Umingmakstrongylus Pallikuukensis (Nematoda: Protostrongylidae) In Gastropods: Larval Morphology, Morphometrics, And Development Rates, Susan J. Kutz, Eric P. Hoberg, Lydden Polley

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Morphological and morphometric aspects of larval development of Umingmakstrongylus pallikuukensis in Deroceras laeve and the effects of temperature on development rates in D. laeve and Deroceras reticulatum were investigated in the laboratory. Larval stages were best differentiated by separation of cuticular sheaths, tail structure, and viability following digestion. Growth in body and esophagus width was observed during the first-stage within the intermediate host, but the major increases in body length and width occurred immediately following the second molt. Larval development in D. laeve and D. reticulatum occurred more rapidly at warmer temperatures. The calculated threshold temperatures were 8.5 and 9.5 …

Worms, Nematoda, Scott Lyell Gardner

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Nematodes are the most speciose phylum of metazoa on earth. Not only do they occur in huge numbers as parasites of all known animal groups, but also they are found in the soils, as parasites of plants, and in large numbers in the most extreme environments, from the Antarctic dry valleys to the benthos of the ocean. They are extremely variable in their morphological characteristics, with each group showing morphological adaptations to the environment that they inhabit. Soil-dwelling forms are extremely small; many marine species have long and complex setae; and parasitic species manifest amazingly great reproductive potential and large …

Phylogenetic Position Of Eimeria Antrozoi, A Bat Coccidium (Apicomplexa: Eimeriidae) And Its Relationship To Morphologically Similar Eimeria Spp. From Bats And Rodents Based On Nuclear 18s And Plastid 23s Rdna Sequences, Xiaomin Zhao, Donald W. Duszynski, Eric S. Loker

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Partial plastid 23S and nuclear 18S rDNA genes were amplified and sequenced from 2 morphologically similar Eimeria species, E. antrozoi from a bat (Antrozous pallidus) and E. arizonensis from deer mice (Peromyscus spp.), as well as some other Eimeria species from bats and rodents. The phylogenetic trees clearly separated E. antrozoi from E. arizonensis. The phylogenies based on plastid 23S rDNA data and combined data of both plastid and nuclear genes grouped 2 bat Eimeria and 3 morphologically similar Eimeria species from rodents into 2 separate clades with high bootstrap support (100%, 3 rodent Eimeria species; …

Phylogenetic Analysis Of The Rhabdocoela (Platyhelminthes) With Emphasis On The Neodermata And Relatives, David Zamparo, Daniel R. Brooks, Deborah A. Mclennan, Eric P. Hoberg

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Phylogenetic systematic analysis of 24 taxa representing the rhabdocoel platyhelminths, based on a suite of 89 morphological characters, produced two equally parsimonious trees, 181 steps long, with a consistency index (CI) of 0.69 and a rescaled consistency index (RCI) of 0.56, differing only with respect to that portion of the tree containing Umagillidae, Acholadidae, Graffillinae, Pseudograffillinae, Pterastericolidae and Hypoblepharinidae. Our results accommodate all previously proposed sister taxa to the Neodermata in a single clade in which ((Dalyelliidae + Temnocephalida) Typhloplanidae) is the sister group of ((Fecampiidae + Urastoma) (Udonella ((Aspidogastrea + Digenea) (Monogenea (Gyrocotylidea (Amphilinidea + Eucestoda)))))). Prevalence Of Eimeria (Apicomplexa: Eimeriidae) In Reintroduced Gunnison's Prairie Dogs (Cynomys Gunnisoni), M. M. Ryan, K. H. Decker, Donald W. Duszynski

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Fecal samples from 54 Gunnison’s prairie dogs (Cynomys gunnisoni) from Albuquerque, NM were analyzed for the presence of coccidia and all were positive. They were then relocated to an abandoned prairie dog town on the Sevilleta Long Term Ecological Research (LTER) site. Six Eimeria species, E. callospermophili, E. cynomysis, E. pseudospermophili (new host record), E. spermophili, E. ludoviciani and E. vilasi (new host record) were found in Albuquerque animals, but only two species, E. callospermophili and E. vilasi were present in relocated hosts. A significant (P < 0.05) reduction was seen in the prevalence of E. vilasi (72% vs. 13%) and in the prevalence of infections …

Phylogey Of Steinernema Travassos, 1927 (Cephalobina: Steinnematidae) Inferred From Ribosomal Dna Sequences And Morphological Characters, S. Patricia Stock, James F. Campbell, Steven A. Nadler Jan 2001

Phylogey Of Steinernema Travassos, 1927 (Cephalobina: Steinnematidae) Inferred From Ribosomal Dna Sequences And Morphological Characters, S. Patricia Stock, James F. Campbell, Steven A. Nadler

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Entomopathogenic nematodes in Steinernema, together with their symbiont bacteria Xenorhabdus, are obligate and lethal parasites of insects that can provide effective biological control of some important lepidopteran, dipteran, and coleopteran pests of commercial crops. Phylogenetic relationships among 21 Steinernema species were estimated using 28S ribosomal DNA (rDNA) sequences and morphological characters. Sequences of the rDNA internal transcribed spacers were obtained to provide additional molecular characters to resolve relationships among Steinernema carpocapsae, Steinernema scapterisci, Steinernema siamkayai, and Steinernema monticolum. Four equally parsimonious trees resulted from combined analysis of 28S sequences and 22 morphological characters. Clades inferred from …

Phylogenetic Relationships Among Species Of Stilestrongylus Freitas, Lent And Almeida, 1937 (Trichostrongyloidea: Heligmonellidae: Nippostrongylinae), Parasites Of Myomorph Rodents (Rodentia: Muridae) In The Neotropics, Gerardo Perez-Ponce De Leon, Scott Lyell Gardner, Jorge Falcon-Ordaz

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The phylogenetic relationships of 14 species of Stilestrongylus were analyzed using the comparative morphology of 21 characters. We obtained 2 shortest trees of 50 steps, with a consistency index of 0.540 and 25 apomorphic character states. Ingroup monophyly was supported on these trees by 2 and 3 synapomorphies, respectively, and the ingroup was defined by the following characters: greater number of ventral ridges relative to the number of ridges dorsally, asymmetric bursa, and externodorsal rays differing in size. Sister-group relationships among Stilestrongylus and the other genera designated as outgroups are relatively consistent with those postulated by Durette-Desset’s in a classification …

Emergence Of Third-Stage Larvae Of Umingmakstrongylus Pallikuukensis From Three Gastropod Intermediate Host Species, Susan K. Kutz, Eric P. Hoberg, Lydden Polley

Harold W. Manter Laboratory of Parasitology: Faculty Publications

We investigated the emergence of third-stage larvae (L3) of Umingmakstrongylus pallikuukensis from the slugs Deroceras laeve, Deroceras reticulatum, and the snail Catinella sp. in the laboratory and from D. laeve on the tundra. Third-stage larvae emerged from 8 of 8 D. laeve and 8 of 8 D. reticulatum housed at 20°C in darkness and from 9 of 10 D. laeve and 5 of 5 Catinella sp. housed at 21°C with 10-12 hours of light/day. Larvae emerged from D. laeve and D. reticulatum over a wide range of infection intensities (2-179 and 20-65, respectively), and the patterns of emergence …

Phylogeny Of The Species Of The Genus Litomosoides (Nematatoda: Onchocercidae): Evidence Of Rampant Host Switching, Sara V. Brant, Scott Lyell Gardner Jun 2000

Phylogeny Of The Species Of The Genus Litomosoides (Nematatoda: Onchocercidae): Evidence Of Rampant Host Switching, Sara V. Brant, Scott Lyell Gardner

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Filarioid nematodes of the genus Litomosoides occur in the abdominal and (or) thoracic cavities of marsupials, rodents, and bats of the Nearctic and Neotropical regions. In this study, the phylogenetic relationships among these nematodes were estimated with a parsimony analysis of morphological characters derived from species descriptions. This nonweighted analysis produced 20 shortest trees. The monophyly of the genus was not supported in that Litomosoides thomomydis and Litomosoides westi failed to group with the other members of the genus. When these 2 taxa (parasites of pocket gophers) were excluded, monophyly of Litomosoides was supported by 2 synapomorphies (structure of the …

Reassignment Of Lamanema From Nematodirinae To Molineinae (Nematoda: Trichostrongyloidea) (Research Notes), Lora G. Rickard, Eric P. Hoberg Jun 2000

Reassignment Of Lamanema From Nematodirinae To Molineinae (Nematoda: Trichostrongyloidea) (Research Notes), Lora G. Rickard, Eric P. Hoberg

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The monospecific Lamanema historically has been assigned to the Nematodirinae within the Molineidae. Inconsistencies in morphological characters, within a phylogenetic context for Nematodirinae, led to a re-evaluation of the putative relationships and taxonomic placement of Lamanema. Among 7 putative synapomorphies for Nematodirinae, Lamanema possesses only one, large eggs. Large eggs, sporadically present in phylogenetically disparate taxa of trichostrongyles, are equivocal with respect to placement of Lamanema; it is considered that possession of this single homoplasious character alone is insufficient justification to retain the genus in Nematodirinae. Affinities with the Trichostrongylidae (Cooperiinae or Haemonchinae) have also been proposed; however, …

Relationships Of Nematodirus Species And Nematodirus Battus Isolates (Nematoda: Trichostrongyloidea) Based On Nuclear Ribosomal Dna Sequences, Steven A. Nadler, Eric P. Hoberg, Deborah S.S. Hudspeth, Lora G. Rickard Jun 2000

Relationships Of Nematodirus Species And Nematodirus Battus Isolates (Nematoda: Trichostrongyloidea) Based On Nuclear Ribosomal Dna Sequences, Steven A. Nadler, Eric P. Hoberg, Deborah S.S. Hudspeth, Lora G. Rickard

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Nuclear ribosomal sequence data from the internal transcribed spacers (ITS-1 and ITS-2), 5.8S subunit, and regions of the 18S and 28S genes were used to investigate sequence diversity among geographic samples of Nematodirus battus, and to infer phylogenetic relationships among Nematodirus species. Phylogenetic analysis of these data yielded strong support for relationships among species, depicting Nematodirus helvetianus and Nematodirus spathiger as sister-taxa and a clade of these two species and Nematodirus filicollis. This tree is consistent with caprine bovids as ancestral hosts, with a subsequent host shift to Bovinae in N. helvetianus. Eleven of 14 N. battus …

Natural Transfer Of Helminths Of Marine Origin To Freshwater Fishes, With Observations On The Development Of Diphyllobothrium Alascence, Robert L. Rausch, Ann M. Adams

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Infective stages of helminths of 5 species that occur as adults in marine mammals were found in burbot, Lota lota (L.) (Gadidae), from the lower Kuskokwim River (southwestern Alaska): Diphyllobothrium alascense Rausch et Williamson, 1958; Pyramicocephalus phocarum (Fabricius, 1780); Corynosoma strumosum (Rudolphi, 1801); Corynosoma semerme (Forsell, 1904); and Pseudoterranova decipiens (Krabbe, 1878). Some larval stages were obtained also from smelt, Osmerus mordax dentex Steindachner, an anadromous fish important as prey of burbot. Burbot, which are freshwater fish, could become paratenic hosts of those helminths by means of at least three interactions: by consuming marine fishes in brackish waters at river …

Helminthoxys Abrocomae N. Sp. (Nematoda: Oxyurida) From Abrocoma Cinerea In Bolivia, Jean-Pierre Hugot, Scott Lyell Gardner

Harold W. Manter Laboratory of Parasitology: Faculty Publications

A new pinworm parasite is described from Abrocoma cinerea, a caviomorph rodent of the superfamily Octodontoidea from the Andes of Bolivia. The new species, Helminthoxys abrocomae n. sp., possesses special secretory mamelons which we consider a synapomorphy of the genus Helminthoxys. Within Helminthoxys, the closest relatives are found in octodontoid rodents: H. gigantea occurs in Octodon degus in Chile and O. bridgesi in Argentina, and H. freitasi is a parasite of Thrichomys aperoides in Brazil. H. abrocomae n. sp. differs from both other species morphometrically in relation to different parts of the body in both sexes, particularly …

A Phylogenetic Hypothesis For Species Of The Genus Taenia (Eucestoda: Taeniidae), Eric P. Hoberg, Arlene Jones, Robert L. Rausch, Keeseon S. Eom, Scott Lyell Gardner Feb 2000

A Phylogenetic Hypothesis For Species Of The Genus Taenia (Eucestoda: Taeniidae), Eric P. Hoberg, Arlene Jones, Robert L. Rausch, Keeseon S. Eom, Scott Lyell Gardner

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Cladistic analysis of a numerical data matrix describing 27 characters for species of Taenia resulted in four most parsimonious phylogenetic trees (174 steps; consistency index = 0.28; homoplasy index = 0.72; retention index = 0.48). Monophyly for Taenia is diagnosed by the metacestode that is either a cysticercus or a form derived from a bladder-like larva; no other unequivocal synapomorphies are evident. Tree structure provides no support for recognition of a diversity of tribes or genera within the Taeniinae: Fimbriotaeniini and Taeniini have no phylogenetic basis. Hydatigera, Fimbriotaenia, Fossor, Monordotaenia, Multiceps, Taeniarhynchus, Tetratirotaenia must …

Meningeal Worm In Deer From Western Nebraska, David W. Oates, Mauritz C. Sterner, Ed Boyd

Harold W. Manter Laboratory of Parasitology: Faculty Publications

One hundred seventy-eight whitetailed deer (Odocoileus virginianus) and 275 mule deer (Odocoileus hemionus) collected from locker plants in the western 2/3 of Nebraska (USA) in November 1997 were examined for the meningeal worm (Parelaphostrongylus tenuis). Parelaphostrongylus tenuis was identified in 17 (10%) of 168 white-tailed deer and in one (<1%) of 273 mule deer. This is the first naturally occurring infection of P. tenuis recorded in a mule deer.

Molecular And Morphometric Evidence For Separate Species Of Uncinaria (Nematoda: Ancylostomatidae) In California Sea Lions And Northern Fur Seals: Hypothesis Testing Supplants Verification, Steven A. Nadler, Byron J. Adams, Eugene T. Lyons, Robert L. Delong, Sharon R. Melin Jan 2000

Harold W. Manter Laboratory of Parasitology: Faculty Publications

California sea lions (Zalophus californianus) and northern fur seals (Callorhinus ursinus) are each believed to host distinct hookworm species (Uncinaria spp.). However, a recent morphometric analysis suggested that a single species parasitizes multiple pinniped hosts, and that the observed differences are host-induced. To explore the systematics of these hookworms and test these competing hypotheses, we obtained nucleotide sequences of nuclear ribosomal DNA (D2/D3 28S, D18/D19 28S, and internal transcribed spacer [ITS] regions) from 20 individual hookworms parasitizing California sea lion and northern fur seal pups where their breeding grounds are sympatric. Five individuals from an …

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The genus Mesocestoides Vaillant, 1863 includes tapeworms of uncertain phylogenetic affinities and with poorly defined life histories. We previously documented 11 cases of peritoneal cestodiasis in dogs (Canis familiaris L.) in western North America caused by metacestodes of Mesocestoides spp. In the current study, DNA sequences were obtained from metacestodes collected from these dogs (n = 10), as well as proglottids from dogs (n = 3) and coyotes (Canis latrans Say, 1823 [n = 2]), and tetrathyridia representing laboratory isolates of M. corti (n = 3), and these data were analyzed phylogenetically. Two nuclear genetic markers, 18S ribosomal …

Phylogeny Of The Ascaridoidea (Nematoda: Ascaridida) Based On Three Genes And Morphology: Hypotheses Of Structural And Sequence Evolution, Steven A. Nadler, Deborah S. S. Hudspeth

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Ascaridoid nematodes parasitize the gastrointestinal tract of vertebrate definitive hosts and are represented by more than 50 described genera. We used 582 nucleotides (83% of the coding sequence) of the mitochondrial gene cytochrome oxidase subunit 2, in combination with published small- and large-subunit nuclear rDNA sequences (2,557 characters) and morphological data (20 characters), to produce a phylogenetic hypothesis for representatives of this superfamily. This combined evidence phylogeny strongly supported clades that, with 1 exception, were consistent with Fagerholm’s 1991 classification. Parsimony mapping of character states on the combined evidence tree was used to develop hypotheses for the evolution of morphological, …

Phylogenetic Relationships Among Species Of Contracaecum Railliet & Henry, 1912 And Phocascaris Høst, 1932 (Nematoda: Ascaridoidea) Based On Nuclear Rdna Sequence Data, Steven A. Nadler, S. D'Amelio, H. P. Fagerholm, B. Berland, L. Paggi

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Nuclear-encoded large-subunit ribosomal DNA sequences were used to infer a phylogenetic hypothesis for 17 taxa (16 nominal species) of the genera Contracaecum and Phocascaris. Phylogenetic trees based on these data have been used to assess the validity of the taxonomic distinction between these genera, which was based on the presence or absence of certain structural features, rather than on explicit hypotheses of evolutionary history. Phylogenetic hypotheses based on parsimony, likelihood, and neighbor-joining analyses of these sequence data strongly support the hypothesis that species of Phocascaris are nested within the clade of Contracaecum species hosted by phocid seals, and are …

Molecular Phylogeny Of Clade Iii Nematodes Reveals Multiple Origins Of Tissue Parasitism, Steven A. Nadler, R. A. Carreno, H. Mejía-Madrid, J. Ullberg, C. Pagan, R. Houston, Jean-Pierre Hugot

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Molecular phylogenetic analyses of 113 taxa representing Ascaridida, Rhigonematida, Spirurida and Oxyurida were used to infer a more comprehensive phylogenetic hypothesis for representatives of ‘clade III’. The posterior probability of multiple alignment sites was used to exclude or weight characters, yielding datasets that were analysed using maximum parsimony, likelihood, and Bayesian inference methods. Phylogenetic results were robust to differences among inference methods for most high-level taxonomic groups, but some clades were sensitive to treatments of characters reflecting differences in alignment ambiguity. Taxa representing Camallanoidea, Oxyurida, Physalopteroidea, Raphidascarididae, and Skrjabillanidae were monophyletic in all 9 analyses whereas Ascaridida, Ascarididae, Anisakidae, Cosmocercoidea, …

A Cox2 Molecular Phylogeny Of The Peronosporomycetes, Deborah S.S. Hudspeth, Steven A. Nadler, Michael E.S. Hudspeth

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The evolutionary history of the mitochondrial COX2 locus has been used to infer the phylogenetic relationships among 15 peronosporomycete and a hyphochytriomycete species. This molecular phylogenetic analysis at both the ordinal and generic levels provides strong evidence for the recognition of the Saprolegniomycetidae and the Peronosporomycetidae as natural groups, and for the monophyly of the Saprolegniales, Leptomitales and Pythiales. A three amino acid insertion/deletion event (indel) has been identified as a putative synapomorphy for the Saprolegniales. Parsimony mapping of 12 morphological and biochemical characters on the COX2 molecular phylogeny yields an hypothesis for peronosporomycete ancestral states and shared-derived features.

Elaeophorosis In Red Deer From Spain, Mónica Santín-Durán, J. M. Alunda, J. M. San Miguel, Eric P. Hoberg, C. De La Fuente

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Elaeophorosis, caused by Elaeophora elaphi, was observed in red deer (Cervus elaphus) from Toledo Province (Spain) for the first time. Adult specimens of Elaeophora elaphi were found in the hepatic vessels of nine of 151 red deer between October 1994 and September 1995; intensity of infection was two to 18 nematodes per host. Adult nematodes were only found during the period from fall through early spring. No differences were present between sex or age groups. Parasites were not found in a limited sample from fallow deer (Dama dama). Blood samples were negative for the presence of …

The authors review the major biological, biochemical, and molecular characters that are used to distinguish the seven Trichinella species (T. spiralis, T. nativa, T. britovi, T. pseudospiralis, T. murrelli, T. nelsoni, T. papuae) and three genotypes whose taxonomic status is yet uncertain (T-6, T-8, T-9). A comparison of host specificity, morphology, reproductive abilities, nurse cell development and freeze resistance is presented, along with useful biochemical and molecular markers. Finally, this information is used to construct a diagnostic key for the species. A phylogenetic classification of the species is needed.

Fresh fecal samples from 5 caecilians (Dermophis mexicanus) were collected and examined for coccidia in the summer of 1998. The caecilians were collected in the Department of San Marcos, Guatemala. Two of the 5 (40%) specimens of caecilians contained an Eimeria species that is described here as new. This represents the first coccidia described from a gymnophionian host. Sporulated oocysts are spheroidal to subspheroidal, 19.5 × 17.7 (16–23 × 15–21) μm, micropyle and oocyst residuum are absent, and 3 (or more) polar granules are always present. Sporocysts are ovoidal, 11.0 × 7.2 (10–12 × 6–9); a Stieda body …

The coccidia (protistan phylum Apicomplexa Levine, 1970) comprise a large group of obligate intracellular parasites commonly found in all classes of vertebrate hosts and in some invertebrates. This review focuses on those species within the Eimeriidae because they are among the most prevalent and speciose of all parasite groups, and there is no taxonomic summation currently available for those species that infect insectivores. All published species descriptions in the genera Cyclospora, Eimeria and Isospora that infect insectivores are reviewed and evaluated. Some of the named species are invalid, either because rules concerning the naming of new species (International …
