Platinum-based chemotherapeutics are used for the treatment of a wide range of cancers and a number of attempts have been made toward developing compounds with better cellular stability and similar or enhanced cytotoxicity as compared to their predecessors. The first part of the work reported here focuses on the cellular effects of the metabolically stable dinuclear platinum compound, BBR3610-DACH. Comet assay showed this compound to form interstrand crosslinks, a highly toxic DNA lesion in HCT116 cells, at equimolar concentrations to its parental compound, BBR3610. The anatomy of the cranial peripheral nervous system has been studied for over a century, yet surprisingly little is known about how the nerves are guided to their targets. The study of the development of these nerves has important implications for our understanding of craniofacial anomalies and possible treatments for both injury and genetic disorders of nerve development such as Goldenhar-Gorlin syndrome. We have discovered that retinoic acid (RA) may play a role in the development of the trigeminal nerve. To date, literature on FHH discussions about cancer have largely focused on patient-physician communication or the dissemination of cancer-specific genetic tests results within the family. When Pliny the Elder wrote Naturalis Historia around 70 A.D., the idea of natural history contained and connected biology, geology, and mineralogy with the history of painting and sculpture. Art was an extension of the natural world as its materials were extracted from plants, animals, and, particularly, mined and quarried pigments, stone, and metals. In my developing body of work, Incidents of Naturalis Historia, Reconstructed, I combine wasps’ nests, architectural fragments, and other found objects excavated from my surrounding environment with elements of glass that resemble lichen, crystallization, and geologic specimens. Advancements in computational capabilities have allowed researchers to turn towards modeling as an efficient tool to replicate and predict outcomes of complex systems. Computational models of the musculoskeletal system have gone through various iterations with early versions employing dramatic simplifications. In this work, a three-dimensional computational model of the wrist joint was developed. It accurately recreated the skeletal structures of the hand and wrist and represented the constraints imposed by soft tissue structures like ligaments, tendons, and other surrounding tissues. Evidence suggests that caregiver health affects patient health. Consistent with the Biopsychosocial Model, mental health (anxiety, depression, satisfaction with life, self-esteem), physical health (bodily pain, general health, performance in physical role, physical functioning), burden, and social support were examined in the present study to understand the relations between these variables and the overall health of 81 caregivers of patients with multiple sclerosis (MS) in Mexico. Individual growth curve (IGC) modeling evaluated longitudinal trajectories of glycemic control and diabetes care of youth with Type 1 Diabetes (T1D) over three years. IGC modeling allowed comparison of confounded sociodemographic predictors of disease outcomes that included ethnicity, SES, parent marital status, family structure, as well as disease duration, to determine the relative impact of these factors in the evolution of HbA1c and diabetes care throughout adolescence. At baseline, participants recruited from two pediatric endocrinology clinics included 198 youth, ages 9-15 (M age = 12.65, 77% Caucasian, 74% lived with married biological parents, M SES = 45.70) with average HbA1c …

The rapid growth in opioid therapy for non-cancer pain has occurred without an adequate appreciation of the consequences of this growth. Few studies provide patient-centered evidence that can be used to inform the current proposed standards for efficacious (safe and effective) opioid prescribing in non-cancer pain. Furthermore, different terms may be used interchangeably in the literature to refer to opioid-taking behaviors, resulting in imprecise or vague interpretation of existing evidence. In this dissertation, different approaches have been employed to address the quest of understanding the formation and growth mechanisms of carbon-containing molecular ions with relevance to astrochemistry. Two-level factorial experiments are widely used in experimental design because they are simple to construct and interpret while also being efficient. However, full factorial designs for many factors can quickly become inefficient, time consuming, or expensive and therefore fractional factorial designs are sometimes preferable since they provide information on effects of interest and can be performed in fewer experimental runs. Heparin is a highly sulfated glycosaminoglycan with potent anticoagulant, antimetastatic, and anti-inflammatory effects. Polymeric and polyanionic nature of heparin makes dosing and side effects a nightmare for healthcare professionals. Our laboratory has proposed appropriately designed, small, highly sulfated aromatic molecules as potential mimetics of heparin. These easier-to-synthesize small molecules have been shown to possess interesting pharmacological and improved toxicological profiles. However, the detection and characterization of these highly sulfated molecules is challenging. Rectangular collimation is used in dentistry to reduce radiation by restricting the x-ray beam to approximately the size of a number 2 intraoral film (3.2X4.1 cm). However, this restricted beam size can lead to exposure errors. The aim of this study was to retrospectively evaluate the number of radiographs exposed and the presence of technical errors by the use of traditional circular or rectangular collimators during endodontic therapy on simulated teeth in manikins. If gutta-percha cartridges are contaminated with bacteria prior to obturation then contamination of the root canal system may result. Successful treatment not only depends on bacterial elimination but also depends on prevention of recontamination. This study was motivated by the observation that endodontists frequently use single use gutta-percha cartridges on multiple patients. The goal of this study was to determine if cross contamination occurred when a single gutta-percha cartridge in the Calamus™ Flow System was used on multiple patients. Urbanization greatly alters plant and pollinator communities and can affect pollinator movement and subsequent gene flow. Plants persisting in urban areas must adjust to local environmental conditions often different from those in which they naturally evolved, and cultivation techniques for landscaping species have developed traits suitable for existence in urban habitats. Cultivated varieties and native conspecifics often exist in geographic proximity, and if pollinator movement is not blocked by urban structural components, functional differences may negatively impact spatially proximate native populations. The overall aim of this work was to apply HINT, an empirical scoring function based on the understanding of hydrophobicity, to analyze and predict the binding affinities and biological activities of colchicine-site anticancer agents. The second, concurrent aim was to improve the scoring function by incorporating tautomerism within the modeling process. Our belief is that proper evaluation of tautomeric forms for small molecules will improve performance of virtual screening. Previous literature provides an overview of the multiple relationships between alcohol use, protective behavioral strategies (PBS), alcohol-related negative consequences, depression, and sleep problems among college students, as well as differences by individual level characteristics, such as age, gender, and race/ethnicity. Several studies have found that specific demographic groups of students are more likely to reach a higher blood alcohol content (BAC) when “partying” or socializing (Turner, Bauerle, & Shu, 2004; Crotty, 2011). Today more than 23 million U.S. children and adolescents are either obese or overweight. Hospital costs from obesity-related diseases in youth have increased from $35 million (0.43% of total hospital costs) during 1979 to 1981 to $127 million (1.79% of total costs) during 1997 to 1999. During the Pathway to Med School program, I collaborated with three other students to develop a research project on childhood and adolescent obesity. It is characterized by necrotic tissue and abnormal vasculature, making it highly resistant to current cancer treatments. A promising alternative to standard cancer therapeutics is the use of drug delivery systems such as polymeric nanoparticles (NP) that deliver silencing RNA (siRNA) exclusively to tumor cells for gene knockdown. Purpose: To assess the impact of dental Medicaid reform in Virginia on dental practice settings (private practice, corporate practice and safety net clinics). Methods: This retrospective cohort study of 16.2 million dental claims is from the Virginia Department of Medical Assistance Services, which included claims for providers participating in Virginia’s Medicaid program during a 10-year period (2002-2012). The dividing date for the reform was July 1, 2005. The outcome measure was mean claims per participating provider. Properties of these clusters can be controlled through size and composition, and such an approach offers a pathway toward designing larger, customized materials. Purpose: The purpose of this study was to create an experimental design to compare the regimen of ketamine-midazolam to chloral hydrate-meperidine-hydroxyzine for moderate oral conscious sedation. Methods: Patients between 36 and 83 months of age have been randomly assigned to receive 1 of the 2 regimens. Dosages, times, and vital signs will be recorded.
