Cave systems and their unique biota are widely viewed as highly endangered, yet very little is known about basic life history, ecology, distributions, habitat requirements, and evolutionary relationships of subterranean species. The crux of the problem in cave studies is the assumption that traditionally defined morpho-species represent distinct evolutionary lineages. Convergence is exhibited in the morphologies of many animal groups, vertebrate and invertebrate, which leads to confusion in diagnosing species' boundaries, geographic distributions, gene flow routes, and imperilment. This dissertation research includes phylogeographic analyses of freshwater cave-dwelling crayfishes in the Southern Appalachians, a global hotspot of subterranean biodiversity. By examining …

The purpose of this study was to examine and describe fundamental literacy messages found within three major science reform documents: Science for all Americans: Project 2061 (AAAS, 1990), Benchmarks for Science Literacy (AAAS, 1993), and the National Science Education Standards (NRC, 1996). A qualitative content analysis was performed in an effort to reveal any messages or statements supporting fundamental science literacy. Results from this study indicate that key science reform documents do in fact contain multiple messages supporting the fundamental sense of science literacy, however, the nature of these messages, the quantity, placement and presence of negative literacy statements may …

This study examined the linkages between marital harmony and conflict and infants' emotional and physiological regulation abilities and developmental status at six- and nine-months of age. Participants included 93 first-time mothers and their infants (43 males, 50 females) from a Mountain West community. Mothers were asked to complete a battery of questionnaires, including a demographic measure and Braiker and Kelly's (1979) marital quality questionnaire. The revised Bayley Scales of Infant Development (BSID II) and Behavior Rating Scales (BRS) 2nd Edition were administered at six- and nine-months to measure infants' mental and motor development status and emotional regulation abilities. This thesis presents three separate manuscripts in chapter format dealing with the ecology of Chukars (Alectoris chukar) in western North America. All three manuscripts have been formatted for publication in professional journals. Chapter one confirms discovery of ingested lead pellets in Chukars across a broad region of western Utah including all four western counties sampled. Prevalence rates were 1.9% (n=105) for crops and 10.7% (n=75) of gizzards showing no evidence of penetration wounds. Ingestion is likely related to grit size preferences that are consistent with common shot sizes. The second chapter describes watering patterns and water-site selection of Chukars. Chukars …

Pka As An Upstream Kinase For Lkb1/Strad/Mo25, Seth Taylor Herway

The LKB1/STRAD/MO25 complex (LSMK) has been identified as the major upstream kinase for AMP-activated protein kinase (AMPK). PKA phosphorylates LKB1 at the Ser428 residue in humans and Ser431 residue in mice. We investigated PKA as an upstream kinase for LSMK. LKB1 that had been incubated with PKA prior to incubation with AMPK experienced up to a 51% increase in AMPK Kinase activity compared to LKB1 alone (p < 0.05). When blocked with a PKA Inhibitor, the kinase effect of PKA on LKB1 was eliminated. Rat epitrochlearis muscle tissue incubated with epinephrine experienced no increase in AMPK activity compared with controls indicating that epinephrine does not cause AMPK activity in this type of tissue. In conclusion, phosphorylation by PKA can increase the AMPKK activity of LKB1-STRAD-MO25 in vitro. Because LKB1 has been found to be constitutively active, it is postulated that phosphorylation by PKA may act to enhance LKB1-AMPK interaction and thus achieve its effect.

Skeletal muscle contraction results in the phosphorylation and activation of the AMP-activated protein kinase (AMPK) by an upstream kinase, AMPKK. The LKB1-STRAD-MO25 complex is the major AMPKK in skeletal muscle; however, LKB1-STRAD-MO25 activity is not increased by muscle contraction. This relationship suggests that phosphorylation of AMPK by LKB1-STRAD-MO25 during skeletal muscle contraction may be regulated by allosteric mechanisms. In this study we tested an array of metabolites including glucose-6-phosphate (G6P), fructose-6-phosphate (F6P), fructose 1,6-bisphosphate (F1,6-P2), 3-phosphoglycerate (3PG), glucose-1-phosphate (G1P), glucose-1,6-bisphosphate (G1,6-P2), adenosine diphosphate (ADP), carnitine (Carn), acetyl-carnitine (Acarn), inosine monophosphate (IMP), inosine, and ammonia for allosteric regulation. Phylogenetic analysis is a central tool in studies of comparative genomics. When a new region of DNA is isolated and sequenced, researchers are often forced to throw away months of computation on an existing phylogeny of homologous sequences in order to incorporate this new sequence. The previously constructed trees are often discarded, and the researcher begins the search again from scratch. The jumpstarting algorithm uses trees from the prior search as a starting point for a new phylogenetic search. This technique drastically decreases search time for large data sets. Carbon nanotubes are appealing materials for nanofabrication due to their unique properties and structures. However, for carbon nanotubes to be used in mass-fabricated devices, precise control of nanotube orientation and location on surfaces is critical. I have developed a technique to align single-walled carbon nanotubes (SWNTs) on surfaces from a droplet of nanotube suspension under gas flow. Fluid motion studies indicate that alignment is likely due to circulation of SWNTs in the droplet. My work provides a facile method for generating oriented nanotubes for nanodevice applications. I have also devised an approach for localizing SWNTs onto 1-pyrenemethylamine-decorated DNA on surfaces. …

Electric field gradient focusing (EFGF) utilizes a hydrodynamic flow and an electric field gradient to focus and concentrate charged analytes and order them in a separation channel according to electrophoretic mobility. Elution can be achieved by decreasing the applied voltage or increasing the hydrodynamic flow. EFGF has the advantages of concentrating a large volume (100 micro-L) of target proteins without significant band broadening and simultaneously removing unwanted components from the sample. It introduces a new perspective on different sources of errors to describe the impact of undermodeling. Our view is that modeling errors can be decomposed into two primary sources of errors: the approximation error and the estimation error. Lyconadin A is an alkaloid possessing a unique structure and antitumor activity. The total synthesis of Lyconadin A was proposed via an acyl radical cascade reaction. To investigate the possibility and stereoselectivity of the cascade cyclization, phenyl selenoester 16 was chosen as a model substrate to study the 7-exo-5-exo radical cyclization. A synthetic route to phenyl selenoester 16 was developed. The 7-exo-5-exo radical cyclization was found to occur with a high yield and excellent stereoselectivty. Proton channels are essential for many of the processes of life. The influenza A viral protein M2 is responsible for sensing the conditions necessary for viral RNA release. The proton-translocating FoF1 ATPase (ATP synthase) uses a proton gradient to drive adenosine triphosphate (ATP) synthesis. We have directly measured proton uptake in vesicles containing reconstituted M2 or FO by monitoring external pH after addition of valinomycin to vesicles with 100-fold diluted external [K+]. This proton flux assay was utilized to quantify proton flux through single M2 and Fo channels. Contrary to previous reports, proton uptake by M2 was not significantly altered …

The purposes of this correlational study were to (a) observe the learning styles of groups of kindergarten students and the teaching styles of their corresponding teachers; and (b) determine the effects that the matching of similar learning and teaching styles had on students' reading performances. It was hypothesized that students whose learning styles matched that of their teachers' teaching styles would do significantly better on their reading scores than those students whose learning styles did not match that of their teachers' teaching styles. The purpose of this thesis is to explore the use of genre writing as an alternative to commonly used expository writing in the seventh grade life science classroom. My research includes student surveys and educator interviews. I surveyed 44 seventh grade science students using a Likert scale. The participating students include 1 eleven-year-old boy, 10 twelve-year-old boys, 10 thirteen-year-old boys, 1 fourteen-year-old boy, 11 twelve-year-old girls, and 11 thirteen-year-old girls. A new geologic map of the Twin Rocks 7.5 minute quadrangle primarily located within Capitol Reef National Park, south-central Utah, provides stratigraphic and structural detail not previously available. This map has also been instrumental in understanding the evolution and development of fluvial terraces associated with Sulfur Creek and the structural geology of the backlimb of the Miners Mountain uplift. Nine bedrock stratigraphic formations and eight types of Quaternary deposits were mapped throughout the quadrangle. Bedrock stratigraphy ranges in age from Permian to Jurassic. Roughly 1 to 4% of the population engages in self-harm. Superficial self-harm is reported by more young women, than young men. Appropriate responses from family, friends, and other important individuals are a key ingredient in facilitating recovery. Non-therapists, such as family, friends, and school personnel often wish to assist young women who self-harm, but the problem is complex and they are often unsure of how to respond. Phosducin-like protein (PhLP) binds G-protein beta gamma subunits and is thought to assist in assembly of the G-protein beta gamma dimer. Phosphorylation of PhLP at serine residues 18-20 by the casein kinase 2 (CK2) appears to play an essential role in this process. PhLP has also been shown to interact with the chaperonin containing TCP-1 (CCT) atop its apical domain, not entering the substrate folding cavity. This study builds on previous research regarding parent-child religious conversations to explore the elements and bidirectional processes of parent-adolescent religious conversations. It employs qualitative analyses of interviews with highly religious parents and adolescents representing the major Abrahamic faiths (Christianity, Judaism, Islam) from New England and Northern California. Variations in content, structure, conversational processes, and bidirectional influence are summarized in a conceptual model. Findings suggest that the quality of conversations is greater for parents and adolescents when they are youth-centered than when they are parent-centered.

The dopamine transporter (DAT) is a transmembrane spanning protein responsible for the clearance of dopamine from the dopaminergic synapse. Uptake blockers such as cocaine inhibit transport resulting in elevated levels of dopamine in the synapse. DAT is predicted to contain twelve transmembrane spanning domains (TMs) connected by alternating intra- and extracellular loops, however, the spatial arrangement of these regions is unknown. There is an abundance of electrophysiological and pharmacological data describing Renshaw cells (RC), however comparatively little is known about their distribution, metabolism, ionic regulation, modulation or exact role in motor behaviors. Implicit family process rules refer to unspoken rules about family beliefs and expectations about communication, closeness, and organization and are an important dimension of family functioning that may have a powerful influence over adolescent psychological well being. This study focused on the relationship between implicit family process rules and adolescent psychological symptoms such as: Hostility, interpersonal sensitivity, depression, anxiety, and somatization. This study utilized paired tones at 1000 Hz with a 500 ms interpair interval and a 10 s interstimulus interval to assess sensory gating. Forty-two participants, ranging from 3-72 years of age were used to observe maturational changes in amplitude, latency, and suppression ratios of the P50 waveform. Previous research has shown that in normal adults the amplitude in response to the second of the paired tones is significantly suppressed compared to the amplitude in response to the first tone. The current study showed amplitude decreased with age to middle adulthood, where it increased slightly to later adulthood. Latencies decreased …

2-Bromoisobutyryl bromide was immobilized on poly(methyl methacrylate) (PMMA) substrates activated using an oxygen plasma. Atom-transfer radical polymerization was then performed to graft poly(ethylene glycol) (PEG) on the PMMA surface. PMMA micro capillary electrophoresis (µCE) devices made with the covalently modified surfaces exhibited substantially reduced electroosmotic flow and nonspecific adsorption of proteins. Both column efficiency and migration time reproducibility were one order of magnitude better with derivatized PMMA µCE devices compared to untreated versions. Fast, reproducible, and efficient separations of proteins and peptides were demonstrated using the PEG-grafted PMMA µCE chips. All analyses were completed in less than 60 seconds, and …

The practice of counseling and psychotherapy is influenced by a number of ideologies, models, and paradigms. Among these, the medical model's influence is particularly salient. The ideology of the medical field pervades the theory, research, and practice of psychology and its influence deserves close examination. The few studies in this area that have been conducted are descriptive and basic in nature. The present study aimed to contribute richness and depth to conversations about medical model influence. The diversity of microbial communities in wetlands has not been fully measured. These communities may offer tools to naturally remediate sites polluted with chlorinated compounds. Polmerase Chain Reaction (PCR) and Restriction Fragment Length Polymorphism (RFLP) are genomic techniques that are capable of rapidly characterizing bacteria communities and can provide valuable information about the diversity of microbial communities.

This research effort develops a program using MATLAB to solve the equations of motion for atmospheric reentry and analyzes the validity of the program for use as a tool to expeditiously predict reentry profiles. The reentry vehicle is modeled as a point mass with constant aerodynamic properties as defined by the user. The equations of motion for reentry are based on the two-body problem. The atmosphere is modeled as a single layer exponentially decreasing in density.
