While the objective of expanding the Oregon Medicaid program is commendable, the means adopted by the Oregon legislature to control the costs of the expansion are problematic. This paper examines the legislature's approach from a legal perspective. The first part of the paper determines whether it is consistent with federal and state law governing the Medicaid program. Oregon is seeking waivers from any conflicting federal requirements, and since there do not appear to be any constitutional impediments, the key question is whether the waivers should be granted as a matter of sound public policy. The 10 ligation species of human cyanomethemoglobin were previously found to distribute into three discrete cooperative free energy levels according to a combinatorial code (i.e., dependent on both the number and configuration of ligated subunits). Analysis of this distribution showed that the hemoglobin tetramer occupies a third allosteric state in addition to those of the unligated (T) and fully ligated (R) species. To determine the nature of the intermediate allosteric state, we have studied the effects of pH, temperature, and single-site mutations on its free energy of quaternary assembly, in parallel with corresponding data on the deoxy (T) and fully …

Measurement Of Acetate Concentrations In Marine Pore Waters By Using An Enzymatic Approach, G. M. King

Acetate concentrations in marine and freshwater matrices were measured by an enzymatic technique which coupled the synthesis of acetyl coenzyme A to AMP production. The resulting AMP was assayed by a sensitive and relatively rapid high-pressure liquid chromatography method, using an aqueous, isocratic mobile phase for elution. The method was insensitive to the presence of seawater salts and required no sample prepurification or distillation. Propionate caused a minor, but statistically insignificant, interference when equimolar with acetate; butyrate caused no interference, even at relatively high concentrations. Detection limits for acetate were approximately 100 nM with a precision of about 5%. Pore …

Specific and sensitive amplification of major outer membrane protein (MOMP) gene DNA sequences of Chlamydia psittaci was achieved in a two-step polymerase chain reaction. First, oligonucleotide primers specific for 5' and 3' nontranslated regulatory regions of the MOMP gene were used in a polymerase chain reaction to amplify a DNA fragment of approximately 1,400 bp. A portion of this DNA fragment was amplified in a second reaction using a degenerate oligonucleotide primer specific for a DNA sequence contained within the 1,400-bp DNA fragment and one of the first-step primers. The entire nucleotide sequences of the spike glycoprotein (S) genes of the highly virulent bovine coronavirus (BCV) strain BCV-LY138, the avirulent BCV-L9 and related Norden Vaccine (BCV-Vaccine) strains were determined using the polymerase chain reaction (PCR) to amplify cDNAs obtained by reverse transcription of viral RNA, and to produce single strand cDNAs for DNA sequencing. The S gene sequences of these viral strains were compared with those of recently published strains BCV-Mebus, BCV-Quebec, and BCV-F15. Monoclonal antibodies (MAbs) against two major structural proteins of the cell-adapted Mebus strain of bovine coronavirus (BCV-L9) were produced and characterized. Seven MAbs reacted with the peplomeric glycoprotein, gp100/S, while three MAbs reacted with the nucleoprotein p53/N in Western blot analysis of BCV polypeptides. MAbs to gp100/S reacted with discontinuous epitopes of gp100/S in Westerns under mild but not under standard denaturing conditions. In contrast, MAbs to p53/N reacted in both types of Westerns, and those epitopes were thus continuous. MAbs to p53/N failed to neutralize BCV infectivity, while 4 MAbs to gp100/S neutralized BCV effectively. Cross reactivity of MAbs …

Sodium zeolite A (SZA), a synthetic sodium aluminosilicate having high ion-exchange capacity, has been shown to increase eggshell specific gravity in laying hens and to improve Ca utilization in chickens. A 4 x 2 factorial arrangement of treatments was used to investigate the effect of dietary Ca (.6, .8, 1.0, and 1.2%) and SZA (0 and .75%) on growth, plasma, and tibia characteristics of chicks from 5 to 15 days of age. Increasing dietary Ca linearly increased (P less than .05) Ca and alkaline phosphatase (AP) in plasma and increased tibia shearing force and percentage ash, Ca, and P in …

A simple procedure that yields highly purified intact chloroplasts from Chlamydomonas reinhardtii is described. This procedure involves breakage of cell wall-deficient cells by passing them through a narrow bore syringe needle. The intact chloroplasts are then purified from the crude homogenate by differential centrifugation and Percoll gradient centrifugation. This procedure generates relatively high yields of chloroplasts capable of CO2 fixation. These chloroplasts were characterized by electron microscopy, marker enzyme analysis, and ferricyanide exclusion. Transmission electron microscopy indicates that these chloroplasts retain their pyrenoids and eyespots. The Soviet-American Gallium Experiment is the first experiment able to measure the dominant flux of low energy p-p solar neutrinos. Four extractions made during January to May 1990 from 30 tons of gallium have been counted and indicate that the flux is consistent with 0 SNU and is less than 72 SNU (68% CL) and less than 138 SNU (95% CL). This is to be compared with the flux of 132 SNU predicted by the Standard Solar Model. © 1991.

The six individual catalytic polypeptide chains within the two catalytic trimers of Escherichia coli aspartate transcarbamoylase (ATCase; EC 2.1.3.2) are folded into two discrete structural domains interconnected in part by helix 12, which comprises residues 285-305 and is located near the carboxyl terminus of the chain. The essential role of this helix in folding of the chains and their assembly into ATCase was demonstrated by introducing a stop codon at the position corresponding to amino acid 284, 291, or 299. Cells containing these mutations are pyrimidine auxotrophs lacking ATCase-like protein in cell extracts. In contrast, stable active enzyme is formed …

Let ℱ be a collection of 3-connected matroids, none a proper minor of another, such that if M is a 3-connected matroid having a proper ℱ-minor and e is an element of M, then M has an ℱ-minor avoiding e. This paper proves that there are precisely two collections ℱ with this property: {U2,4} and {U2,4, M(K4)}. Several extensions of this result and some similar results for 2-connected matroids are also established. © 1991, Academic Press Limited. All rights reserved.

Let n be an integer exceeding one and M be a matroid having at least n + 2 elements. In this paper, we prove that every n-element subset X of E(M) is in an (n + 1)-element circuit if and only if (i) for every such subset, M X is disconnected, and (ii) for every subset Y with at most n elements, M Y is connected. Various extensions and consequences of this result are also derived including characterizations in terms of connectivity of the 4-point line and of Murty's Sylvester matroids.
