Systematic Analysis Of Carbon Nanotubes Toxicity In Cellular Systems, Gabrielle Rogers-Nieman
Systematic Analysis Of Carbon Nanotubes Toxicity In Cellular Systems, Gabrielle Rogers-Nieman
Graduate Theses, Dissertations, and Problem Reports (ETD)
As the applied use for carbon nanotubes (CNTs) increases across engineering and biomedical sectors, a clear understanding of the deleterious effects surrounding CNT-induced toxicity, namely genotoxicity, is required. Currently, the exact genotoxic mechanisms that surround CNT-induced mitotic disruption that result in aneuploidy are not completely understood. Thus, there is a need for clear mechanical models to be formulated. Herein a comprehensive overview of the nanotube toxicity is provided insisting on the aspects related to CNTs genotoxicity.;In Chapter 1, I am providing a review of the current research in the CNT-s induced toxicity. I start by defining these members of the …
From Cellular Transport To Synthetic Biomimetic Transport Using Carbon Nanotube - Actin Hybrid Assemblies, Zahra Ronaghi
From Cellular Transport To Synthetic Biomimetic Transport Using Carbon Nanotube - Actin Hybrid Assemblies, Zahra Ronaghi
Graduate Theses, Dissertations, and Problem Reports (ETD)
One of the many interesting materials that have emerged in the field of nanotechnology in the last quarter century is Carbon Nanotube (CNT). CNTs have been explored in a broad range of fields from electronic devices and biosensors, to bioimaging and tissue engineering. However, as stand-alone materials CNTs have limited capabilities in the field of biology and medicine unless they are combined with biological agents. Due to the similarity in diameters, CNTs can be combined with biomolecules such as enzymes, antibodies, antigens, DNA, etc. These "hybrid" assemblies will combine the properties of the CNTs with the recognition characteristics and functions …

Graduate Theses, Dissertations, and Problem Reports (ETD)
Breast cancer comprises about 29% of all types of cancer in women worldwide. This type of cancer caused what is equivalent to 14% of all female deaths due to cancer. Nowadays, tissue biopsy is routinely performed, although about 80% of the performed biopsies yield a benign result. Biopsy is considered the most costly part of breast cancer examination and invasive in nature. To reduce unnecessary biopsy procedures and achieve early diagnosis, ultrasound elastography was proposed.;In this research, tissue displacement fields were estimated using ultrasound waves, and used to infer the elastic properties of tissues. Ultrasound radiofrequency data acquired at consecutive …
Characterization And Application Of Self-Assembled Nano-Materials For Fluid Steering And Separation In Microfluidics, Xingwei Wu
Graduate Theses, Dissertations, and Problem Reports (ETD)
Aqueous phospholipid preparations comprised of 1,2-dimyristoyl- sn-glycero-3- phosphocholine (DMPC) and 1,2-dihexanoyl-sn-glycero-3-phosphocholine (DHPC) are prevalent materials for biological characterization. The viscosity behavior of DMPC/DHPC was characterized as a function of q value (q= [DMPC]/[DHPC] ), hydration as well as temperature firstly on bench-top and then in microfluidic chip. The rheology of 20% phospholipid preparations of [DMPC]/[DHPC] = 2.5 reveals that, under conditions utilized for fluid steering, the materials are shear-thinning power-law fluids with a power-law index ranging from 0.30 through 0.90. Phospholipid preparations are utilized to steer fluids in microfluidic chips and support hydrodynamic delivery of sample across a double T …
Surfaces Modulate Β-Amyloid Peptide Aggregation Associated With Alzheimer's Disease., Elizabeth Anne Yates
Surfaces Modulate Β-Amyloid Peptide Aggregation Associated With Alzheimer's Disease., Elizabeth Anne Yates
Graduate Theses, Dissertations, and Problem Reports (ETD)
A hallmark of Alzheimer's disease, a late onset neurodegenerative disease, is the presence of neuritic amyloid plaques deposited within the brain composed of β-amyloid (Aβ) peptide aggregates. Aβ can aggregate into a variety of polymorphic aggregate structures under different chemical environments, specifically affected by the presence of differing surfaces. There are several point mutations clustered around the central hydrophobic core of Aβ (E22G Arctic mutation, E22K Italian mutation, D23N Iowa mutation, and A21G Flemish mutation). These mutations are associated with hereditary diseases ranging from almost pure cerebral amyloid angiopathy to typical Alzheimer's disease pathology with both plaques and tangles. To …
Mechanisms Of Intracellular Calcium Homeostasis In The Developing And Mature Bovine Corpora Lutea An In-Depth Study To Promote Scientific Literacy Through The Use Of Primary Literature In An Introductory Biology Course, Marietta Felicidad Wright
Graduate Theses, Dissertations, and Problem Reports (ETD)
Although it is generally accepted that calcium is an intracellular mediator of prostaglandin F2 alpha (PGF2alpha) actions on luteal cells, studies attempting to define the mechanisms of calcium homeostasis in bovine corpora lutea (CL) are lacking. The increase in intracellular calcium concentration ([Ca2+]i) induced by PGF2alpha in steroidogenic cells from mature CL is greater than in those isolated from developing CL. Our long-term hypothesis is that differences in signal transduction associated with developing and mature CL contribute to the increased efficacy of PGF2alpha to induce a calcium signal capable of mediating luteal regression in mature CL. Specifically, these studies were …

An In Vitro Microenvironment For Nucleus Pulposus Regeneration, Mark S. Shoukry
An In Vitro Microenvironment For Nucleus Pulposus Regeneration, Mark S. Shoukry
Graduate Theses, Dissertations, and Problem Reports (ETD)
Intervertebral disc degeneration begins early in life, increases with age, and can ultimately result in disc failure. At the center of the disc, the nucleus pulposus (NP) resides in a hypoxic environment where degeneration is thought to begin. Initially, nucleus pulposus cells (NPCs) display altered morphology and function, along with increased rates of cellular senescence and apoptosis. Though no widely accepted treatment exists, autologous NP implantation techniques have shown promise. However, the process is flawed as a result of the long in vitro expansion required to obtain sufficient numbers of cells. Prolonged expansion on plastic produces negative changes such as …

Graduate Theses, Dissertations, and Problem Reports (ETD)
Communicating and interacting with others is an essential part of our daily routines as humans. Performing these actions appropriately requires the ability to identify, extract, and process salient social cues from the environment. The subsequent application of such knowledge is important for inferring and predicting the behavior of other people. The eyes and brain must work together to fixate and process only the most critical social signals within a scene while passing over and / or completely ignoring other aspects of the scene. While brain activation to isolated presentations of objects and people presentations have been characterized, information about the …

An Evaluation Of The Occurrence Of Micronuclei And Other Nuclear Abnormalities In Fishes From The Great Lakes Basin, United States, Ryan Patrick Braham
Graduate Theses, Dissertations, and Problem Reports (ETD)
Biological markers (biomarkers) sensitive to genotoxic and mutagenic contamination in fishes are widely used to identify contamination in the aquatic environment. The "fish tumors or other deformities" biological use impairment (BUI) occurs at 18 of the 30 areas of concern (AOC) located in the Great Lakes basin within the United States. As each AOC evaluates this specific BUI for possible delisting, biomarkers sensitive to both genotoxic and mutagenic chemicals can be integrated as a possible criterion for delisting. The micronucleus assay identifies genotoxic contamination by observing the presence of a micronucleus (MN), along with a primary nucleus within the cellular …

Stream restoration is estimated to be a multi-billion dollar industry and includes techniques such as planting vegetation, modifying channel form, and installing in-stream structures. These techniques potentially alter the velocity field and flow complexity. The specific objective of this research was to characterize near-boundary flow within the three restored reaches of varying intensities located in the Stream Research, Education, and Management (StREAM) Laboratory at Virginia Tech. Three restoration techniques were applied in 2009-10 to consecutive reaches of Stroubles Creek: 1) cattle restriction only (0.5 km); 2) cattle restriction, banks reshaped to 3:1 slope and re-vegetated (0.6 km); and, 3) designed …
Riparian Ecological Community Assessment With An Emphasis On Wood Turtles (Glyptemys Insculpta) In The Cacapon River Watershed, West Virginia, Kathryn R. P. Mccoard
Riparian Ecological Community Assessment With An Emphasis On Wood Turtles (Glyptemys Insculpta) In The Cacapon River Watershed, West Virginia, Kathryn R. P. Mccoard
Graduate Theses, Dissertations, and Problem Reports (ETD)
Riparian zones serve to maintain high water quality, low water temperatures, and structural complexity in aquatic and terrestrial environments, among other beneficial services. Riparian buffers provide habitat and corridors linking forest patches for terrestrial wildlife. High riparian vegetative structure and complexity attract a high diversity of wildlife, including birds, herpetofauna, and small mammals. Devegetated riparian zones, often a result of developmental and agricultural practices, lose their beneficial functions and require restorative actions to regain them. The Cacapon River watershed in West Virginia is agriculturally-dominated with many areas of riparian zone degradation. A section of the Cacapon River was selected for …

Public Land Management Values: A Delphi Study Of Austrian And Pacific Northwest United States Recreation Resource Managers, Jessie Meybin
Graduate Theses, Dissertations, and Problem Reports (ETD)
There is a continuing global increase in recreational visitor use of public land. This rise in visitation can be a good thing if a proactive management plan utilizing strong methodology to anticipate impacts to an area and sustain its natural and social resources is in place. Although a wide spectrum of issues is presented to each management, few of the concepts are new to any of the administrators. With increasing duties, newly emerging concerns, and decreasing time, there is little room or patience for error concerning the implementation of any management derivatives or techniques. If the existing research on all …
