Stability And Controlled Release Of Lutein Loaded In Zein Nanoparticles With And Without Lecithin And Pluronic F127 Surfactants, Thanida Chuacharoen, Cristina M. Sabliov

The ability of zein nanoparticles, made with and without surfactants, to protect lutein from oxidation and to control its release was studied under various conditions. A combination of lecithin and pluronic F127 as surfactants was used to promote physicochemical stability of the nanoparticles and entrapped bioactives. Surfactant use resulted in an increased size, improved polydispersity index, decreased zeta potential, and improved entrapment efficiency. A two-phase lutein release profile was observed in phosphate-buffered saline (PBS) that consisted of an initial-burst release at 24 h, decreased in the presence of surfactants, followed by a gradual zero-order release profile for both systems. Correct developmental timing is essential for plant fitness and reproductive success. Two important transitions in shoot development—the juvenile-to-adult vegetative transition and the vegetative-to-reproductive transition—are mediated by a group of genes targeted by miR156, SQUAMOSA PROMOTER BINDING PROTEIN (SBP) genes. To determine the developmental functions of these genes in Arabidopsis thaliana, we characterized their expression patterns, and their gain-of-function and loss-of-function phenotypes. Our results reveal that SBP-LIKE (SPL) genes in Arabidopsis can be divided into three functionally distinct groups: 1) SPL2, SPL9, SPL10, SPL11, SPL13 and SPL15 contribute to both the juvenile-to-adult vegetative transition …

© 2016 by The American Society for Biochemistry and Molecular Biology, Inc. Oncostatin M (OSM) is a multifunctional gp130 cytokine. Although OSM is produced in adipose tissue, it is not produced by adipocytes. OSM expression is significantly induced in adipose tissue from obese mice and humans. The OSM-specific receptor, OSM receptor ô (OSMR), is expressed in adipocytes, but its function remains largely unknown. To better understand the effects of OSM in adipose tissue, we knocked down Osmr expression in adipocytes in vitro using siRNA. Agricultural crop rotations have been shown to increase soil carbon (C), nitrogen (N), and microbial biomass. The mechanisms behind these increases remain unclear, but may be linked to the diversity of crop residue inputs to soil organic matter (SOM). We used a residue mixture incubation to examine how variation in long-term diversity of plant communities in agroecosystems influences decomposition of residue mixtures, thus providing a comparison of the effects of plant diversification on decomposition in the long term (via crop rotation) and short term (via residue mixtures). © 2016 American Chemical Society. The amyloid-β (Aβ) peptide of Alzheimer's disease (AD) forms polymorphic fibrils on the micrometer and molecular scales. Various fibril growth conditions have been identified to cause polymorphism, but the intrinsic amino acid sequence basis for this polymorphism has been unclear. Several single-site mutations in the center of the Aβ sequence cause different disease phenotypes and fibrillization properties. The E22G (Arctic) mutant is found in familial AD and forms protofibrils more rapidly than wild-type Aβ. © 2016 American Chemical Society. Plasminogen activator inhibitor type 1 (PAI-1) regulates the fibrinolysis pathway by inhibiting the protease activity of plasminogen activators. PAI-1 works in concert with vitronectin (VN), an extracellular protein that aids in localization of active PAI-1 to tissues. The Peterson laboratory demonstrated that Cu(II) and other transition metals modulate the stability of PAI-1, exhibiting effects that are dependent on the presence or absence of the somatomedin B (SMB) domain of VN. © 2016 American Chemical Society. Ammonia binds to two sites in the oxygen-evolving complex (OEC) of Photosystem II (PSII). The first is as a terminal ligand to Mn in the S2 state, and the second is at a site outside the OEC that is competitive with chloride. Binding of ammonia in this latter secondary site results in the S2 state S = 5/2 spin isomer being favored over the S = 1/2 spin isomer. Using electron paramagnetic resonance spectroscopy, we find that ammonia binds to the secondary site in wild-type Synechocystis sp. PCC 6803 PSII, but not in D2-K317A mutated …

A three-dimensional coupled physical-biogeochemical model was used to simulate and examine temporal and spatial variability of sea surface pCO2 in the Gulf of Mexico (GoM). The model was driven by realistic atmospheric forcing, open boundary conditions from a dataassimilative global ocean circulation model, and observed freshwater and terrestrial nutrient and carbon input from major rivers. A 7-year model hindcast (2004-2010) was performed and validated against ship measurements. Model results revealed clear seasonality in surface pCO2 and were used to estimate carbon budgets in the Gulf. Based on the average of model simulations, the GoM was a net CO2 sink with …

Quantifying the role that freshwater ecosystems play in the global carbon cycle requires accurate measurement and scaling of dissolved organic carbon (DOC) removal in river networks. We reviewed reach‐scale measurements of DOC uptake from experimental additions of simple organic compounds or leachates to inform development of aquatic DOC models that operate at the river network, regional, or continental scale. Median DOC uptake velocity (vf) across all measurements was 2.28 mm min−1. Measurements using simple compound additions resulted in faster vf (2.94 mm min−1) than additions of leachates (1.11 mm min−1). We also reviewed published data of DOC bioavailability for ambient …

A large hypoxic zone forms every summer on the Texas-Louisiana Shelf in the northern Gulf of Mexico due to nutrient and freshwater inputs from the Mississippi/Atchafalaya River System. Efforts are underway to reduce the extent of hypoxic conditions through reductions in river nutrient inputs, but the response of hypoxia to such nutrient load reductions is difficult to predict because biological responses are confounded by variability in physical processes. We investigated the effects of the root endophyte Piriformospora indica on interactions between rice (Oryza sativa) plants and its root herbivore rice water weevil (RWW; Lissorhoptrus oryzophilus), and how plant jasmonic acid (JA) and GA regulate this tripartite interaction. The ability of adipose tissue expansion in the offspring of obese mothers is limited and is associated with metabolic dysfunction of adipose tissue when challenged with a high-fat diet. Maternal obesity induces DNA demethylation in the promoter of zinc finger protein 423, which renders progenitor cells with a high adipogenic capacity. Maternal obesity demonstrates long-term effects on the adipogenic capacity of progenitor cells in offspring adipose tissue, demonstrating a developmental programming effect. Natural fires ignited by lightning strikes following droughts frequently are posited as the ecological mechanism maintaining discontinuous tree cover and grass-dominated ground layers in savannas. Such fires, however, may not reliably maintain humid savannas. We propose that savanna trees producing pyrogenic shed leaves might engineer fire characteristics, affecting ground-layer plants in ways that maintain humid savannas. We explored our hypothesis in a high-rainfall, frequently burned pine savanna in which the dominant tree, longleaf pine (Pinus palustris), produces resinous needles that become highly flammable when shed and dried. The crucifer flea beetle, Phyllotreta cruciferae (Goeze), is a key pest of canola (Brassica napus L.) in the northern Great Plains of North America. The efficacies of entomopathogenic nematodes (Steinernema spp. and Heterorhabditis spp.), a sprayable polymer gel, and a combination of both were assessed on canola for flea beetle management. Plots were treated soon after colonization by adult flea beetles, when canola was in the cotyledon to one-leaf stage. Objective: To compare levels of 25-hydroxyvitamin D (25OHD) associated with a plateauing of intact parathyroid (iPTH) across latitudes among adults with African ancestry. Methods: This study included approximately 500 adults of African ancestry ages 25 to 45 years living in 4 sites: Chicago, Illinois (41°N), Jamaica (17°N), Ghana (6°N), and South Africa (34°S). Multivariate linear regression models, a nonlinear logistic growth curve model, and piecewise linear models with a single knot were fitted to estimate the 25OHD level associated with a plateauing of iPTH with adjustment for covariates. Strontium and barium to calcium ratios are often used as proxies for tracking animal movement across salinity gradients. Many estuarine rivers face saltwater intrusion due to sea level rise, potentially causing changes in mobility and distribution of these metals upstream. From May 2013 to August 2015, monthly water samples were collected and in-situ measurements were performed at six sites along an estuary strongly affected by saltwater intrusion, the Calcasieu River, with salinity ranging from 0.02 to 29.50 Parts Per Thousand (ppt). Carcinogenesis induced by environmental metal exposure is a major public health concern. The exact mechanisms underlying metal carcinogenesis remain elusive. In the past few decades, the relationship between metal-induced generation of reactive oxygen species (ROS) and the mechanism of metal carcinogenesis has been established. The carcinogenic process is a very complex one. Ecological factors such as habitat and food availability affect the social structure of bottlenose dolphins (Tursiops spp.). Here, we describe the social structure of bottlenose dolphins (T. truncatus) in Golfo Dulce, Costa Rica, a semi-enclosed, fjord-like tropical embayment resembling a pelagic system. We also examine behaviour-linked social strategies by comparing social structure relative to behavioural state: feeding versus non-feeding. We analysed 333 sightings over 210 days from boat-based surveys. Despite the uniqueness of the area, the 47 analysed adults had a social structure similar to other populations: a well-differentiated fission-fusion society with sex-specific patterns of associations and aggression. © The Animal Consortium 2015. The abundance and cross-linking of intramuscular connective tissue contributes to the background toughness of meat, and is thus undesirable. Connective tissue is mainly synthesized by intramuscular fibroblasts. Myocytes, adipocytes and fibroblasts are derived from a common pool of progenitor cells during the early embryonic development. It appears that multipotent mesenchymal stem cells first diverge into either myogenic or non-myogenic lineages; non-myogenic mesenchymal progenitors then develop into the stromal-vascular fraction of skeletal muscle wherein adipocytes, fibroblasts and derived mesenchymal progenitors reside. Stabilization In A Chemostat With Sampled And Delayed Measurements, Frédéric Mazenc, Jérome Harmand, Michael Malisoff

We study chemostat models with constant substrate input concentrations. We allow growth functions that are not necessarily monotone. The measurement is the substrate concentration, which is piecewise constant with a nonconstant delay, so only sampled observations are available. Under new conditions on the size of the delay and on the largest sampling interval, we solve the problem of asymptotically stabilizing a componentwise positive equilibrium point with the dilution rate as the control. We use a new Lyapunov approach.

Gamma-aminobutyric acid (GABA) is a widely conserved signaling molecule that in animals has been adapted as a neurotransmitter. GABA is synthesized from the amino acid glutamate by the action of glutamate decarboxylases (GADs). Two vertebrate genes, GAD1 and GAD2, encode distinct GAD proteins: GAD67 and GAD65, respectively. We have identified a third vertebrate GAD gene, GAD3. This gene is conserved in fishes as well as tetrapods. We analyzed protein sequence, gene structure, synteny, and phylogenetics to identify GAD3 as a homolog of GAD1 and GAD2. Interestingly, we found that GAD3 was lost in the hominid lineage. Herein, we report the first example of a crystalline metal–donor–fullerene framework, in which control of the donor–fullerene mutual orientation was achieved through chemical bond formation, in particular, by metal coordination. The 13C cross-polarization magic-angle spinning NMR spectroscopy, X-ray diffraction, and time-resolved fluorescence spectroscopy were performed for comprehensive structural analysis and energy-transfer (ET) studies of the fulleretic donor–acceptor scaffold. Furthermore, in combination with photoluminescence measurements, the theoretical calculations of the spectral overlap function, Förster radius, excitation energies, and band structure were employed to elucidate the photophysical and ET processes in the prepared fulleretic material. Numerous functionalization methods have been employed to increase the solubility, and therefore, the processability of fullerenes in composite structures, and of these radical addition reactions continue to be an important methodology. C60 and Sc3N@C80 derivatives were prepared via radical addition of the photodecomposition products from the commercial photoinitiator TMB-PPO, yielding C60(TMB-PPO)5 and Sc3N@C80(TMB-PPO)3 as preferred soluble derivatives obtained in high yields. The Mississippi (MS) barrier island chain along the northern Gulf of Mexico coastline is subject to rapid changes in habitat, geomorphology and elevation by natural and anthropogenic disturbances. The purpose of this study was to compare habitat type coverage with respective elevation, geomorphic features and short-term change between the naturally-formed East Ship Island and the man-made Sand Island. Ground surveys, multi-year remotely-sensed data, habitat classifications and digital elevation models were used to quantify short-term habitat and geomorphic change, as well as to examine the relationships between habitat types and micro-elevation. Mass spectrometry imaging (MSI) was introduced more than five decades ago with secondary ion mass spectrometry (SIMS) and a decade later with laser desorption/ionization (LDI) mass spectrometry (MS). Large biomolecule imaging by matrix-assisted laser desorption/ionization (MALDI) was developed in the 1990s and ambient laser MS a decade ago. Although SIMS has been capable of imaging with a moderate mass range at sub-micrometer lateral resolution from its inception, laser MS requires additional effort to achieve a lateral resolution of 10 μm or below which is required to image at the size scale of single mammalian cells.
