Several factors contribute to the failure of the central nervous system (CNS) to regenerate after injury. These include inhibition of axonal growth by myelin and glial scar associated molecules, as well as the intrinsic inability of adult CNS neurons to grow long axons in environments that are permissive for younger neurons. Neurons in the peripheral nervous system (PNS) display a much higher capacity to regenerate after injury than CNS neurons, as shown by conditioning lesion experiments and by microtransplantation of dorsal root ganglia neurons into CNS white matter tracts. This dissertation examines aspects of the relationship between connectivity and the development of genetic structure in subdivided coral reef populations using both simulation and algebraic methods. The first chapter develops an object-oriented, individual based method of simulating the dynamics of genes in subdivided populations. The model is then used to investigate how changes to different components of population structure (e.g., connectivity, birth rate, population size) influence genetic structure through the use of autocorrelation analysis. The autocorrelograms also demonstrate how relationships between populations change at different spatial and temporal scales. Integrated sequence stratigraphy and geochemistry has significantly improved our knowledge of the formation and distribution of early and late diagenetic products in North American Mississippian carbonates. Deposition of the Madison Limestone occurred in concert with a major perturbation to the global C-pool, the timing of which was constrained by comparing secular variations in the delta13C values from the Madison Limestone with limited biostratigraphy. These early constraints were then improved by peak matching of 87Sr/86Sr values from this study with European brachiopod 87Sr/86Sr. The secular variations in the delta13C values were then applied as a chronostratigraphic tool to outcrops and subsurface …

Evolutionarily conserved Pumilio (Pum) RNA-binding proteins act as translational repressors during embryo development and cell fate specification. Previous work in the lab has shown that over-expression of Pum2 (Pum2-EML) supports maintenance and suppresses mutilineage differentiation of murine multipotent HSC/MPP-like cell line EML. The subsequent analysis of HSC markers and functional analysis has revealed that wt EML cells share the LKS CD34 positive phenotype, whereas the majority Pum2-EML cells are similar to LKS CD34 negative. The CD34 positive wt EML cells can be divided into CD34low, CD34med and CD34high subpopulations, whereas Pum2-EML CD34 positive cells correspond to CD34low subpopulation. Colony forming …

Infidelity in romantic relationships can be devastating, and can cause many complex emotional reactions. The Jealousy as a Specific Innate Module (JSIM) hypothesis posits that due to differing reproductive pressures over evolutionary history, men and women have evolved different mechanisms to respond to infidelity. JSIM proposes that men, due to fears of cuckoldry, will respond with intense jealousy to a partner's sexual infidelity. Cognitive behavioral violence prevention (CB-VP) parenting groups are commonly used for the primary and secondary prevention of violence. These groups use persuasive messages that target violence-related attitudes and cognitions, with the expectation that this will result in behavior change. Despite their frequent use as family violence prevention strategies, little is known about the actual messages being exchanged in CB-VP groups and how participants perceive and recall these messages. Polycyclic aromatic hydrocarbons (PAHs) are environmental and occupational carcinogens that are produced by the incomplete combustion of organic material, such as from the burning of tobacco, coal, and petroleum products. In addition to causing cancer, exposure to PAHs is hypothesized to contribute to atherosclerosis and to lead to increased incidence rates of cardiovascular disease in populations. Considering the number of deaths attributable to tobacco smoke exposure, ambient air pollution, and occupational hazards, PAHs may be a significant contributor to the prevalence of cardiovascular disease in human populations. Dolphinfish (Coryphaena hippurus) is a pelagic species that is ecologically and commercially important in the western Atlantic region. This species has been linked to dominant oceanographic features such as sea surface temperature (SST) frontal regions. This work first explored the linkages between the catch rates of dolphinfish and the oceanography (satellite-derived SST, distance to front calculations, bottom depth and hook depth) using Principal Components Analysis (PCA). It was demonstrated that higher catch rates are found in relation to warmer SST and nearer to frontal regions. The goal of this research was to determine the relative quality of near shore marine areas by investigating their influence on Haemulidae community structure, distribution pattern, condition, and growth. Habitat was defined at the small spatial scale of individual habitat types such as seagrass beds, mangroves and coral reefs, and at the broader spatial scale of the interconnection of these individual habitat types within a mosaic (IHM). The processes influencing larval fish survival in the low-latitude open ocean are poorly understood, especially with regard to feeding. Foraging presents a significant challenge for neonatal predators. Both physical maturation and predatory experience may improve predatory abilities. To tease apart which improvements in predatory abilities were due to increased maturation and which to increased experience, several experiments that isolated the effects of maturation and experience were conducted. Individual whitespotted bamboosharks, Chiloscyllium plagiosum, improved predatory efficiency with both maturation and experience. Physical maturation, when isolated from experience, improved predatory efficiency of naïve sharks foraging on shrimp, an elusive prey, but not of sharks foraging on worms, a non-elusive prey. Spinal Muscular Atrophy (SMA) is an autosomal recessive motor neuron disease. SMA is associated with homozygous mutations in the Survival of Motor Neuron gene I (SMN1). SMN protein does not appear to exist in cells in isolation but associates with several proteins to form a large multi-protein complex. Over the last decade our understanding of mercury cycling has dramatically changed. Evidence of rapid atmospheric oxidation has been observed in the Arctic, Antarctic, the MBL, coastal environments, saline lakes, and the upper troposphere/lower stratosphere. These results show that, Hg0, can undergo rapid gas-phase oxidation under standard atmospheric conditions. However, the mechanism and importance of this transformation is still unclear. This dissertation explores the spatial patterns and place-based characteristics of colorectal cancer (CRC) late stage incidence and CRC-specific mortality in Miami-Dade County. Because CRC is the second leading cause of death among all cancers and is almost 90 percent preventable through medical screenings, investigations of CRC disparities across groups and communities are extremely relevant in the fight against cancer. Pathogenesis of enterotoxigenic Escherichia coli (ETEC) and Shigella flexneri relies predominantly on members of the AraC/XylS family of transcriptional regulators, Rns (or its homolog, CfaD) and MxiE, respectively. Rns/CfaD regulate the expression of pili, which allow the bacteria to attach to the intestinal epithelium. Better understanding of the role Rns plays in virulence was attained by expanding our knowledge of the Rns regulon, revealing that it functions as an activator of cexE, a previously uncharacterized gene. By in vitro DNase I footprinting two Rns-binding sites were identified upstream of cexEp, both of which are required for full activation of cexE. …

A new strategy was developed to switch the luminescence of semiconductor quantum dots with chemical stimulations. It is based on the photoinduced transfer of either energy from CdSe-ZnS core-shell quantum dots to [1,3]oxazine ligands or electrons from the organic to the inorganic components. Upon addition of base or acid, energy or electron transfer pathways respectively become operative, leading to changes in the luminescence of the nanoparticles. Activated Heat Shock Transcription Factor 1 (HSF1) has received attention in recent literature as a therapeutic effector in diseases of protein misfolding, as an immune modulating adjuvant in tumor regression, and as a trigger for gene therapy transcription. In its normal function, activated HSF1 enhances heat shock protein (Hsp) expression when additional molecular chaperoning is required (i.e., in situations of proteotoxic stress, including thermal stress) in a process known as the heat shock (HS) response. But whether or not arbuscular mycorrhizal fungi (AMF) provide any benefit to M. cerifera is not known. "Why have separate sexes?" is a fundamental question in biology and has been investigated intensively since Darwin first proposed two hypotheses: Separate sexes evolve 1) to avoid detrimental effects of self-fertilization or 2) to improve allocation of scarce resources to each sex's reproductive function. In animal-pollinated plants dioecy is hypothesized to be favored when small, generalized pollinators predominate because they increase self-fertilization rates of plants. I concurrently tested Darwin's hypotheses using Echinocereus coccineus (Cactaceae). I found, as predicted, when specialized pollinators (hummingbirds) were excluded from plants and only generalized pollinators (bees) visited, selfing rates of plants increased 238%. High selfing …

Recent evidence suggests that overfishing of large predatory fishes has resulted in substantial population declines and pelagic longline (PLL) fishing is a major contributor. The primary objective of this dissertation is understanding factors that affect the interactions between marine fish PLL fishing. These factors are important to determine vulnerability of bycatch and target species, especially when PLL catch and effort data are used to estimate stock abundance. Located at the northern edge of the Northern Gulf of Alaska (NGOA), Prince William Sound(PWS) is a small, two-strait semi-enclosed sea. The general ocean circulation pattern inside PWS is significantly affected by external forcings, for instance, the large-scale circulation in NGOA, atmospheric pressure and surface winds, surface heating/cooling, runoff, and tides. Motivated by multi-year experience with a well-validated, quasi-operational ocean circulation nowcast/forecast system for PWS (viz., Extended PWS Nowcast/Forecast System (EPWS/NFS)), the present study addresses some aspects of the PWS response to various external forcings, via numerical simulations. This study confirmed whether children on the current treatment of choice for HIV infection, Highly Active Antiretroviral Treatment (HAART), exhibit better immune functioning than children on earlier forms of treatment, including sole exposure to Monotherapy/Combination Therapy (Mono-Combo) and "upgrading" from Mono-Combo to HAART (Conversion). It explored whether HAART protects areas of neuropsychological functioning previously found to be compromised in children perinatally infected with HIV more effectively than these earlier treatments. This study includes a unique population (i.e., predominantly minority, low SES status, and largely bilingual), and controls for a number of treatment variables that have not been previously considered. World-wide coral reefs are in a state of decline as a result of many local and regional factors. Recent global mass mortalities of reef corals due to record warm sea temperatures have led researchers to consider global warming as one of the most significant threats to the persistence of coral reef ecosystems over the next 100 years. It is well established that elevated sea surface temperatures (SSTs) cause widespread coral bleaching, yet confusion lingers as to what facet of extreme temperatures is most important.
