AD. Therefore, molecular pathways underlying the neurotoxicity and generation of Aβ represent promising therapeutic targets for AD. Recent studies have shown that actin severing protein, Cofilin plays an important role in synaptic remodeling, mitochondrial dysfunction, and AD pathogenesis. However, whether Cofilin is an essential component of AD pathogenesis and how Aβ induced neurotoxicity impinges its signals to Cofilin are unclear.

Diabetes has afflicted 8.3%, approximately 25.8 million, of the United States population and is the seventh leading cause of death [1]. Type I diabetes (T1D) accounts for 5 to 10% of all diagnosed cases of diabetes in the United States [2]. If present trends continue, the rate of T1D incidence among children under the age of 14 will increase by 3% globally [3]. T1D is an autoimmune disorder in which the β-cells of the pancreatic islets are destroyed, leading to high blood sugar. Numerous studies suggest that modulation of the cholinergic system through the use of nicotinic agonists can improve motor function in humans or animals with motor disorders. Specifically, although there are no approved therapeutics for patients with ataxia, the nicotinic receptor agonist varenicline has demonstrated efficacy to improve coordination and gait in several groups of patients with different subtypes of ataxia. Leptospirosis, a zoonotic disease caused by pathogenic spirochete bacteria (family Leptospiraceae, genus Leptospira), is endemic in developing tropical regions of the world. It occurs in epidemics and is endemic in Ecuador where environmental conditions are ideal for maintenance. The role of soil as a long term reservoir has been previously been documented. Geographic Information System (GIS) and Remote Sensing (RS) technology was used in our study to further explore the role of soil as an environmental reservoir and its potential use as a static risk indicator for disease.
