Ambrosia beetles, a polyphyletic group of weevils, utilize pouch-like structures at various locations on the body to transport for their sole food source, their fungal partner. Ambrosia fungi are known to originate from multiple unrelated fungal lineages with no consensus of traits required to become a fungal cultivar. While the biology and ecology of both partners is in many cases well understood, the genetic underpinnings of this fascinating mutualism remain a mystery. In this study of the place of Cherokee forced removal, I use a Third Space approach to blend Indigenous Methodologies, Deep Mapping, Geospatial mapping and analysis, and 3D Geography. Using a hybrid platform that provides a mélange of media types, storytelling forms, and interactive tools, I reimagine and retell the story of the Cherokee “Trail of Tears.”

This dissertation (a) provided a comprehensive review of the known therapeutic challenges in nicotine delivery system users with lung cancer and (b) investigated the impact of electronic cigarettes on blood-brain/blood-tumor barrier function and efficacy of small molecule KRAS inhibitors in a syngeneic lung cancer brain metastasis model. The blood-brain barrier is a physiochemical barrier that limits systemic delivery of therapeutics into brain. When tumors invade the central nervous system, the blood-brain barrier is disrupted and subsequently forms a blood-tumor barrier. Fecal coliform contamination is a major contributor to surface water pollution in the United States’ (U.S.) waterways, whether from combined sewer overflows (CSOs) (point source pollution), runoff from precipitation and snowmelt (nonpoint source pollution), or exfiltration from aging and failing infrastructure. In addition to fecal bacteria, chloride contamination is also a major, prevalent surface water contaminant impacting U.S. waterways. Chloride contamination is predominantly released into U.S. waterways as nonpoint source pollution, endemic to northern areas that use industrial brine and salt as a form of mitigation for ice accumulation on roadways. Historical mining, intensive timber harvesting, agriculture, and atmospheric acid deposition have left lasting impacts on soil health throughout the Central Appalachian region, reducing soil productivity, carbon storage, and ecosystem resilience. This thesis evaluated restoration strategies across two long-term research systems in West Virginia to determine how land-use history and soil amendments influence soil health, carbon dynamics, and ecosystem recovery. The first study examined reclaimed mine lands and marginal agricultural soils planted with switchgrass (Panicum virgatum) or willow (Salix spp.) and treated with biochar or inorganic fertilizer. Identifying drivers of variation in the timing of breeding behaviors, such as courtship activities and nesting chronology, is important for the effective management of avian species. Likewise, productivity is an important component of game bird population dynamics and can be broken up into multiple components such as the probability of nesting, clutch size, and nest success. Feed and harvested forage represent a major share of production costs in Appalachian cow-calf systems, where a pronounced summer forage gap forces producers to rely on purchased hay. This thesis evaluates whether adopting diverse forage seeding strategies reduces production costs relative to the existing pasture management practice, accounting for the spatial heterogeneity of West Virginia’s production environments. The analysis proceeds in two steps. This research project compares Romantic instrumental narrative in Robert Schumann’s Papillons, Op. 2; Fantasiestücke, Op. 12; and Kinderszenen, Op. 15, with particular attention to how pianists communicate cyclic continuity in performance. In this study, narrative continuity refers to relationships formed across ordered short pieces through contrast, interruption, return, literary association, fantasy, memory, tonal and motivic recall, and the shaping of sound and time. Comparative score analysis shows that Schumann creates a distinct form of continuity in each cycle. Influenza viruses (IV) affect populations globally, with the World Health Organization (WHO) estimating that IV are responsible for an estimated 3-5 million infections and 290,000 – 650,000 deaths annually1–3. More specifically, the 2019-2020 IV season is associated with an estimated 38 million illnesses and 22,000 deaths in the United States alone4. Behavior intervention plans are used to increase desired behavior and decrease problem behavior, and these plans require the use of effective reinforcers. High-quality reinforcers that are delivered immediately tend to be the most effective, but optimal reinforcement parameters are not always feasible or appropriate in applied settings. Some reinforcers may be disruptive, teachers’ competing responsibilities may introduce delays to reinforcer delivery, and reinforcers may not be immediately accessible. Background: Approximately 1 billion people currently live with obesity. Driven by factors such as a high-fat diet (HFD), this pathology can have drastic effects throughout the body, with the liver in particular being a main target of its downstream consequences. Although it is capable of storing lipids, excess fat can conglomerate in the liver and contribute to various hepatic pathologies. A specific pattern of hepatic dysfunction seen in response to an HFD is increased acetylation of mitochondrial proteins. Burnout remains a widely studied phenomenon in the healthcare literature, with much of the focus placed on reactive mitigation strategies that lack meaningful engagement with moral development. Healthcare education programs rarely incorporate formal learning experiences that promote moral development as a means of addressing burnout symptoms. This qualitative single case study examined how health professions students describe the impact of experiential learning on their moral development. For over a century, cardiovascular disease (CVD) has been and remains the leading cause of death globally. CVD risk and severity increase significantly with advanced age, and mitochondrial dysfunction has been implicated in the pathogenesis of aging. Furthermore, mitochondrial health is crucial for cardiac function, as one-third of total cardiomyocyte volume is occupied by mitochondria in order to meet the high energy demand of contractile and relaxational function. Jurors’ perceptions of child witnesses can have significant influence in judicial outcomes, particularly in cases where children’s testimony is central (Goodman et al., 1997; Goodman et al., 1998; Castelli et al., 2005). The role of gesture in shaping jurors’ perceptions is underexplored. Both animal and human research has identified early life adversity (ELA) as a prominent risk factor for substance use disorder (SUD). However, the underlying mechanisms linking ELA to SUD development remain unclear. Animal studies point to differences in reward responsiveness as the primary explanation; specifically, that ELA could lead to individual differences in the attribution of incentive salience to reward-predictive cues, or the “sign-tracking” behavioral phenotype. Sign-tracking has been linked to addiction-related behaviors in both animal and human studies, but no human studies have examined whether sign-tracking could partially explain the link between ELA and SUD. Lyme disease is the most common vector-borne disease in the United States with an estimated yearly incidence rate of more than 300,000 cases. Borreliella burgdorferi, the etiological agent of Lyme disease, is transmitted by the bite of an infected Ixodes scapularis tick and can cause debilitating pathologies affecting numerous body systems. Symptoms include fever, fatigue, joint pain, and a characteristic bull’s eye rash (erythema migrans) at the site of the tick bite. The timely and appropriate use of antibiotics effectively treats early Lyme disease; however, uncomfortable side effects reduce treatment compliance. The purpose of this research is to explore the expectations and experiences of program directors of campus support programs which center wardship experienced students. This research question guided the current study: What expectations and experiences influenced program directors’ development and reform of a wardship-centered campus support program? This inquiry applies Ürie Bronfenbrenner’s Person–Process–Context–Time (PPCT) bioecological model. Semi-structured interviewing was utilized for this case study. In Chapter 2, an experiment was conducted to determine if the inclusion of 1.50% mixer-added water (MAW) and/or 0.25% Azomite® (AZM) would be suitable in alleviating concerns regarding pellet production rate and pellet quality when manufacturing feed with low-moisture corn. Furthermore, the authors aimed to determine if these additives had varying effects when utilizing different pellet die thicknesses. This thesis and accompanying exhibition explore the translation of panenhenic mystical experience and the collaboration with Appalachian natural spaces into abstracted landscape images. This work aims to “map” a sensation of fundamental unity and gesture toward the ineffable by utilizing found-object stamp printing, and automatic mark-making. Inspired by practical mystic traditions and contemporary landscape painting, this body of work, entitled Orogenesis, combines intricate, abstracted ink landscape drawings and photographs to create images suggestive of the mystical patterns in the process.

This work develops computational methods for identifying molecular signatures from high-throughput genomic data and for modeling long non-coding RNA (lncRNA) sub-cellular localization. The response of multiple myeloma to CB-6644, a selective RUVBL1/2 complex inhibitor with potential anti-tumor activity, is analyzed to identify drug-responsive pathways and molecular signatures. Conventional gene set enrichment analysis (GSEA) often excludes low-expression genes. Here, phenotype comparison is reformulated as a supervised machine learning problem: genes most informative for discrimination are first selected using a machine learning approach, and GSEA is then applied to these machine-learning derived gene sets. Habitat fragmentation has the potential to isolate localized populations, particularly those which are understood to have limited dispersal capabilities. This isolation can reduce genetic diversity through the effects of inbreeding depression, and small populations are more susceptible to the impacts of genetic drift. Invasive plants alter forest ecosystems by reducing species richness, habitat heterogeneity, and nutrient cycling as they rapidly displace native plants. Invasive plant composition can influence soil health through altering soil microbial associations and communities. By studying the mechanisms in which invasives alter above- and below-ground communities, we can better understand whole-ecosystem impacts and inform management decisions. The Norman invasion of Italy in the eleventh century was a process that required political support and legitimacy for its leaders, the Hauteville dynasty. The members of this dynasty either commissioned or financed three individuals as they wrote three respective histories of the Norman conquest in order to accomplish this legitimation. In these sources, each author took inspiration from the classic works of history and poetry of the Greek and Roman world and applied symbolism and direction from their understanding of medieval Christian theology. Maternal electronic cigarette (ECIG) use during pregnancy raises concerns about potential risks to offspring, yet its behavioral effects remain unclear. The present studies were designed to investigate how prenatal exposure to ECIG aerosols, with and without nicotine, influences the behavior of Sprague-Dawley rat offspring. Dams were exposed to aerosols containing base liquid alone, aerosols containing base liquid with 18 mg/ml nicotine, or filtered air (control) throughout gestation. Following postnatal day 65, male offspring from 17 litters completed lever-press acquisition (Experiment 1), sustained-attending (Experiment 2), and nicotine self-administration (Experiment 3) tasks. Volunteers are the heart of the land-grant outreach model. They bring education and resources to communities across West Virginia. Volunteers have been relied upon in many aspects of Extension programming and current budget conditions have reduced the level of staffing within WVU Extension. There may be an increased demand for volunteers to help with programming. The purpose of this study was to investigate the relationship between motivation and the perceived level of well-being within the WVU Extension volunteer programs. Robots provide a unique proving ground for testing scientists’ understanding of neuromuscular systems. Synergistically, mimicking biological control laws and mechanics could offer robotics the robust and adaptive locomotion of animals. Previous work has utilized the wealth of research on insect biomechanics to develop scaled robotic models but doing so presented a problem: while these robots’ larger scale has simplified manufacturing, the scale has also shifted its mechanics from those of the modeled organisms.
