In immune-competent individuals, tumor growth occurs due to a failure of the immune system to recognize and destroy malignant cells. Immune surveillance can be curtailed by the presence of immunosuppressive cells such as myeloid-derived suppressor cells (MDSCs) and T-regulatory cells (Tregs) which both serve to dampen antitumor immunity. These studies utilize a metronomic low-dose therapeutic approach to examine the effects of anticancer drugs against murine breast cancer and the effects on the immune cell compartments. Postpartum mental health is a public health priority, particularly for mothers with infants in the Neonatal Intensive Care Unit (NICU), who may have experienced a precipitous birth, followed by a traumatic NICU hospitalization. Autosomal dominant polycystic kidney disease (ADPKD) is one of the most common heritable diseases in the world, characterized by the development of large fluid filled cyst. Treatments for this disease are limited due to a lack of understanding of disease pathogenesis. The increasing use of quantitative real-time polymerase chain reaction (qPCR) as a method for quantifying gene expression has led to an increased demand for standardization of data analysis methods to ensure accurate reporting and robust, reproducible results. The exponential nature of qPCR amplification results in the potential magnification of what are usually very small sources of error. Relative gene expression calculations circumvent this issue by normalizing target gene expression data to within-sample expression of a previously validated, stably expressed reference gene or genes. Humans undergo two developmental switches in the predominantly expressed β-like globin chain during embryogenesis and fetal development. The first switch from embryonic (ε) to fetal (γ) occurs around week 5 of embryonic development, while the second switch from fetal to adult (β) globin occurs shortly after birth. By adulthood, fetal hemoglobin represents only 1-2% of total hemoglobin in the blood. As sufficiently elevated levels of fetal hemoglobin are beneficial for improving clinical outcomes in sickle cell disease and β-thalassemia, the mechanisms that enforce silencing of fetal hemoglobin expression postnatally are of great clinical significance. The research of this thesis investigated the mechanism of action for the therapeutic compound CLP290, which our laboratory has previously demonstrated to reverse degradation of the protein potassium-chloride co-transporter 2 (KCC2) and restore neuronal function following modeled traumatic brain injury (TBI) in the rat. The loss of membranous KCC2 from the plasma membrane after TBI worsens the excitation/inhibition imbalance common in neurotrauma. The Dimerization Partner, Rb-like, E2F, And MuvB (DREAM) complex was identified in humans due to homology of the genes involved in the D. melanogaster dREAM complex. It assembles during cellular arrest, or G0, to function as a transcriptional repressor of cell cycle genes. It is known that phosphorylation of LIN52 at serine 28 is necessary for the critical DREAM forming interaction between LIN52 and the pocket protein p130. Our laboratory has previously shown that gene editing of LIN52 to replace serine 28 with alanine (S28A) prevents phosphorylation by the kinase DYRK1A and inhibits DREAM formation. Rivers play a critical role in global carbon (C) budgets despite their comparatively small surface area. A significant portion of the terrestrial C that they receive is transformed, re-mineralized, or stored during transit to the ocean. Radiocarbon (∆14C) data show that a fraction of riverine organic C (OC) has been pre-aged in the terrestrial environment. Lateral export of carbon from these aged pools may contribute to atmospheric carbon dioxide emissions through microbial and photochemical oxidation. Purpose: Compromised tooth structure on permanent and deciduous teeth in the pediatric population is increasingly being restored with tooth-colored prefabricated zirconia crowns. These restorations may need to be removed or replaced with permanent crowns. The purpose of this in vitro study was to explore the use of an Er,Cr:YSGG laser for removal of prefabricated zirconia crowns cemented with RMGI as a non-invasive alternative to rotary instruments.

Work Hope And Work Volition: Exploring The Influence Of Community College Students’ Rurality And Socioeconomic Status, Jesse A. Wingate

Data from a sample of community college students (N = 478) in Virginia were used to examine relations among rurality, socioeconomic status, work hope, and work volition. Socioeconomic status, work hope, and work volition were positively associated. However, rurality, measured both as a continuous and categorical variable, was not correlated with work hope or work volition. Additional analyses showed convergence between measures of work hope and work volition confirming construct similarity. Results, limitations, implications, and recommendations for future study are included.

The purpose of this research is to establish a correlation between the integration of Social Cognitive Neuroscience (SCN) frameworks (e.g., The SCARF model) into day-to-day social interactions in the workplace. The study also sought to determine whether SCN practices can increase prosocial behaviors and improve a leader's influence. This qualitative study had four phases: an educational video, a self-assessment, a learning workshop, and an experiential learning exercise. Data were collected through eight interviews. This qualitative study explored female leaders' experiences with gender norms, implicit bias and microaggressions that they have experienced over the course of their careers. Research questions explored what gender norms exist, how they show up behaviorally in the workplace, and how gender norms, implicit bias and microaggressions impact women. 12 women participated in the study and were asked 12 interview questions. Participants' answers to these questions were coded to highlight themes. Monitoring land use and land cover (LULC) change and its effect on habitat availability and connectivity is crucial for species conservation and management. This is most efficiently accomplished at a sufficiently broad scale using remote sensing technology. Current approaches benefit from freely available data sources such as National Land Cover Database (NLCD), derived from 30 m Landsat imagery. However, temporal frequency and spatial resolution of NLCD may be insufficient for some applications. Breeding mountain lion (Puma concolor) populations currently inhabit the western portion of North Dakota. In recent years, apparent increases in human-lion interactions across North Dakota, and other Midwest states, have challenged wildlife managers who strive to balance biological and social carrying capacities of these animals. In 2019, we surveyed 2,000 residents of North Dakota, USA, using a self-administered mail survey to discern knowledge of and attitudes towards mountain lions, and to explore whether the presence of these animals would influence decisions to recreate in western North Dakota. Sensorimotor gating impairments are observed across a range of neuropsychiatric conditions. The prepulse inhibition of the acoustic startle response (PPI) is a validated measure of sensorimotor gating. Genetic and pharmacological manipulations in rodents have shown PPI is regulated by specific brain monoaminergic systems. Using genetically heterogeneous NIH-HS rats, we stratified individuals by %PPI. In low PPI animals, we observed elevated mRNA levels of certain neurotransmitter receptors, including metabotropic glutamate receptor Grm2, dopamine receptors Drd1 and Drd2, serotonin receptors Htr1a and Htr2a, and scaffolding protein Homer1, in the frontal cortex (FC) and striatum (STR).
