Voltage Gated Ion Channel Function: Gating, Conduction, And The Role Of Water And Protons, Alisher M. Kariev, Michael E. Green

Ion channels, which are found in every biological cell, regulate the concentration of electrolytes, and are responsible for multiple biological functions, including in particular the propagation of nerve impulses. The channels with the latter function are gated (opened) by a voltage signal, which allows Na+ into the cell and K+ out. These channels have several positively charged amino acids on a transmembrane domain of their voltage sensor, and it is generally considered, based primarily on two lines of experimental evidence, that these charges move with respect to the membrane to open the channel. At least three forms of motion, with …

Xylophagous insects derive nutrients from intractable substrates by producing or ingesting cellulolytic enzymes, or by maintaining associations with symbiotic microbes. Wood-boring cerambycid beetle larvae sometimes house maternally-transmitted endosymbiotic yeasts that are presumed to provide their hosts with nutritional beneÞts. These are thought to be absent from species in the large subfamily Lamiinae; nevertheless yeasts have been repeatedly isolated from the guts of neotropical lamiines. The objective of this study was to conduct transmission electron microscopy (TEM) studies of cerambycid larval midgut tissues to determine if gut yeasts were intracellular, or simply present in the gut lumen. Nine cerambycid larvae were …

Autism spectrum disorders (ASDs) are highly heritable developmental disorders caused by a heterogeneous collection of genetic lesions. Here we use a mouse model to study the effect on cortical connectivity of disrupting the ASD candidate gene PTEN (phosphatase and tensin homolog deleted on chromosome 10). Through Cre-mediated recombination, we conditionally knocked out PTEN expression in a subset of auditory cortical neurons. Analysis of long-range connectivity using channelrhodopsin-2 revealed that the strength of synaptic inputs from both the contralateral auditory cortex and from the thalamus onto PTEN-cko neurons was enhanced compared with nearby neurons with normal PTEN expression. Laser-scanning photostimulation showed …

Background Transposition in IS3, IS30, IS21 and IS256 insertion sequence (IS) families utilizes an unconventional two-step pathway. A figure-of-eight intermediate in Step I, from asymmetric single-strand cleavage and joining reactions, is converted into a double-stranded minicircle whose junction (the abutted left and right ends) is the substrate for symmetrical transesterification attacks on target DNA in Step II, suggesting intrinsically different synaptic complexes (SC) for each step. Transposases of these ISs bind poorly to cognate DNA and comparative biophysical analyses of SC I and SC II have proven elusive. We have prepared a native, soluble, active, GFP-tagged fusion derivative of the …

Vitamin A deficiency is a serious global health problem that can be alleviated by improved nutrition. Development of cereal crops with increased provitamin A carotenoids can provide a sustainable solution to eliminating vitamin A deficiency worldwide. Maize is a model for cereals and a major staple carbohydrate source. Here, we discuss maize carotenogenesis with regard to pathway regulation, available resources, and current knowledge for improving carotenoid content and levels of provitamin A carotenoids in edible maize endosperm. This knowledge will be applied to improve the nutritional composition of related Poaceae crops. We discuss opportunities and challenges for optimizing provitamin A …

Aberrant expression of the presynaptic serotonin 1A receptor (5-HT1A-R) because of a polymorphism in the 5-HT1A-R gene is associated with severe depression in human, whereas its absence up to postnatal day 21 (P21) in the forebrain of mice results in heightened anxiety in adulthood. These observations collectively indicate that the 5-HT1A-R has a crucial role in brain development. To understand the mechanistic underpinnings of this phenomenon, we used organotypic slice cultures of hippocampi from C57BL6 mice (C57) at P15, which coincides with the peak of neonatal synaptogenesis. Stimulation of the hippocampal 5-HT1A-R caused a dramatic increase …
Evidence For A Grooming Claw In A North American Adapiform Primate: Implications For Anthropoid Origins, Stephanie Maiolino, Doug M. Boyer, Jonathan I. Bloch, Christopher C. Gilbert, Joseph Groenke
Evidence For A Grooming Claw In A North American Adapiform Primate: Implications For Anthropoid Origins, Stephanie Maiolino, Doug M. Boyer, Jonathan I. Bloch, Christopher C. Gilbert, Joseph Groenke
Publications and Research
Among fossil primates, the Eocene adapiforms have been suggested as the closest relatives of living anthropoids (monkeys, apes, and humans). Central to this argument is the form of the second pedal digit. Extant strepsirrhines and tarsiers possess a grooming claw on this digit, while most anthropoids have a nail. While controversial, the possible presence of a nail in certain European adapiforms has been considered evidence for anthropoid affinities. Skeletons preserved well enough to test this idea have been lacking for North American adapiforms. Here, we document and quantitatively analyze, for the first time, a dentally associated skeleton of Notharctus tenebrosus …

Relationships Between Urinary Biomarkers Of Phytoestrogens, Phthalates, Phenols, And Pubertal Stages In Girls, Tandra R. Chakraborty, Eilliut Alicea, Sanjoy Chakraborty
Publications and Research
Phytoestrogens, phthalates, and phenols are estrogen-disrupting chemicals that have a pronounced effect at puberty. They are exogenous chemicals that are either plant-derived or man-made, and can alter the functions of the endocrine system and cause various health defects by interfering with the synthesis, metabolism, binding, or cellular responses of natural estrogens. Phytoestrogens, phthalates, and phenols are some of the potent estrogens detectable in urine. Phytoestrogens are plant-derived xenestrogens found in a wide variety of food products, like soy-based food, beverages, several fruits, and vegetables. Exposure to phytoestrogens can delay breast development and further lead to precocious puberty. The effect of …

Notes On The Diet Of The Malay Civet (Viverra Tangalunga) And Other Civets In Logged And Unlogged Lowland Dipterocarp Rain Forests In Sabah, Borneo, Christina P. Colon, John B. Sugau
Publications and Research
Civet diets were examined in a logged and unlogged Bornean rain forest. Malay civets (Viverra tangalunga) consumed invertebrates, fruit, rodents, insectivores, birds, snakes and lizards, and appear to show preference for centipedes and scorpions. Other civet species consumed fruit, such as figs, Connarus sp. and Annona sp., particularly in the unlogged forest, but also consumed invertebrates and vertebrates. Reduced fruit consumption observed in the logged forest may be due to lower availability and may be offset by increased consumption of invertebrates. The increased overlap in diet between Malay civets and other civets in disturbed areas may lead to …

Plasticity Of Corticospinal Neural Control After Locomotor Training In Human Spinal Cord Injury, Maria Knikou
Plasticity Of Corticospinal Neural Control After Locomotor Training In Human Spinal Cord Injury, Maria Knikou
Publications and Research
Spinal lesions substantially impair ambulation, occur generally in young and otherwise healthy individuals, and result in devastating effects on quality of life. Restoration of locomotion after damage to the spinal cord is challenging because axons of the damaged neurons do not regenerate spontaneously. Body-weight-supported treadmill training (BWSTT) is a therapeutic approach in which a person with a spinal cord injury (SCI) steps on a motorized treadmill while some body weight is removed through an upper body harness. BWSTT improves temporal gait parameters, muscle activation patterns, and clinical outcome measures in persons with SCI. These changes are likely the result of …
Culture Ontogeny: Lifespan Development Of Religion And The Ethics Of Spiritual Counselling, Glen Milstein, Amy Manierre
Culture Ontogeny: Lifespan Development Of Religion And The Ethics Of Spiritual Counselling, Glen Milstein, Amy Manierre
Publications and Research
The counsellor has an ethical obligation to treat the whole person. Humans are cultural beings and the foundation of most cultures is religion. Religion and culture are received from our early relation~ ships and modified through later relationships across the lifespan. The paper introduces the term "culture ontogeny" to emphasize that this is a biological process wherein abstract ideas of culture and religion become material in the developing neurophysiology of each brain. A framework and methods are offered to examine the changing roles of religion in clients' emotional self~ structure, inclusive of those who describe themselves as spiritual, not religious. …
Estimating Climate Trends: Application To United States Plant Hardiness Zones, Nir Krakauer
Estimating Climate Trends: Application To United States Plant Hardiness Zones, Nir Krakauer
Publications and Research
The United States Department of Agriculture classifies plant hardiness zones based on mean annual minimum temperatures over some past period (currently 1976–2005). Since temperatures are changing, these values may benefit from updating. I outline a multistep methodology involving imputation of missing station values, geostatistical interpolation, and time series smoothing to update a climate variable’s expected value compared to a climatology period and apply it to estimating annual minimum temperature change over the coterminous United States. I show using hindcast experiments that trend estimation gives more accurate predictions of minimum temperatures 1-2 years in advance compared to the previous 30 years’ …
Good Men Grow Corn: Embodied Ecological Heritage In A Belizean Mopan Community, Kristina Baines
Good Men Grow Corn: Embodied Ecological Heritage In A Belizean Mopan Community, Kristina Baines
Publications and Research
Recent developments in land rights and land use in the Toledo district, Belize has generated anthropological and activist interest surrounding traditional ecological knowledge and practice, and the role of heritage in communities. This study explores the connection between ecological knowledge and practices, and the concurrent construction of heritage, and community health and wellness, broadly defined. Developing and using the concept of “embodied ecological heritage,” this dissertation takes a phenomenological approach to understanding the convergence of ecological heritage and health in multiple realms of everyday life, arguing that lived experience of participating in “traditional”practices is fundamentally connected to wellness in the …

The Effect Of Dissolved Humic Acids On Aluminosilicate Formation And Associated Carbon Sequestration, Ashaki A. Rouff, Brian L. Phillips, Stacey G. Cochiara, Kathryn L. Nagy
Publications and Research
Allophane and imogolite neogenesis in soils may occur in the presence of organic matter. To understand this process under conditions relevant to soils, the influence of dissolved organic carbon (DOC) as humic acid (HA), on aluminosilicate formation was studied at 25◦C, pH 6, and low-DOC concentrations. For solutions with initial Al/Si ratios of 1–2.1, and 0–6mg/L DOC, precipitates recovered after 20h had Al/Si ratios of 2.2–2.7. The formation of allophane, imogolite-like material, and aluminosilicate gel was confirmed by XRD, FTIR, and NMR. The effect of DOC was to produce a small, but systematic increase in imogolite-like Si in the precipitate, …

Interrogating Texts: From Deferent To Efferent And Aesthetic Reading Practices, Cheryl Hogue Smith
Publications and Research
This article offers a revised version of transactional reading theory to explain how students classified as basic writers tend to employ counterproductive reading and thinking processes that inhibit them from full participation in academic life. Louise Rosenblatt proposes that readers have two main positions or purposes in reading—the efferent stance, where readers focus on the information in a text, and the aesthetic stance, where readers focus on the experience they have with a text. This article describes a third, deferent stance of reading that many basic writers adopt when they defer their interpretations of texts to other readers or defer …

Standardised Classification Of Pre-Release Development In Male-Brooding Pipefish, Seahorses, And Seadragons (Family Syngnathidae), Stefan Sommer, Camilla M. Whittington, Anthony B. Wilson
Publications and Research
Background: Members of the family Syngnathidae share a unique reproductive mode termed male pregnancy. Males carry eggs in specialised brooding structures for several weeks and release free-swimming offspring. Here we describe a systematic investigation of pre-release development in syngnathid fishes, reviewing available data for 17 species distributed across the family. This work is complemented by in-depth examinations of the straight-nosed pipefish Nerophis ophidion, the black-striped pipefish Syngnathus abaster, and the potbellied seahorse Hippocampus abdominalis.
Results: We propose a standardised classification of early syngnathid development that extends from the activation of the egg to the release of newborn. The classification consists …

In photosynthetic organisms, carotenoids serve essential roles in photosynthesis and photoprotection. A previous report designated CruP as a secondary lycopene cyclase involved in carotenoid biosynthesis [Maresca J, et al. (2007) Proc Natl Acad Sci USA 104:11784–11789]. However, we found that cruP KO or cruP overexpression plants do not exhibit correspondingly reduced or increased production of cyclized carotenoids, which would be expected if CruP was a lycopene cyclase. Instead, we show that CruP aids in preventing accumulation of reactive oxygen species (ROS), thereby reducing accumulation of β-carotene-5,6-epoxide, a ROS-catalyzed autoxidation product, and inhibiting accumulation of anthocyanins, which are known chemical …

Justice In Context: Assessing Contextualism As An Approach To Justice, Michael Buckley
Justice In Context: Assessing Contextualism As An Approach To Justice, Michael Buckley
Publications and Research
Moral and political philosophers are increasingly using empirical data to inform their normative theories. This has sparked renewed interest into questions concerning the relationship between facts and principles. A recent attempt to frame these questions within a broader approach to normative theory comes from David Miller, who has on several occasions defended ‘contextualism’ as the best approach to justice. Miller argues that the context of distribution itself brings one or another political principle into play. This paper examines this claim. It considers several plausible strategies for carrying out Miller’s general project and argues that each strategy fails. Nevertheless, the author …

Policing Gender, Sexuality And ‘Muslimness’, Anissa Helie
Publications and Research
In her Introduction to the book Sexuality in Muslim Contexts: Restrictions and Resistance (ZED Book, UK), Anissa Helie discusses the notion of "Muslimness" and how such a homogenizing notion, if unchallenged, may serve to justify the policing of sexuality in various Muslim countries and communities.
The book itself looks at emerging trends that affect women’s sexuality, with a particular focus on Asia and the Middle East, and documents both the curtailing of sexual rights occurring in diverse Muslim societies and the strategies designed to counter these developments.
Yet this focus in no way suggests that the policing of gender and …
