Bipolar disorder (BD) and attention deficit/hyperactivity disorder (ADHD) share an overlapping number of symptoms. These shared symptoms may result in the misdiagnosis or over diagnosis of these two disorders. The purpose of this study was to look at the diagnostic practices of clinicians and clinicians-in-training to see what diagnosis they would give to a hypothetical patient who presents with ambiguously ADHD and bipolar disorder symptoms. Clinicians and clinicians-in-training (N = 40) read two vignettes, one child and one adult, where the patient presented with both ADHD and BD overlapping symptoms, and then were asked to provide one primary DSM-IV diagnosis, …

Due to the inherently error-prone nature of RNA replication, mutations to genomes of RNA viruses occur frequently and accumulate. We hypothesized that RNA versions of nucleoside analogues that increase mutation rates in DNA could cause increases in the mutation rate of a model RNA virus, Reovirus, and decrease the fitness of the virus in vitro due to the accumulation of deleterious mutations. After conducting multiple passages of Reovirus in mouse L929 cells in the presence of these potential RNA mutagens in four separate trials, there were not only virus samples with the expected decreased infectivity, but surprisingly, samples with marked …

DNA polymerase I from Rhodothermus marinus is a high-fidelity DNA polymerase capable of operating at high temperatures and incorporating 2´,3´-dideoxynucleotides. The R. marinus DNA Polymerase I active site contains an unusual proline in the middle of a mobile “O helix.” This proline residue is hypothesized to decrease the relative free energy of a kinetic checkpoint termed the ajar conformation, and thereby slow the incorporation of incorrect nucleotides. One-pot reactions were studied in order to develop procedures for the formation of important carbon-carbon and carbon-nitrogen bonds that are easily reproducible. In the presence of trimethylsilyl triflouromethanesulfonate (TMSOTf), Mukaiyama-aldol and aldol-like reactions occur in one-pot due to TMSOTf acting as a Lewis acid and silylating agent. A variety of reactions, including methoxyalkylation, were performed to form carbon-nitrogen bonds. Here, research involving TMSOTf-mediated additions to acetals, nitrones, and aminals will be discussed.

After the widespread infection of the American chestnut tree (Castanea dentata) due to the fungus Cryphonectria parasitica the species was nearly brought to extinction and now merely exists as stump sprouts. Since the fungus infects the bark of the tree but not the root systems new shoots are able to emerge and grow before once again being girdled by the fungus. Cryphonectria parasitica is believed to have been brought to America on Asian species of chestnut which possess resistance to the blight caused by the fungus. Laromustine is a sulfonylhydrazine anticancer prodrug whose cytotoxicity results from the formation of interstrand cross-links caused by the synergistic action of co- generated 2-chloroethylating and carbamoylating species. The cytotoxic activities of Laromustine involve the induction of apoptosis. Described herein is an investigation into this drug’s effects on apoptotic gene expression in HL-60 cells using qRT-PCR. Significant changes in the expression levels of 13 genes were observed, most dramatically in the upregulation of the bcl2-associated athanogene 3 (BAG3) gene. Recognition of structural features of DNA such as gaps, nicks and abasic sites is critical for many DNA-binding proteins, such as those involved in DNA damage repair. This study explores a novel strategy to model these protein-DNA interactions using phage display. Phage display has been commonly used to identity zinc fingers that bind to DNA sequences, but selection of peptides that binding to specific DNA structures has not been reported in the literature. Choline is essential to the development and function of the central nervous system. Supplemental choline is neuroprotective against a variety of insults, including neurotoxins like dizocilpine (MK-801). MK-801 is an NMDA receptor antagonist that is frequently used in rodent models of psychological disorders. At low doses, it causes cognitive impairments, and at higher doses it induces motor deficits, anhedonia, and neuronal degeneration. Amphibian populations have been declining for several decades, in part due to the emerging fungal skin disease, chytridiomycosis, caused by the chytrid fungus Batrachochytrium dendrobatidis (Bd). Chytridiomycosis has caused extinctions and extirpations in many parts of the world, but its influence varies across species, populations, and individuals. The Green Frog (Lithobates clamitans), which inhabits the eastern half of the United States, seems capable of sustaining Bd infections without experiencing die-offs. Mechlorethamine (HN2), a nitrogen derivative of mustard gas, was the first synthetic anti-tumor chemotherapeutic because it forms covalent cross-links between strands of duplex DNA. HN2 represents a class of bifunctional alkylating agents that are both chemotherapeutic and carcinogenic: diepoxybutane (DEB), the active form of the pro-drug treosulfan, and epichlorohydrin (ECH), a structural hybrid of HN2 and DEB, also form covalent cross-links between DNA. Comprised mostly of university-educated women, FEMEN has received international media coverage, encountered controversy, and received death threats for their topless protests. For their activism, French magazine Madame Figaro ranked one of the founding members Inna Schevchenko on the 13^th position in their list of Women of the Year (Madame Figaro 2012). They have delivered lectures on their movement at international forums and universities. FEMEN now has chapters in Brazil, Tunisia, France and Germany. Conservation easements are legal agreements through which landowners donate or sell some or all of their properties' development rights to conservation entities, typically government entities or land trusts. Scytovirin is a novel anti-viral protein isolated from the cyanobacterium Scytonema varium. This protein has produced great scientific interest for its anti-human immunodeficiency virus (HIV) activity. Scytovirin disrupts the mechanism of HV infection by binding to HIV-1 proteins glycoprotein 120, glycoprotein 160, and glycoprotein 41. It does not bind to the CD4 receptor on human immune cells. The wild-type protein consists of a single 95 amino acid chain. When folded, Scytovirin's amino acid sequence is arranged into two distinct domains, each with a high degree of sequence conservation. The goal of this thesis is to analyze the effects of hydrogen bonding on liquid 1H- 1,2,3-triazole. Spectra of neat 1H-1,2,3-triazole are compared to simulated spectra and are found to differ greatly. The experimental spectrum, however, matches the simulated spectra of 1H-1,2,3-triazole dimers very well. This suggests that dimers play a large role in the liquid structure of 1H-1,2,3-triazole. Though tautomerism is thought to play a large role in the vibrational behavior of 1,2,3- triazole, the Raman spectra obtained here suggest that molecular dimers dominate the liquid state much more so than tautomerism. As a dual behavioral and computational neuroscience research project, this study first tested reproductive effects on attention, learning, and decision making using the Attentional Set-Shifting Task (AS-ST) and then a Machine Learning model was constructed to simulate perceptual judgments and decision making through reinforced learning. In the behavioral task, response times and errors from 5 primiparous (one-time mothers) and 4 nulliparous (never pregnant) Sprague-Dawley rats were recorded during four increasingly complex attention modulation and paired associative learning tasks. This thesis examined the roles of speech-language pathologists (SLPs) within the Neonatal Intensive Care Unit (NICU). In 2004, the American Speech-Language-Hearing Association published a set of policy documents regarding speech-language pathology practice in the NICU. Research compiled in the literature review explained and supported each of the roles listed in the ASHA policy documents. The literature also revealed that very few sources specific to the speech-language pathology field addressed roles other than those related to feeding and swallowing evaluation and intervention.
