13C Nmr Investigation Of Nonenzymatic Glucosylation Of Protein, Carolyn I. Neglia, Helga J. Cohen, Albert R. Garber, Paul D. Ellis, Suzanne R. Thorpe, John W. Baynes
Faculty Publications
Nonenzymatic glucosylation of protein is initiated by the reversible condensation of glucose in its open chain form with the amino groups on the protein. The initial product is an aldimine (Schiff base) which cyclizes to the glycosylamine derivative. The aldimine can undergo a slow Amadori rearrangement to yield the relatively stable ketoamine adduct which is structurally analogous to fructose. 13C NMR has been used to characterize these early products of nonenzymatic glucosylation, using RNase A as a model protein. C-1 of the beta-pyranose anomer of the glycosylamine was identified at 88.8 ppm in the spectrum of RNase glucosylated approximately 1:1 …

Current Trends In The Planning And Development Of Northern European Collections, Richard Hacken
Current Trends In The Planning And Development Of Northern European Collections, Richard Hacken
Faculty Publications
Current planning and development of collections in the social sciences and humanities for German-speaking Europe, the Netherlands, and Scandinavia reflect the changing needs of research, strict budgetary limits, the prevailing publishing market in those countries, and certain innovations in library automation. Librarians responsible for supporting an area study of Northern Europe may shape the trends to their advantage by careful policy planning, by informed financial choices, by the use of data bases and resource sharing, by privately-nurtured channels of acquisition and support and by a continuing self-education program that might include participation in the activities of the CES and WESS.

Nonenzymatic Glucosylation And Glucose-Dependent Cross-Linking Of Protein, Albert S. Eble, Suzanne R. Thorpe, John W. Baynes
Faculty Publications
A model system using RNase A has been established for studying the nonenzymatic glucosylation and glucose-dependent cross-linking of protein (Maillard reaction) under physiological conditions in vitro. The rate of glucosylation of RNase was first order in glucose. Glucosylation was accompanied by a comparable decrease in primary amino groups in the protein and lysine recoverable by amino acid analysis. Analysis of glucosylation reaction mixtures by sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the presence of mercaptoethanol revealed the time-dependent formation of RNase dimer and trimer. The polymerization reaction was mixed order with respect to glucose concentration, but was approximately first order with …

The Mr of spinach chloroplast coupling factor 1 has been determined by sedimentation equilibrium and by light scattering to be 400 000 ± 24 600 and 407 000 ± 20 000, respectively. These values differ substantially from that obtained previously (325 000) and are consistent with an α3β3γδε{lunate} subunit stoichiometry. © 1983.

Association Between Coronary Heart Disease Risk Factors And Physical Fitness In Healthy Adult Women, Larry W. Gibbons, Steven N. Blair, Kenneth H. Cooper, Mike Smith
Association Between Coronary Heart Disease Risk Factors And Physical Fitness In Healthy Adult Women, Larry W. Gibbons, Steven N. Blair, Kenneth H. Cooper, Mike Smith
Faculty Publications
We examined associations between physical fitness and risk factors for coronary heart disease in healthy women ages 18-65 years. Physical fitness was objectively determined by the duration of a maximal treadmill exercise test. Six physical fitness categories (very poor to superior), specific within 10-year age increments, were established. Mean risk factor levels varied across categories, but so did potential confounders such as age and weight. Multiple linear regression modeling was used to control for the effects of age, weight and year of exam on coronary risk factors. After adjustment, physical fitness was independently associated with triglycerides (p < 0.001), high-density lipoprotein cholesterol (HDL-C) (p less …

Studies showing an increase in plasma concentration of high density lipoprotein cholesterol (HDL-C) with moderate exercise have usually rejected the role of body weight change in the HDL-C raising process, ostensibly because the amount of weight lost has been negligible. To investigate HDL-C changes more thoroughly, we followed initially sedentary middle-aged men randomly assigned to either a moderate running (N=36) or a sedentary control (N=28) group for one year. Among runners, one-year changes in plasma HDL-C concentrations correlated strongly with their body weight changes (r = -0.53, P < 0.001). Curve-fitting procedures and regression analysis suggested that processes associated with weight change produce much of the plasma HDL-C changes induced by moderate exercise and that changes in HDL-C concentration predominantly reflect changes in the reputedly anti-atherogenic HDL2 sub-component. Further, the interaction between weight change and plasma HDL-C concentration was significantly different (P

Molecular Cloning And Analysis Of The Cry1 Gene: A Yeast Ribosomal Protein Gene, John C. Larkin, John L. Woolford
Faculty Publications
Using cloned DNA from the vicinity of the yeast mating type locus (MAT) as a probe, the wild type allele of the cryptopleurine resistance gene CRY1 has been isolated by the technique of chromosome walking and has been shown to be identical to the gene for ribosomal protein 59. A recessive cryRl allele has also been cloned, using the integration excision method. The genetic distance from MAT to CRY1 is 2.2 cM, while the physical distance is 21 kb, giving a ratio of about 10kb/cM for this interval. The phenotypic expression of both plasmid borne alleles of the gene can be …

Library Use Patterns Among Full- And Part-Time Faculty And Students, Jo Bell Whitlatch
Faculty Publications
Trends in higher education are toward part-time students and majors in professional fields which are associated with low library use. Research findings at San Jose State University, California and other academic libraries, demonstrate that declining library use may be a future trend. Declining use will further erode administrative support for library budgets. Libraries must become effective in identifying and meeting the information needs of new nontraditional students. User surveys establishing service priorities can be an effective tool in planning library service for future student and faculty populations.

From the outset, the reader should be familiar with our script. Our threefold purpose in writing this essay is to be at once descriptive, evaluative, and prescriptive. We shall describe briefly the decisions of the United States Supreme Court during the 1981 Term in the broad field of federal jurisdiction, and shall compare and contrast those decisions with the past decisions of the United States Court of Appeals for the Eleventh Circuit.

The Federal Trade Commission, along with other administrative agencies, has been especially affected by the current emphasis on deregulation. Some proponents of the free market system view the Commission as an unnecessary and costly impediment to market functioning. In this article, Professor Koch and Ms. Martin review past FTC regulatory efforts in light of the FTC's mandate: to maintain an efficient, competitive free market Although the FTC has deviated from its market maintenance goal, this deviation should not serve as a basis for constricting Commission rulemaking activity. Rather, the authors demonstrate that past Commission practices illustrate the need for an …

Studies of six 8-9-month-old rams showed that the specific activities of myoinositol-1-phosphate synthase (EC 5.5.1.4) were highest in epididymal spermatozoa, intermediate in testis and lowest in epididymal tissue. The activity per spermatozoon decreased from caput to cauda. The levels of activity of myo-inositol-1-phosphate synthase in ejaculated spermatozoa from four 3-year-old rams and the seminal vesicles of two 3-year-old rams were insignificant, but in pooled Sertoli cells from the testes of young lambs, the specific activity was much lower than in epididymal spermatozoa although activity per cell was of the same order of magnitude. We conclude that epididymal spermatozoa contain a …

Pigeons acquired a different four‐response chain each session by responding sequentially on three keys in the presence of a sequence of four colors. The response chain was maintained by food presentation under a fixed‐ratio schedule. Errors produced a brief timeout but did not reset the chain. Each day there were four 15‐minute sessions, with a 10‐minute intersession interval. Cumulative dose‐effect curves for phencyclidine, pentobarbital, and d‐amphetamine were obtained by giving an injection before each of the four sessions; successive injections increased the cumulative dose in equally spaced logarithmic steps. For comparison, non‐cumulative doses of each drug (i.e., doses not preceded …

A systemic streptococcal infection in cultured bullfrogs in Brazil was characterized by necrotizing splenitis and hepatitis with hepatic and renal hemorrhage. A non-hemolytic Group B Streptococcus appeared to be the cause of the lesions, and the stimulus for the splenic reticuloendothelial hyperplasia observed in the animals. Stress may have been a factor in the development of the pathological condition.

Urinary Sodium Excretion And Blood Pressure In Children: Absence Of A Reproducible Association, Richard Cooper, Kiang Liu, Maurizio Trevisan, William Miller, Jeremiah Stamler
Faculty Publications
A previous report by our group described a cross-sectional association between urinary sodium excretion and blood pressure (BP) in children. Attempts were made to reproduce these findings in two subsequent studies carried out with an identical protocol. Although descriptive results were similar and the correlations between measures of body size and blood pressure were reproduced, a clear-cut association between 7-day mean urinary sodium excretion and BP was observed only in the first of the three studies. These results could imply inadequate study methods, an inconsistent and weak relationship, or the absence of a relationship altogether. © 1983 American Heart Association, …

A population-based survey of 134 white men, recruited from the Chicago People’s Gas Company labor force, was carried out to examine the association between sodium-lithium (Na-Li) countertransport and hypertension. Of the 134 participants in this industry-sponsored periodic health examination, 64 were normotensive and 70 were either taking antihypertensive medications or had a systolic pressure ≥ 140 or a diastolic pressure ≥ 90 mm Hg. The hypertensives were older and more overweight. Countertransport was significantly higher in hypertensives than in normotensives. Among the three subgroups of hypertensives - untreated borderline (140/90 to 160/95 mm Hg), untreated definite (over 160/95 mm Hg), …

Age-standardized death rates from all causes for persons age 35-74 fell 23 per cent, 1968-1979. The monotonic decline was interrupted by only one period of excess influenza mortality. In the late 1970s, however, the decline in mortality decelerated and no further decrease was observed, 1979-1981. The recurrence of epidemic influenza contributed significantly to this trend although it would appear that the underlying mortality pattern has begun to change.
