Proton Coupled Electron Transfer (PCET) reactions play a vital rote in a variety of biological and chemical processes. Prior work by E. Laviron developed the theory of proton coupled electron transfer kinetics based on a stepwise mechanism with the transfer coefficient for electron transfer remaining constant at all overpotentials. The present work explores the consequences of a potential-dependent transfer coefficient as applied to redox couples attached to electrodes. Specifically, this work focuses on the 1-electron, 1-proton (1e1H) system. Neurons using gamma-aminobutyric acid (GABA) as a neurotransmitter are critically important as inhibitory neurons in the CNS. A major hindrance to further studies of this important and highly diverse class of neurons is their relative scarcity, which makes it difficult to identify and target them for morphological and physiological studies. To overcome this difficulty, we developed a novel protocol for visualizing detailed morphology of GABAergic neurons in living mouse brain slice cultures, using biolistic transfection with GAD67-GFP. The morphological, immunocytochemical and physiological characteristics of GAD67-GFP expressing neurons indicated that they were functional GABAergic neurons. This dissertation consists of three essays on trade policy and factor mobility. The first essay of the dissertation deals with issues on trade policy. It focuses on the possibility of recipient immiserization under optimal export taxation in a multi-country framework. We find asymmetry between two nations is very crucial for recipient immiserization. The interesting finding is that being more aggressive in taxation is a disadvantage in gaining from receipts of transfers. The larger exporter exercises more market power and is also more vulnerable to the adverse welfare effects of transfer.;The second and third essays explore the issues of trade policy …

Blast resistant structures are gaining increased need in today's society. This research addresses adding composite reinforcement to existing concrete structures to decrease blast damage. An upgraded material model for concrete is presented. This model, used in LS-DYNA3D, incorporates tensile damage, which allows the concrete to degrade as it is loaded. The concrete model is tested using existing unreinforced and reinforced concrete beam tests. Some beams were plain, others were externally reinforced with composites, bonded to the exterior of the beams. There is a very good correlation between the predicted and existing data. Glass fibers are commonly added to thermoplastics by the process of extrusion compounding for a variety of reasons, mainly to enhance their strength and make them dimensionally stable. Since the extruder has to be flushed out each time product composition is changed, a large amount of incompatible polymeric waste is generated. This waste material is usually landfilled even though the polymers contained in it are valuable and worth being recycled. Uric acid is an important antioxidant due to its role as a free radical scavenger, and methods to elevate its plasma concentration may be important in animal health. In a preliminary study, the effect of dietary protein on plasma uric acid (PUA) was determined in 3-wk-old chicks. Twenty-four broiler chicks were randomly assigned to 4 diets: low protein (LP) containing 10% casein, medium protein (MP) containing 20% casein, high protein (HP) containing 45% casein and a commercial diet (COD) for 2-wks. The objective of this research was to determine if disease on susceptible tomato cultivars is reduced by intercropping with resistant cultivars. A second objective was to investigate the mechanism of the disease reduction observed. Early blight (caused by Alternaria solani) rate of disease increase and lesion expansion were lower on susceptible tomato cv. 'Brandywine' when intercropped with resistant cv. 'Juliet' than with 'Brandywine' monoculture in both the field and greenhouse. Yield from 'Brandywine' plants was 17.3% greater when intercropped with 'Juliet' than when grown in monoculture. Reduction in lesion expansion on 'Brandywine' when intercropped with 'Juliet' suggests an interaction initiating …

The heat shock proteins (hsps) are believed to provide protection against a variety of insults such as elevated temperature, heavy metals and chemical agents. The goal of this study was to determine if the immortalized HK-2 human proximal tubule cell line could provide a model system to study the stress response of the proximal tubule cell. Heat stress, elevated temperature at 42.5°C for 1 hr, caused a marked increase only in hsp 70 mRNA and protein, but not that of hsp 27 or hsp 60 mRNA and protein. Two first to second order streams (Kent Run and Munnell Hollow) at Ryerson Station State Park, Greene County, Pennsylvania, were compared as to water parameters (dissolved oxygen, hardness, temperature, pH, and current velocity), species diversity, and abundance of both immature and adult insects. Samples were taken biweekly for one year from May, 1999 through April, 2000 for a total of 26 sampling periods. Data were analyzed using a repeated measures ANOVA. Kent Run had lower dissolved oxygen, higher pH and hardness, lower temperatures, and higher current velocity. A total of 137 taxa were collected at both streams. Of the total …

Despite the successes, there are threats to the long term stability of THA, chief among them aseptic loosening of the femoral stem. A Florida dolomitic phosphate pebble sample was processed using a 1.2 liter stirrer-tank flotation cell and 2-in ID packed flotation column aiming at achieving a phosphate concentrate containing more than 30% P2O 5 and less than 1.0% MgO contents at high P2O5 recovery.;The characteristics of fatty acid collector, FA-12 and a newly developed PA-31 collector were evaluated for frothability and dolomite flotation rate. The evaluation results show that both reagents have high frothability, but PA-31 has much stronger frothability than FA-12, and higher selectivity in separating dolomite from phosphate mineral.;The mineralogical study indicates that the phosphate pebble sample needs to …

A model single phase loop thermosiphon was designed and built such that velocity profiles and heat transfer rates could be studied by obtaining temperature data, along with a detailed set of non-intrusive Laser Doppler Velocimetry measurements. The quantities measured were the instantaneous velocity profiles for the water flow. These velocities were then reduced into sets of mean and root mean square velocities, for different angles of tilt of the experimental setup. The present study assessed the construct validity of the Sport Disengagement Questionnaire (SDQ) through a series of factor analyses and examined demographic variables in relation to total SDQ scores. The overall sample was comprised of 400 collegiate athletes representing five National Collegiate Athletic Association (NCAA) Division I universities and one NCAA Division II university across a total of 17 varsity sports. This study examined the relationship between the preferred leadership styles, the levels of computer anxiety, and the selected personal attributes of sex, age, and computer training upon the computer use of West Virginia public school principals. A one shot case study design was used to examine the relationships. As there are over twenty potential PKC phosphorylation sites in AFAP-110 structure, deletional mutagenesis was employed to determine which regions of AFAP-110 are most likely to contain crucial sites for PKC phosphorylation. Our data indicate that functional PKC phosphorylation sites must be localized to the amino acid stretch 266--294 within STK region of AFAP-110. There are four PKC candidate phosphorylation sites in this region; they are being substituted by alanine residues in order to determine their significance in serine/threonine phosphorylation regulation of AFAP-110 function.;Analysis of AFAP-110 amino acid sequence has also revealed that AFAP-110 contains twelve potential 14-3-3 binding sequences. …

Soil series occurring above 1020 m in elevation in the Monongahela National Forest in West Virginia were sampled and analyzed for soil organic and soil microbial biomass carbon as well as various fertility parameters. Additionally, vegetative biomass estimates were made at the sampling sites. The soil series sampled (established and tentative) are frigid, well and moderately-well drained inceptisols and spodosols, covered by northern hardwood or spuce-dominated forests. A longtitudinal study was performed in prepubertal heifer calves to determine when the individual components of the hypothalamic-pituitary-ovarian axis were capable of functioning. In study 1, the hypothalamus and anterior pituitary of prepubertal dairy heifers were challenged with exogenous hormones (estradiol and GnRH, respectively) to determine at what age gonadotropin surges could be induced. Two-, four-, six-, and eight-month-old heifer calves were challenged with GnRH or estradiol. Blood was sampled from the jugular vein every 15 minutes for 6 hours or hourly for 30 hours, for the two treatments, respectively. Heifers in each age group released LH in response to …

More specifically, this researcher investigated the social contagion of eating attitudes and behaviors among first year college women living in residence hall communities. The results demonstrated that college women developed eating attitudes and behaviors similar to those of other women in their small friendship clusters. Similarities in eating attitudes and behaviors were not found among participants in the study when residence hall floor membership was used as the independent variable. During 1998--2000, I examined the relationship between the reproductive success of a declining Neotropical migrant songbird, the wood thrush ( Hylocichla mustelina), and surrounding habitat within the highly forested region encompassing the northwestern portion of the Monongahela National Forest, West Virginia, USA. I measured nest fate, food delivery rates, and nest attendance rates of 56 videotaped nests and then related these variables to habitat measured at three concentric scales: nest site (11.3 m radius), territory (100 m), and landscape (1000 m). The study was conducted to investigate and model phosphorus sorption in soils and to study the effectiveness of acid mine drainage (AMD) floc in controlling phosphorus (P) loss to surface runoff from soils. Phosphorus sorption capacity of surface and subsurface horizons of four benchmark soil series - Berks, Huntington, Lindside, and Monongahela - of West Virginia was measured. The soils of these four series were loaded with increasing amounts of P up to 60% of their P retention capacity.
