The hepatitis c virus (HCV) core gene has been chemically synthesized and used to direct the expression of core protein in Escherichia coli (E.coli). When cloned downstream of the inducible T7 promotor the core gene directed the expression of a soluble protein with a molecular weight of 22 kDa. Under native conditions the protein behaved in a manner consistent with the formation of a multimeric structure, which may represent assembled core particles. Further, when examined by electron microscopy a heterogeneous mixture of nucleocapsid-like particles were visible. Core protein was specifically recognized by antibodies present in HCV infected serum, suggesting that …

Florine Stettheimer was an independent painter and eminent "art hostess" among the avant-garde in New York City during the years between the World Wars. In 1916, Stettheimer rather suddenly affected a naive or unschooled style that did not fit within any academic or vanguard movement. This new style, what I have termed conscious naiveté, can be considered the genesis of Stettheimer's mature works. Contemporaneous critical appraisals after the shift in style undervalued the inventive modernity of her work and unfairly “feminized” her style.

Wheat germ agglutinin (WGA) belongs to a family of dimeric chitin binding lectins specific for N-acetyl-D-glucosamine (GlcNAc) and N-acetyl-D-neuraminic acid (NeuNAc). The polypeptide chain consists of a tandem repeat of four conserved 4.3 kDa domains (A, B, C, D) stabilized by four disulfide bonds. Saccharide binding occurs in the dimer interface where domains of different subunits are in contact (A with D, B with C). An aromatic amino acid rich pocket on one domain (sufficient for saccharide binding) and a polar region on the contacting domain constitute a complete binding site. A correlational approach was utilized in this study to investigate the relationship between adoption related task performance and perceived quality of life of families with an adopted special-needs child. Additionally, a set of contextual variables suggested by the literature to influence family functioning with an adopted special-needs child were also studied. Purposive and availability sampling approaches were employed to identify the sample of special-needs adoptive families (N = 289) to whom a survey questionnaire was sent. Both mothers and fathers were asked to complete the Survey.

A machine which can read unconstrained words remains an unsolved problem. For example, automatic entry o handwritten documents into a computer is yet to be accomplished. Most systems attempt to segment letters o a word and read words one character at a time. Segmenting a handwritten word is very difficult and often, the confidence of the results is low. Another method which avoids segmentation altogether is to treat each word as a whole. This research presents a multiresolution wavelet analysis tool for analyzing motion in time sequential imagery. A theoretical framework is developed for constructing an L2R wavelet multiresolution analysis from three non-identical spatial and temporal L2R wavelet multiresolution analyses. This framework provides the flexibility to tailor the spatio-temporal frequency characteristics of the three dimensional wavelet filter to match the frequency behavior of the analyzed signal. An unconventional, discrete multiresolution wavelet decomposition algorithm is developed which yields a rich set of independent spatio-temporally oriented frequency channels for analyzing, the size and speed characteristics of moving objects. This thesis effort, the Synthetic Battle Bridge SBB , provides a real-time simulated environment for military commanders to observe on-going computer simulations of varying participation levels and helps them wring the most from a simulation. A two-dimensional, X-Y addressable, multiplexed array of 256 electrodes (16 x 16) has been fabricated using conventional semiconductor processing techniques. The individual electrodes are 16O microns x 160 microns, approximating the size of the cortical columns; the overall array size is 3910 microns x 3910 microns. The array has been fitted to a chronically implantable package and tested for several days in a simulated neural environment. EEG-like data were collected successfully from individual electrodes in the array. This study investigated the stress-strain response and failure modes of Graphite/Polyetheretherketone (Gr/PEEK) tension specimens containing a central elliptical hole. Both experiments and finite element numerical analyses were conducted. Test specimens included seven layups and three hole geometries; three of each combination were tested. The holes consisted of an ellipse with major axis aligned with the load axis, an ellipse with major axis transverse to the load axis, and a circle. Strain gages were located adjacent to the hole and midway between the hole and tab on all specimens. Strain data was used to plot stress-strain response. The analysis utilized a …

The purpose of this study was to compare demographic and personality characteristics of a group of inpatients at a psychiatric hospital in New York State who were sexually abused and who were self-mutilators (N = 64) with: a group of sexually abused inpatients (N = 42); a group of self-harming inpatients (N = 25); and a group of selected general psychiatric inpatients (N = 25). All subjects were 18 years of age or older at the time of the study and over 90% of the participants were females.

This report considers the use of chlorine dioxide as an alternate disinfectant for the removal of biota from Garrison Diversion water. Laboratory testing has shown that chlorine may not be an acceptable disinfectant because it can produce carcinogens from side reactions with constituents found in natural surface waters.

This study used Lake Sakakawea water collected at the Snake Creek Pumping Station located on the eastern shore. A Parallel Discrete Event Simulation Coprocessor was designed to off- load the synchronization overhead from the processors executing the application. In a multiprocessor architecture, one coprocessor executes the synchronization routines for each host processor. Speedup can be achieved when the host processor executes the application and the coprocessor concurrently executes synchronization routines. The coprocessor uses a programmable microcode control store to guarantee flexibility in the synchronization routines. The coprocessor uses an Extreme Search Associative Memory to support fast Next Event Queue NEQ management. This research designed and implemented a reusable Distributed Interactive Simulation DIS visual simulation architecture for Silicon Graphics platforms. The goal was to research software architecture technologies and to create a design and implementation using these ideas. The architecture was designed using object oriented techniques to provide the ability to customize it via inheritance extension. The term "learned helplessness" has been used to describe behavioral deficits in an organism which has learned that the termination of aversive events occurs independently of its responding (Overmier & Seligman, 1967). An appetitive analogue to learned helplessness, referred to as "learned indolence," results when an organism receives noncontingent reinforcement (Bordages, unpublished thesis). This study sought to compare deficits produced by both phenomena in order to argue for a central underlying mechanism. The basis of this study was the prediction that bovine anterior lens capsules (LCs) and Descemet's membranes (DMs) would accumulate Maillard reaction products with age and that these products could be detected by immunoelectron microscopy. In addition, it was predicted that Maillard reaction product content could be related to alterations in basement membrane (BM) morphology. Antiglucitollysine and antipyrraline antibodies were used to detect early and advanced Maillard reaction products in intact glucose-treated and untreated bovine anterior LCs and DMs. Light microscopy revealed that old LCs and DMs were significantly thicker than young LCs and DMs. The distribution and content of …

This research accomplished three major tasks. First it examined familiarity and usage rates for fifteen published R&D project selection methods in the context of a larger general issue, the Air Force's ability to develop and exploit technology. Wright Laboratory served as the focus for the research effort and displayed a greater tendency to use formal methods in 1993 than was shown in prior research. The laboratory's overall preference for simpler models like Checklist, Scoring, and Sorting led to a recommendation that authors familiar with the other techniques communicate them in engineering and management vernacular. Cleaning up hazardous and toxic wastes left from past activities has become one of the major goals of the Department of Defense. Within the Air Force, individual major commands (MAJCOMs) annually review numerous projects submitted under the Defense Environmental Restoration Program (DERP) and forward these projects to HQ USAF/CEVR for funding. Until recently, adequate resources have been available to fully fund all projects. However, within the last two years, funding shortfalls have been experienced, and projects have received only partial funding. This study evaluated the TPH (total petroleum hydrocarbon) cleanup standard for petroleum contaminated soils (PCS). A survey of 13 state regulators was performed to characterize current standards and regulatory viewpoints on the use of a TPH versus a BTEX cleanup standard. The regulatory community considers the BTEX constituents the greatest threat to groundwater, yet expressed concern that the use of a compound specific standard, without an accompanying analysis for TPH, might result in residual soil contamination that may present risk. This study refines risk analysis procedures for trichloroethylene (TCE) using a physiologically based pharmacokinetic (PBPK) model in conjunction with the Monte Carlo method. The Monte Carlo method is used to generate random sets of model parameters, based on the mean, variance, and distribution types. The procedure generates a range of exposure values for human excess lifetime cancer risk of lxl0 (exp-6), based on the upper and lower bounds and the mean of a 95% confidence interval. This thesis produced a model of the Army airdrop segment of the Pope AFB deployment system. The original intent was to model the entire system and simulate the simultaneous deployment of the 23rd Wing and 82nd Airborne Division; however, time constraints and incomplete data forced a reduction in scope. The study provides an excellent foundation for further research into the use of simulation to develop a generalized deployment sizing model. The goal of this study was to develop a physiologically based pharmacokinetic (PBPK) model that predicts mammalian blood concentrations of dibromomethane following exposure by dermal absorption more accurately than a previously developed Homogeneous Dermal Model. The Homogeneous Dermal Model contains a dermal compartment with no dermal sub-compartments. The 1:1 Dermal Model developed in this research contains a dermal compartment with a stratum corneum and a composite dermal sub-compartment of equal volume. This model yields predictions which are 21.4 percent more accurate than the original homogeneous model. In order to represent skin anatomy more accurately, the 1:10 Dermal Model variation was …

This thesis assesses the risk of the health liabilities from exposure to toxic metals found in the composted material of Air Force municipal solid waste (MSW). The goal is to determine the probability that the composted MSW could be a health hazard if it were used as a soil amendment. The research limited the assessment of the exposure risk to heavy metals found in raw MSW and its resulting compost. A cost-effectiveness prioritization methodology was developed using concepts from the cost-effectiveness technique. The new methodology called the Cost-Effectiveness methodology (CEM) allows the prioritization of mitigation projects across broad categories of hazardous sites as would be ranked among the offices of the Environmental Protection Agency. Two performance criteria were incorporated into the process--rank projects that save the most lives per dollar and enhance general applicability across different categories of risk. A metric was developed that represents the ratio of lives saved per dollar spent on mitigation alternatives of contaminated environmental sites.
