Workplace violence is increasing in the healthcare and emergency department (ED) violence is escalating to the point where nurses experience violent assaults at 12 times the national average (U. S. Department of Labor, Bureau of Labor Statistics, 2008). Suicide is a national public health concern which can result in emotional and physical devastation for individuals, families, and communities. Suicide leads to unanswered questions for families and communities directly impacted by suicide. Childhood obesity has become epidemic in the United States. One of the best places to combat this problem is within the school system. The purpose of this study was to examine changes in exercise self-efficacy, science interest, and science knowledge of children who participated in a school-based anatomy and healthy lifestyle intervention called Anatomy Academy. In this pilot study, 212 study participants were recruited from 5th and 6th grade children enrolled in one of three charter schools who participated in our 7-week intervention, Anatomy Academy. Past research has demonstrated that the motivational effects of opiates causes a change in ventral tegmental area (VTA) γ-amino butyric acid (GABA) subtype A receptor [GABA(A)R] complexes in opiate-dependent animals, which switch from a GABA-induced hyperpolarization of VTA GABA neurons to a GABA-induced depolarization. Previously shown in naïve animals, superfusion of ethanol (IC50 = 30 mM) and the GABA(A)R agonist muscimol (IC50 = 100 nM) decreased VTA GABA neuron firing rate in a dose-dependent manner. Mass spectrometry facilitates cutting edge advancements in many fields. Although instrumentation has advanced dramatically in the last 100 years, data processing algorithms have not kept pace. Without sensitive and accurate signal segmentation algorithms, the utility of mass spectrometry is limited. iquid sloshing and propellant distribution is an important field of research for aerospace applications such as launch vehicles and spacecraft. The propellant mass behavior can greatly influence vehicle dynamics, altering trajectories and structural loading distributions. In order to combat these concerns some propellant tanks employ thin elastomeric diaphragms to separate the fuel from the gas volumes and restrict the fluid's motion. The diaphragm’s flexible behavior is generally highly nonlinear, and various propellant fill levels and acceleration conditions can cause large deflections resulting in complex buckling and folding patterns. Pulsed lasers are known for their spatial and temporal specificity in delivering heat energy to the tissues. This is useful in laser ablation treatment mechanism where damage to the healthy tissues is highly undesired. However, the efficacy of the process is limited by the damage caused by the pulsed laser. A pulsed laser has both photothermal and photomechanical interaction with tissues. Photothermal interaction is caused by the rise in temperature due to the laser irradiation. This includes the denaturation of proteins, increased mitochondrial membrane permeability and ultimately vaporization. In general, ionizing radiation promotes cytoprotective autophagy in a majority of breast tumor cells. Previous studies from our laboratory indicated that radiation (5x2 Gy) induces cytoprotective autophagy in MCF-7 cells. In the current work, inhibition of autophagy by silencing of Beclin-1 in MCF-7 cells resulted in an increase in sensitivity to radiation based both on cell number and clonogenic survival; however, there was no increase in apoptosis and the basis for this sensitization is currently under investigation. Unexpectedly, enhancement of autophagy by silencing of Bcl-2 also led to an increase in sensitivity to radiation, possibly through the conversion of cytoprotective …

The feedback literature is based on the premise that given a favorable context and a drive to reduce uncertainties, all individuals should seek self-relevant performance information. However, this framework has only been empirically examined using Western samples and has failed to consider the influence of the broader context, with the exclusion of Sully de Luque and Sommer’s (2000) cultural framework. The phenomenon of group affect has been well documented, however the group level conditions through which affective convergence emerges has received very little research. The current study helps to fill this gap by using agent-based modeling to examine affective convergence under varying group conditions. Prompted by increased interest in understanding microporosity, recent efforts at describing and classifying pore types in mudstones have focused primarily on siliceous, gas producing unconventional reservoirs with little attention being paid to carbonate, mixed oil-and-gas producers. The Niobrara Formation in the Denver-Julesburg Basin is a self-sourced resource play producing oil and natural gas from low permeability chalks. Key reservoir lithologies consist of chalk, chalky marl and marl. These lithologies contain flattened chalk fecal pellets which play a significant role in providing porosity. Introduction: Cancer-related fatigue is often described by patients as a lack of energy, mental or physical tiredness, diminished endurance, and prolonged recovery after activity. Etiologic mechanisms underlying CRF are not well understood. Methods: A literature review was conducted to examine studies that had investigated the association of mitochondrial dysfunction with fatigue. The major conclusion from this review was that alterations in energy metabolism may contribute to fatigue. Therefore, the dissertation study focused on laboratory techniques for measuring mitochondrial oxidative phosphorylation enzymes (complexes I-V) and a mitochondrial-specific oxidative stress marker (superoxide dismutase 2 [SOD2]). Many organisms use both vegetation structure and social cues in selecting habitats. Many species of songbirds use the presence of breeding conspecifics as a social cue and sign of habitat quality, and can be induced to settle in unoccupied habitats by artificially broadcasting breeding song, a process referred to as “conspecific attraction”. In our study, we tested response to conspecific attraction a low-density population of the threatened Golden-winged Warbler (Vermivora chrysoptera) in Highland County, VA. Intercellular junctions and cell-matrix adhesions play important roles in the maintenance of epithelial integrity. Assembly and remodeling of the plasma membrane complexes are regulated by membrane trafficking and fusion. This thesis is aimed to elucidate the roles of an important membrane fusion protein, Ykt6, in the regulation of epithelial cell adhesion and migration. For the first time, we show that Ykt6 is essential for assembly of adherens junctions and tight junctions in human prostate epithelial cells. We also observed that Ykt6 negatively regulates both collective epithelial cell migration and cell invasion into Matrigel. The effects of YKT6 on epithelial junctions …

High-throughput technologies for measuring gene expression made inferring of the genome-wide Gene Regulatory Networks an active field of research. Reverse-engineering of systems of transcriptional regulations became an important challenge in molecular and computational biology. Because such systems model dependencies between genes, they are important in understanding of cell behavior, and can potentially turn observed expression data into the new biological knowledge and practical applications. Prevailing community practice models, approaches, and frameworks remain primarily based on practice wisdom, experience, and intuition. Difference, pervasive in various contemporary contexts of practice, largely mediates interactions at the community level. For this thesis, two studiesinvolving leading edge vortex generation for lift production as observed in nature were explored intheir respective flow regimes. Through interrelated experiments in writing, video, sound, and movement, this research investigates the intersections of intimacy, ecology, and performance. Integrating an autoethnographic approach to gender and sexuality with creative practice and theoretical inquiry, this layered investigation offers implications that are both deeply personal and widely accessible. White-tailed deer are ecosystem engineers that dramatically alter forest understory vegetation. Consequently, deer can impact many species in a forest through both direct and indirect effects. One species that deer may indirectly affect is the gypsy moth, whose pupae are preyed upon by the white-footed mouse. Through alterations to understory habitat of mice, deer may reduce mouse predation on gypsy moth pupae. In this study, I tested for indirect effects of deer on the gypsy moth by comparing mouse abundance, vegetation properties, and predation on pupae inside, and outside, of long-term deer exclosures. The global carbon (C) balance is vulnerable to disturbances that alter terrestrial C uptake and loss. Moderate disturbances that kill or defoliate only a subset of canopy trees such as insect defoliation, drought, and age-related senescence are increasing in extent and frequency; yet, little is known about the effect of moderate disturbance on forest production and the mechanisms sustaining or supporting the recovery of the C cycle across a range of moderate disturbance severities.
