The overall focus of this thesis is on separating and utilizing fractions and co-products from food manufacturing as functional ingredients. Therefore, the objective of this research was to 1) evaluate the effectiveness of water degumming to separate omega-3 PUFA-rich phospholipids from other lipids found in krill oil and 2) determine how different organic solvents would affect physiochemical and functional properties of egg yolk protein recovered from spray-dried egg yolk using a novel one-step organic solvent extraction method.

The insecticide imidacloprid is widely used to mitigate hemlock (Tsuga spp.) mortality resulting from the invasive hemlock woolly adelgid (HWA), but evidence suggests that
imidacloprid can have negative effects on adjacent stream systems. Imidacloprid exposure causes physiological changes, bioaccumulation, and reduced survival in anurans, and induces physiological impairment and reduced survival in benthic macroinvertebrates. In this thesis, we assessed whether HWA treatments using imidacloprid affect stream salamander abundances and benthic macroinvertebrate community composition. We also investigated whether imidacloprid or its metabolites bioaccumulate in benthic macroinvertebrates or salamanders and cause sublethal effects. We conducted this study in two National Park …

Pertussis is a human respiratory disease, primarily caused by the Gram-negative pathogen Bordetella pertussis. The infection is most severe and can be life-threatening in young children and infants where it manifests as a series of paroxysmal coughs. The disease is more commonly known as whooping cough, due to the whoop omitted during a massive inspiratory effort to bring air back into the lungs. Amphibians are facing global declines due to climate change, loss and degradation of habitat, invasive species, and disease. The Appalachian region of the eastern USA is a global biodiversity hotspot for salamanders, which are considered keystone species that influence nutrient dynamics in terrestrial and aquatic food webs. There are high rates of salamander endemism in the Appalachian region, with many species restricted to isolated, high elevation areas. Lung cancer is a leading cause of cancer-related death worldwide. Every year it claims more lives than other common malignancies, breast, prostate, and colorectal cancers, combined. And despite recent improvements in disease diagnostics and treatment, drug resistance and tumor relapse remain major contributors to low patient survival. Both issues have been largely attributed to the existence of cancer stem-like cells (CSCs) or tumor initiating cells (TICs). This study explores a problematic disconnect associated with virtual heritage and the immersive 3D computer modeling of cultural heritage. The products of virtual heritage often fail to adhere to long-standing principles and recent international conventions associated with historic preservation, heritage recording, designation, and interpretation. Though the duodenum is the primary site of iron absorption, the colon also contains iron transporters and participates in iron absorption. The purpose of this study was to characterize colonic iron absorption in control patients and those with ulcerative colitis (UC), a disease that frequently results in iron deficiency. Using RT-qPCR and western blot analysis, we found that colonic expression of the apical iron importer, divalent metal transporter 1 (DMT1), is equal to duodenal expression. However, colonic expression of ferroportin 1 (FPN1), an iron exporter, is only about one-quarter that of the duodenum. Patients with active ulcerative colitis were found …

Mountains are important areas for avian diversity and conservation since they have steep elevational and environmental gradients. In the central Appalachian Mountains, several songbird species have trailing edge populations that are restricted to high elevations. Climate change and other factors can cause species distributions to change, which is of particular concern for birds in restricted trailing edge populations. Sensory systems are often uniquely tailored to encode behaviorally relevant signals and comparative studies across species can thus reveal how evolutionary changes shape sensory functions. The structure of communication signals varies widely between ghost knifefish species. Recent findings suggest that the nervous system co-adapted to the various signal structures observed across species to support different sensory behaviors. In underground coal mines, mining activity disturbs the natural equilibrium state of in-situ stresses. The induced and in-situ stresses deform the rockmass surrounding the mine openings. Primary roof supports impede the deformation of the rockmass overlying the entries. However, failure can occur in the bolted rockmass, causing fatalities and injuries. Coenzyme A (CoA) is an essential cofactor required for hundreds of metabolic processes. Because it is such a critical cofactor, CoA levels are tightly regulated. In the fasted state and in diabetic mice, the concentration of CoA increases dramatically in the liver. This phenotype is associated with constitutively low CoA degradation, a process that is emerging as a potentially important mechanism for CoA regulation. Nudt7 and Nudt19 are two mammalian peroxisomal enzymes with CoA-degrading activity, which are highly expressed in the liver and kidney, respectively. Experiments were conducted to investigate the effects of exogenous enzymes in poultry diets. In Chapter 4, a study was conducted to determine the effects of a corn-expressed recombinant carbohydrase (AC1) on broiler performance and digesta viscosity in high non-starch polysaccharide (NSP) diets through 21 days of age. One day-old Hubbard × Cobb 500 chicks were assigned to 6 dietary treatments. Each treatment consisted of 12 replicate pens of 10 birds. The positive control diet (PC) was a corn and soybean meal formulation. The negative control diet (NC) included 10% wheat and 10% corn distiller’s dried grains with solubles (DDGS). The …

Ramps have been used in Appalachian folk medicine for its health promoting roles in lowering blood pressure and cholesterol. Information on the chemical composition of bioactive components responsible for the potential health benefits are scarce. Analysis of the chemical composition in similar Alliumplants showed that flavonols, induced similar health benefits. Context: Iron deficiency is a concern in distance runners due to the implications that hemoglobin production becomes impaired limiting oxygen uptake and tissue oxidation in the body. Human skeletal muscle is a highly metabolic tissue necessary for mobility and coordination. Responsible for approximately one-fifth of the resting human metabolism, skeletal muscle is also an important regulator of metabolites like glucose and contributes to the regulation of body temperature. Although there is a gradual decline in muscle mass associated with aging, a certain percentage of the population suffer from severe muscle mass and strength deterioration, classified as sarcopenia (5-13% for people aged 60-70 years old, 11-50% for those 80 or older). Resurgence is the recurrence of a previously reinforced response after a more recently reinforced response is placed on extinction. Resurgence may explain the recurrence of socially appropriate behavior, including academic responding, but this had not yet been empirically demonstrated. The aim of this study was to determine if resurgence would occur when a participant solved quadratic equations using multiple methods. Each participant was taught two methods of solving quadratic equations across experimental phases, followed by a phase in which neither method resulted in the correct solution. In this research, the artificial immune system (AIS) paradigm is used for the development of a conceptual framework for autonomous flight when vehicle position and velocity are not available from direct sources such as the global navigation satellite systems or external landmarks and systems. The population of eastern wild turkeys in northeastern South Dakota appeared to be expanding soon after reintroduction in the 1990s, however, recent harvest trends suggest declining abundance in the region. Spring turkey hunting expenditures are projected to be around $6.5 million in South Dakota, with about $3 million of those expenditures occurring within the northeastern prairie region. However, the number of birds harvested during the spring prairie firearm season has been declining since 2010. Proteostasis is maintained by several systems in the cell including the ubiquitin-proteasome system (UPS), chaperones, chaperone-mediated autophagy, and macroautophagy. The UPS is the principle route for the degradation of intracellular misfolded, damaged, or unneeded cellular proteins and has a critical role essential every cell process, including: cell cycle progression, transcriptional regulation, genome integrity, apoptosis, immune responses, and neuronal plasticity. Sensory systems have to extract useful information from environments awash in noise and confounding input. Studying how salient signals are encoded and filtered from these natural backgrounds is a key problem in neuroscience. Communication is a particularly tractable tool for studying this problem, as it is a ubiquitous task that all organisms must accomplish, easily compared across species, and is of significant ethological relevance. The purpose of this research was to determine whether differences in the way the media frames an act of violence leads to different reactions by consumers. In particular, it was hypothesized that the ideology and race of the perpetrator would lead to differences in perceptions of whether or not the attack was terrorism. A vignette-style experiment was performed using respondents recruited via MTurk. Four versions of the vignette were evenly distributed to 441 respondents, changing whether the frame contained a photo, the ideology, and the name of the perpetrator. In this work we demonstrate the power of compound-specific isotope analysis (CSIA) to analyze proteinaceous biological materials in three distinct forensic applications, including: 1) linking necrophagous blow flies in different life stages to their primary carrion diet; 2) identifying the harvesting area of oysters for food authentication purposes; and 3) the ability to predict biometric traits about humans from their hair.
In the first application, we measured the amino-acid-level fractionation that occurs at each major life stage of Calliphora vicina (Robineau-Desvoidy) (Diptera: Calliphoridae) blow flies. Adult blow flies oviposited on raw pork muscle, beef muscle, or chicken liver. Larvae, pupae …
