I studied evolutionary history in the angiosperm order Apiales, with a special emphasis on interactions between form, time, and space. Four broad categories of problems were addressed: interfamilial relationships in Apiales, the assignment of genera traditionally assigned to the Apiaceae subfamily Hydrocotyloideae, the estimation of divergence times of the major clades, and the reconstruction of the biogeographic history of Apiales. We used molecular markers with different evolutionary properties and rates derived from the plastid (trnD-trnT and rpl16), nuclear (RPB2), and mitochondrial (nad1 intron 2) genomes, from more than 250 species representing all major clades in the order. Mutations in the breast cancer susceptibility 1 (BRCA1) gene are linked to breast as well as ovarian cancers. However, most cancer-causing mutations within the BRCA1 gene have been found in the N’ and C’ terminal regions of the BRCA1 protein, both believed to be important for DNA double-strand break (DSB) repair. The BRCA1 C’ terminal (BRCT) repeats have been implicated in phospho-serine protein binding whereas the N’ terminal RING domain interacts with the BARD1 protein to form a hetero-dimeric complex with E3 ubiquitin ligase activity. We worked on finding a new kinase regulator to develop basic data to be used for cancer prevention. Our work found a link between three previously unrelated proteins involved in cancer, ERK2 and ERK6 and Scribble. The MAP kinase cascade is involved in cell proliferation, which is highly deregulated in cancer. Through the screening of ERK6 associating molecules, we found the cell polarity, and cell cycle related molecule Scribble. Furthermore, we found that Scribble was a dual-specific kinase regulator. We clearly demonstrated that these ERKs interact with Scribble through the LRR and the PDZ domains of Scribble. We hypothesize that …

Meta-Analysis: Racial Disparities in Prostate Cancer Survival: Prostate cancer is the second leading cause of cancer-related mortality in men. Previous studies have drawn inconsistent conclusions on racial differences in prostate cancer survival. This meta-analysis was conducted to investigate the relationship between race and survival from prostate cancer. A systematic review of published articles from 1968 to 2007 assessing survival from prostate cancer among African American and White men was conducted. The glutamate system dysfunctions present in schizophrenia raise new questions about possible glutamatergic actions of the atypical antipsychotic clozapine. While clozapine has been shown to partially substitute for the discriminative stimulus of the glutamate agonist N-methyl D-aspartate (NMDA) in rats, NMDA discrimination has not previously been established in mice. The present study was designed to explore the possible role of NMDA activity in clozapine’s discriminative stimulus. Two groups of C57BL/6 mice were trained to discriminate either 2.5 mg/kg CLZ from vehicle or 30 mg/kg NMDA from vehicle in a standard two-lever drug discrimination task. Tidal freshwater marshes (TFMs) are unique ecosystems that bridge the gap between terrestrial and aquatic ecosystems and are important in the sequestration of soil organic carbon. Smith-Magenis syndrome (SMS) is a complex disorder caused by haploinsufficiency of RAI1 and characterized by sleep disturbances, behavioral abnormalities, mental retardation, and obesity in teens and adults. Rai1+/- mice are obese after 20 weeks. Dup(17)(p11.2) syndrome is a complex disorder associated with overexpression of RAI1. A transgenic mouse model of dup(17)(p11.2) syndrome overexpresses Rai1 and results in a mouse that is growth delayed. Two studies were conducted to investigate how the use of different types of language affects attitudes. Participants scrutinized arguments supporting a hypothetical toothpaste that differed in terms of argument strength (strong versus weak) and linguistic abstractness (abstract versus concrete) and subsequently evaluated the toothpaste. In addition, half of the participants in the second study were subjected to a cognitive load manipulation (i.e., rehearsing a ten-digit number) in order to limit their level of cognitive elaboration. Results indicated that strong arguments and those containing concrete descriptions led to more positive attitudes about the toothpaste, whereas weak messages comprised of abstract terms …

A study was conducted to optically determine the viability of the Gram +, endospore-forming bacterial genera Bacillus using a hyperspectral reflectance spectrometer. Endospores are a dormant, differentiated cellular capsule form taken by select bacteria to ensure survival when environmental conditions become unfavorable. Two species of Bacillus were used for this study, Bacillus megaterium and Bacillus subtilis. These endospores were killed using a chemical treatment of sodium hypochlorite or a physical treatment of heat and pressure in an autoclave. Electrospinning can be used to selectively process a variety of natural and synthetic polymers into highly porous scaffolds composed of nano-to-micron diameter fibers. This process shows great potential as a gateway to the development of physiologically relevant tissue engineering scaffolds. In this study we examine the structural and functional considerations regarding electrospun scaffolds for dermal template applications using novel quantification techniques. Shrub expansion in herbaceous ecosystems is emerging as an important ecological response to global change, especially in mesic systems where increases in canopy biomass are greatest. Two consequences of woody encroachment are increases in belowground resources, such as carbon and nitrogen, and reductions in above-ground resources such as light, which affect diversity, community trajectory, and ecosystem function. As a consequence, how these neurons convert convergent sensory input into multisensory information is an important question facing neuroscience today. Psychiatric patients are particularly affected by symptoms of insomnia. Because insomnia is often secondary to other conditions and was once thought to be less treatable, this condition has received little attention in terms of treatment and research. Additionally, psychiatric patients have typically fewer resources to seek treatment. Generally, insomnia is treated with medications that may have biological side effects and offer little restorative sleep. Behavioral or cognitive interventions have often been overlooked. The present research examined respiratory responses of blue crabs to long term (4, 13, and 21 days) hypercapnic hypoxia in freshwater at 23 C. Hypoxic conditions (50-60 & 75-85 mmHg O2) were induced by allowing the crabs to consume their oxygen supply, resulting in a hypercapnic induced decrease in pH that remained through the exposure. Postbranchial hemolymph responses to hypoxia/hypercapnia in freshwater demonstrate decreases in PO2, increases in PCO2, and decreases in pH. Lactate levels decreased over time, but hemocyanin concentration was highly variable with no trends. PH, lactate, and hemocyanin observations also demonstrated high variability and a variety of …

Removal of apoptotic cells by macrophages is required in order to prevent autoimmune responses. Previous studies have reported that group IIA secretory phospholipase A2 (sPLA2) preferentially binds to apoptotic cells and induces a chemotactic factor which promotes the migration of macrophages to apoptotic cells. Based on these observations, we hypothesized that the pool of sPLA2 trapped on apoptotic cells produces chemoattractant lipids that recruit macrophages to phagocytose the apoptotic cells. To test this hypothesis, Jurkat cells were cultured under normal conditions and induced to undergo early apoptosis through treatment with D,L-threo-dihydrosphingosine. Glucocorticoids (GC) are common components of many chemotherapeutic regimens for lymphoid malignancies. GC-induced apoptosis involves an intrinsic BCL-2 family-regulated pathway. It has been shown that BIM (BCL-2 interacting mediator of cell death), a BH3-only pro-apoptotic protein, is up-regulated by dexamethasone (Dex) treatment in acute lymphoblastic leukemia (ALL) cells. Furthermore, BIM is inactivated by extracellular signal-regulated kinase (ERK)-mediated phosphorylation. We therefore hypothesized co-treatment with Dex and MEK/ERK inhibitors would promote apoptosis in ALL cells through BIM up-regulation and activation. The purpose of this thesis project was to examine the function of connexin-36 (Cx36) gap junctions (GJs) in producing alcohol's intoxicating and rewarding effects. GABA neurons are thought to inhibit dopamine (DA) neurotransmission in the mesocorticolimbic system, which originates in the midbrain ventral tegmental area (VTA) and projects to limbic structures such as the nucleus accumbens (NAcc). The mesolimbic DA system is believed to be the neural substrate of alcohol intoxication and addiction (Tepper, Paladini, & Celada, 1998). RNAs have become increasingly recognized as possible drug targets due to their involvement in important biochemical functions, as well as their unique but well-defined structures. Recently published crystal structures depict the binding of a series of aminoglycosides- or more specifically- 2-deoxystretamine (2-DOS), the most preserved central scaffold of aminoglycosides, to a conserved 5'-GU-3'region on their target RNAs. A novel unnatural γ-amino acid, 1, has been synthesized using 2-deoxystreptamine as a template through structure-based rational design. I investigated seasonal and inter-site variation in factors influencing diel dissolved oxygen (DO) metabolism at four streams in the Virginia Coastal Plain. Data were collected monthly from January to August 2008 to characterize DO metabolism (diel DO amplitude, production, respiration), incident solar radiation (PAR), dissolved nutrients, fine benthic materials (mass, C:N, Chlα) and hydrology (discharge, water transit time, transient storage). Reach-scale metabolism estimates were determined using the one-station diurnal DO technique and transit time and transient storage were determined by conservative tracer additions. This study aims to determine species-level relationships within the genus Heteromys, as well as generic-level relationships among members of the subfamily Heteromyinae using a phylogenetic framework. Molecular sequence data were generated from two mitochondrial genes (cytochrome b and cytochrome oxidase I) and three nuclear gene segments (β-fibrinogen, engrailed protein II, and myosin heavy chain II), and analyzed under maximum parsimony, maximum likelihood, and Bayesian optimality criteria to infer relationships. Chapter 1 focuses on the phylogenetic and taxonomic implications for Heteromys from the analyses of sequence data. Parkinson’s disease (PD) is a devastating neurodegenerative disorder characterized by progressive loss of dopaminergic neurons located in the substantia nigra. Accumulating evidence indicates that microglia-driven neuroinflammation contributes significantly to chronic neurodegeneration in PD. The matrix metalloproteinases (MMPs) play an important role in several neuroinflammatory paradigms; however, their relationship to dopaminergic neurodegeneration in PD remains relatively unexplored. To address this, the temporal relationship between MMP-2, MMP-3, MMP-9, and MMP-13 expression and dopaminergic neurodegeneration was compared in the 6-hydroxydopamine (6-OHDA) and lipopolysaccharide (LPS) models of PD. Abstract Proof of concept for a continuous environmental sampling methodology that employs common terrestrial arthropods as environmental samplers was tested by analyzing pitfall, malaise and black light captures over a six month period over a replicated urban-suburban-rural gradient in Central Virginia. All arthropods captured at the nine sites were identified and assigned to aquatic, vegetation, or soil groups based on their association with these microhabitats. This study investigated the validity of the Reading Level Achievement Test of the English Language Center (ELC) of Brigham Young University. Test validation is a complicated process that involves evaluation of various types of validity. It was beyond the scope of this study to investigate different types of validity of the Reading LAT. One of the primary goals of active research in molecular biology is to better understand the process of transcription regulation. An important objective in understanding transcription is identifying transcription factors that directly regulate target genes. Identifying these transcription factors is a key step toward eliminating genetic diseases or disease susceptibilities that are encoded inside deoxyribonucleic acid (DNA). There is much uncertainty and variation associated with transcription factor binding sites, requiring these sites to be represented stochastically.
