Genetic epidemiology has shown a large role for genetic influences on schizophrenia. However, the nature of the variants involved is debated. The common disease-common variant (CDCV) hypothesis suggests that schizophrenia is caused by common alleles with small effect sizes. According to the common disease-rare variant (CDRV) hypothesis, schizophrenia is caused by rare variants with large effect sizes. In recent years, evidence has been found for both common and rare variants in schizophrenia. Several SNPs have been associated with schizophrenia through genome-wide association studies (GWAS), supporting the CDCV hypothesis. Treatment with buprenorphine represents a promising alternative for pregnant opioid addicts but there is a need to understand potential effects on nervous system development. We previously showed effects of perinatal exposure to buprenorphine on axonal caliber and myelination in 26-day-old rat corpus callosum. These changes, detected at the end of rapid brain myelination and accompanied by earlier oligodendrocyte maturation, suggested interference with mechanisms coordinating axonal growth and myelination. Naegleria fowleri (N. fowleri) and Naegleria lovaniensis (N. lovaniensis) are closely related amebae found in the environment. N. fowleri causes Primary Amebic Meningoencephalitis (PAM), a fatal disease of the central nervous system, while N. lovaniensis is nonpathogenic. N. fowleri infection occurs when amebae enter the nasal passages, and migrate to the brain. The molecular mechanisms involved in the pathogenesis of PAM are not well-defined. Therefore, the purpose of this study was to define phenotypic characteristics that may be functionally linked to the pathogenicity associated with N. fowleri. MicroRNAs (miRNAs) are a family of small non-coding RNAs that regulate gene expression at both the mRNA and protein levels. MiRNAs have been shown to affect neuronal differentiation, synaptosomal complex localization and synapse plasticity, all functions thought to be disrupted in schizophrenia. We investigated the expression of 667 miRNAs (miRBase v.13) in the prefrontal cortex of individuals with schizophrenia (SZ, N = 35) and bipolar disorder (BP, N =35) using a real-time PCR-based Taqman Low Density Array (TLDA). Examining older adults’ experiences with and response toward hurricanes within the context of the community of residence is key to both understanding their experiences and planning for future hurricanes. Specific to this research, the objective was to understand the current social network of older adults, or who provides care for whom under what circumstances, using the social constructionist perspective. Grounded theory combined with action research was the theoretical orientation guiding the study. Thermal stability of theG37 tRNA methyltransferase proteins from Thermotoga maritima and Aquifex aeolicus have been compared using Differential Scanning Calorimetry. It was shown that the Thermotoga protein is remarkably stable and is denatured at temperatures in excess of 100 degrees Centigrade. The Aquifex aeolicus protein was less stable, denaturing broadly at temperatures between 55oC and 100oC. In contrast, the mesophilic E. coli protein was completely denatured at 55oC. Enzymatic activity of the proteins was measured at various temperatures. Both the Thermotoga and Aquifex enzymes are active at ambient temperatures, and display a significant decrease in activity when the temperature is …

Lyme disease is an emerging infection that is caused by the Borrelia burgdorferi sensu lato complex. These bacteria exist in nature through an enzootic life cycle involving Ixodes ticks and various reservoir hosts. One way that this bacterium adapts to the different hosts in the enzootic cycle is through the expression of outer surface protein C (OspC). OspC is a surface exposed lipoprotein encoded on circular plasmid 26 that forms homodimers on the bacterial surface and has distinct conserved and variable portions of sequence. The efficacy of peat humic substances in enhancing the degradation of fats, oils and greases (FOG) was investigated under controlled laboratory conditions using bench scale well-mixed bioreactors. An experimental design was used to evaluate the effects of temperature and peat humic substance (PHS) concentration on FOG degradation in domestic wastewater for a temperature range from 10 deg.C to 30 deg.C and a PHS concentration range from 0 to 20 ppm(v). Staphylococcus aureus is one of the leading causes of nosocomial and community acquired infections. Many of the virulence factors are encoded on staphylococcal mobile genetic elements. Members of the SaPI family of S. aureus mobile elements encode superantigens and are mobilized at high frequency by specific helper bacteriophages. One remarkable feature of helper phage exploitation by SaPIs is remodeling of the normal T=7 bacteriophage capsid to produce smaller T=4 phage-like particles. This study is aimed at developing modeling methodologies for simulating the flow of air and aerosol particles through fibrous filter media made up of micro- or nano-fibers. The study also deals with modeling particle deposition (due to Brownian diffusion, interception, and inertial impaction) and particle cake formation, on or inside fibrous filters. Nicotine is the primary addictive substance in tobacco, and most smokers who quit will relapse within a year. Heparin is a complex mixture of sulfated polysaccharides derived from animals and one of the oldest drugs in use. While an efficacious anticoagulant, heparin is beset by side effects and pharmacokinetic difficulties. Low molecular weight heparins (LMWH) are made by depolymerizing unfractionated heparin (UFH) and present improvements in these areas. Obesity results from an excess of adipose tissue and is a major risk factor for type 2 diabetes, cardiovascular disease, and cancer. Adipose tissue exists in two main forms: white adipose tissue (WAT), which stores energy as triglycerides, and brown adipose tissue (BAT), which dissipates stored energy as heat. White adipose tissue is composed of several subcutaneous and visceral depots, each possessing distinct molecular and functional characteristics. Preeclampsia (PE), characterized by hypertension and proteinuria after 20 weeks of gestation, affects 5-8% of pregnancies worldwide. Although preeclampsia is a significant cause of maternal and perinatal mortality and morbidity, its etiology remains to be elucidated. Racial differences have been observed for preeclampsia, with U.S. Blacks having higher rates and more severe disease, compared to U.S. Whites and Hispanics. One potential source of racial differences in preeclampsia is genetic variation between populations. The chemokine receptor CCR5 has been implicated in the pathogenesis of cancers and AIDS. A series of novel piperidine derivatives were designed, synthesized, and evaluated as CCR5 antagonists. The ability of the new ligands to inhibit the increment of intracellular calcium level stimulated by endogenous ligand CCL5 was measured in the calcium mobilization assay as an indication of its CCR5 receptor antagonism. The anti-proliferation assay was performed to measure the ability of these new compounds to inhibit the proliferation of prostate cancer cell lines, PC-3 and M12. Investigations of host-associated differentiation of parasitoids have largely focused on the degree of molecular genetic differentiation, but a true test of species status must examine the mating patterns of differentiated populations to determine if they can interbreed in the wild and produce viable offspring. We examined possible mechanisms of isolation between two genetically distinct host-plant complex races of the braconid, Cotesia congregata, originating from hosts on tobacco and catalpa. Recent attention to these species has resulted in the development of many DArT -based markers in the tetraploid A. magna Murphy et Terrill (2n = 28, CCDD genomes), along with numerous RFLP's, SSR's, DArT's, and EST-based SNPs in hexaploid A. sativa L. Advancements in micro air vehicles can benefit applications such as surveillance, reconnaissance, and search and rescue. In this research, flapping kinematics of a ladybug was explored using a direct linear transformation. A flapping mechanism design is presented that was capable of executing ladybug or other species-specific kinematics. Lysophosphatidic acid (LPA) is a bioactive lipid with a plethora of biological functions, including roles in cell survival, proliferation, and migration. Although high-performance liquid chromatography electrospray ionization tandem mass spectrometry (HPLC ESI-MS/MS) technology has been used to measure the levels of LPA in human blood, serum and plasma, current methods cannot readily detect the minute levels of LPA from cell culture. In this study, a novel HPLC ESI-MS/MS method with enhanced sensitivity was developed which allows accurate measurements of LPA levels with a limit of quantitation at approximately 10 femtomoles. A qualitative study was conducted investigating the perceptions of seven parents of high functioning adolescents with autism spectrum disorders (ASD) regarding incidents of bullying in which the child was involved. Results of the study indicated that all of the children had been bullied at one time or another, and that the majority had difficulty in demonstrating appropriate social skills. The parents viewed supportive peers to be a positive factor, fostering the children's satisfactory social adjustment. Parent interventions for their child included social skills instruction and empathy training. The purpose of this study was to examine how multi-sensory education impacts learning and community in a second grade general education classroom. Lesson plans that targeted multiple senses - and oftentimes included team-building activities - were developed and implemented in the classroom. Qualitative data was collected through interviews, focus groups, student work, and a teacher research journal. After analyzing the data, it was revealed that multi-sensory education can lead to student motivation and engagement, as well as provide a generative space for the use of technology and making connections to the outside world. Learning and memory in the brain are thought to be dependent on synaptic plasticity. In response to sensory input, synapses can be strengthened or weakened, known as long-term potentiation or long-term depression (LTD), respectively. Transient receptor potential vanilloid 1 (TRPV1) has been shown to mediate a novel form of presynaptic LTD in hippocampal interneurons. TRPV1 is currently being heavily studied in the PNS and being targeted by pharmaceuticals for its anti-nociceptive and anti-inflammatory properties. Although it is well documented how introduced species can negatively affect native species, we only poorly understand how they may alter ecosystem functions. We investigated how an invasive fish affected the flux of aquatic insects to terrestrial food webs using mesocosms in a desert spring ecosystem. We compared aquatic insect emergence between alternative community states with monocultures and polycultures of two native species of fish, least chub (Iotichthys phlegethontis) and Utah chub (Gila atraria) plus, introduced western mosquitofish (Gambusia affinis).
