Aging is an inevitable process that occurs as humans grow older. It is characterized by the chronological accumulation of cellular damage over time leading to functional decline as an organism grows older. Several processes are thought to contribute to the aging phenomenon, but one of the most prolific of these is the disruption of protein homeostasis (proteostasis). The collapse of proteostasis can lead to accelerated aging and the development of age-related diseases including devastating neurodegenerative diseases (NDs) like Alzheimer and Parkinson disease. The bobcat (Lynx rufus) is an important furbearer in South Dakota. However, management of bobcats can be difficult because of their elusive nature and lack of demographic information. In particular, managers lack information on abundance, survival, and reproductive rates necessary to ensure sustainable harvests and stable populations through time. Smallmouth Bass (Micropterus dolomieu) population characteristics were examined within the Tygart and Cheat River watersheds of north-central West Virginia. Growth, mortality and recruitment were examined for each individual population prior to modeling potential effects of various management schemes. Smallmouth Bass were sampled for total length and weight, aged using sagittal otoliths and sexed through observations of gonadal or ovarian development. Growth for each population was examined using von Bertalanffy growth models and assessed for variation among populations. Background: Diabetic ketoacidosis (DKA) is a serious complication of diabetes that occurs when the body compensates for insufficient insulin, producing dangerously high levels of ketones. Without proper treatment, DKA can be fatal. The primary objective of my thesis was to develop and utilize a biocompatible multifunctional trityl spin probe for concurrent measurement of pO2, pHe, and [Pi] in vivo by electron paramagnetic resonance (EPR) spectroscopy (Chapter 2). My first goal was to synthesize the proposed probe we are terming HOPE71. Secondly, HOPE71 was characterized by X-band and L-band EPR spectroscopy. Next, the biocompatibility of HOPE71 was assessed through an albumin binding test, cytotoxicity assays, and in vivo intravenous tolerance. Mass Spectrometry (MS) is one of the widely used tools for chemical detection down to attomole concentrations for certain proteins. Ionization sources are a critical component for MS analysis. One of the major ionization methods is electrospray ionization (ESI), which remains a gold standard for proteins and peptides. ESI uses voltages up to ~4 kV to produce ions from solubilized molecules. According to the latest statistics from the National Cancer Institute (NCI), about 1 in 8 U.S. women (~13%) will develop invasive breast cancer over the course of their lifetime. This translates to an estimated 268,600 new cases of breast cancer for the year 2019, and these diagnoses will collectively make up 15% of all new cancer cases across all cancer types. This dissertation investigates the effects of designer opioids on the development of blow fly (Lucilia sericata) larvae and the ability to detect and quantify the drugs and/or metabolites present in each stage of the fly’s life cycle. Reinforcer magnitude is one of several parameters of reinforcement. In the present study, it referred to the quantitative value of the reinforcer in terms of duration of access to a finite supply of grain pellets. Pigeons responded on concurrent-chain schedules earning one of several reinforcer durations depending on response allocation and experimental conditions. Experiment 1 consisted of relatively long reinforcer durations available for completing one chain compared to relatively short durations available on the other. Pellet mill production rate aids are also utilized to reduce feed manufacture costs and may have additional benefits when fed to poultry that are associated with the increase in production rate. Child-caregiver attachment is an important factor in healthy child development and is often targeted by early intervention programs. To assess the efficacy of these interventions, attachment must be accurately measured across the toddler years in populations referred for treatment of externalizing behavior problems. The Strange Situation Procedure (SSP; Ainsworth et al., 1978) and Attachment Q-Set (AQS; Waters, 1987) are empirically-validated measures of child-caregiver attachment, each with unique strengths and weaknesses. The purpose of this study was to determine the level of adoption of West Virginia agricultural educators’ usage of internet-based educational technology for instruction. The population for this study included 103 educators during the 2020-2021 school year. The study yielded thirty-two responses from the population. Respondents indicated they use internet-based educational technology for their classroom instruction. Respondents indicated they use internet-based educational technology in a variety of different ways for instruction. Respondents adopted internet-based educational technology at varying degrees depending on the technology, some respondents being early adopters, and others later adopters.

Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen which can cause severe, recurrent, and chronic infections. The pathogen is highly adaptable, and pneumonia caused by it is associated with high morbidity and mortality, especially in individuals with the genetic disorder cystic fibrosis. Antibiotic resistance amongst clinical isolates of P. aeruginosa is steadily increasing, and multi-drug resistant strains are prevalent. Risky choice can be operationally defined as the choice for a larger, uncertain reinforcer over a smaller, certain reinforcer. Research suggests smokers engage in more risky or maladaptive decisions when compared to nonsmokers. The relation between nicotine and risky choice could benefit from further investigation, since nicotine is the active substance of tobacco products that maintains tobacco addiction. Acute nicotine administration has shown to alter risky choice; however, since the everyday smoker uses nicotine repeatedly, more research on chronic administration is warranted and would allow for assessment of tolerance or sensitization of these effects. A focus on classroom integration of science, technology, engineering, and math (STEM) concepts has resulted in secondary teachers broadening the scope and delivery of STEM principles in their classes. Because agriculture classes allow students to convert abstract ideas into concrete ideas, they provide a useful setting to teach STEM concepts. Using secondary agricultural classes to teach science is effective and has contributed to higher standardized science test scores, as opposed to a science class alone.
