The Role Pde11a4 Signaling And Compartmentalization In Social Behavior, Kaitlyn Pilarzyk Oct 2021

The Role Pde11a4 Signaling And Compartmentalization In Social Behavior, Kaitlyn Pilarzyk

Theses and Dissertations

People tend to be social by nature. Being socially connected not only helps people live longer and healthier lives, but being an engaged and contributing member of society strengthens our communities overall. Maintaining intact social behaviors is key to maintaining this wellbeing, with disruptions negatively affecting both mental and physical health. Indeed, social isolation and feelings of loneliness can significantly increase a person’s risk of premature death, heart disease, and stroke as well as depression, anxiety, suicide, and dementia. Maintaining the ability to create and store social memory with age is, therefore, key to maintaining proper social behaviors. Unfortunately, individuals …

Impact Of Acetylcholine On Internal Pathways To Basal Amygdala Pyramidal Neurons, Tyler Daniel Anderson-Sieg Oct 2021

Impact Of Acetylcholine On Internal Pathways To Basal Amygdala Pyramidal Neurons, Tyler Daniel Anderson-Sieg

Theses and Dissertations

The basolateral nuclear complex of the amygdala (BNC) – consisting of the lateral (LA), basolateral (BL), and basomedial nuclei (BM) – detects salient environmental stimuli (cue-detection) and motivates appropriate behavioral responses to their implied meaning (cue-guided behavior) via a precise pattern of internal circuitry. Glutamatergic signals representing environmental stimuli enter the LA and split into parallel streams that activate pyramidal neurons (PNs) in the anterior and posterior BL (BLa and BLp), which putatively mediate negative and positive emotions, respectively. The BL is the most densely innervated target of the cholinergic basal forebrain (CBF) and ACh transients (phasic ACh) in cortex …

Impact Of Acetylcholine On Amygdala Network Oscillations, Joshua Xavier Bratsch-Prince Oct 2021

Impact Of Acetylcholine On Amygdala Network Oscillations, Joshua Xavier Bratsch-Prince

Theses and Dissertations

The ability to appropriately respond to and store memories of emotionally salient events is a critical feature of survival. The basolateral amygdala (BLA) is a temporal lobe structure that is essential in mediating emotional behaviors, learning, and memory. Important to circuit function are synchronized neuronal oscillations. Different oscillatory signatures are implicated in mediating different aspects of behavior and learning and memory processes: theta frequency (3-12 Hz) oscillations represent “online” states and are important for information encoding and learning, and sharp wave ripples (SWRs) represent more “offline” states that are important for the consolidation of memory. In the BLA, both of …

Investigating The Chemistry Of Harmful Algal Blooms And Microbial Communities In The Natural Environment, Samuel Patrick Putnam Oct 2021

Investigating The Chemistry Of Harmful Algal Blooms And Microbial Communities In The Natural Environment, Samuel Patrick Putnam

Theses and Dissertations

Cyanobacteria has existed on the planet for over 3 billion years, and as technology, science, and industry have allowed us to shape our world, these changes have caused a proliferation of biomass. Higher carbon dioxide levels in the atmosphere, warmer temperatures, and rapid increases in the nutrient cycle have created excellent growing conditions for cyanobacteria, making them endemic in fresh waters throughout the world. The work in this dissertation was directed towards the secondary metabolites produced by cyanobacteria and other surrounding microbes and understanding their concentrations and release in natural systems. Analytical methods were developed for the extraction and quantification …

Advancing Knowledge Of Exercise As A Therapeutic Management Strategy For Women With Polycystic Ovary Syndrome, Pamela J. Wright Oct 2021

Advancing Knowledge Of Exercise As A Therapeutic Management Strategy For Women With Polycystic Ovary Syndrome, Pamela J. Wright

Theses and Dissertations

Polycystic ovary syndrome (PCOS) is the most common endocrinopathy among premenopausal women with a prevalence that ranges 15-21%. The estimated financial burden in the United States for evaluating and treating premenopausal women with PCOS was over $8 billion in 2020. PCOS etiology is complex and poorly understood, as is the optimal treatment and management. Endocrine Society Clinical Practice Guidelines recommend exercise as first-line treatment. Yet, the optimal exercise type and “dosing” are not defined. As last reported for women with PCOS, less than 60% are regularly physically active and more than 25% are sedentary. Additionally, little to no published data …

“Before The World Gets Them”: The Impact Of Racialized Parenting On Black Mothers, Mia Brantley Oct 2021

“Before The World Gets Them”: The Impact Of Racialized Parenting On Black Mothers, Mia Brantley

Theses and Dissertations

This dissertation explores the distinct practices Black women implement to protect their children from both actualized and anticipatory experiences of racism, as well as its effects on their mothering experiences, health and well-being, as well as how they manage the emotional and mental toll of their children’s experiences. Race plays an integral role in shaping mothering practices. More specifically, motherwork examines how Black mothers ensure the physical, mental, and emotional survival of their children in the face of micro-and macro-level structures that perpetuate racism and inequality. However, much is left to explore regarding the interconnectedness between Black women’s motherwork, linked …

Real Time Neurochemical Analysis Of The Brain For Pharmacological Treatments In Mood Disorders And Neurodegeneration, Anna Marie Buchanan Oct 2021

Real Time Neurochemical Analysis Of The Brain For Pharmacological Treatments In Mood Disorders And Neurodegeneration, Anna Marie Buchanan

Theses and Dissertations

The monoamine, serotonin, is an important modulator in the central nervous system. It is believed to participate in a variety of functions ranging from moderating stress, promoting brain plasticity, and regulating sleep and appetite. Dysfunction of the serotonergic system has also been implicated in mood disorders and neurodegenerative diseases. While serotonin is of great interest to the research community, real-time in vivo serotonin dynamics remain understudied. This is because direct measurement of fast serotonergic transmission is analytically challenging. In 2009, the Hashemi lab pioneered the direct measurement of endogenously evoked serotonin using a method called fast scan cyclic voltammetry (FSCV). …

Daphnia Pulex: The Mixed Messages Of Mutations , Matthew Randall Bruner Oct 2021

Daphnia Pulex: The Mixed Messages Of Mutations , Matthew Randall Bruner

Theses and Dissertations

While mutations are almost universally considered to be more often deleterious than beneficial, their precise interactions between different populations and individual lines have been largely overlooked. Using mutation accumulation lines of four clones of obligately asexual Daphnia pulex, this research is intended to investigate the degree to which spontaneous mutation would affect fitness-related traits after roughly 100 generations. The expectation was that there would be a visible decrease in juvenile specific growth rate, the surrogate measurement used for fitness, across all four clones due to the deleterious nature of mutation in a selection free environment. Through measuring birth mass in …

Helping High School Students Succeed Academically By Meeting Their More Basic Needs, Karen Eyler Martin Oct 2021

Helping High School Students Succeed Academically By Meeting Their More Basic Needs, Karen Eyler Martin

Theses and Dissertations

In this action research, I seek to show the connection between breakfast consumption and student academic achievement. Students skip breakfast for a variety of reasons, including not allowing enough time and not having enough resources for it. Others eat what they can obtain, including snack foods that are highly processed. In addition, school districts have gotten away from making their meals from scratch, and instead, use a third-party catering service that often serves food that is cheap and easy to heat up, which often means it is highly processed and does not have the same nutritional content as whole foods. …

Like many countries of the world, Ghana is experiencing a nutrition transition and rising non-communicable diseases. Adolescents are susceptible to diet-related health risks as they experience significant physical and psychological changes, which are happening in tandem with food environment changes, including widespread proliferation of large portion and package sizes of energy-dense, nutrient-poor foods. Both local and multinational food and beverage companies have encouraged consumption of their products through various marketing tactics targeted directly to adolescents. Some of these tactics include the use of characters or celebrity endorsement, promotional discounts, and appeal to sociocultural values, including messages about body size preferences. …

Fate And Transport Of The Microbiome In Built Environments, Dahae Seong Oct 2021

Fate And Transport Of The Microbiome In Built Environments, Dahae Seong

Theses and Dissertations

Buildings are complex ecosystems evolving continuously. Indoor components such as occupants, ventilation system, and building structures influence the microbiome. To shed light on how the microbial population in a building will respond to such a fluid system, the fundamental interactions between microbes and the indoor ecosystem must be understood. Buildings are being designed as air-tight structures to save energy; however, this could lead to degradation of indoor air quality through indoor sources of contaminants and/or containment of pollutants in the room or introduce outdoor pollutants indoors depending on the ventilation conditions. In addition, transmission of infectious microbes in indoors threaten …

A Novel Model To Study Adipose-Derived Stem Cell Differentiation, Austin N. Worden Oct 2021

A Novel Model To Study Adipose-Derived Stem Cell Differentiation, Austin N. Worden

Theses and Dissertations

The use of three-dimensional (3D) culture systems (hydrogels) and adipose-derived stem cells (ADSCs) in regenerative medicine to advance early-stage investigation and modeling of the mechanisms of diseases, treatments, targets, etc. has recently increased. ADSCs, specifically, are utilized due to their innate programming during embryogenesis and in adult tissues in addition to their ability to differentiate into mesodermal, endodermal, and ectodermal cell-specific lineages. Of importance is that these advancements do not involve a model specimen (i.e. mice or rats) and simulate the numerous conflicting signals a migrating cell is exposed to in vivo such as chemokines, extracellular matrix (ECM), growth factors, …

Multi-Fidelity Surrogate And Reduced-Order Model-Based Microfluidic Concentration Gradient Generator Design, Haizhou Yang

Theses and Dissertations

The microfluidic concentration gradient generator (μCGG) is an important device to generate and maintain concentration gradients (CGs) of biomolecules for understanding and controlling biological processes. However, determining the optimal operating parameters of μCGG is still a significant challenge, especially for complex CGs in cascaded networks. To tackle such a challenge, this study presents multi-fidelity surrogate and reduced-order model-based optimization methodologies for accurate and computationally efficient design of μCGGs.

The surrogate-based optimization (SBO) method is first proposed for the design optimization of μCGGs based on an efficient physics-based component model (PBCM). Various combinations of regression and correlation functions in Kriging and …

The Influence Of Adhd On Concussion In Ncaa College Athletes, Brett Steven Gunn Oct 2021

The Influence Of Adhd On Concussion In Ncaa College Athletes, Brett Steven Gunn

Theses and Dissertations

Evidence suggests attention deficit/hyperactivity disorder (ADHD) may be associated with an increased incidence of concussion; however, no study has cross-sectionally (Aim 1) and longitudinally (Aim 2) assessed the associations of ADHD with concussion. Additionally, there is reason to expect cognitive alterations following concussion in athletes with ADHD. Further, there is currently no study that has evaluated the influence of common medications often taken to mitigate some symptoms of ADHD, such as methylphenidate and amphetamines. Therefore, we sought to longitudinally assess cognitive profiles (Aim 3) of athletes with ADHD taking medication (ADHD+Rx) and not taking medication (ADHD_uRx) at baseline, 24-48 hours …

Let’S Be Friends: Examining Consumer Brand Relationships Through The Lens Of Brand Personality, Engagement, And Reciprocal Altruism, Daniel D. Haun Oct 2021

Let’S Be Friends: Examining Consumer Brand Relationships Through The Lens Of Brand Personality, Engagement, And Reciprocal Altruism, Daniel D. Haun

Theses and Dissertations

Brands have always exuded personality and status to consumers in order to promote a competent impression and induce positive perceptions and behaviors (Griskevicius, et al., 2007). In order for individuals to identify with brands, the brands must become more personable by consciously embodying a specific personality trait. Research has found that when a brand embodies a personality, there is an associated increase in consumer preference and usage (Sirgy, 1982), purchase choices for that brand (Ha & Janda, 2014; Gordon et al., 2016; Guèvremont & Grohmann, 2013; Swaminathan et al., 2009), and consumer preference and brand loyalty (Louis & Lombart, 2010; …

Utilization Of Remote Sensing To Analyze Aedes Mosquito Egg Abundance With Combined Weather Variables In Two Southeastern Coastal Cities, Danielle M. Johnson

Theses and Dissertations

Several vector-borne diseases have recently resurfaced, and many have suddenly spread to new areas in the United States (U.S.) due to climate change and tropicalization. Two vectors of public health importance are Aedes aegyptiand Aedes albopictus. The Southeastern region of the U.S. appears to be a preferential ecological niche for both Aedes species. Aedes aegyptiand Aedes albopictus are highly susceptible to environmental conditions, such as humidity, precipitation, temperature. Remote sensing technology has proven to be useful for estimating vector populations. Miami-Dade County is an established hotspot for disease transmission. Charleston County is newly vulnerable due to similar …

Robot Area Coverage Path Planning In Aquatic Environments, Nare Karapetyan Oct 2021

Robot Area Coverage Path Planning In Aquatic Environments, Nare Karapetyan

Theses and Dissertations

This thesis is motivated by real world problems faced in aquatic environments. It addresses the problem of area coverage path planning with robots - the problem of moving an end-effector of a robot over all available space while avoiding existing obstacles. The problem is considered first in a 2D space with a single robot for specific environmental monitoring operations, and then with multi-robot systems — a known NP-complete problem. Next we tackle the coverage problem in 3D space - a step towards underwater mapping of shipwrecks or monitoring of coral reefs.

The first part of this thesis leverages human expertise …

The concerns-based assessment model (originally proposed by Hall et al., 1973, and further developed by Hall & Hord, 1987; 2011) was used to investigate the degree to which language instructors adopted the innovation. An adapted form of the concerns-based assessment model’s levels of use focused interviews and instructional artifacts were used to measure the level of implementation of the task-based approach. Analysis of the data identified categoricalchanges as instructors implemented the new curriculum. The category of sharing revealed higher levels of use due …

Assessing Longitudinal Impacts Of Mentor Role On Student Outcomes, Jody Lanza-Gregory Oct 2021

Assessing Longitudinal Impacts Of Mentor Role On Student Outcomes, Jody Lanza-Gregory

Theses and Dissertations

Most youth form relationships with extra-familial adults, sometimes called natural mentoring relationships, and these connections appear to benefit youth in several ways. Previous research demonstrates that the presence of a mentor can positively impact student outcomes including educational expectation, educational attainment, and social success. However, little research has considered how the impact of a natural mentorship may differ based upon the role of the adult in the youth’s life. Using data from three waves of the National Longitudinal Study of Adolescent Health, this study investigates if the role of a mentor (i.e. teacher, coach, religious leader) influences the types of …

Satellite Observations And Spatiotemporal Assessment Of Salt Marsh /Dieback Along Coastal South Carolina (1990-2019), Huixuan Li Oct 2021

Satellite Observations And Spatiotemporal Assessment Of Salt Marsh /Dieback Along Coastal South Carolina (1990-2019), Huixuan Li

Theses and Dissertations

Coastal wetland mapping is often difficult because of the heterogeneous vegetation compositions and associated tidal effects. Past studies in the Gulf/Atlantic coast states have reported acute marsh dieback events in which marsh rapidly browned and thinned, leaving stubble of dead stems or mudflad with damaged ecosystem services. Reported marsh dieback in South Carolina (SC), USA, however, have been limited. Previous studies have suggested a suite of possibly abiotic and biotic attributes responsible for salt marsh dieback. However, there are no consensus answers in current literature explaining what led to marsh dieback in past decades, especially from the spatiotemporal perspective. In …

Variations In Er Stress And Unfolded Protein Response Predict Disease Susceptibility, Youwen Zhang Oct 2021

Variations In Er Stress And Unfolded Protein Response Predict Disease Susceptibility, Youwen Zhang

Theses and Dissertations

Numerous studies linked endoplasmic reticulum (ER) stress and unfolded protein response (UPR) to physiological responses to stress and various human pathologies, including metabolic disorders. Although variability in stress responses among individuals has been widely recognized, how such variations in the UPR affect susceptibility to diseases remains unknown. Using outbred deer mice as a model, we explored the consequences of the differential propensity to UPR at the transcriptomic, cellular, and organismic level.

Initially, we analyzed the correlation between several UPR-related genes in primary fibroblasts, at the whole transcriptome level, after ER stress induction. The analysis of correlated transcripts revealed that their …

The Development Of Novel Cdk8 And Cdk19 Inhibitors And Degraders As Potential Anti-Cancer Agents, Li Zhang Oct 2021

The Development Of Novel Cdk8 And Cdk19 Inhibitors And Degraders As Potential Anti-Cancer Agents, Li Zhang

Theses and Dissertations

CDK8 and its homolog CDK19 are essential for transcription regulation and their dysregulation has been identified in numerous diseases, especially in cancers. CDK8 and CDK19 have been implicated in cancer development through their kinase activity and kinase independent functions. Despite the existing CDK8/19 inhibitors, there is still a need to develop novel CDK8/19 inhibitors with improved potency and PK profile. It is also necessary to develop CDK8/19 degraders that are able to eliminate the kinase-independent functions. Although two CDK8-degrading PROteolysis Activating Chimeras (PROTACs) have been reported, they have limited CDK8 degradation efficacy and no effect on CDK19. PROTACs with improved …

Theses and Dissertations

Lower back pain effects up to 80% of people at some point in their life, a majority of cases being the result of degenerative disc disease. Treatment options for degenerative disc disease are limited, jumping from physical therapy to major spinal fusion and total disc replacement surgery with little to no approaches in between. Furthermore, surgical treatments have not shown to be more effective than conservative treatments and reducing pain and disability over the long term. Hydrogels have shown promise as a potential nucleus pulposus replacement device. Their properties are controllable and can be implanted into the body through minimally …

Behavioral Effects Of Novel Treatments For Pain And Alcohol Use Disorder, Christa Marie Donegan Sep 2021

Behavioral Effects Of Novel Treatments For Pain And Alcohol Use Disorder, Christa Marie Donegan

Theses and Dissertations

Opioid use disorder (OUD) and alcohol use disorder (AUD) are pressing public health problems in the United States that require new pharmacotherapies to be explored. Current FDA-approved treatment options for these two disorders are only moderately effective. Thus, there is a demand for the identification of new targets for drug development. Previous research initiatives have shown that when morphine is co-administered with the novel imidazodiazepine, MP-III-024, synergistic effects in models of analgesia and antinociception are produced. Our research efforts were concerned with understanding if morphine in combination with MP-III-024 produced synergistic effects in measures of undesirable pharmacological responses: opioid side …
