This study compared five solvents - urea, carbonate-bicarbonate, citrate, phosphate, and water - for their ability to extract proteins from beech (BM) (Hypsizygus marmoreus) and Swiss brown (SB) (Agaricus bisporus) mushrooms. Using mass spectrometry-based proteomics and bioinformatics, we found that urea produced the highest total protein identifications by data-dependent acquisition at 1% false discovery rate (FDR): BM: 1739, SB: 1907; and by data-independent acquisition: BM: 1969, SB: 2172 proteins, especially enriching cytosolic and ribosomal proteins. Carbonate-bicarbonate favoured the extraction of metabolic and translational proteins, while citrate targeted extracellular and membrane-associated proteins related to cell wall dynamics. Phosphate-enriched catalytically active proteins, …

Habitat condition and area shape global species distributions, with shallow-water reefs hosting a disproportional share of marine biodiversity. Although reef area is a well-established predictor of marine species richness, its historical context is less well understood. Here, we show that the rise of tropical marine biodiversity is closely tied to reef expansion in space and time. During the Early/Mid Miocene (23 to 11.6 million years ago), Indo-Pacific reefs reached unprecedented size and thickness, surpassing any reef systems in the past 66 million years. These massive reefs, likely driven by unique environmental, biotic, and tectonic conditions, fostered the expanding diversity and …

Research outputs 2022 to 2026
Amyloidogenic proteins, such as amyloid-β (Aβ), self-assemble into cross-β fibrils whose accumulation is central to Alzheimer's disease (AD). Measuring Aβ aggregation and clearance in living cells remains challenging using current cell-based assays, which are often low-throughput or not suited for real-time monitoring. This study aimed to (1) develop a robust, quantitative, and scalable fluorescence-based assay using Amytracker to monitor Aβ accumulation and clearance in an Aβ-producing neuronal cell model, and (2) validate its utility for mechanistic studies and therapeutic screening. We established a plate-based fluorescence assay using Amytracker in MC65 neuronal AD model expressing the Amyloid precursor protein C-terminal fragment …

Skeletal muscle exhibits remarkable plasticity following injury, yet most research has focused on responses to a single bout of eccentric contractions. This review addresses adaptations to repeated eccentric contraction–induced injuries across physiological and pathological conditions, with emphasis on insights from preclinical rodent models. In healthy muscle, the repeated bout effect (RBE) reduces strength loss and accelerates recovery after subsequent bouts. However, these adaptations are highly condition dependent. Aging can attenuate the RBE, while dystrophic muscle remains vulnerable to repeated injury despite compensatory remodeling. Other factors, including but not limited to, chronic alcohol intake and malignant hyperthermia can influence these responses, …

NRAS mutations occur in 10%–30% of cutaneous melanomas and are associated with high tumor mutational burden. Mutant NRAS signaling drives aberrant cell growth and proliferation, in part, through activation of the RAF–MEK-ERK1/2 kinase pathway; however, targeted therapies to this pathway have limited effectiveness in patients with NRAS mutant melanoma. The role of other targetable signaling pathways in NRAS mutant melanoma is poorly characterized. Here, we demonstrated that one isoform of diacylglycerol kinase, diacylglycerol kinase eta (DGKη), a lipid signaling regulator, was highly expressed in NRAS mutant melanoma patient samples. Knockdown of DGKH in NRAS mutant melanoma cell lines resulted in …

Thermo-Hydro-Chemical Coupled Numerical Modeling Of Hydrogen-Thermal Co-Development In Hydraulically Fractured Peridotite, Jiacheng Dai, Zhonglin Ma, Yunzhi Yue, Yujie Yuan, Shouceng Tian
Research outputs 2022 to 2026
Hydrogen generation through serpentinization reactions in peridotite formations under high temperature conditions represents a promising avenue for subsurface hydrogen production. However, the thermal energy in the formation environment have not been sufficiently considered. To integrate hydrogen production with thermal energy development, this study develops a thermo-hydro-chemical coupled numerical model, which is used to investigate hydrogen-thermal co-development in hydraulically fractured peridotite. Given that the hydrogen-thermal co-development process involves fluid flow, heat transfer and serpentinization reactions, the governing equations are formulated based on Darcy flow and energy conservation equations, with serpentinization kinetics incorporated through the reactive source terms. To evaluate hydrogen production …

Dual History Enhancement With Hybrid Hypergraph-Graph Networks For Temporal Knowledge Graph Reasoning, Kailun Ye, Xiangjie Kong, Yuchao Zhang, Xuan Wang, Linan Zhu, Jiaxin Du, Guojiang Shen, Jianxin Li
Research outputs 2022 to 2026
Temporal Knowledge Graph (TKG) reasoning seeks to predict future events by analyzing historical data, where the effective leverage of both local and global historical facts proves crucial. Existing approaches employ graph neural networks (GNNs) and recurrent neural networks (RNNs) for local evolution patterns, complemented by statistical methods to enhance attention to global facts, demonstrating efficient predictive capabilities. However, traditional GNNs, constrained by their low-order neighborhood aggregation design, inherently fail to model potential high-order dependencies among facts. Furthermore, existing global history modeling approaches may introduce irrelevant historical information that interferes with prediction tasks. To address these limitations, we propose a Dual …

Mood Monitoring In Schools: A Promising Alternative To Single-Time-Point Screening, Shane L. Rogers, Brown, Kathryn Campbell, Matthew Goulding
Research outputs 2022 to 2026
School-based mental health screening typically relies on single-time-point assessments, which assume that students’ emotional well-being is sufficiently stable for classification based on a single measurement. The present study examined this assumption by evaluating the stability of emotional well-being classifications under repeated mood monitoring. Students from two secondary schools (United Kingdom, n = 413; Australia, n = 354) completed the Brief Emotional Experience Scale weekly across six to seven weeks. Emotional well-being classifications were examined relative to a predefined low well-being threshold to assess stability across time, and a post-monitoring survey examined students’ self-reported perceptions of the monitoring experience. Most students …

Global warming is primarily driven by the rapid accumulation of carbon dioxide (CO₂) in the atmosphere. Metal–organic frameworks (MOFs) have emerged as promising materials for mitigating CO₂ emissions due to their tunable porosity, large surface area, and structural flexibility. Among them, MIL-53(Al) has attracted widespread interest owing to its excellent thermal and chemical stability. Recent studies show that modifying MOFs with secondary metals can significantly enhance their CO₂ adsorption performance. In this work, a one-pot synthesis method was employed to incorporate calcium (Ca), functioning as a Lewis basic metal, into the MIL-53(Al) framework to produce a series of bimetallic materials: …

Background: Respiratory fitness and sarcopenia status have been reported to be cross-sectionally linked with each other and increase mortality risk. However, little is currently known regarding the connection between impaired respiratory function and sarcopenia and their joint effect on future death risk. Methods: This study included 12 027 participants (50.8% were women; mean [SD] age, 58.9 [9.3] years) from a nationwide, prospective cohort in China (China Health and Retirement Longitudinal Study). Respiratory function was assessed by peak expiratory flow (PEF). Sarcopenia status was assessed according to the Asian Working Group for Sarcopenia 2019 (AWGS 2019) criteria. Time-to-event survival analyses and …

Research outputs 2022 to 2026
Rechargeable zinc–air batteries (ZABs) are promising next-generation energy storage systems due to their high energy density, intrinsic safety, and low cost. Nevertheless, commercial deployment of Zn anodes is hampered by persistent challenges like dendrite formation, deformation, passivation, corrosion, and the hydrogen evolution reaction. This review systematically highlights the critical relationship between anode morphology and electrochemical performance and discusses recent advances in advanced manufacturing strategies for Zn anodes. Additive manufacturing approaches, such as 3D printing, enable the fabrication of porous and tunable Zn electrodes with enhanced surface area and uniform ion flux distribution. Furthermore, advanced materials processing techniques like alloying, coatings, …

AbstractKoalas (Phascolarctos cinereus) give birth to highly altricial young that rely on maternal milk for nutritional support and immune protection in the first year of life. This study applied proteomics to analyse stage-specific shifts in the koala milk proteome across early-, mid-, and late-lactation. Proteins were extracted from seven samples and analysed using liquid chromatography-tandem mass spectrometry. Quantitative profiling identified 874, 1220, and 498 proteins in early-, mid- and late-lactation stages, respectively. The most abundant proteins—including β-lactoglobulin, caseins, clusterin, trichosurin, albumin and lactotransferrin—were consistently detected across all stages, reflecting crucial roles in nutrition, immune function, and detoxification. Protein–protein interaction analyses …

Introduction: Recent advances allow blood tests to detect key proteins linked to Alzheimer's disease (AD). Methods: In this before-and-after study across three Australian memory and cognition clinics, we evaluated the impact on clinicians' diagnostic confidence and management following disclosure of routine patients' AD probability, using predefined plasma phosphorylated tau 181 (pTau181) thresholds set at 90% sensitivity and 90% specificity for amyloid-beta (Aβ) positron emission tomography (PET) positivity. Results: One hundred thirteen participants (mean age: 71.2 ± 8.4; mean Mini-Mental State Examination score: 27.7 ± 2.5) with dementia (n = 17, 15.0%), mild cognitive impairment (n = 48, 42.5%), and subjective …
