The American chestnut tree (Castanea dentata) has been reduced from a key canopy hardwood to an understory shrub by the chestnut blight disease. C. dentata shares similar morphological characteristics with the Allegheny chinquapin (C.pumila variety pumila) and the Ozark chinquapin (C.pumila variety ozarkensis). Before the chestnut blight epidemic, the easiest and most obvious means of separating species of American chestnut and chinquapins was based on habit or flower morphology. C.dentata existed as a large tree, while varieties of C. pumila existed as shrubs or small trees; C. dentata contains three nuts per bur, while varieties of C. pumila contain one …

The purpose of this study was to assess whether high sulfur inputs in a local wetland had an effect on the functions of this wetland, i.e. an increased sulfate reduction rate and an increased sulfate concentration compared to a control wetland. Sulfate reduction rates were compared in two wetlands in Chattanooga, Tennessee during July and December 2007.These wetlands were known to have bad different histories of sulfur input via sulfates in precipitation or runoff.Through use of a 35S tracer technique, overall sulfate reduction rates could be compared for the two wetlands.Replicated sediment cores were collected from each wetland on the …

Work conducted in the Millard Biochemistry Research Laboratory examines the dual nature of molecules as carcinogens and anti-tumor agents through the molecular mechanisms of duplex DNA damage by bifunctional alkylating agents. Diepoxybutane (DEB) and epichlorohydrin (ECH) are polar molecules that form covalent DNA interstrand lesions by cross-linking the N7 position of deoxyguanosine residues. A recent experiment indicated that ECH preferentially targets nuclear DNA over mitochondrial DNA, whereas DEB shows similar rates of lesion formation for both loci. It was concluded that preferential targeting of nuclear DNA results from relatively poor uptake of ECH across the mitochondrial membrane. The objective of …

The aim of this project is to provide an explanation for recently obtained binding constants for two similar guest molecules, NDMG and N-MAP, with a p-sulfonatocalix[6]arene host in ammonium acetate buffer. This work was done primarily using pressure perturbation calorimetry, which is a technique that determines the coefficient of thermal expansion, α, which is in turn related to the solute molecule's effect on the order of the surrounding water molecules. Stomatal frequencies of fossil-plant species are used to estimate past pCO2 levels based on the physiological functions of living taxa. Numerous studies have shown that there is an inverse relationship between pCO2 and stomatal frequency parameters. As levels of pCO2 increase, the Stomatal Density (SD) and Stomatal Index (SI) decrease. However, pCO2 is not the only factor affecting SD and SI values, which are a product of leaf growth and expansion. Stomatal characteristics differ between genera, and studies also have shown that SD and light intensity have a positive correlation. The present study hypothesizes that SD and SI are not …

Pollinator visitation rates over the life of a flower are determined by pollinator abundance and floral longevity. If flowers are not visited frequently enough, pollen limitation may occur, favoring the evolution of self-compatibility (SC). In plant species with varying SC levels, central populations often are self-incompatible (SI) and peripheral populations are SC. Witheringia solanacea (Solanaceae) is a species that follows this trend with the exception of one population in the Monteverde Cloud Forest Reserve, which is peripheral yet SI. I investigated this population using multiple techniques including floral bagging, pollinator observations, microsatellite analysis, and floral longevity manipulations. My results confirmed …

Effects Of Alcohol And Training On Exercise Performance And Muscle Recovery Biochemistry In The Brown Anole (Anolis Sagrei), Maryclaire Mcgovern

Contrary to previous research, training may improve exercise performance in a lizard, the brown anole. A brief, two-week training period resulted in increased performance speed and distance before exhaustion in trained lizards. Trained lizards were also able to more effectively use leg glycogen stores, however each of these improvements were not found in lizards treated with alcohol. Liver glycogen concentrations were also lower in alcohol-treated lizards, and patterns of liver glycogen concentrations during recovery indicate some hepatic lactate gluconeogenesis.

Microhabitat selection of the limpet Tectura testudinalis, an important grazer along the rocky Maine coast is examined using both mensurative and manipulative experiments. T. testudinalis substrate selection is essential to its survival in the unpredictable and harsh regime of the intertidal zone. At three sites studied in the Gulf of Maine, T. testudinalis selectively inhabits tidal pools and vertically oriented substrates. Both of these microhabitats reduce the degree of environmental stress (desiccation, extreme temperature, and hypersaline conditions) and predation experienced by individuals. Differences among sites are common, indicating the importance of varying environmental factors in regulating and influencing habitat selection …

Large carnivores are some of the world’s most difficult species to conserve because of their large spatial requirements, low population density, and high potential for conflict with humans. However, large carnivores are valuable for ecological, cultural, and ethical reasons. Carnivore management can take a wide variety of forms, including translocation, sterilization, livestock compensation programs, regulated hunting, improved livestock husbandry practices, and zoning, among others. The objective of this thesis was to conduct a comparative analysis of examples of zoning from around the world to examine whether it is an effective carnivore management strategy.

Recently, it has been demonstrated that an individual’s rhythmic limb movements exhibit greater variability when viewing spatially incongruent biological limb movements, but not when viewing spatially incongruent non-biological movements. Some researchers have concluded that a ‘mirror-neuron system’ might mediate the intrinsic bidirectional link between perception and action underlying interpersonal, but not environmental, coordination (e.g., Kilner et al., 2003; Tognoli et al., 2007). The current study aimed to: 1) contest this recent finding; and 2) demonstrate that the self-organized entrainment processes of coupled oscillators can explain the differing influences of biological and non-biological movements. From a social constructionist perspective linked with a feminist standpoint, I examine three forms of adolescent self-destructive behaviors: eating disorders, self-mutilation and substance abuse. The social construction of adolescents’ norms, values, and beliefs, as based upon their interactions with family, peers, and the media, helps explain these self-destructive actions. In addition to a comprehensive literature review, I interviewed five adults who work with adolescents in the state of Maine, and used these professionals’ experiences and knowledge to support the current theories pertaining to these acts of self-harm. Through an interdisciplinary analysis of children’s homes in San Pedro Sula, Honduras, this project reflects on how conceptions of family affect the prioritization of the needs of the homes’ beneficiaries. One consequence of the impoverished nature of Honduras are orphaned, abandoned, mistreated, malnourished, abused, neglected and disowned children who have come to live in group children’s homes that, for the most part, meet their short-term physical needs. Calix[n]arenes, cyclIc molecules composed of "n" benzene rings meta-linked by a methylene to form a macrocycle, are very prevalent jn the literature and enjoy a wide range of applications. However, the oxygen bridged and nitrogen bridged systems, oxacalllx[n)arenes and azacalix[n]arenes, respectively, are far less common because they are synthetically challenging. Furthermore, the selective synthesis of oxacalix[6]arene systems has not previously been well documented, but the systems can now be accessed in a facile approach disclosed here. The insight and knowledge gained from the synthesis of oxacalix{6]arenes has provided a foray into the synthesis of azacalix[nlarenes.

The carbamolyating activity of Cloretazine, a novel anticancer prodrug, modifies protein function and acts synergistically with the choroethyalting species, which a1kylales DNA. The reactive subspecies responsible for carbamolylating activity is methyl isocyanate, which reacts with sulfhydryl groups and amines. DNA repair enzymes have been identified as potential targets for the modification by methyl isocyanate; in particular, base excision repair enzymes. Enzymes in DNA base excision repair include DNA polymerase beta (Pol ?) and apurinic/apyrimidic endonucease-l (APE/Ref-1). APE/Ref-1 hydrolyzes the 5' -phosphodiester DNA backbone from an abasic DNA template, where the base was excised by a DNA glycosylase. The entire nucleotide …

This thesis is the product of my five weeks of ethnography in three classrooms in Waterville, Maine and the surrounding area, in addition to individual and group interviews with both students and the educators themselves. It seeks to understand why, in a culture so saturated with images of sexuality and naked bodies, the teachers I observed were largely unwilling or unable to discuss human sexuality in public schools as anything more than a public health issue. Studies have been conducted examining the impact of commodity bundling on company profits and the premium prices charged for these bundles. With recent news of the skyrocketing costs of higher education, it is critical to highlight the specific effects of commodity bundling on the prices of textbooks. A study performed by the Government Accountability Office in 2005 cited that a main cause of increasing textbook prices is the recent inclusion of textbook supplements such as software and workbooks. We conducted an experiment to assess the effect of presidential rhetoric in a time of crisis. Our study was based in part on terror management theory, which posits that subtle reminders of death (mortality salience) lead to increased support of leaders an authority figures. Subjects were randomly placed in either a mortality salient condition or control condition. We also composed two speeches - one charismatic and one non-charismatic - and subjects were randomly assigned to hear one of the two. Potassium channel interacting proteins (KChIPs) co-assemble with Kv4 a subunits to form native complexes that encode considerable components of neuronal somatodendritic A- type K+current (Is,J in neurons, and transient outward current (ho) in cardiac myocytes. The binding of KChIPs to the cytoplasmic N-termini of Kv4 a subunits enhances surface expression by facilitating intracellular trafficking, stimulates subunit assembly, and regulates the functional gating properties of Kv4 channels. KChIPs, like many other neuronal calcium sensor proteins, contain four EF-hand calcium binding sites; two of which (EF-3 and 4) have particularly high affinity for calcium. Sponges produce an impressive variety of secondary metabolites that perform a variety of ecological functions. Many marine sponges even harbor diverse carotenoid compounds, an unusual class of secondary metabolites that animals are incapable of producing. Furthermore, sponges serve as hosts to an astonishingly diverse microbial community that can occupy up to sixty percent of a sponge's biomass. Kahalalide F (KF, 1) is the largest and most biologically active cyclic peptide of the naturally occurring kahalalide family of depsipeptides. Previous evaluations of the compound have shown significant in vitro and in vivo activity against various forms of cancer. KF has been investigated in phase I clinical trials and is being evaluated in phase II clinical studies. Sulfonylhydrazines are a class of DNA alkylating drugs which also produce carbamoylating activity in situ. The carbamoylating species is of significance because it has shown to inhibit important thiol-containing enzymes such as glutathione reductase (GR) and thioredoxin reductase (TrxR). TrxR catalyzes dithiol-disulfide exchange reactions on thioredoxin (Trx), which in turn catalyzes other reductive processes such as deoxyribonucleotide biosynthesis. In this study, we demonstrate that TrxR activity is strongly inhibited by the anticancer prodrug Cloretazine both in purified form and in leukemia cell lysates. This inhibition is specific to the carbamoylating activity of methylisocyanate (MiC). In contrast, another important oxidoreductase, glutathione …

The purpose of my thesis was to create merchandising and promotional artwork for the Oxford, Mississippi-based band RiverLeft and their album “Stranger Things.” I used the design software Adobe Illustrator and Adobe Photoshop to create album artwork, posters, and other illustrations to be implemented onto promotional materials. By tying all of the artwork together around a central theme, I created an identity for the band’s marketing and advertisement around the south and beyond. 1

Tectura testudinalis is a limpet that lives in the mid-intertidal zone along the coast of Maine and grazes on a variety of encrusting algae. A previous study asserted that T. testudinalis preferred to feed and rest on the encrusting alga Clathromorphum circumscriptum and that this species of limpet displayed homing behavior. However, I show that T. testudinalis does not home or return to any specific substrate while resting. Conclusive evidence was found for nocturnal movement. I show that C. circumscriptum was the preferred food source for this limpet, closely followed by Hildenbrandia rubra, another encrusting alga. Field and lab experiments …

Mitragynine is a major alkaloid extracted from the leaves of the Thai traditional herb, Mitrcigymi speciosa Korth (Rubiaceae). The extracts of this plant have been used as substitutes for opium and recent studies have reported that mitragynine possesses antinociceptive actions mediated via the mu opioid receptors. Nonetheless, structural features of mitragynine do not fit the classical opiate ligands. Osteopathic medicine has quickly grown into a successful and respectable branch of medical practice. It was created out of a necessity for change in the late 1800s, and continues to be a credible form of alternative medicine. After reviewing literary material, journal articles, and numerous osteopathic college sources it was seen that osteopathic medicine can be practiced in numerous settings and will become integral in the near future due to the shortage of physicians in the United States. Today, osteopaths work closely with all types of medical personal including medical doctors.
