DNA isolations from the whole blood are commonly performed to obtain DNA for molecular research and diagnostics. Generally, blood samples are taken into anticoagulant tubes and stored in deep freezers until DNA isolation. In fresh blood, pretreatments or leukocytes preparations can be performed and suggested for advanced DNA isolation. However, similar applications in freeze-thawed blood (FTB) have not been shown yet. The aim of this study is to estimate lactation milk yield of Holstein cattle using first calving age, lactation period, and service period with (ANFIS) adaptive neuro-fuzzy inference system. The input variables for the system in the study were first calving age, lactation period, and service period. The output variable from the system was lactation milk yield. Predicted values obtained from the ANFIS were compared with the observed values. Ticks are well known as hematophagous ectoparasites, which cause different diseases in humans and animals. This investigation was designed in central Iran to assess the prevalence, intensity, and abundance of tick infestation in stray dogs and detection of Ehrlichia canis. Overall, 962 ticks were collected from 260 stray dogs between April and August 2018. DNA samples with the goal of amplification of pathogen were screened by real-time PCR assay. The prevalence of tick infestation on stray dogs was 60.3%. Rhipicephalus sanguineus sensu lato (s.l.) was only species found in stray dogs. Out of 42 tick pools, 4 (9.5%) were positive …

Mastitis is an inflammation of the mammary gland whose diagnosis is based on clinical criteria and the determination of inflammatory markers. The disease is mainly caused by bacteria causing intra-mammary infections. The etiological agent/s are detected by milk culture or by molecular biology approaches. However, these methodologies are unpractical for on-farm diagnosis, where rapid decisions must be made when infected animals are detected. For this reason, the levels of glycosaminoglycans (GAGs) in milk were evaluated as a tool for the diagnosis of mastitis. But, till now, comparative assessment of the effects on the improvement of geese economic traits with these strategies is largely unknown. Two fat sources, soybean oil (SO) and refined poultry oil (RPO), and 4 emulsifiers (control, lysolecithin, lysophospholipid, and bile acids) were used in this experiment. Thirteen hundred and sixty (1360) birds (ROSS - 308) were divided into 8 groups having 5 replicates, 34 birds in each. Energy efficiency, feed conversion (FCR), and protein efficiency ratios were improved in birds receiving SO than RPO diets. Weight gain, FCR, feed intake, and dressing percentage were increased in broiler birds receiving bile acid than those given lyso-phospholipid and lysolecithin (p < 0.05). Cooking loss was greater (p < 0.05) in broiler birds given RPO than those fed SO. Birds receiving bile acid had greater (p < 0.05) water holding capacity of chest meat than other fat emulsifiers. Bile acids in broiler diet presented higher (p < 0.05) digestibility of crude protein and ether extract as compared to other treatments. Bile acid supplemented diets had a minimum (p < 0.05) cost of production per kg live weight than other fat emulsifiers. Therefore, the use of bile acids with RPO diets can increase nutrient digestibility, growth performance, meat quality, and economic efficiency in broiler production systems.

Some Hematological Parameters And Enzyme Histochemistry Of Peripheral Bloodleukocytes In Kivircik Sheep, Mehmet Faruk Aydin, İhsan Kisadere

The aim of the present study was to determine some hematological parameters (RBC, WBC, HCT, HGB, MCH, MCHC, MCV, and PLT), the presence and percentages of ANAE and AcP-ase positivities in leukocytes and the differential leukocyte count in Kivircik sheep. Blood samples were taken from the jugular vein of 40 (rams = 20; ewes= 20) healthy, 1-year-old sheep. MCH and MCHC values were found statistically higher in rams than ewes (p < 0.05). Mean percentages of ANAE-positive PBL were defined as 74.16 ± 11.94% in ewes and 70.33 ± 8.39% in rams. It was not determined any significant changes according to sex. On the other hand, the percentages of ACP were determined as 67.12 ± 6.54% in ewes and 66.71 ± 6.58% in rams. It was not found any significant differences according to sex. For ANAE enzyme activity, lymphocytes, monocytes, neutrophils, and eosinophilic granulocytes showed a positive reaction. For ACP enzyme activity, although lymphocytes presented positivity, monocytes, neutrophils, eosinophils were negative. The ratio of lymphocyte, monocyte, neutrophile, eosinophile, and basophile of Kivircik ewes and rams were found as 64.35 ± 5.74%, 60.64 ± 8.85%; 3.41 ± 1.27%, 3.82 ± 1.42%; 27.58 ± 5.46%, 30.94 ± 8.71%; 2.41 ± 0.93%, 2.88 ± 1.45%; 1.76 ± 0.75%, 1.29 ± 0.58%, respectively. The difference between mean values was not found significant according to sex (p > 0.05). The present study evaluated the effect of selection for higher body weight (weight based selection = WBS) and egg numbers (egg based selection = EBS) on productivity, egg quality, and hatching traits of Japanese quail for three generations. From a base population of 1125 day-old chicks of Japanese quail, best performing families were allowed to propagate for getting next generation. WBS in G3 presented higher values of feed intake, egg weight, hatching weight. However, egg production and feed conversion ratio were better in EBS during G3. Studies involving molecular genetic characterization of silkworms are quite limited. Therefore, with this study, pure and hybrid silkworm lines raised in Turkey was characterized for the first time by microsatellite DNA markers. Samples were collected from 7 different silkworm lines, of which the pure lines were obtained from the Provincial Directorate of Agriculture and Forestry in Bursa and the hybrid ones from Silkworm Cocoon Association of Turkey (Kozabirlik). This study was carried out to investigate the species identification and enumeration of some pathogenic microorganisms before and after feeding the full ration contaminated by predatory starling bird herds on 5 different days in the dairy cattle farm in Aksaray. The aim of this study was to investigate the distribution and density of IgG, vimentin, and CD45 in mouse placenta on the fourth, tenth, and seventeenth days of pregnancy. Strept avidin-biotin-peroxidase complex (streptABC) staining method was used. On the fourth day of pregnancy, the expression of IgG, vimentin, and CD45 was more intense around the uterine glands. On the tenth day, IgG, vimentin and CD45 positive cells were demonstrated in decidua basalis and mesometrial parietal trophoblast giant cells (P-TGCs) and mesometrial lymphocyte groups (MLAp) portions. In this study, monensin (MON) was investigated for its apoptotic and genotoxic effects on chick liver tissue and cytotoxic and apoptotic effects on chick primary hepatocytes isolated from perfused liver tissue. The aim of the study was to assess the ability of horses to learn how to open feeder boxes in a direction associated with the time of day. The study involved 10 horses. The first stage of the study consisted in learning how to complete the task. The second stage involved the test and was followed by a control stage. Malassezia pachydermatis is an opportunistic yeast found in the ear canal of small animals; however, the current azole-based therapy applied to it has failed to achieve clinical success due to the antifungal resistance. This issue has encouraged the studies in natural products, such as Origanum vulgare (oregano), Origanum majorana (marjoram) and Rosmarinus officinalis (rosemary) essential oils, although their mechanism of action remains unclear. Malassezia pachydermatis specimens deriving from otitis cases in dogs (n = 22) and cats (n = 2) were subjected to CLSI M27-A3.
