Dopaminergic neurotransmission is critically involved in the etiology and treatment of many psychiatric and neurological disorders. One modulator of dopaminergic neurotransmission is the kappa opioid receptor (KOR) -- a G protein-coupled receptor (GPCR) that is densely expressed within dopaminergic neurons and circuits. GPCRs are tightly regulated by a variety of intracellular signaling molecules, including Regulator of G Protein Signaling (RGS) proteins. Canonically, RGS proteins act as GTPase accelerating proteins (GAPs) on GTP-bound Ga subunits following GPCR activation, thereby hastening the rate at which GPCR-mediated G protein signaling is terminated. Pregnancy loss in cattle is greatest in the first 30 days of gestation. Near d15 of gestation, bovine conceptuses elongate and express LGALS1, an immunosuppressive glycan-binding protein believed to be essential for implantation. However, other members of the bovine galectin family, including LGALS3, LGALS4, LGALS7, LGALS8, LGALS9 and a galectin-13-like gene (LGALS13L), also may be expressed by the elongating conceptus. Therefore, galectin expression in elongating, d16 bovine conceptuses and intercaruncular endometrium as well as elongating, d14 ovine conceptuses and intercaruncular endometrium was assessed by RT-qPCR. Hetero(element) carboxylation is an appealing transformation that involves the installation of CO2 and another hetero(element) in one step. This transition metal-catalyzed one-pot synthesis provides a route to achieve highly functionalized carboxylic acid products with an abundant and cheap C1 feedstock. This difunctionalization also avoids the use of reactive metal hydride species, which is a prominent limitation of hydrocarboxylation chemistry. A copper-catalyzed regioselective boracarboxylation of vinyl arenes has been developed to access pharmaceutically relevant β-boryl-α-aryl propionic acid products. Forests in the western United States have experienced a shift from historical disturbance regimes in the past century. Many of these changes were induced by European settlers logging the forests and suppressing fires. In the past, the dry mixed conifer forests of California’s Sierra Nevada mountains experience frequent, low to mixed severity fires. This fire regime helped maintain a heterogeneous landscape comprised of groups of trees and openings. However, due to fire suppression and high grading logging, forest structure has changed; there are less openings and more small, fire-intolerant trees that can carry a fire into the forests crown. In the past decade, there has been an exponential increase in the study of the Behavioral Immune System (BIS), a suite of psychological processes that serves an infectious disease avoidance function. The BIS is proposed to be inherently intertwined with the physiological immune system. However, very few studies have actually demonstrated this fundamental, theoretical assumption. The purpose of this thesis was to test the possible relation between the physiological and behavioral immune systems in two studies. Cannabinoids have analgesic and anti-inflammatory properties. For example, in preclinical models of acute inflammatory pain, cannabinoid receptor subtype 2 (i.e., CB2, Table 1) agonists decrease paw inflammation and resulting pain. Yet, the anti-inflammatory properties of cannabinoid receptor manipulation in chronic inflammatory pain states are unknown. Monoacylglycerol lipase (MAGL) is the primary catabolic enzyme of the endocannabinoid (i.e., endogenous cannabinoid ligand) 2-arachidonoyl glycerol (i.e., 2-AG). Compounds that inhibit MAGL indirectly increase tissue levels of 2-AG by preventing its catabolism. MAGL inhibitors demonstrate efficacy in acute models of inflammatory/rheumatic disease. The Kanawha River in West Virginia is a historically diverse system in terms of ichthyofauna although, like many other large rivers, it has been subjected to a suite of anthropomorphic perturbations. These include this system being modified to allow the passage of commercial vessels for industry thus altering flow regimes, increased sedimentation, water depth, and channelization, and decreased interactions with its floodplain. Endogenous opioids mediate both analgesic and affective responses to stress. While the mu opioid receptor (MOR) produces the reinforcing euphoric effects responsible for the high abuse potential of traditional opioid analgesics, the kappa opioid receptor (KOR) is hypothesized to mediate the dysphoric component of stress. With increasing rates of opioid addiction, extensive research efforts are focused on better understanding each of these systems and how they interact, with the goal of developing less addictive pain management strategies and better approaches to addiction treatment. The natural structure of porous lignocellulosic structure was preserved during activation with further developed porosity by the activation. The activated carbon can be well applied to electrochemical double layer capacitor for transportation storage of ions as well as adsorbent materials for metal ion removal from wastewater.

Swimming exercise (bodylenghts/sec), and dissolved oxygen (DO), are important environmental variables in aquaculture. While there is an obvious physiological association between these two parameters, their interaction has not been adequately studied in Atlantic salmon, Salmo salar and Rainbow trout, Oncorhynchus mykiss. Since both variables can be easily manipulated in modern aquaculture systems, we sought to assess the impact of these parameters, alone and in combination, on the performance, health, and welfare parameters of the two species. Les Six, formally established in 1920, included a group of young French composers: Georges Auric (1899-1983), Louis Durey (1888-1979), Arthur Honegger (1892-1955), Darius Milhaud (1892-1974), Francis Poulenc (1899-1963) and Germaine Tailleferre (1892-1983). The primary motivation for their formation was the reaction against foreign influences, especially German Romantic music, in French music. Jean Cocteau, inspired by Erik Satie’s style, proposed a new French musical aesthetic: simplicity, directness, clarity, and terseness. Les Six followed these ideals in their formative years. Their artistic association was short-lived, but while it lasted it produced Album des Six, their sole joint production, a published …

Peripheral viral infections are po­tent comorbid factors that exacerbate neuro­degene­ration. Although the under­lying mecha­nisms have not been defined, neuronal hyperexcitability has been established as an underlying feature. Our lab has developed a preclinical model in which a viral mimetic, poly­inosinic-poly­cytidylic acid (PIC) is injected in­tra­peritoneally to induce an anti-viral acute phase response (APR). APR in turn elicits robust neuronal hyperexcitability. The present study was undertaken to characterize molecular mechanisms that mediate the development of hyperexcitability in response to PIC challenge. This study examines the usage of English swearwords by L1 German speakers and poses the following three questions: (1) Which English swearwords do native-speakers of German use? (2) In which situations do German speakers use English swearwords? Why do they use English swearwords instead of their German counterparts? (3) What emotional weight do they apply to swearwords in English (i.e. How taboo are they)? Using narrative inquiry, this case study explored how one teacher, who had deep connections to place, learned and utilized the place-based Enrichment Triad in her rural Appalachian elementary gifted program. Data collected included teacher, student, and parent interviews and classroom observations. Data were analyzed using a socio-cultural narrative analysis and narrative thematic analysis. Results from analysis created thick descriptions of the teacher’s journey to teaching, her existing practices, her experiences learning and implementing the framework, and her future curricular intentions. Many allergenic metals are being manufactured in nanoparticulate forms. Although the decreased size profile of metal nanomaterials has been consistently associated with more pronounced lung toxicity compared to larger materials, it is unclear if metal-induced immunotoxic effects exhibit a similar size-dependency.

The central hypothesis of these studies was that metal nanoparticles cause more pronounced immunomodulatory effects on allergic processes than larger forms of respective metals. A detection and identification scheme for abnormal conditions was developed for an unmanned aerial vehicle (UAV) based on the artificial immune system (AIS) paradigm. This technique involves establishing a body of data to represent normal conditions referred to as “self” and differentiating these conditions from abnormal conditions, referred to as “non-self”. Data collected from simulation of the UAV attempting to autonomously fly a pre-decided trajectory were used to develop and test a scheme that was able to detect and identify aircraft sensor and actuator faults. Earlier evidence indicates exposure to propanil, a post-emergent herbicide used on rice and wheat fields, increases the number of antibody-secreting cells (ASC) in the spleen of mice after immunization with heat-killed Streptococcus pneumoniae (HKSP, 2x108 CFU). Interestingly, an intact hypothalamic-pituitary-gonadal (HPG) axis is required for this immune response assessed on day 7-10 after immunization in female mice. Hence, an overarching premise is that propanil arbitrates the connection between the reproductive and immune system by altering the concentration of hormones involved in the HPG axis, which may then affect the populations of splenic cells. The National Institute of Occupational Safety and Health (NIOSH) is the organization in charge of approving respiratory devices in the United States, as permitted by 42 CFR Part 84. Standards that NIOSH uses to certify powered air-purifying respirators (PAPRs) are based on respiratory data collected before the 21st century. With new technologies, it is possible to gain a more accurate understanding of breathing flow of healthcare workers (HCWs). A new portable breathing flow recording device developed by The S.E.A. Growth of an individual is the end result of many physiological processes stemming from the consumption of food items. These processes can be influenced by many variables including temperature. Current bioenergetics models for Brook Trout Salvelinus fontinalis suggest growth increases with temperature until an inflection point of 20.2°C. With a warming climate, and water temperatures expected to rise, the ability of a population to adapt to the warming environment is crucial to the survival of the species. This dissertation is composed of three separate but related manuscripts with the common theme of using historic log buildings from the central Appalachian Mountain region of eastern North America as ecological archives. In Chapter 1, I explore the biases, limitations, and ecological applications of tree-ring data from historic log buildings. European immigrants selected trees from a forested stand based on species, log sizes, and construction locations. Despite this selection bias, ecological information can be gleaned from historic log buildings, which offer a complementary record of past forest ecology and represent a site type that is not often associated with old-growth …

Despite the overwhelming research that suggests visual artmaking experiences can produce positive outcomes for individuals, many state art education policies fail to implement and integrate the arts into core academic curriculums. Specifically in the state of West Virginia, kindergarten through twelfth grade students often lack the access to a quality and consistent art education. With high teacher-pupil ratios and inclusive learning environments, educators face the difficult task of meeting all the individual needs of their students’ in the classroom.
