Cognitive theory attributes social anxiety to a host of perceptual biases that transform harmless interpersonal stimuli into threats. Socially anxious college students were expected to misinterpret visual and auditory cues of emotion in others more readily than their more confident counterparts. This group of socially anxious men and women were also expected to perceive negative emotion from visual and auditory cues that were determined to be neutral or positive through an external validation process. Bacterial Persistence In Escherichia Coli: Examination Of The Role Of Hipba, Shaleen B. Korch

Persistence is the ability of a small fraction of bacteria (one-in-a-million) to survive antibiotic exposure, yet the survivors are as susceptible to antibiotic as the parental cells. "High persistence" in Escherichia coli is linked to mutations in hipA, the toxin entity of the toxin-antitoxin module hipBA. The wild type hipA gene encodes a "toxin" while hipB encodes a DNA-binding protein that autoregulates hip operon expression and binds to HipA to nullify its effects. The hipA7 allele confers persistence to 1:100 cells and serves as a model system to study persistence. Wild-type HipA, when in excess of HipB, causes culture growth …

Chronic portal hypertension is an end-state clinical condition associated with cirrhosis and other diseases that elevate portal vascular resistance. An intriguing vascular consequence of chronic portal hypertension is impaired vasoconstrictor function which favors a chronically vasodilated condition. This vasoconstrictor dysfunction has been linked to prolonged activation of protein kinase A (PKA). However the cellular mechanism(s) responsible for the altered vasoconstrictor function is still not clear. The present studies tested my hypothesis that PKA mediates impaired vasoconstrictor function by affecting the actin filament dynamics via two small heat shock proteins, HSP20 and HSP27. The analysis of proteins in biological fluids by capillary electrophoresis (CE) is of interest in clinical chemistry. However, due to low analyte concentrations and poor concentration limits of detection (CLOD), protein analysis by this technique is frequently challenging. Coupling preconcentration techniques with CE greatly improves the CLOD. An on-line preconcentration-CE method that can selectively preconcentrate any protein for which an antibody is available would be very useful for the analysis of low abundance proteins and would establish CE as a major tool in biomarker discovery. Dynamic pricing is a difficult problem for machine learning. The environment is noisy, dynamic and has a measurable cost associated with exploration that necessitates that learning be done in short-time horizons. These short-time horizons force the learning algorithms to make pricing decisions based on scarce data. In this work, various machine learning algorithms are compared in the context of dynamic pricing. These algorithms include the Kalman filter, artificial neural networks, particle swarm optimization and genetic algorithms. The retinal pigment epithelium (RPE) is a monolayer of cells located directly behind the photoreceptor cells in the retina. This dissertation presents the fabrication of hollow optical waveguides integrated on planar substrates. Similar in principle to Bragg waveguides and other photonic crystal waveguides, the antiresonant reflecting optical waveguide (ARROW) is used to guide light in hollow cores filled with liquids or gases. Waveguides with liquid or gas cores are an important new building block for integrated optical sensors. The fabrication method developed for hollow ARROW waveguides makes use of standard microfabrication processes and materials. Dielectric layers are deposited on a silicon wafer using plasma-enhanced chemical vapor deposition (PECVD) to form the bottom layers of the ARROW waveguide. A sacrificial …

By seeking to miniaturize and automate separation-based analysis, microfluidic research seeks to improve current methods through decreased cost, analysis time, and sources of contamination. The objective of this research study was to explore whether valid Multiple Intelligence profiles could be created for preschool children. This study was conducted over a three month period and included 16 preschool-aged participants. Research strategies included surveys (teacher, paraprofessional, and parent), naturalistic observations, and participant interviews. There were four key findings in this study. First, the teacher, the paraprofessional, and the parents had differing perceptions about which intelligences were dominant for each participant. Second, participants appear to have multiple, equally dominant intelligences. Cyclooxygenases (COXs) are of important therapeutic value as they are the target site of aspirin-like drugs. Here I report nine new COX-1 splice variants in chapter 1, which I characterized with regard to heme-binding and other properties. Inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) are co-inducible in many tissues following mitogenic and proinflammatory stimulation. In chapter 2, I investigate the physical and enzymatic properties of human COX-2 and iNOS and demonstrate that, despite reports to the contrary by another laboratory, they do not interact. This investigation provides a comprehensive analysis of entrained-flow biomass combustion processes. Experimental and theoretical investigations indicate how particle shape and size influence biomass combustion rates. Experimental samples include flake-like, cylinder-like, and equant (nearly spherical) shapes with similar particle masses and volumes but different surface areas. Samples of small (less than 500 µm) particles were passed through a laboratory entrained-flow reactor in a nitrogen/air atmosphere and a maximum reactor wall temperature of 1600 K, while large samples were reacted in suspension in a single particle furnace operated at similar conditions as the entrained-flow reactor. Background: Cartilage disorders affect millions of people in the United States alone, with effects ranging from poor skeletal development and joint pain to shortened lifespan and perinatal lethality. Many of these disorders have their root in defects of collagen, type XI collagen being among the most important. A mouse model of such a type XI collagen defect is the chondrodysplasia (cho) mutant. Mice homozygous for this null mutation in the Col11a1 gene do not express the α1 chain of type XI collagen. This results in a functional knockout of type XI collagen, leading to insufficient skeletal development and perinatal lethality. …

Ferritins are ubiquitous iron storage proteins in living systems. Although much is known about the iron deposition process in ferritin and a mechanism has been developed, several important issues still remain unknown. One lingering question is the less than stoichiometric quantities of hydrogen peroxide detected in previous studies on animal ferritins. The mountain whitefish (Prosopium williamsoni) has been largely overlooked in population genetic analyses despite its wide distribution in discrete drainage basins in western North America for over four million years. Its closest sister taxa the Bear Lake whitefish (P. abyssicola), Bonneville cisco (P. gemmifer), and Bonneville whitefish (P. spilonotus) are found only in Bear Lake Idaho-Utah and were also included in the analyses. Recent research shows that liquid crystals can be used to report the presence of different types of substances through optical amplication of ligand-receptor binding. In this work, simulations based on a coarse-grained method have been performed to study a class of liquid-crystal-based sensors. A tensor order parameter was used to model the liquid crystalline system and the Beris-Edwards formulation was employed to obtain the time evolution of a liquid crystal medium containing particles. Paddlefish are long-lived large river fish which are declining in many areas of their range due to habitat modifications and overfishing. A framework for management of paddlefish in Mississippi is proposed and a case study of its application to the paddlefish population of the Tennessee-Tombigbee Waterway (TTW) is presented. The framework includes four phases: (I) distribution and stock assessment; (II) determination of limiting factors; (III) design and implementation of management actions; and (IV) review and monitoring. The unmanned aerial vehicle?s design departs from existing configurations in utilizing a keel beam fuselage which replaces the enclosed fuselage by a flat keel beam on which the sensors, the autopilot, their related power sources, and flight control systems are mounted. Woodland-period archaeological sites in the North Central Hills physiographic region of Mississippi appear to be highly variable in occupation size, site function, duration, and occupational intensity. To better understand the occupational history and settlement patterns of such sites, several characteristics of four Woodland-period sites located within the Tombigbee National Forest, Ackerman Unit, Mississippi, are evaluated and compared to a larger, previously investigated site, 22WI516. From the data obtained in the field and laboratory, a determination of the causes of the variability among Woodland-period sites in the study area is attempted. To prove this formulation experimentally, I hypothesized that cartilaginous phenotype can be induced in vitro in a seemingly non-cartilaginous cell source from fibrous tissue. A non-stationary Krylov subspace based iterative solver for the three dimensional finite element-boundary integral (FE-BI) method for implantable antennas is presented. The present method numerically solves the frequency domain Maxwell?s equations in the variational form to formulate the finite element solution using hexahedral discretization elements in conjunction with the appropriate boundary integral equations. Four different solvers are used to investigate the convergence behavior of the FE-BI technique on the design of the antennas. In this research, the adsorptive capacities of kenaf in the forms of chopped whole stalk, chopped core, and bast materials were evaluated for the removal of lead, zinc, and toluene from contaminated synthetic waste streams using traditional adsorption isotherm techniques. The effect of surface oxidation using ozone was observed with respect to the adsorption of metals. In this study, we present two new frequency domain tests for testing the Gaussianity and linearity of a sixth-order stationary univariate time series. Both are two-stage tests. The first stage is a test for the Gaussianity of the series. Under Gaussianity, the estimated normalized bispectrum has an asymptotic chi-square distribution with two degrees of freedom. If Gaussianity is rejected, the test proceeds to the second stage, which tests for linearity. Under linearity, with non-Gaussian errors, the estimated normalized bispectrum has an asymptotic non-central chi-square distribution with two degrees of freedom and constant noncentrality parameter. The effects of deionized water and PSE pork percentage on the quality of smoked deli ham and retorted pouched ham with and without non-meat adjuncts were evaluated. Product quality was determined through evaluation of water holding capacity, cooked color, protein-protein bind, and sensory quality. A randomized complete block design with either three or four replications was utilized to test treatment effects in three separate experiments.
