Effect Of Concentrations Of Progesterone On Follicular Development And Fertility In Beef Cows, Jason Brian Candler

The present study was performed to investigate the effects of low ascending concentrations of progesterone on fertility in lactating beef cattle. Cows on the low ascending progesterone treatment received PGF2alpha from days 3 to 4 after estrus. All cows received PGF2alpha on day 9, and estradiol benzoate on day 11. Cows were inseminated artificially 12 hours after estrus. Concentrations of progesterone were lower in treated versus control cows. Pregnancy rate did not differ between treated and control cows. Pregnancy rate was greater in cows that ovulated a follicle from the right (78%) versus left (52%) ovary (p < 0.01). Low ascending progesterone early in the estrous cycle did not decrease fertility to the same extent seen with extended periods of low progesterone.

Skeletal fragility is an important orthopedic concern including the prevention of osteoporosis, long-term stability of prosthetic implants and stress fractures. Damage in human cortical bone has been implicated as a cause of increased fragility and is thought to initiate bone remodeling. Therefore, characterization of the mechanisms of damage initiation and accumulation in bone is challenging not only from the engineering prospective but also has a potential of revealing new insights in its physiology. In the ewe, anestrus is caused by increased sensitivity to E2 negative feedback that is mediated by A15 dopaminergic neurons that are only responsive to E2 in anestrus. Experiment 1, tested the hypothesis that synaptic input on A15 neurons varies with season by examining synapsin-positive close contacts on A15 cells using immunofluorescence and confocal microscopy. The number of contacts on dendrites, but not somata, increased during anestrus. There were also corresponding changes in dendritic morphology including an increase in dendritic length, surface area, and number of bifurcations. To demonstrate that alveolar macrophages (AM) from rats exhibit pepT1-like transporter for the uptake of small arginine-containing peptides (ACPs) and utilized these peptides as direct substrates for nitric oxide (NO) production.;Method. A HPLC assay was developed for quantitative measurement of Arg and ACPs in rat plasma and bronchoalveolar lavage (BAL) fluid. The uptake of small peptides by rat AM was evaluated using fluorescein isothiocynate (FITC)-labeled (*) peptides (Arg-Lys*, beta-Ala-Lys*, and Gly-Sar-Lys*), HPLC analysis of potential peptide degradation, and known inhibitors on arginine (Arg) and PepT1 transport. Reverse-transcription polymerase chain reaction (RT-PCR) was performed to measure myostatin, follistatin, activin-B gene expressions during chicken embryonic development. Strong myostatin expression was found in the early chicken embryos (E 0, E 1) and the developmental expression pattern of myostatin mRNA coincided with the periods of primary and secondary muscle fiber formation. Follistatin transcripts followed a linear expression pattern from E 0 to E 20, while activin-B had a quadratic pattern. The ontogeny of myostatin gene expression was nearly identical in satellite cells isolated from pectoralis major (PM) and biceps femoris (BF) muscles of chicken. The quantity of progesterone (P4) delivered by a new P 4 releasing intravaginal insert (polycapralactone [PCL] insert), was inadequate to increase serum concentrations of P4 to that observed in cycling ewes during the luteal phase. Percentage of ewes in estrus was greater in P4 treated than in control ewes. Estrous response was greater after treatment with P4 for 5 than for 12 d. Progesterone increased the percentage of ewes lambing to both service periods by 20 percentage points. Prolificacy to the first service period in ewes treated with FSH was greater than the prolificacy in control and all ewes lambing …

Nitrogen (N) and defoliation effects on productivity, root growth and soil nitrate (NO3-) in a Kentucky bluegrass and white clover pasture were examined. Nitrogen was applied annually in two equally split applications at rates of 0, 80 or 160 kg N ha -1. After reaching 12.7 cm, plants were cut to 2.5, 5.1 or 7.6 cm. Herbage was analyzed for yield, crude protein (CP), neutral detergent fiber and acid detergent fiber. The purpose of this study was to determine the frequency of unethical practices in exhibiting animals observed by West Virginia extension agents and high school teachers. A researcher-created demographic sheet and a fifty-eight-statement Lickert type questionnaire were sent to all extension agents and high school agriculture teachers in West Virginia (N = 154). Descriptive data were analyzed using the Statistical Package for Social Sciences (SPSSpc). This study compared nutrient content and intake of student school and home-packed lunches. Food intake was estimated by weighing the food and leftovers of 114 elementary school children. Eighty-one percent (n = 92) consumed school lunches and nineteen percent (n = 22) consumed packed lunches. A nutrient analysis was conducted using Nutritionist V. Significant differences between the two types of lunch were determined using ANOVA and Tukeys Test (HSD.UEN), p < 0.05.;School lunches were nutritionally superior (p < 0.01) for protein, vitamin A, vitamin D, riboflavin, vitamin B6, folate, vitamin B 12, pantothenic acid, phosphorus, magnesium, zinc, and dietary fiber; (p < 0.05) for calcium and niacin.;After considering food waste, intake from school lunch remained significantly higher (p < 0.01) for vitamin D, vitamin B12, pantothenic acid, phosphorus, magnesium, and zinc; (p < 0.05) for riboflavin, vitamin B 6, folate, and calcium.;It was also noted that intake of milk and other dairy foods was higher (p < 0.01) from school lunches.

Previous qualitative studies about the lived experience of persons with rheumatoid arthritis (RA) have yielded common themes, such as becoming aware, seeking help, learning to live with the disease, and uncertainty (Brown & Williams, 1995; Ryan, 1996; Shaul, 1995, 1997; Yoshida, 1996). Gaps identified included lack of personalized guidance and support from health care providers, gender differences, and the active role of the patient in managing the disease (Brown & Williams, 1995; Ryan, 1996; Shaul 1995, 1997). The river continuum concept (RCC) asserts that the biological community in a stream system is largely regulated by geomorphological factors (Vannote, et al., 1980, 131). The concept states that community structure in natural stream systems is structured in order to capitalize on the most efficient energy utilization throughout. The objective of this research is to test the RCC in the Wheeling Creek watershed of northern West Virginia and southwestern Pennsylvania and to compare the results with those hypothesized under the RCC. The purpose of this study was to evaluate the impact of an intensive one-week Lifestyle Management course on behavioral, psychological, physiological, and psycho-behavioral factors. The current study contained forty-five participants (N = 45) which included twenty-four individuals enrolled in the Lifestyle Management course (experimental group) and twenty-one matched individuals (control group). The experimental group experienced high-level wellness in the intensive one-week course of Lifestyle Management. In normotensive rats fed high salt, arteriolar responsiveness to endothelium-dependent dilators is reduced due to suppression of local nitric oxide (NO) bioavailability. In the first study we evaluated the possibility that an increase in microvascular reactive oxygen species (ROS) may account for this decrease in NO activity. Arcade arterioles in the spinotrapezius muscle of Sprague-Dawley rats fed either a low (0.45%, LS) or high (7%, HS) salt diet for 4–5 weeks were examined. This study analyzes food security and coping mechanisms in semi-arid West Pokot between 1920–1995. Based on a wide array of primary and secondary sources, the study analyzes the historical role of indigenous irrigation, specifically furrow irrigation, rain-fed farming and livestock keeping, in food production and food security among the Pokot of northwest Kenya. Besides farming and herding, the study also analyzes trade, hunting and gathering, as well as lending and borrowing (for example of food) based on reciprocity, and their contribution to food security in the area. Triiodothyronine (T3) stimulates a 70-fold or greater increase in transcription of the malic enzyme gene in chick embryo hepatocytes. Previous work has shown that the effects of T3 on malic enzyme transcription are mediated in large part by sequence elements located within a liver-specific and T3-inducible DNase I hypersensitive region in the 5′-flanking DNA of the malic enzyme gene. Within this DNase I hypersensitive region is a cluster of nuclear T3 receptor (TR) binding sites that together constitute a T3 response unit (TAU). This study examined relationships among the way teachers communicate in the classroom, teachers temperaments, and perceptions of teachers by students. Specifically, the first goal was to determine to what extent a teacher's levels of extraversion, neuroticism, and psychoticism can predict students perceptions of task attraction, immediacy, assertiveness, responsiveness, and credibility. The second goal was to determine the relationships between teacher temperament and teacher evaluations, affective learning, and cognitive learning. Self-report surveys of 52 teachers and 1242 students across seven disciplines at West Virginia University were collected. Instructors received measures of extraversion, neuroticism, and psychoticism. I analyzed community structure of ectomycorrhizal fungi on white pine (Pinus strobus) on two reclaimed surface mines in eastern Ohio. One site was acidic and metal-stressed, the other was alkaline. Completely different community composition was found on the two sites by PCR/ITS/RFLP analyses of field collected mycorrhizae. However, Shannon-Wiener index values indicated that the two communities were equally diverse. A population of isolates of one species common to both sites was obtained from trap cultures planted with white pine. The identity of this species as Wilcoxina mikolae was established through DNA sequencing of the ITS region. The W. mikolae populations …
Increasing Nutrition Knowledge And Food-Related Behavior Practices Among Participants In The "Dining With Diabetes" Program In West Virginia, Amy B. O'Dell

Diabetes is a serious disease that effects a large number of West Virginia's population. A study was conducted using 591 diabetic and non-diabetic subjects who attended the "Dining with Diabetes" program. Subjects completed a demographic form, pretest, and posttest. Pearson's correlation and ANOVA were used to determine the effect of a diabetes intervention program on nutrition knowledge, diabetes behaviors, and practices. The results indicated that there was no significant difference in nutrition knowledge between diabetic and non-diabetic subjects. Interaction was noted when subjects were placed into groups based on their pretest score (p < 0.07). The change in score (posttest score-pretest score) was negatively correlated with pretest score (r = -0.75, p < 0.00). Significant association was also noted between nutrition knowledge score and several behavior and diabetic practices. The results conclude that diabetes education programs may significantly affect some behaviors and diabetic practices.

The examination of aquatic environments for bacterial indicator organisms has proven to be a useful and well-established practice for the purpose of monitoring microbiological water quality. An ideal indicator of fecal pollution should be present in sufficient density to allow detection, present simultaneous with pathogen(s), incapable of aftergrowth in external aquatic environments, easy to enumerate, and exclusively of fecal origin. Unfortunately, none of the indicator organisms presently in use today meet all of these requirements. This dissertation examined roles of arbuscular mycorrhizal fungi, of which Glomus clarum is the primary species, in broomsedge (Andropogon virginicus) plants under limited Pi and elevated Al conditions in four studies. The inoculum originated from an acidic coal-mining site in Morgantown, WV. The inoculum of Glomus clarum from INVAM collection, which originated from an alkaline coal-mining site in WV, was also used in two of the studies. Many women do not initiate breastfeeding although breastfeeding promotes their baby's health. Three reasons why women do not breastfeed are women's mental organization, cultural beliefs about breastfeeding, and women's breastfeeding personal efficacy beliefs. The conceptual framework for personal efficacy beliefs is Bandura's social-cognitive theory (1992, 1995, 1997). This dissertation used a new research instrument to examine women university students' breastfeeding personal efficacy beliefs. The instrument was tested by mail survey techniques that resulted in a 70.6% respondent rate. University women's personal efficacy beliefs about breastfeeding factored into five factor subscales. Experiments were performed to determine if a short-lived corpus luteum was embryotoxic in ewes given supplemental progesterone and the route of transfer of embryotoxic products. Mated ewes were lutectomized or sham lutectomized on day 4 after estrus. Following surgery, ewes received saline or PGF 2alpha through days 6 or 7. Pregnancy rate was reduced after treatment with PGF2alpha (P < 0.001) compared to controls and was increased in PGF2alpha-treated ewes by lutectomy (P < 0.05). Mated ewes with bilateral ovulations were unilaterally lutectomized, and each uterine horn was ligated to eliminate intercornual flow on day 4 after estrus. Treatment with PGF2alpha on day 5 through 8 reduced pregnancy rates (P < 0.01), but embryonic survival did not differ in the uterine horns ipsilateral or contralateral to the regressing CL. The regressing CL is embryotoxic in progesterone-supplemented ewes, and embryotoxic luteal products have been shown to act by non-local pathways.

The purpose of this study was to demonstrate associations between perception of body image among women and their body image behaviors. A study was needed to determine the extent to which subjective norms, attitudes, beliefs, Body Mass Index, and self esteem were associated with body image behaviors. The target population in this study was female students at West Virginia University. Data collection was facilitated through an anonymous mailed questionnaire sent to 400 female students at West Virginia University. The intent of this study was to examine parental attitudes toward parenting and parenting styles before and after participation in a rural parent education program. The parental attitudes toward parenting were measured by the Parental Attitude Research Instrument (PARI). The three parenting styles; authoritarian, authoritative, and permissive, were measured by the Parenting Style Dimension (PSD). Efforts toward greater inclusion of children with a range of presenting problems have resulted in increasingly more children with difficult behavior in non-specialized classrooms. Unfortunately, teachers report that they have not been trained adequately to work with children who exhibit extreme behaviors. It is important, therefore, that effective methods of training teachers are empirically investigated. In clinical settings, strategies for training parents of young children with disruptive behavior disorders have substantial empirical support. Therefore, using these techniques to train teachers is a logical step. Parent-Child Interaction Therapy is one empirically validated parent training approach that uses coaching as a means …
