Neurochemistry, Physiology, And Behavior In A Model Of Gulf War Illness, Victoria A. Macht Jan 2018

Neurochemistry, Physiology, And Behavior In A Model Of Gulf War Illness, Victoria A. Macht

Theses and Dissertations

Upon returning from the first Gulf War, soldiers cited a plethora of unexplained physical and cognitive deficits which have since been termed Gulf War Illness (GWI). Presentation of GWI is positively correlated with pyridostigmine bromide (PB) use, which was prophylactically administered to soldiers in response to threats of chemical warfare. To test the overarching hypothesis that PB interacted with stress of deployment to alter neural, endocrine, and immune systems, the following studies used a 2×2 rodent model with 14 days of drug treatment (vehicle; PB) and 10 days of repeated restraint stress (stressed) or non-stressed-control conditions. Results indicate that PB …

Theses and Dissertations

Dietary composition can impact colonization and composition of gut microbiota. The first aim of this dissertation was to evaluate the effect of dietary Se deprivation, sex and, aging on taxonomic composition of gut microbiota in mice. We have previously shown that long-term dietary Se deprivation promotes health span deteriorate including type-2 diabetes like symptoms in later state Terc-/- mice carrying humanized telomeres. In the present study, the data suggested that the dietary Se deprivation and aging significantly and comparably altered the gut microflora composition. The ratio of Firmicutes/Bacteroidetes was decreased by dietary Se deprivation or aging in both sexes. Abundance …

Perception Vs. Reality: Improving Mission Commander Decision-Making Capabilities By Use Of Heart Rate Zone Feedback In Training Environments, Caitlin M. Oviatt Mar 2017

Perception Vs. Reality: Improving Mission Commander Decision-Making Capabilities By Use Of Heart Rate Zone Feedback In Training Environments, Caitlin M. Oviatt

Theses and Dissertations

In the military environment, it is common for commanders to rely exclusively on perceptional information (e.g., visual observations) to make decisions on their personnel’s physical capabilities. There is little evidence to support the idea that the information provided by physiological feedback technology, typically made available to the individual visually, could improve assessments and decision-making capabilities of outside observers (e.g., mission commander in a remote location). Through experimental examination this research shows that commanders who have Heart Rate Zone (HRZ) biofeedback information about their Airmen by use of physiological technology were able to more accurately predict the level of effort and …

Induced Pluripotent Stem Cells Derived From Mouse Germ Cells, Nora Khan Feb 2017

Induced Pluripotent Stem Cells Derived From Mouse Germ Cells, Nora Khan

Theses and Dissertations

While embryonic stem cells are well known to give rise to tissues comprising all three germ layers, only recently was it found that embryonic-like stem cells could be derived from the postnatal mice testis in culture. Embryonic-like stem cells from postnatal testes have shown that they can undertake most if not all, the functions of embryonic stem cells. Most recent reports have demonstrated that somatic cells can be induced to pluripotent stem cells, mostly with the addition of genes. However, the aim of this work is to explore the potential of testicular cells to become pluripotent, including expression of pluripotency-associated …

Nitrosative Stress Sensing In Porphyromonas Gingivalis: Structure And Function Of The Heme Binding Transcriptional Regulator Hcpr, Benjamin R. Belvin Jan 2017

Nitrosative Stress Sensing In Porphyromonas Gingivalis: Structure And Function Of The Heme Binding Transcriptional Regulator Hcpr, Benjamin R. Belvin

Theses and Dissertations

Porphyromonas gingivalis, a Gram negative anaerobe implicated in the progression of periodontal disease, is capable of surviving and causing infection despite high levels of reactive nitrogen species found in the oral cavity due to its efficient nitrosative stress response. HcpR is an important sensor-regulator that plays a vital step in the initiation of the nitrosative stress response in many Gram negative anaerobic bacteria. We employ a combination of X-ray crystallography, SAXS, resonance Raman spectroscopy, UV-Vis spectroscopy, and molecular biology techniques to better understand this key regulator. Knockout of the hcpR gene in W83 P. gingivalis results in the inability of …

Hydrogen Sulfide Regulation Of Kir Channels, Junghoon Ha Jan 2017

Hydrogen Sulfide Regulation Of Kir Channels, Junghoon Ha

Theses and Dissertations

Inwardly rectifying potassium (Kir) channels establish and regulate the resting membrane potential of excitable cells in the heart, brain and other peripheral tissues. Phosphatidylinositol- 4,5-bisphosphate (PIP2) is a key direct activator of ion channels, including Kir channels. Gasotransmitters, such as carbon monoxide (CO), have been reported to regulate the activity of Kir channels by altering channel-PIP2 interactions. We tested, in a model system, the effects and mechanism of action of another important gasotransmitter, hydrogen sulfide (H2S) thought to play a key role in cellular responses under ischemic conditions. Direct administration of sodium hydrogen sulfide (NaHS), as an exogenous H2S source, …

Computational Fluid Dynamics In A Terminal Alveolated Bronchiole Duct With Expanding Walls: Proof-Of-Concept In Openfoam, Jeremy Myers Jan 2017

Computational Fluid Dynamics In A Terminal Alveolated Bronchiole Duct With Expanding Walls: Proof-Of-Concept In Openfoam, Jeremy Myers

Theses and Dissertations

Mathematical Biology has found recent success applying Computational Fluid Dynamics (CFD) to model airflow in the human lung. Detailed modeling of flow patterns in the alveoli, where the oxygen-carbon dioxide gas exchange occurs, has provided data that is useful in treating illnesses and designing drug-delivery systems. Unfortunately, many CFD software packages have high licensing fees that are out of reach for independent researchers. This thesis uses three open-source software packages, Gmsh, OpenFOAM, and ParaView, to design a mesh, create a simulation, and visualize the results of an idealized terminal alveolar sac model. This model successfully demonstrates that OpenFOAM can be …

Role Of Selenium In Age-Related Degeneration: Selenotranscriptome Hierarchy And Characterization Of Selenoprotein H, Lei Cao

Theses and Dissertations

Selenium (Se) is an essential trace element exerting its biological functions mainly through selenoproteins. Our lab has recently shown a paradoxical role of dietary Se at nutritional levels of intake in the delay of age-related degeneration while reducing longevity in mice carrying humanized telomeres. The first aim of this dissertation was to evaluate the effects of long-term dietary Se deficiency, aging, and sex on selenotranscriptome hierarchy in tissues. Four unique patterns in selenotranscriptomic changes were summarized. First, the responses of selenotranscriptomes to dietary Se deprivation and aging were sexually dimorphic. Second, a few selenoproteins responded to dietary Se deficiency and …

Governing The Rurals: Food Security And Governance In The 21st Century, Gina Paola Rico Mendez Dec 2016

Governing The Rurals: Food Security And Governance In The 21st Century, Gina Paola Rico Mendez

Theses and Dissertations

This dissertation develops a theory about the consolidation of state legitimacy given transformations in food security. Food security has moved from a national food maximization effort to the provision of food to households and individuals, regardless of the production source. This definitional change was triggered by urbanization, industrialization of agriculture and liberalization of trade. These dynamics altered the formula for balance and control of a country’s territory; as agricultural output moved into a global context, urban centers became less reliant on its periphery for sustenance and thus governments have fewer incentives to provide governance of any quality to rural areas. …

In this complex and ever changing operating environment, there remains an essential need to better understand how these developments, and …

Multiscale Mechanics Of Bone: Examining The Influence Of Genetic Alterations, Brendyn James Miller Dec 2016

Multiscale Mechanics Of Bone: Examining The Influence Of Genetic Alterations, Brendyn James Miller

Theses and Dissertations

This MSc thesis investigates how genetic alterations involved in the synthesis of proteins or in the regulation of tissue remodeling affects the composition, structure and mechanics of bone at the multiscale levels. This was accomplished by performing two mouse model studies. In the first study, bone properties of TIMP-3 knock-out (Timp-3⁻ˊ⁻) mice were examined by performing mechanical testing, µCT analysis, and FTIR analysis. Results demonstrated that the mechanical integrity of the Timp-3⁻ˊ⁻ bone was compromised. Decreased values for the yield and ultimate load, stress capacity, and fracture toughness were observed in both the TIMP-3 deficient bones. These reductions in the …

Fundamental Principles Of Tremor Propagation In The Upper Limb, Andrew Doran Davidson Aug 2016

Fundamental Principles Of Tremor Propagation In The Upper Limb, Andrew Doran Davidson

Theses and Dissertations

Although tremor is the most common movement disorder, there exist few effective tremor-suppressing devices, in part because the characteristics of tremor throughout the upper limb are unknown. To clarify, optimally suppressing tremor requires a knowledge of the mechanical origin, propagation, and distribution of tremor throughout the upper limb. Here we present the first systematic investigation of how tremor propagates between the shoulder, elbow, forearm, and wrist. We simulated tremor propagation using a linear, time-invariant, lumped-parameter musculoskeletal model relating joint torques and the resulting joint displacements. The model focused on the seven main degrees of freedom (DOF) from the shoulder to …

Neck Injury Criteria Development For Use In System Level Ejection Testing; Characterization Of Atd To Human Response Correlation Under -Gx Accelerative Input, Craig M. Zinck

Theses and Dissertations

The use of Helmet Mounted Displays is ubiquitous in the field of aviation, adding operational capability, increasing head-supported weight and potential neck injury risk. Developing neck injury criteria to evaluate and quantify neck injury is important to ensure ejection systems are produced within acceptable standards. An ATD to human transfer function is developed that quantifies the difference between ATD and human neck response from -Gx accelerative tests, and demonstrates how this function can be applied to ATD test data to make previously developed human risk functions directly applicable to interpreting ATD data with a human based neck injury criterion. A …

Comparison Of The Effects Of Energy Flux On Metabolic Conditions And Satiety In Young Adults, Molly Madison Demello Jan 2016

Comparison Of The Effects Of Energy Flux On Metabolic Conditions And Satiety In Young Adults, Molly Madison Demello

Theses and Dissertations

Within the United States, 68% of all adults have been classified as overweight to obese, as defined by 25 kg/m2 ≤ BMI ≤ 35 kg/m2. Research has shown that overweight/obese individuals can still gain health benefits through participation in aerobic activity without losing weight. These improvements include improved glucose control, CRF, HDL and quality of life. More research is necessary to understand the health benefits associated with a chronic aerobic intervention under weight stable conditions. We therefore examined the health benefits of an aerobic 6-month intervention under weight stable conditions on body composition, cholesterol and measurements of appetite and satiety …

The Study Of The Regulon Of Oxyr In Escherichia Coli And Porphyromonas Gingivalis, Christopher K. Pham Jan 2016

The Study Of The Regulon Of Oxyr In Escherichia Coli And Porphyromonas Gingivalis, Christopher K. Pham

Theses and Dissertations

The facultative anaerobe, Escherichia coli and the obligate anaerobe, Porphyromonas gingivalis are two bacteria that reside in our body. Although they reside in separate environments, they are both subject to hydrogen peroxide stress and have mechanisms to regulate the stress. OxyR is the primary transcriptional regulator/sensor of oxidative stress response caused by hydrogen peroxide. OxyR in P. gingivalis is not well-characterized compared to OxyR in E. coli. We sought to characterize and compare the two forms of OxyR in order to gain a better understanding of the protein. We determined the oligomeric state of both proteins: primarily a tetramer for …

Robust Models For Operator Workload Estimation, Andrew M. Smith Mar 2015

Robust Models For Operator Workload Estimation, Andrew M. Smith

Theses and Dissertations

When human-machine system operators are overwhelmed, judicious employment of automation can be beneficial. Ideally, a system which can accurately estimate current operator workload can make better choices when to employ automation. Supervised machine learning models can be trained to estimate workload in real time from operator physiological data. Unfortunately, estimating operator workload using trained models is limited: using a model trained in one context can yield poor estimation of workload in another. This research examines the utility of three algorithms (linear regression, regression trees, and Artificial Neural Networks) in terms of cross-application workload prediction. The study is conducted for a …

Clergy Sexual Abuse, Robert P. Allred Jan 2015

Clergy Sexual Abuse, Robert P. Allred

Theses and Dissertations

Sexual abuse perpetrated by trusted members of the clergy presents unique challenges to clinicians and yet the current literature on the effects of clergy sexual abuse is sparse. The vast majority of current research on clergy sexual abuse is based on the perspective of the perpetrators and not the survivors. Some literature suggests that clergy sexual abuse is equivalent to incest due to the level of betrayal trauma associated with each form of abuse. The current study seeks to examine the effects of clergy perpetrated sexual abuse on survivors and examine those effects in the context of the general literature …

Regulation Of Tie2 Extracellular Complex Formation In Angiogenesis, Annamarie Dalton Jan 2015

Regulation Of Tie2 Extracellular Complex Formation In Angiogenesis, Annamarie Dalton

Theses and Dissertations

Pathological angiogenesis is an essential component of tumor growth, development, and metastasis for which few effective therapeutic options exist. Though many cancer therapies target the function of cell surface receptors, mechanisms regulating membrane receptor crosstalk remain unclear. Two important families of receptors in angiogenesis, the Ties and Integrins, respond to the extracellular environment via outside-in and, in the case of Integrins, also inside- out signaling. Recent reports showed that the endothelial specific tyrosine kinase receptor, Tie2, forms complexes with two of the endothelial Integrin heterodimers, α5β1 and αVβ3, providing a convenient mechanism for …

Inquiry-Based Teaching In The College Classroom: The Nontraditional Student, Daniel A. Kiernan Jan 2015

Inquiry-Based Teaching In The College Classroom: The Nontraditional Student, Daniel A. Kiernan

Theses and Dissertations

Decline in the economic realm often bolsters an increase of nontraditional student enrollments in colleges and universities (Windolf, 1992). Many of these students, who do not desire to major in some scientific area, find themselves struggling in required science courses. Over the last decade, science departments of higher education have been adjusting their curriculum to include inquiry in the college science classroom. Although inquiry-based teaching has been shown to be very academically positive in science classrooms from K-12, “at the college level the data are mixed as to whether increasing inquiry instruction can significantly change students’ learning or attitudes toward …

A Perception Of Change, A Change Of Perception, Christopher D. White Jan 2015

A Perception Of Change, A Change Of Perception, Christopher D. White

Theses and Dissertations

Change is a constant reminder that permanence is the ultimate illusion. It is through the creation of hyper-realistic, ceramic sculpture that I explore the relationship between nature, humans, and the phenomenon of impermanence. I seek to expose the beauty that often results from decay while, at the same time, making my viewer question their own perception of the world around them. The juxtaposition of natural and man-made features in combination with the skewing of scale, proportion, and material, creates an altered perspective – forcing the viewer to look closer. By combining both human and natural elements within my work I …
