This study was conducted to determine the effects of chromium picolinate (CrPi) added into diet containing 710.3 ppb chromium on yield and nutrient digestibility of laying hens at low temperature. Forty-six-week-old laying hens were randomly assigned to four groups of 30 hens per group. Treatment groups were fed different supplemental dietary chromium levels. Thus, hens were fed diets with no supplemental chromium (Control Group), 100 ppb of supplemental chromium (100 Group), 200 ppb of supplemental chromium (200 Group) and 400 ppb of supplemental chromium (400 Group). In this study, our aim was to diagnose lymphoid leukosis and Marek's disease, which are neoplastic diseases of chickens, by ELISA and Indirect Immunofluorescence Test (IIF), respectively. Samples (eggs and tissues) were selectively taken from the flocks suspected of having lymphoid leukosis and Marek's disease. The tissues were macroscopically and microscopically examined. Tissues belonging to some flocks showed typical lesions for lymphoid leukosis and Marek's disease. Seven broiler breede flocks and three commercial layer flocks were tested for avian leukosis virus (ALV) group specific antigen (p27) in the albumen. Most animal breeding experiments are based on more than one economically important trait measured in each individual. In the statistical analysis of the resulting data, traits recorded in the same individuals are often considered one at a time. Usually we are interested, however, not only in the mode of inheritance of a particular trait but also in its relationships with other traits. Multivariate analyses are required to make inferences about genetic and phenotypic correlations between traits, but point estimates of such parameters can be poor even when breeding data on hundreds of animals are used. In this study, the effects of experimentally induced hyperthyroidism and hypothyroidism on the erythrocyte, leucocyte and haematocrit values, the amount of haemoglobin and plasma glucose, cholesterol, triglyceride, creatinine and lactate dehydrogenase (LDH) levels were investigated. A total of 45 4-week-old female Japanese quail (Coturnix coturnix Japonica) were used. The quails were divided into 3 groups as hyperthyroid, hypothyroid and the control group, with 15 quails in each. A bifunctional gene xylanase of B. subtilis RSKK243 encoding detectable beta(1,4) glucanase (CMCase) activity in addition to a high level of main xylanase activity was cloned and expressed in Escherichia coli with pUC18 plasmid and in various Bacillus subtilis strains with pUB110 vector. For enzymatic analysis and plasmid isolation, the gene was also expressed in xylanase and beta glucanase negative B.subtilis YB886 strain with pUB110 vector. In the survey study, a total of 19 canine mast cell tumors were examined with regard to sex, age, breed of dogs, tumor location, and pathological findings in the period of 1960-1999. The majority of tumors occurred in males (78%) and between the ages of 6 and 11 years (67%). The pointer was predisposed to development of tumors (35%). The hindquarters and hind legs were most commonly involved, followed by the extragenital inguinal area and trunk. The tumors clearly had a site preference for the posterior region of the body (79%). The study was carried out to investigate the effects of sunflower meal with an enzyme mixture containing \alpha-amylase, ß-glucanase, cellulase, lipase and protease substituted for soybean meal in broiler diets on performance, intestinal weight, viscosity and pH, as well as the frequency of sticky droppings and abdominal fat. Survey Of European Foulbrood Disease In Bee Farms Located In Elazığ And The Surrounding Provinces, Hali̇l Şi̇mşek, Canan Özcan

Turkish Journal of Veterinary & Animal Sciences

A total of 250 specimens from 110 bee farms located in Elazığ and the surrounding provinces were taken in 1999 and 2000. The specimens were examined for European Foulbrood Disease. Under macroscopic examination clinical symptoms were noted in 12 specimens, and Paenibacillus alvei was isolated and identified from 8 of these specimens. As a result of the antibiotic sensitivity test, the isolate was found sensitive to enrofloxacin, streptomycin and erythromycin 75%, 75%, and 62,5% of the time respectively. In this study, the presence of European Foulbrood Disease was determined in 3.2% of cases in Elazığ and the surrounding provinces.

The objectives of this study were to determine the effects of copper sulphate administration on body weight gain and some biochemical, haematological parameters in lambs which had serum copper levels at subclinical values. This study was performed on 30 Ivesi male lambs (control group: 12, experimental group: 18), aged with 15-20 days. A total of 473 sera from hens of different age groups and different breeds and a total of 430 eggs were collected from 11 breeders and egg producers in the districts of Adapazarı, Afyon, Ankara and Konya. Lipopolysaccharide (LPS), heat-extract (HE) and flagella antigens were tested in the standardizing ELISA technique. Flagella antigen was found useful in the screening of field samples. Of the 48 (10.2%) rapid plate agglutination positive sera, 7 were negative in ELISA. Exposure of buffalo liver and kidney tissue arginases to a 150 w light source from a distance of 8-10 cm, in the presence of 0.05mM methylene blue, caused significant inhibition of arginase activity. It was determined that this inhibition occurred differently in liver and kidney tissues. Arginase activity was spectrophotometrically measured using the Thiosemicarbazide diacetyl-monoxime urea (TDMU) method. Two studies were conducted to evaluate if Fertirelin acetate (an analogue of GnRH) and Gentamycin sulphate would improve pregnancy rate in healthy cows or in nonpregnant cows although they were inseminated three times or more. This investigation was carried out to demonstrate some production traits and egg quality characteristics of Japanese quails (Coturnix coturnix japonica). One hundred female quails were used in the study. The experiment was carried out for six months. Egg production, egg weight, feed consumption and some egg quality characteristics were taken into consideration. Egg quality characteristics were measured at 8-24 weeks of age. The study was conducted at Alata Horticultural Research institute between 1994 and 1996. The aim of the study was to obtain a strong colony by way of supersedure and empty frames, feeding with saccharose syrup and encouraging egg laying of queen bees at the end of summer. According to the results, there were significant differences among treatments in terms of the development of colony population, brood rearing activity and honey yield. Treatments had no significant effect on wax building activity and swarming tendency.

This investigation was carried out to determine the effect of amitraz poisoning on the the formation of methemoglobin and osmotic fragility of erytrocytes. in this study, 12 guinea-pigs, weighing 250-300 gr were used. They were equally divided into two groups, control and experimental. The animals in the experimental group were intraperitoneally injected with amitraz at a dose of 1 mg/kg body weight. One hour after amitraz injections, erytrocyte, hematocrit, hemoglobin levels and also percent hemolysis of erytrocytes were measured in the blood samples taken from the control and experimental groups. Trichloroethylene (TE) is an environmental toxic solvent hazardous to human and domestic animals and well known in the industrial sector. The purpose of this study was to determine whether oral TE plays a role in lipid oxidation and tissue damage. Fourteen dogs were treated with an oral toxic dose of 0.5 ml/kg T.E. The acute changes that occurred in creatine kimase (CK), malondialdehyte (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were investigated in blood samples taken before application and 8, 24, 48, 72 and 168 hours after treatment. Acid glycine extract (AGE) and whole cell (WC) antibodies wer e used in ELISA testing for the detection of Campylobacter fetus subsp. venerealis and subsp. fetus antibodies in the blood sera of 150 female cattle and 100 bulls which wer e one year old or older . This study was carried out to investigate the prevalance of leptospirosis in the urine of cattle slaughtered in three major abattoirs in the east of Turkey. A polymerase chain reaction (PCR) based on a pair of genus-specific primers was used to detect leptospiral DNA in the urine samples of 473 cattle, 284 of which were from Elazig, 112 from Malatya and 77 from Diyarbakir. The detection limit of the method was determined to be approximately five bacteria per ml of urine. Development Of An Enzyme Immunoassay For The Determination Of Ovine Growth Hormone In Plasma, Behi̇ç Serpek, Seyfullah Hali̇loğlu

A competitive enzyme immunoassay for the determination of ovine growth hormone (oGH) was developed. The assay is based on (anti-rabbit IgG) goat IgG-coated microtitreplates, anti-oGH from rabbits, biotinyl-oGH and streptavidin-horseradish peroxidase. The assay is done directly with 40 ml of plasma and the calibration graph (90% relative binding at 0.38 ± 0.02 ng ml -1 and 50% relative binding at 3.80 ± 0.18 ng ml -1 ) is prepared in plasma without measurable endogenous oGH. Recovery in different plasma samples with added oGH amounted to 96.78 - 100.03 %, and all variabilities were < 10.44. The assay shows no significant binding of oFSH, oLH, oTSH and oPRL. The assay detects high and low plasma oGH levels within the physiological variation as well as changes in plasma oGH after stimulation with growth hormone releasing factor. Hence, it offers a reliable alternative radioimmunoassay. However, a reference method providing evidence of the identity of exogenous oGH will be required for proving illegal treatment with oGH and for forensic purposes.

One hundred pneumonic lungs from calves were examined by an immunoper oxidase technique using avidin-biotin- per oxidase complex method in formalin-fixed, paraffin-embedded sections for Haemophilus somnus. In the histological examination of tissue sections, strong positive reactions against H. somnus were detected in 14% of the lungs. H. somnus antigens were observed at the surface and/or within the epithelial cells, macrophages, necrotic alveolar walls, alveolar -bronchial/br onchiolar exudates, fibrins, distended interlobular septa and perivascular areas. In addition, dense antigen accumulations were detected in the necrotic macrophages and neutrophils.

This research was carried out on mirror carps ( Cyprinus carpio, L.,1758) taken from Seyhan Dam Lake and Seyhan River between January and October 1996 to determine some blood parameters. Erythrocyte, leucocyte count, leucocyte cell formulas, total serum protein and serum glucose values of mirror Carps ( Cyprinus carpio, L.,1758 ) caught from Seyhan Dam Lake and Seyhan River were also compared statistically. Different values were recorded in erythrocyte and serum protein amounts for both environments (P0.05).
