Altered Na, K- Atpase Isoform Expression In Artemia Franciscana In Response To Hypersaline Environments, Jessica Drenth

Theses and Dissertations

The Na,K-ATPase (NKA) is an essential membrane pump that helps to establish cell ion gradients, and regulate intracellular salt in many organisms. One such species, Artemia franciscana (brine shrimp), extreme halophiles which live in hypersaline environments, expresses 2 distinct α-catalytic subunits of the NKA. One of these subunits, α2-(KK), has two key lysine substitutions located within the cation binding sites. Prior work has demonstrated this specific subunit may be directly involved in brine shrimp adaptation to their extreme environments. However, the precise molecular and physiological effects of α2-(KK) have not been entirely elucidated. I determined through immunohistochemistry that my initial …

Theses and Dissertations

Soybean (Glycine max) yield is significantly affected by soybean cyst nematode (SCN), Heterodera glycines, and causes an annual loss of billions of US dollars. In this study, Glycine max xyloglucan endotransglycosylase/hydrolase gene (Gm-XTH52) was transformed into a nematode susceptible G. max [Williams 82/PI 518671] variety of soybean to test whether the protein expression has a role in resistance to H. glycines, and possible chemical changes the expression may cause in the plant composition. Expression level of the Gm-XTH52 gene was three times higher than in controls. Significant reduction in the number of SCN cysts suggested suppression of H. glycines parasitism …

Biopolitics In J.M. Coetzee's Disgrace And Waiting For The Barbarians: Archives Of Bodily Movements In Modernity, Gregory Marcus May 2017

Biopolitics In J.M. Coetzee's Disgrace And Waiting For The Barbarians: Archives Of Bodily Movements In Modernity, Gregory Marcus

Theses and Dissertations

By examining the allegorical ways which bodies are produced and their movement controlled within J.M. Coetzee’s works, Waiting for the Barbarians and Disgrace, this thesis follows the shifting paradigm of power from apartheid’s presumption of unity of the state to the rainbow nation’s constitutional declaration that its citizens are “unified in [their] diversity” and equal under the law. In my chapter on Waiting for the Barbarians, I argue that the state creates a fiction of unity which allows the state to invoke claims of emergency and suspend the law; within this suspension, an economy of sacrifice functions to cleanse the …

Deprotonation As A Unified Pathway For Organothiol Binding To Citrate- And Borohydride Reduced Gold Nanoparticles, Sandamini Heshani Alahakoon Ulpanhewa Vidanalage May 2017

Deprotonation As A Unified Pathway For Organothiol Binding To Citrate- And Borohydride Reduced Gold Nanoparticles, Sandamini Heshani Alahakoon Ulpanhewa Vidanalage

Theses and Dissertations

The mechanism of organothiol (OT) binding to gold has remained controversial for decades. There are three mechanisms proposed for OT binding to gold surfaces. The first is the radical pathway in which the sulfur-bound hydrogen atoms (RS-H) are released as hydrogen atoms which eventually converted into hydrogen gas. Second is the deprotonation pathway in which the sulfur-bound hydrogen atoms leave as protons. Third is direct adsorption in which the RS-H bonds remain intact on the gold surface. This study demonstrates a combined pH and surface enhanced Raman spectroscopic study of organothiol binding to citrate- and borohydride-reduced gold nanoparticles (AuNPs) in …

The objective of this dissertation was to explore the interaction of canine platelets and tumor cells in vitro. The main hypothesis was that canine platelets were similar to human and murine platelets, and would interact with tumor cells, resulting …

Effect Of Sub-Lethal Chlorine Stress On The Homologous Stress Adaptation, Antibiotic Resistance, And Biofilm Forming Ability Of Salmonella Enterica, Tomilola O. Obe May 2017

Effect Of Sub-Lethal Chlorine Stress On The Homologous Stress Adaptation, Antibiotic Resistance, And Biofilm Forming Ability Of Salmonella Enterica, Tomilola O. Obe

Theses and Dissertations

The effect of exposure to sub-lethal chlorine stress on Salmonella enterica serotypes Typhimurium and Heidelberg was examined in this study. Both serotypes demonstrated an acquired tolerance to chlorine with the adapted cells growing in concentrations above the MIC. Chlorine induced a morphological change to the rugose variant in Salmonella. The biofilm formation of the adapted and control cells was tested on food-contact surfaces at room temperature and 37°C. The chlorineapted rugose formed stronger biofilms (P < 0.05) when compared to smooth (adapted and control) on both surfaces tested and at both temperatures. The possibility of crossaptation to antibiotics and low pH was evaluated. Adapted rugose showed reduced susceptibility against some of the antibiotics tested. Chlorine does not aid in the survival of Salmonella enterica at low pH. Chlorine stress can select for tolerant Salmonella cells that attach strongly to food-contact surfaces and after some time may become less susceptible to antimicrobials.

Effects Of Dietary Plasticity And Landscape Heterogeneity On Brown Bears, Lindsey Stutzman May 2017

Effects Of Dietary Plasticity And Landscape Heterogeneity On Brown Bears, Lindsey Stutzman

Theses and Dissertations

The brown bear (Ursus arctos) is a generalist omnivore that occupies diverse habitats and displays high levels of behavioral plasticity. Plasticity in foraging behavior can result in variable diets across a population, and allow individuals to use a greater variety of resources. The distribution of food in space and time may also be an important factor in determining brown bear resource selection as it can greatly influence foraging efficiency. Our objectives were to determine if seasonal brown bear home ranges were influenced by temporal availability and spatial distribution of resources and if deviation from an optimal diet influenced brown bear …
