Nosocomial Respiratory Tract Infections Associated With The Use Of Ventilatory Support Systems: Epidemiological And Bacteriological Study Of The Effect Of Changing Breathing Circuits At 24 Or 48 Hours, Virginia Archer Lamb
Theses and Dissertations
Nosocomial (hospital-acquired) pneumonia (HAP) continues to be an important cause of morbidity and mortality in the hospital. HAP is the third most common nosocomial infection after urinary tract and surgical wound infections. In addition, HAP is the nosocomial infection with the highest mortality rate. These infections are often difficult to treat, because most are caused by Gram-negative bacilli (GNB) that may be highly resistant to antimicrobial agents. HAPs frequently occur in intensive care patients with underlying lung and/or systemic diseases. Many patients are intubated and are on assisted ventilation. Several sources of infection associated with ventilators or respirators have been …

Effects Of Zinc Deficiency On Sertoli Cell Number And Ultrastructure: An In Vivo And In Vitro Study., Gro Thorne-Tjomsland
Effects Of Zinc Deficiency On Sertoli Cell Number And Ultrastructure: An In Vivo And In Vitro Study., Gro Thorne-Tjomsland
Theses and Dissertations
Hypogonadism in the male is a well known clinical symptom of zinc deficiency. The testicular lesions have been primarily attributed to the effects of decreased zinc on germ cells. This study assessed possible effects of zinc deficiency on Sertoli cell number and ultrastructure.Long-Evans dams and their newborn litters were placed on zinc deficient (ZD), pair-fed or ad libitum fed (AL) diets. All dams received a 20% egg white based diet containing less than one ppm zinc. ZD dams received distilled, deionized water (<1 ppm ZN), while PF and AL dams received the above water fortified with 25 ppm zinc. Male pups were sacrificed day 10 and their testes were processed for light (LM) and transmission electron microscopy (TEM).LM cell counts were performed on one (mu)m sections. Average number of germ cells on the basement membrane (BM) (=GC2) per tubular cross-section was reducted by two-thirds in ZD and PF compared to AL animals (p < 0.0001). Average number of germ cells not on the BM (=GC1) and average number of Sertoli cells per tubular cross-section were not statistically different in the three diet groups.Volume fractions of TEM subcompartments/total cytoplasm were determined by morphometric methods. The lysosome-like fraction was doubled in ZD and PF compared to AL animals (p < 0.05). Endoplasmic reticulum, Golgi and mitochondrial fractions were not statistically different in the three diet groups.Five day old Sertoli cell cultures derived from ZD, PF and AL pups were volumetrically analyzed as in vivo. The endoplasmic reticulum fraction was approximately 100% lower in ZD- than in AL-derived cultures (p < 0.01). Golgi, mitochondrial, dense body and inclusion body fractions were similar in ZD-, PF- and AL-derived cultures. These data suggest that Sertoli cells, although not significantly reduced in number in ZD and PF animals, were affected ultrastructurally by dietary treatment. The volumetric increase in lysosome-like bodies in vivo suggested that Sertoli cells participate in phagocytosis of germ cells degenerating as a result of dietary treatment. The decreased ER volume fraction of ZD-derived tissue cultured Sertoli cells suggested that Sertoli cell secretory potential may be altered as a result of zinc deficiency.

Comparative Studies Of The Metal Activation Of The Cytoplasmic And Mitochondrial Forms Of Phosphoenolpyruvate Carboxykinase In Rabbit Liver (Gluconeogenesis)., Gertrude Hanke Jacoby
Theses and Dissertations
The cytoplasmic and mitochondrial isozymes of phosphoenolpyruvate carboxykinase (PEPCK) catalyze the reversible decarboxylation and phosphorylation of OAA to PEP with GTP or ITP serving as the phosphoryl donor. The forward reaction is an essential component of the gluconeogenic pathway. The purpose of this research was to compare the effects of metal activation on the forward (decarboxylating) and reverse (carboxylating) reactions of the PEPCK isozmes purified from rabbit liver.A purification procedure for rabbit cytoplasmic PEPCK was developed. The final enzyme preparation was homogeneous as established by SDS-PAGE. Pure cytoplasmic PEPCK and pure mitochondrial PEPCK from rabbit liver exhibit identical electrophoretic behavior …

Regulation Of The Solubility Of Zinc In Milk (Calcium-Phosphate, Co-Precipitation, Iron)., Lowell S. Nelson Jr.
Regulation Of The Solubility Of Zinc In Milk (Calcium-Phosphate, Co-Precipitation, Iron)., Lowell S. Nelson Jr.
Theses and Dissertations
Humans absorb zinc, iron and copper more readily from human milk than from bovine milk or infant formulae. Given that solubility is requisite to absorption, the solubilities of select components, including zinc, were examined in preparations of bovine and human milk and in model solutions based on the low-molecular-weight components of the two milks. Solubility was examined over a pH range similar to that found in the human digestive system. Factors which could alter zinc solubility were examined by changing the composition of the model solutions and by fortifying milks with various compounds. Each aliquot of the models or milks was …
Part I: The Intraluminal Chemistry Of Calcium, Magnesium And Zinc In Human And Bovine Milks. Part Ii: Complexed Metal Ion Binding To Poly(Acrylic Acid): A Simple Model For In Vivo Metal Ion Binding., Richard Warren Ames
Theses and Dissertations
Part I. The bioavailability of calcium, magnesium, and zinc is greater from human milk than bovine milk, even though individual concentration magnitudes are reversed. Reasons for this are investigated by studying the complexation and precipitation reactions of these minerals and the low molecular weight ligands present in both milks at pHs ranging from two to eight. These reactions control the solubility of these minerals and we assume that for a mineral to be bioavailable it must be in solution and arrive at the appropriate intestinal segment for absorption. Techniques used to study mineral solubilities are: computer modelling, synthetic models, and …

A Genetic, Ultrastructural, And Tissue Culture Study Of Five Embryo-Lethal Maize Mutants (Desk, Developmental)., Janice Kay Clark
Theses and Dissertations
The genetics and development of 5 embryo-lethal maize mutants defective in early embryogenesis were studied by genetic, histological, ultrastructural, and tissue culture techniques. The mutants examined were cp*-E1113A (dek22) on 1L, dcr*-E1428 (dek23) on 2L, rgh*-E1210, fl*-E1253B, and cp*-E1399A. They were found to be non-allelic. Long term studies of chromosome arm location by the Twaxy translocation stocks and mapping of cp*-E1113A and dcr*-E1428 with multiple linkage marker stocks were initiated. Three mutants lacked leaf primordia at kernel maturity, while two made leaf primordia but were unable to germinate. Embryos of cp*-E1113A became blocked at the transition stage but remained morphologically …
