Situated in the Appalachian Mountains, one of the oldest mountain ranges on Earth, West Virginia waters boast rich ichthyofauna including native sportfish species walleye (Sander vitreus), largemouth bass (Micropterus nigricans), and muskellunge (Esox masquinongy). These three species are all native to the contemporary Ohio River watershed and play a major recreational and ecological role in local fisheries. Ecologically, all three species are apex predators and play a key role in ecosystems by directly influencing local fish assemblages. Tsetse flies (Diptera: Glossinidae) are the vector of trypanosomes (Trypanosoma spp.), which are responsible for African trypanosomiasis, also known as sleeping sickness. African trypanosomiasis is a significant health and economic burden yet remains a neglected tropical disease. As an obligate blood feeder, tsetse flies are reliant on B vitamin supplementation from intracellular Gammaproteobacteria, Wigglesworthia glossinidia. Folate (B9) is essential for tsetse development and reproduction, as well as DNA methylation in animals. As a reversible modification, DNA methylation typically occurs in a CpG dinucleotide context and can alter gene expression via the restriction of transcription. Across insects, the …

The ability for beef cattle to utilize feed efficiently is a critical determinant for both economic and environmental sustainability in livestock production. However, despite extensive research, the complex microbial ecosystem within the rumen and its relationship to feed efficiency remain incompletely understood. This dissertation explores the physiological and microbial foundations of feed efficiency by characterizing how rumen microbial communities and their functions differ among beef cattle with divergent residual feed intake (RFI) phenotypes and how the RFI status of beef cattle influenced their response to dietary interventions such as supplementation with a Bacillus-based feed additive. Virtual flight test aircraft provide good opportunities to test new parameter estimation methods. However, good estimates for the parameters in VIRTTAC Castor, a virtual test aircraft, are unavailable. This thesis uses two well-established time domain methods, the Output Error Method and the Filter Error Method, to estimate the parameters of VIRTTAC Castor. Three standard maneuvers were used: Elevator 3-2-1-1, Rudder Doublet, and Aileron Multi-Step. The parameters were estimated across three different flight conditions: weight, altitude and air speed, which resulted in 27 tests for each maneuver. All parameters, and standard deviations estimated are included in the appendix as a table.

Endodontic treatment aims to prevent or address apical periodontitis by reducing intracanal bacterial populations to promote tissue healing, requiring thorough debridement and disinfection of the canal space. This study aims to examine the presence of trace metal elements and chemical compounds in over-the-counter (OTC) and medical-grade sodium hypochlorite (NaOCl) solutions.
Method
Over-the-counter NaOCl brands (Clorox® and Great Value®) and medical grade 6% NaOCl solutions (CanalPro® and Chlor-Xtra®) were purchased, stored in amber glass bottles, and tested in triplicate. Each was analyzed using mass spectrometry, identifying 7881 chemical compounds with statistical comparisons via adjusted p-values, fold-change, and heat maps. …

The family Nectriaceae includes numerous phytopathogenic fungi that cause canker diseases on both angiosperm and conifer hosts worldwide. Numerous Neonectria spp. cause cankers on hardwoods and conifers in North America, but their roles in contributing to tree decline and mortality outside of beech bark disease are largely understudied. One such pathogen, Neonectria magnoliae, causes perennial cankers on two native hosts in central Appalachia: Fraser magnolia (Magnolia fraseri) and tulip-poplar (Liriodendron tulipifera). Resurgence is a type of relapse that consists of the recurrence of a previously eliminated response following worsening reinforcement conditions for an alternative response. Resurgence can occur following a history of positive or negative reinforcement; however, no previous evaluations have directly compared resurgence following these processes. In the present set of experiments, college students responded on a computer program to earn points (positive reinforcement) and to avoid losing points (negative reinforcement). Cosmic rays and solar energetic particles (SEP) bombard the Earth’s geomagnetic field, posing a threat to satellites, space stations, and human space exploration. These rays and particles also produce radiocarbon (14C) in Earth’s atmosphere, thus records of past 14C concentration in the atmosphere may be indicative of past cosmic and solar activity. Ecosystem services provided by dung-associated beetles, such as reducing pasture fouling, parasite suppression, supporting nutrient recycling, and increasing water infiltration, contribute to animal and pasture health. However, parasiticides used to manage cattle parasites, particularly flies and helminths, in West Virginia, can adversely affect these non-target beetles. We assessed the exposure and impact of some parasiticides used in WV including Clarifly® (active ingredient [a.i.] diflubenzuron), Cydectin® (a.i. moxidectin), LongRange® (a.i. eprinomectin), and Safe-guard® (a.i. fenbendazole). Both age and gender are personal characteristics that are at least in part outwardly visible and act as a category for social judgments (Cuddy & Fiske, 2002). The current study examined associations among individuals’ views on their aging, their gender typicality, well-being, and perceived discrimination. Research Question 1 asked whether views on aging and gender typicality interact to influence perceptions of discrimination; Research Question 2 explored the same potential interactions’ influence on well-being. This study investigated clothing preferences among cisgender queer women within Appalachia and whether they used these preferences to identify their own peers. It also further investigated whether they use clothing as a tool to conceal or reveal their own LGBTQ+ identity. Participants were invited for two interviews each. Between interviews, there was a one-week rumination period for each woman. The women reported themes of using clothing to blend in with their hetero-normative surroundings as they grew up and now lean into a more alternative style and less narrow-minded view of clothing. Multiple sclerosis (MS) is a cell-mediated, demyelinating autoimmune disease that affects the central nervous system (CNS). T helper (Th) 17 cells mediate the pathogenesis of MS and its animal model, experimental autoimmune encephalomyelitis (EAE), in part by infiltrating the CNS and interacting with astrocytes, a resident glial cell. Astrocytes are central to coordinating several inflammatory processes in the CNS and can respond to cytokines and chemokines produced by encephalitogenic Th17 cells. Capillary Vibrating Sharp-edge Ionization device (cVSSI device) was developed by our laboratory with a low cost and an easy fabrication procedure. The generation of small liquid droplets by this technique has been used as an ionization source for mass spectrometry analysis platforms. Lung cancer remains one of the deadliest cancers. Novel, paradigm shifting treatments including immunotherapy and targeted therapies have recently been developed to cull the deadly effects of lung cancer, but many challenges remain. There remains a significant unmet need to accurately predict and optimally select for patients who will respond to immune checkpoint inhibitors (ICI) treatment. In Chapter 2 of this dissertation, we investigated a novel live single cell cytokine profiling lab-on-chip platform, IsoLight, using peripheral CD4+ and CD8+ T-cells for ICI biomarker development. Tree-of-heaven (Ailanthus altissima) is a highly invasive tree species with a cosmopolitan distribution, including an expansive range in North America. In the U.S., tree-of-heaven has established populations in more than 40 states and is considered among the most notorious of invasive plant species. The discovery of Verticillium nonalfalfae and V. dahliae causing acute wilt and mortality of Ailanthus in the mid-Atlantic in the 2000s has resulted in numerous studies evaluating the utility of these fungi as potential biocontrol agents. Whooping cough, or pertussis, is a highly infectious respiratory disease caused by infection with the Gram-negative bacterial pathogen Bordetella pertussis. Pertussis affects all age groups and populations; however, it is most severe in infants who are too young to receive vaccinations and remains a global health concern with more than 200,000 cases reported globally in 2023. Vaccines against pertussis have been available in the United States since the 1940’s with the development and widespread use of whole cell pertussis vaccines. This thesis explores ideas and practices in the French penal system during the Revolutionary era, especially during the Reign of Terror, considering the experiences of Revolutionary leaders and the executioner of Paris during the Revolution. It argues that while in some ways Revolutionary leaders used Enlightenment penal laws to justify their actions during the Reign of Terror, in other ways the Reign of Terror was a departure from Enlightenment penal reform laws.
