Tissue Engineering Cellularized Silk-Based Ligament Analogues, Scott Sell Jun 2009

Tissue Engineering Cellularized Silk-Based Ligament Analogues, Scott Sell

Theses and Dissertations

The resurgence, and eventual rise to prominence in the field of tissue engineering, that electrospinning has experienced over the last decade speaks to the simplicity and adaptability of the process. Electrospinning has been used for the fabrication of tissue engineering scaffolds intended for use in nearly every part of the human body: blood vessel, cartilage, bone, skin, nerve, connective tissue, etc. Diverse as the aforementioned tissues are in both form and function, electrospinning has found a niche in the repair of each due to its capacity to consistently create non-woven structures of fibers ranging from nano-to-micron size in diameter. These …

Participating students were required to complete a consent form and obtain parental permission. Students completed a creative writing activity and a kinesthetic activity in which …

Transcriptional, Epigenetic, And Signal Events In Antifolate Therapeutics, Alexandra Racanelli Jun 2009

Transcriptional, Epigenetic, And Signal Events In Antifolate Therapeutics, Alexandra Racanelli

Theses and Dissertations

A targeted approach to the development of antifolate therapies has been sought for many years. Central to the success of such development is an understanding of the molecular mechanisms dictating the sensitivity of cells to antifolates and the fundamental differences of these processes between normal and neoplastic phenotypes. This dissertation addressed transcriptional mechanisms and cell-signaling events responsible for the efficacy of antifolate therapies. Transcriptional processes and cell signaling pathways are often aberrant in neoplastic tissues, providing a potential point of distinction between a normal and neoplastic cellular state. Folylpolyglutamate synthetase (FPGS) catalyzes the formation of poly-γ-glutamate derivatives of folates and …

Investigating The Response Of Oligodendrocyte Progenitor Cells To The Cuprizone Model Of Demyelination, David Moffatt

Theses and Dissertations

Multiple sclerosis and other myelin diseases affect the quality of life many people. In the United States alone, multiple sclerosis afflicts as many as 400,000 individuals. Myelin, which is attacked by multiple sclerosis, plays a critical role in maintaining the healthy function of the adult nervous system. There are many model systems that study myelin and its formation and loss. Our lab investigates the cuprizone model of demyelination and remyelination. The cuprizone model is commonly believed only to affect adult oligodendrocytes, which it kills. The current study investigates whether other cells in the oligodendrocyte line, such as oligodendrocyte progenitor cells, …

Novel Cinchona Alkoloid Derived Ammonium Salts As Phase-Transfer Catalysts For The Asymmetric Synthesis Of Beta-Hydroxy Alpha-Amino Acids Via Aldol Reactions And Total Synthesis Of Celogentin C., Bing Ma

Theses and Dissertations

Project I. Cinchona alkaloid-derived quaternary ammonium salts have been successfully used as phase-transfer catalysts, particularly in asymmetric alkylations. Our group applied this type of catalyst in the synthesis of β-hydroxy α-amino acids via aldol reactions and discovered that the Park-Jew catalyst afforded good yields and good enantiomeric excess of the syn diasteromers, but negligible diastereoselectivity. This project was therefore focused on the synthesis of novel cinchonidine-derived catalysts with the Park-Jew catalyst as the lead structure. The C3 position of cinchonidine nucleus was modified to achieve dimers and catalysts possessing electron-deficient alkyne and alkene moieties. Synthesized catalysts were tested in the …

Roles Of Krüppel Like Factors Klf1, Klf2, And Klf4 In Embryonic Beta-Globin Gene Expression, Yousef Alhashem

Theses and Dissertations

Krüppel like factors (KLFs) are a family of 17 proteins whose main function is gene regulation by binding to DNA elements in the promoters of various genes. KLF transcription factors recognize CACCC-elements and act as activators or repressors of the gene expression. Among the 17 family members, KLF1, KLF2, and KLF4 share high homology to each other. KLF1 is the founding member of the family and is an erythroid-specific protein. KLF2 is expressed in erythroid, endothelial, and other cells. KLF4 is expressed in endothelial, smooth muscle, and other cells. In this thesis, the functions of these KLFs were reviewed in …

Investigating The Role Of Electric Fields In Directing Schwann Cell Behavior, Nishant Magar Jun 2009

Investigating The Role Of Electric Fields In Directing Schwann Cell Behavior, Nishant Magar

Theses and Dissertations

This study examines the potential of Schwann cells (SCs) to be manipulated by electric fields (EFs) in order to improve recovery from spinal cord injury (SCI). It had long been believed that the central nervous system (CNS) is incapable of regeneration, but recent studies have proven otherwise. SC transplants are known to be useful in promoting axon regeneration after SCI, but is not sufficient for functional recovery. EFs are known to exist in vivo, and have been known to drastically affect the morphology and behavior of cells in various tissues. It was the hypothesis of this study that the conditioning …

Production Of Recombinant Injectosome And Outer Membrane Proteins From Yersinia Pestis Kim5, Jeremiah N. Betz Jun 2009

Production Of Recombinant Injectosome And Outer Membrane Proteins From Yersinia Pestis Kim5, Jeremiah N. Betz

Theses and Dissertations

Molecular biology techniques and low cost reagents have lowered the barriers to entry for development of biological arsenals by non-state and state groups. Additionally, genetic engineering of epitope targets from such pathogens as Y. pestis would annul current detection methods and therapeutic treatments. Vaccines often have short shelf lives and are of minimal utility if not used prior to exposure to BW agents. Camelidae, including camels and llamas produce unique antibodies termed Nanobodies® (Nbs) or antigen specific fragments (VHH) which are much smaller than traditional antibodies (15 vs. ~150 kDal) yet seem to attach with the same …

Expressive Writing As A Therapeutic Process For Drug Dependent Women, Meshberg Cohen Sarah Jun 2009

Expressive Writing As A Therapeutic Process For Drug Dependent Women, Meshberg Cohen Sarah

Theses and Dissertations

Women with Substance Use Disorders (SUD) have high rates of trauma and PTSD, which is linked to greater physical and mental health problems and poorer SUD treatment outcomes. While research affirms trauma should be addressed during SUD treatment, the majority of addiction programs do not offer such services. One promising intervention is Pennebaker’s expressive writing paradigm, which includes disclosure of traumatic/stressful experiences through 20-minute writing sessions over 3-5 consecutive days. While expressive writing has been linked to improvements in mental and physical health, the intervention has not been studied in persons with SUDs. The present study was a randomized clinical …

Expressive Writing With University Students With Disabilities, Geraldine Lotze Jun 2009

Expressive Writing With University Students With Disabilities, Geraldine Lotze

Theses and Dissertations

Research suggests college students with high incidence disabilities experience more distress than their peers without disabilities as they adapt to college. The expressive writing paradigm developed by Pennebaker and Beall (1986) effectively reduced distress in college students and other nonclinical samples when participants wrote about emotions they experienced surrounding an upsetting event. Previous research on expressive writing has not addressed the effectiveness of the paradigm with students with disabilities. A randomized control trial study examined changes in distress and daily hassles for participants with disabilities who engaged in expressive writing compared to a control condition in which participants wrote about …

An Irresistible Invitation: Enhancing Academic Publication In Rhetoric And Composition By Inviting Online Peer Commentary, Sarah L. Cutler Jun 2009

An Irresistible Invitation: Enhancing Academic Publication In Rhetoric And Composition By Inviting Online Peer Commentary, Sarah L. Cutler

Theses and Dissertations

In many ways the current publishing system in rhetoric and composition, which centers on the peer-reviewed journal, undermines core values we hold for ideal scholarly communication. These values include collaboration, dialogue, participation, and public engagement. Though the current system's methods of preserving, distributing, and maintaining quality control of scholarly work contradict our values, technological developments have made possible alternative publishing models that could better uphold our values. Developing a preprint archive where scholars develop and share ideas before submitting them for publication in traditional peer-reviewed journals would bring our publishing process closer to our ideals.

The Relationship Between Early Maladaptive Schemas, Chronic And Acute Depression, And Help Seeking Behaviors, Jason Walker May 2009

The Relationship Between Early Maladaptive Schemas, Chronic And Acute Depression, And Help Seeking Behaviors, Jason Walker

Theses and Dissertations

Depression is a disorder that affects many people's lives; approximately 24% of Americans are diagnosed with some form of depression in their lifetime. In order to effectively treat depression we must first understand the disorder and how the symptoms affect people. This study aims to investigate unipolar depression in two ways through the use of a questionnaire. One is to understand the etiology of depression, specifically the role of Early Maladaptive Schemas in the etiology of chronic and acute depression. Hierarchical regressions on 88 participants indicated that EMS and anxiety significantly predict acute and chronic depression. The other aim of …

“Design And Synthesis Of Molecular Probes For The Study Of 5-Ht2a And H1 Receptors”, Jitesh Shah May 2009

“Design And Synthesis Of Molecular Probes For The Study Of 5-Ht2a And H1 Receptors”, Jitesh Shah

Theses and Dissertations

The serotonin (5-HT) receptors, with seven subtypes and at least fifteen distinct members, mediate a wide range of physiological functions both in the central nervous system and in the periphery. All members of the 5-HT family except the 5-HT3 subtype belong to the family of aminergic G protein-coupled receptors (GPCRs). Over the years, various molecules have been reported which act selectively at 5-HT2 receptors. However, there are no ligands that exhibit complete selectivity for one subpopulation of 5-HT2 receptors. Insight into how drugs bind to 5-HT2 receptors could contribute significantly to the development of subtype-selective agents with enhanced therapeutic effects. …

Development Of Ppar-Γ Receptor Agonists As Therapeutic Agents For Diabetes, Ashwini Goswami May 2009

Development Of Ppar-Γ Receptor Agonists As Therapeutic Agents For Diabetes, Ashwini Goswami

Theses and Dissertations

The peroxisome proliferator-activated receptors (PPARs) are the transcriptional regulators of glucose, lipids and cholesterol metabolisms. It has been established that PPAR-γ is the receptor for thiazolidinediones (TZDs) class of type II anti-diabetic drugs. These compounds act as agonists of PPAR-γ. They may delay the development of type II diabetes in individuals at high risk of developing the condition, and have been shown to have potentially beneficial effects on cardiovascular risk factors. PPAR-γ receptor activation by TZDs improves insulin sensitivity by promoting fatty acid uptake into adipose tissue, increasing production of adiponectin (responsible for glucose regulation and fatty acid metabolism) and …

Expression Of Chemokines And Vegfs In Hnscc, Crystal Cunningham May 2009

Expression Of Chemokines And Vegfs In Hnscc, Crystal Cunningham

Theses and Dissertations

Head and neck squamous cell carcinoma (HNSCC) is the sixth most common malignancy worldwide. The 5-year survival rate when the cancer remains as a primary tumor is 81% but when it metastasizes to distant sites, defined as a metastatic cancer, it decreases dramatically to 26%. Approaches to prevent these cancers from undergoing these metastatic changes can greatly improve the survival and outcome of these cancer victims. This current study is examining the expression profiles of chemokines and VEGFs in HNSCC. By investigation the underlying pathways involved in the expressions of chemokines and VEGFS we hope to sort out the transcriptional …

Genetic Analysis Of Ribosome Stalling And Rescue, Douglas Ray Tanner May 2009

Genetic Analysis Of Ribosome Stalling And Rescue, Douglas Ray Tanner

Theses and Dissertations

In eubacteria, ribosome stalling on broken messenger RNA transcripts can lead to cell death. The trans-translation quality control mechanism rescues many of these stalled ribosomes. In this process, tmRNA enters stalled ribosomes by mimicking a transfer RNA, accepting the stalled nascent peptide. The ribosome then releases the broken mRNA and resumes translation on a coding region within tmRNA itself. Translation of tmRNA marks the nascent peptide for destruction by the addition of a short proteolysis tag and the ribosome is released at a stop codon within the tmRNA open reading frame. An intriguing aspect of trans-translation is that …

Heritage Braided, Akiko B. Jackson May 2009

Heritage Braided, Akiko B. Jackson

Theses and Dissertations

The awareness of marginality, oppression, and hierarchy at an early age directly influences the creation of work specific to my identity as a woman of color. Born and raised on an island in the Pacific Ocean, I was aware of my sense of location and space relative to the world. The vast ocean separating me from the ‘mainland’ created a specific understanding of marginality, of what is “main” and what is “minor,” and how these categorical placements continued from my youth to ongoing perpetuation. The work I create has a subtle and persistent investigation into my identity as a socio-economic …

Synthesis And Evaluation Of Stimulatory Properties Of Glycolipids For Natural Killer T Cells, Xiangtian Long May 2009

Synthesis And Evaluation Of Stimulatory Properties Of Glycolipids For Natural Killer T Cells, Xiangtian Long

Theses and Dissertations

Natural killer T cells (NKT cells) are a subset of T cells. They regulate a wide range of diseases including infection, tumor growth, and autoimmune diseases, through recognizing glycolipid antigens in the context of CD1d. An understanding of the scope of glycolipid antigens would facilitate use of this cell type in controlling immune responses. Till today, a lysosomal glycolipid, isoglobotrihexosylceramide (iGb3), is the only natural glycolipid that has been found to be recognized by both human and mouse NKT cells. To elucidate the molecular basis of this specific recognition, iGb3 variants were designed and prepared: i) replacement of the C26 …

Structure And Repetition, Jon Sutter May 2009

Structure And Repetition, Jon Sutter

Theses and Dissertations

This work is the result of an exploration into the relationship of form and pattern. Its inspiration began as an experiment where certain two dimensional graphical patterns were interpreted as three dimensional elements in a lattice-like assembly. The resulting compositions are built entirely of MDF, and each piece is made from multiple copies of one or two identically shaped parts that are joined with shallow grooves. Some of the pieces resemble Asian architectural lattices as well as the scientific models of molecules in crystals. Often, the flat components seem to undulate in space or twist into impossible shapes. Although the …
