As translocations increase in popularity for use as a biodosimeter for prior radiation exposure, information on the possible decline of these 'stable' aberrations over time is desperately needed. This study is the first to assess the stability of radiation-induced chromosome aberrations in vivo over the life span of a rodent model. C57BL/6 female mice were given an acute dose of 0, 1, 2, 3 or 4 Gy {dollar}\\sp{lcub}137{rcub}{dollar}Cs of 8 weeks of age. The darter fauna in the Elk River, West Virginia, is extremely diverse (18 species); however, few studies exist on its ecology. This dissertation, which consists of three manuscripts, provides data on the ecology of darters within the Elk River. In the first manuscript, I estimated allozyme variation in the Tippecanoe (Etheostoma tippecanoe) and golden (Etheostoma denoncourti) darters, fishes with extremely fragmented distributions. Despite our findings of low variation at 22 loci, frequency differences at Pgm and Pgdh separated E. tippecanoe and E. denoncourti. Many small communities located in rural areas of the United States lack adequate domestic wastewater treatment plants. Constructed wetlands are being promoted as low-cost, low-maintenance systems for treatment of domestic wastewater in areas where standard septic tanks fail. In order to evaluate the efficacy of small constructed wetlands, 20 small-scale wetlands mesocosms (100 gal each) containing five plant treatments (no plants/control, Typha, Scirpus, and Juncus monocultures, and all three species combined) were planted into pea gravel at either 18 or 24 inches in depth. Antinociceptive and nociceptive responses can be elicited by the administration of endorphin within the periaqueductal gray region of the midbrain. The pain inhibitory and facilitory systems exist in a state of balance such that under most conditions, the combined activation of these processes by the administration of -endorphin results in the expression of antinociception. However, during other situations, such as stress, the delicate balance between these endogenous systems is disrupted and alterations in the overall response to opioids occurs. The purpose of this study was to determine if spatial intelligence contributes to a student's success in a computer science major or if mathematical-logical intelligence is sufficient data on which to base a prediction of success. The study was performed at a small university. The sample consisted of 15 computer science (CS) majors, enrolled in a computer science class, and 15 non-CS-majors, enrolled in a statistics class. Seven of the CS-majors were considered advanced and seven were considered less advanced. We have found that Host Factor I (HF-I), the product of the hfq gene, is required for efficient expression of the RpoS stationary phase sigma factor in Salmonella typhimurium. HF-I is an RNA-binding protein originally characterized for its role in replication of phage Q{dollar}\\beta{dollar} of E. coli. Its role in the uninfected bacterial cell has previously been unknown. Assays of {dollar}\\beta{dollar}-galactosidase in strains with rpoS-lac fusions, Western blot analysis, and pulse-labeling and immunoprecipitation of both fusion proteins and native RpoS show that an hfq mutant has a four- to seven-fold reduction in the translation of rpoS. This dissertation is composed of two topics that share the focus of placental vascular tone control. In the first investigations, we looked at the effect of caffeine (Caff; 10nmol/L-10{dollar}\\mu{dollar}mol/L), nicotine (Nic), and cotinine (Cot) (1nmol/L-100{dollar}\\mu{dollar}mol/L for both) administration on isolated placental blood vessels, to see if these substances altered placental vascular tone. We found that moderate relaxations were initiated by all three alkaloids in precontracted placental blood vessels. We also found that pretreatment of placental blood vessels and Caff, Nic, and Cot did not alter responses to a variety of contractile agents. In conclusion, it would seem that the adverse …

Propanil (3,4-dichloropropionanilide) is a herbicide that is used for post-emergence control of weeds on rice, wheat, and potato production in the United States. The first hypothesis of our study is: propanil significantly inhibits cytokine production by macrophages which may contribute to its overall immunotoxicity. We demonstrate that i.p. propanil exposure significantly inhibited IL-6, TNF-{dollar}\\alpha{dollar} and IL-1{dollar}\\beta{dollar} production by macrophages. Acute oral exposure with propanil demonstrates that both 3 and 7 day post-exposure cytokine production by macrophages was reduced. However, only 7 day post-exposure with the primary metabolite of propanil, 3,4-dichloroaniline (DCA), resulted in the inhibition of cytokine production. Therefore, we …

Using a combination of in-vitro and in-vivo experimental techniques, the maturational changes in arteriolar structure and blood flow regulation were explored in the spinotrapezius muscle of weanling, juvenile and mature rats. In the first study, morphometric analysis of the arteriolar network architecture revealed that mean arcade segment length and arcade loop circumference had significantly increased from weanling to mature groups, whereas total number of arcade arteriole segments remained fairly constant. The number of transverse arteriole subnetworks per muscle remained constant from weanling to mature rats, but there were significant increases in total number of vessels in each branch order, total …

Dissolution of the Soviet Union in 1991 changed the government bureaucracy and dismantled the centrally controlled economic system. Directors of zapovedniki (strict nature preserves) and lespromkhozi (forest industry dependent communities) are experiencing a challenge to their ability to manage, protect and/or study the lands under their supervision. We use dynamical Monte Carlo simulations to investigate instabilities of solids in the presence of externally applied stresses or strains. One of the instabilities is the classical Euler buckling instability of compressed rods. We address, for the first time, the dynamical aspects of this instability, that is, we study how an initially straight flexible chain reaches the final buckled configuration at long times. We find that this process has much in common with coarsening phenomena such as the spinodal decomposition or unstable mound growth in molecular beam epitaxy. The present study investigated differences in the conceptualization of death among children and adults with and without mental retardation (MR). Thirty-five adults (17 adults without MR, 18 adults with MR) and 29 children (21 children without MR, 8 children with MR) participated in the study. The study employed a 2 x 2 design with Age and MR as the factors. Participants completed several questionnaires, cognitive developmental tasks, and intelligence assessments. Results indicated that diagnosis of mental retardation was slightly more influential than chronological age in determining participants' responses about death. The chicken very low density apolipoprotein II (apoVLDLII) gene encodes an egg yolk protein that is synthesized in the liver in response to estrogen. Regulation of this gene is mediated by the estrogen receptor via two estrogen response elements (EREs) located in the promoter. Following the administration of estrogen, there is a four hour lag period prior to the detection of apoVLDLII mRNA. To examine the events that occur during this lag period, in vivo footprinting and run-on transcription assays were performed. Clomipramine (CLI) administered to neonatal rats has been purported to induce behavioral abnormalities in adulthood analogous to human endogenous depression (Vogel et al., 1990). Whether growth of large, ovulatory follicles differed after different modes of stimulation with FSH and whether growth was enhanced by exogenous LH were examined in prepubertal gilts. Although not as commonplace today, it still has significant clinical relevance. More importantly, however, is the large body of knowledge that has been generated which addresses the pharmacokinetic and pharmacodynamic characteristics of this {dollar}\\beta{dollar}-blocker. Plant species that develop resistance to one herbicide (e.g., s-triazine) may simultaneously exhibit resistance to other herbicides to which they have never been exposed. The objectives of this study were to compare the response of atrazine resistant and susceptible biotypes of redroot pigweed (Amaranthus retroflexus L.) and velvetleaf (Abutilon theophrasti Medic.) to the herbicides alachlor (2-chloro-N-(2,6-diethylphenyl-N-(methoxymethyl) acetamide), atrazine (6-chloro-N-ethyl-{dollar}N\\sp\\prime{dollar}-(1-methyl-ethyl)-1,3,5-triazine-2, 4-diamine), linuron ({dollar}N\\sp\\prime{dollar}-(3,4-dichloro-phenyl)-N-methoxy-N-methylurea), and metribuzin (4-amino-6-(1,1-dimethylethyl)-3-(methylthio)-1,2,4-triazin-5 (4H)-one). Stimulation of malic enzyme transcription by triiodothyronine (T3) is robust ({dollar}>{dollar}60-fold) in chick embryo hepatocytes, weak (5-fold) in chick embryo fibroblasts that stably overexpress the nuclear T3 receptor-{dollar}\\alpha,{dollar} and still weaker (1-fold) in chick embryo fibroblasts which contain nuclear T3 receptor levels that are similar to those of CEH. Directed by the DNase I hypersensitive regions in the malic enzyme gene, functional transfection and in vitro DNA-binding analyses were performed and four cis-acting elements were identified in the malic enzyme 5{dollar}\\sp\\prime{dollar}-flanking DNA. A pilot study was completed to evaluate the data collection techniques. Objective. To develop a targeting delivery system for lung cancer prevention and treatment using tumor cells as the drug carrier. Methods. Doxorubicin has been loaded into B16-F10 murine melanoma. This drug-loaded tumor cell system (DLTC) has been characterized, and its anticancer activity evaluated on C57BL/6J mice bearing pulmonary B16-F10 melanoma. Results. Up to 96 {dollar}\\rm\\mu g{dollar} of doxorubicin could be loaded into {dollar}10\\sp6{dollar} cells. The release of drug from the DLTC was both cell concentration and temperature dependent. Acute exposure to propanil central nervous system depression, loss of a righting reflex, and cyanosis. The immunotoxic effects of propanil on animals include reduction of thymus weight and cellularity with increase of spleen weight and cellularity, inhibition of humoral and cell-mediated immune response and alteration NK cell activity and macrophage cytotoxicity. In contrast to CA, a TA model fragments the problem space along restricted routes. All cells on the routes are treated as equal and controlled by rules. To date, no standard dissolution methodology is recognized in the official compendia for protein-based injectable suspensions. Development of a sensitive dissolution method would have utility as an in vitro quality control tool and an indicator of associated in vivo product performance. During periods of egg laying in the chicken, when circulating levels of estrogen are increased, the liver-specific estrogen-dependent very low density apolipoprotein II (apoVLDL II) gene is expressed. Transcription is accompanied by estrogen-dependent, liver-specific changes in the chromatin conformation within and surrounding the gene. One of the regions that changes in conformation is in the promoter where the estrogen receptor along with multiple liver-specific and ubiquitous transcription factors interact in a cooperative manner to assemble a functional transcriptional initiation complex. The purposes of this study were to gain an understanding of the Gypsy culture and to discover the relationship, within the school domain, between the Gypsy children and the non-Gypsy teachers. The study was conducted in Malaga, Spain and lasted two and a half months. The research design for this study was qualitative. Data for this ethnographic research were gathered using three strategies: (1) participant observations, (2) interviews, (3) collecting school magazines.
